Influence of adenosine triphosphate on the isolated perfused mesenteric artery of the rabbit

The vascular effects of adenosine triphosphate (ATP) were examined in the isolated perfused mesenteric arteries of the rabbit. Bolus injections of ATP (1 × 10−8 to 10−6 mol) induced a dose-dependent vasoconstrictor response at resting perfusion pressure, while continuous perfusion with ATP briefly elicited a vasoconstrictor response which was not maintained. Perfusion with phentolamine (2.65 × 10−6 M, an α-adrenergic receptor blocker), indomethacin (8.37 × 10−6 M, an inhibitor of cyclooxygenase), atropine (1 × 10−7 M, a muscarinic receptor blocker), and hydralazine (2 × 10−4 M, a vascular smooth muscle inhibitor) for a period of 1 h had no effect on vasoconstrictor responses to ATP. However, pretreatment with reserpine (2 mg∙kg−1∙day−1 for 2 days), an agent which depletes catecholamines, potentiated responses to ATP. On the other hand, when vascular tone was increased with an isoosmotic 60 mM K+ depolarizing Krebs bicarbonate solution, bolus injections of ATP elicited a prominent dose-dependent vasoconstriction followed by a prominent vasodilation. The degree of vasodilation but not of vasoconstriction elicited by ATP was greater in small terminal arteries with branches (<0.5 mm outside diameter (o.d.)) than in the medium size arteries (≤1 mm o.d.) without terminal branches. Both the vasoconstrictor and vasodilator responses were unaffected by a perfusion with atropine, indomethacin, or eicosatetraynoic acid (ETYA, 1 × 10−4 M) for 1 – 2 h. The vasoconstrictor responses were potentiated while the vasodilator responses were inhibited significantly by perfusion with propranolol (3 × 10−6 M) and phentolamine (2.65 × 10−6 M) together for 1 h or by pretreatment with reserpine followed by cold storage at 2 °C for 24 h. Perfusion with 8-phenyltheophylline (4 × 10−6 M (8-PT), an adenosine receptor blocker) for 1 h significantly inhibited by the vasodilator responses to bolus injections of adenosine but not to ATP. Further, ATP but not adenosine elicited a much more prominent vasodilator response on norepinephrine (NE) induced tone than on 60 mM K+. These studies suggest that ATP may induce vasoconstriction independent of activation of α-adrenergic or muscarinic receptors or enhanced synthesis of prostaglandins. This vasoconstriction is resistant to the inhibitory influence of hydralazine. On the other hand, the vasodilator response to ATP may be mediated through its interactions with released or circulating norepinephrine.

The total arbutin content in the leaves of all the studied Bergenia plants ( B. crassifolia, B. ciliata and B. x ornata) was determined. The highest values of the arbutin content have been established for B. crassifolia (58.9 ± 0.7 mg.g−1 DW) and B. x ornata (51.0 ± 1.21 mg.g−1 DW), and the lowest for B. ciliata (5.9 ± 0.6 mg.g−1 DW). Arbutin concentration in the Bergenia leaves was the lowest in spring, in the autumn, on the contrary it increased. All the tested aqueous extracts caused a dose-dependent increase in diphenolase activity of fungal tyrosinase in a similar way as arbutin. On the other hand, all the ethanol extracts inhibited the diphenolase activity of tyrosinase.

Comprehensive assessment of the medium size rich-dome engines was conducted leading to the following emissions correlations: (1) LTO NOx = 1.129 × OPR 1.0899 with R 2 = 0.9248 Takeoff NOxEI given by (2) NOxEI = 0.0729 × OPR 1.7197 with R 2 = 0.9603 COEI idle = 396.42 NOxEI Takeoff 0.814 These correlations may be compared with the following for the CFM56 Tech Insertion: Takeoff NOxEI CFM_TI = 0.0744 × OPR 1.7151 Idle COEI CFM_TI = 396.42 Takeoff NOxEI 0.814 Idle HCEI CFM_TI = 0.1609 × Idle COEI - 3.1959 TALON II takeoff NOxEI data are reproduced well by: NOxEI TALON II = 0.0167 × OPR 2.1403 TALON II gives 10% lower NOx at 26 OPR and its NOx is comparable with the CFM_TI at 34 OPR. The CFM DAC technology is competitive with LEC’s for the low rated thrust engines. However, interaction between the two domes leads to early quenching with resultant higher idle COEI plateau. On the other hand, the 40 OPR lean DAC gave 25% higher NOx than LEC. Moreover, lean DAC (Gen-1) impacted fuel burn adversely making its likelihood to continue as product discouraging. The second generation lean dome technology initially kicked off under NASA sponsorship with significantly larger funding support from the CFMI and GE Aviation (GEA) led to successful introduction of TAPS into products (GEnx-1B and Gen-2B) with potential applications in other future GEA engines.

Detached leaves of Echeveria elegans Bgr. produce both roots and shoots. However, when their bases are severed, they tend to produce only roots. It was observed that roots had to be present on detached leaves, bases removed or not, in order to have an increase in size and also in dry weight. The growth in size was found to be due mainly to enlargement of cells, and the increase in dry weight was presumably due to maintenance of normal metabolic activity in the detached leaves. Thus detached leaves of E. elegans have a greater potential for growth than is normally realized by the leaves that are mature and still attached to the parent axis. The vigorously growing shoots seem to have an inhibitory influence on growth in the detached leaves. Roots on detached leaves, on the other hand, seem to play an important role not only in the delaying of senescence but also in reducing the inhibitory influence of shoots. Roots alone appeared to bring about "uncontrolled" enlargement of cells and consequently lesions were formed on the leaves, which finally died off. Thus growth, regeneration, and senescence in the detached leaves appear to be correlative phenomena.

Background: Although surviving sepsis campaign guidelines recommend the use of inotropes in the presence of myocardial dysfunction, the effects of inotropes, including epinephrine, dobutamine, and milrinone, on in-hospital mortality in patients with septic shock remains unclear. Materials and Methods: We conducted an international,2-center, retrospective cohort study. The Cox proportional hazards regression model with time-varying covariates was used to investigate whether epinephrine, milrinone, or dobutamine reduces in-hospital mortality in patients with septic shock. Sensitivity analysis was performed using propensity score matching. The primary outcome was in-hospital mortality. The secondary outcome included atrial fibrillation (Afib) with a rapid ventricular response (RVR) in the intensive care unit (ICU) and ICU-free days. Results: A total of 417 patients with septic shock were included, 72 (17.3%) of whom received inotropes. The use of epinephrine and dobutamine was associated with significantly higher in-hospital mortality (epinephrine, hazard ratio [HR]: 4.79, 95% confidence interval [CI]: 2.12-10.82, P = .001; dobutamine, HR: 2.53, 95% CI: 1.30-4.95, P = .046). The effects of epinephrine and dobutamine were time- and dose-dependent. The use of milrinone was not associated with increased mortality (HR: 1.07, 95% CI: 0.42-2.68, P = .345). The use of epinephrine, dobutamine, and milrinone was associated with significantly increased odds of Afib with RVR (epinephrine, odds ratio [OR]: 3.88, 95% CI: 1.11-13.61, P = .034; dobutamine, OR: 3.95, 95% CI: 1.14-13.76; and milrinone, OR: 3.77, 95% CI: 1.05-13.59). On the other hand, the use of epinephrine, dobutamine, and milrinone was not associated with less ICU-free days (epinephrine, adjusted OR: 0.30, 95% CI: 0.09-1.01, P = .053; dobutamine, adjusted OR: 0.91, 95% CI: 0.29-2.84; and milrinone, adjusted OR: 0.60, 95% CI: 0.19-1.87). Conclusion: The present study showed that the use of epinephrine and dobutamine was associated with significantly increased in-hospital mortality in patients with septic shock. These effects were both time- and dose-dependent. On the other hand, the use of milrinone was not associated with increased in-hospital mortality.

Trehalose has been described to protect several enzymes against destabilizing conditions. This sugar is naturally accumulated by yeast as a stress protectant. A common stress condition that yeast is normally submitted is the presence of ethanol, the by-product of fermentation process of several yeast. In this paper we show the effects of trehalose and ethanol, alone or together, on yeast pyrophosphatase, and the effects of these compounds on inhibition and unfolding of pyrophosphatase promoted by urea. We show that both trehalose and ethanol inhibit pyrophosphatase in a dose-dependent manner, and that the presence of ethanol does not modify the inhibition promoted by trehalose as well as the presence of trehalose does not modify the inhibition promoted by ethanol. The effects of trehalose on pyrophosphatase are completely reversible, but the inhibition caused by ethanol was only partially reversible. Incubation of pyrophosphatase with 10% (v/v) ethanol promoted an inhibition of 15%, and the control activity was completely recovered after removal of ethanol. On the other hand, when pyrophosphatase was incubated with 20% (v/v) ethanol an inhibition of 40% of the control activity was observed which persisted after removal of ethanol. Ethanol also potentiates the inhibition of pyrophosphatase promoted by urea, and contributes for an irreversible inactivation and unfolding of pyrophosphatase in the presence of urea. Trehalose, that protects this enzyme against the inhibition and unfolding promoted by the chaotropic compound urea, was inefficient to protect against the effects of ethanol. Trehalose was also efficient to prevent an irreversible inactivation induced by urea.

21079 Background: Aminobisphosphonates (ABPs) has a definite direct anti-tumour activity but a limited activity in vivo. Their molecular targets are still not completely defined. Therefore, we have studied the effects of zoledronic acid (ZOL) addition to prostate cancer PC3 cells on gene expression profile. Methods: We have treated PC3 cells with 100 μM ZOL for 24 hours, extracted mRNAs and probed on Affimetrix HG-U133. Thereafter, we have identified down modulated and upregulated genes and checked for modulation of mRNA with RT PCR and of the relative encoded proteins with western blotting. Results: We have found 6 down modulated and 32 upregulated genes. We have focused our attention on NDRG1 associated to the androgen-differentiation and on Cysteine rich 61 (CYR61) involved in the regulation of proliferation and angiogenesis. NDRG1 mRNA was up-regulated and CYR61 mRNA was downregulated by ZOL in a dose-dependent manner. Similar effects were observed at protein product levels with an about 2-fold change recorded already in cells treated with 50 μM ZOL. Interestingly, also Gefitinib, Sorafenib and Tipifarnib used at their IC:50s could induce changes in both NDRG1 and CYR61 expression, but 50 μM ZOL was about 2-fold more potent. On the other hand, cytotoxic agents such as docetaxel did not have any effect. The addition of farnesol (FOH) or geranylgeraniol (GGOH) to ZOL-treated cells was able to counteract the effect of ZOL on CYR61 expression partially or completely, respectively. On the other hand, both FOH and GGOH had poor effect on the regulation of NDRG1 expression induced by ZOL. Conclusions: ZOL induces a strong regulation of the expression of NDRG1 and CYR61 at both mRNA and protein levels that appears to be dose-dependent and specific. CYR61 modulation seems to be more dependent from the inhibition of geranylgeranylation processes while NDRG1 changes could be at least in part independent from the inhibition of isoprenylation induced by ZOL. The study of the biological relevance of these effects on the anti-cancer effects of ZOL is ongoing with small interference RNA approaches. No significant financial relationships to disclose.

The effects of isoproterenol and forskolin on cyclic AMP levels and tension were compared in helical strips of bovine coronary artery. Both compounds produced time- and dose-dependent increases in cyclic AMP levels and both compounds relaxed potassium-contracted arteries. However, in contrast to the results with isoproterenol, cyclic AMP elevation and relaxation were not well correlated at lower concentrations of forskolin. For example, 0.1 μM forskolin increased cyclic AMP levels in the arteries by approximately 5.5-fold but did not relax the muscles. A smaller elevation of cyclic AMP produced by 1.0 μM isoproterenol, on the other hand, was accompanied by an almost complete relaxation of the arteries. These results suggest that either cyclic AMP is not responsible for the relaxation caused by these agents, or some form of functional compartmentalization of cyclic AMP exists in this tissue.

A hospital based cross sectional study was carried out among the under-five children admitted to the longer stay wards (LSU) of the hospital of ICDDR,B to investigate the relationship between children’s nutritional status and clinical recovery as well as sock, economic condition. Nutritional status was determined by anthropometric measurements. The study conducted on 94 children of 6-59 months, of whom 49 were malnourished and 49 were well nourished 64 of them were male and 35 were female. (It means that boy’s are more sufferers in diarrhea than girls.) (Children’s age come within a range of 6-59) month. Mother’s education level was lower, 32.6% mothers were illiterate and 40% mothers of malnourished children were illiterate where, 25% mothers having well nourished child were illiterate. Most of the malnourished children’s Family income was below TK. 5,000 per month for majority of the parents whereas, most of the well nourished children’s family income was 8,000tk. It shows that, children having low family income are more likely malnourished than well nourished children. Family member was within 4 persons for majority of the well nourished children’s households’. On the other hand family member was more than 5 in out of the malnourished households. Within their limited income; it was difficult to maintain a medium size family properly. Our study reveals that knowledge, attitude and practices of dietary pattern, health, sanitation and immunization of the malnourished children’s family were lower than well-nourished children’s family. Their nutritional knowledge was too low, compared with well nourished children’s parents. In our study, Most of the parents of well-nourished children used to feed colostrums (80.6%) but Parents having malnourished children discarded colostrums (93.6%). Our study reveals that 64.4% malnourished children’s mother has started complementary food in <2 months of age ,on the other hand majority of well-nourished children’s mother gave their child complementary food at the age of 6-9 months. It has been seen that, the majority of well nourished children (62%) were immunized by taking all doses of vaccines. where as most of the malnourished children (49%) has not taken all doses as a result there immunity was lesser than well-nourished children. Most of the caregivers of well-nourished children (43.5%) knows that diarrhea causes by not giving immunization, on the other hand majority of caregiver of malnourished patients (31.9%) knows that main cause of diarrhea is infection by germs. The immunization status of well nourished children was quite satisfactory. 62% well nourished children and only 40% Mal nourished children was completely immunized. It has been seen that, ORS intake was higher in well-nourished children (172.2lml-on the 1st day of hospitalization, compare to malnourished children. Stool output was higher in malnourished children 235.48 ml-on the 1st day after admission. It shows that there is a co-relation between ORS intake, stool output and recovery period. It has been seen that, mean duration of diarrhea before hospitalization was lower in well-nourished children group, but duration of fever, vomiting and abdominal pain rate was higher in this group compared to malnourished group. Algae were supplied continuously to four-armed pluteus larvae of the sea urchin Hemicentrotus pulcherrimus until the adult rudiment reached stage g, which is the initial stage of rudiment formation (Chino et al., 1994) After stage g, one group of larvae was reared without the addition of algae for comparison of the development of the adult rudiment with that in larvae given algae. Three days later, only 9.1 ± 0.3% of larvae without algae had reached stage j, while 76.8 ± 0.6% of larvae with algae had reached a stage beyond j, which indicates the formation of the complete adult rudiment.When larvae at rudiment stage g were reared in a medium supplemented with T4 or its derivatives, such as T3, 3,3′,5′-L-triiodothyronine (rT3) or triiodothyropropionine (Tp3), in place of algae, the adult rudiment developed in a dose-dependent manner. T4 was the most effective and induced formation of the adult rudiment in more than 70% of specimens at 1 nM and in almost 100% at 100 nM. T3 was one-tenth as effective as T4. Other derivatives were still less effective. On the other hand, casein, ovalbumin, tyrosine and a mixture of 20 amino acids had no effect on the development of larvae and adult rudiments, suggesting that they were not available as nutrients and sources of thyroid hormones.

AbstractWe have developed a model for theH2OandOHevolution in a comet outburst, assuming that together with the gas, a distribution of icy grains is ejected. With an initial mass of icy grains of 108kg released, theH2OandOHproductions are increased up to a factor two, and the growth curves change drastically in the first two days. The model is applied to eruptions detected in theOHradio monitorings and fits well with the slow variations in the flux. On the other hand, several events of short duration appear, consisting of a sudden rise ofOHflux, followed by a sudden decay on the second day. These apparent short bursts are frequently found as precursors of a more durable eruption. We suggest that both of them are part of a unique eruption, and that the sudden decay is due to collisions that de-excite theOHmaser, when it reaches the Cometopause region located at 1.35 × 105kmfrom the nucleus.
