Introduction: Tendinopathy is a pathology of tendons of multifactorial origin displayed clinically as chronic degenerative tissue resulting in dysfunction and painful musculoskeletal system. Genetic implications in tendinopathy are not fully elucidated. Haptoglobin (Hp) polymorphism has been investigated for potential role in several diseases such as cardiovascular diseases. This study aims at investigating Hp polymorphism potential association with chronic tendinopathy in Jordan. Materials and Methods: A hospital-based case-control study, of 101 patients diagnosed with chronic tendinopathy and 104 age and gender matched controls, was employed and Hpphenotypes distributions were determined using Sodium Dodecyl Sulphate Polyacrylamide gel electrophoresis technique. Results: showed a significant association between tendinopathy cases and controls in relation to Hpphenotypes frequencies (Fisher’s Exact Test, [Formula: see text]). Odd ratios showed that the odds of tendinopathy cases in Hp2.1 group were reduced while odds in Hp2.2 and Hp1.1 were increased by 3.22 and 7.34 times, respectively, as likely compared to the controls. Conclusion: Results suggested that Hppolymorphism might play a role in chronic tendinopathy genesis.

SummaryElevated plasma levels of lipoprotein(a) [Lp(a)] represent a significant independent risk factor for the development of atherosclerosis. Interindividual levels of apo(a) vary over 1000-fold and are mainly due to inheritance that is linked to the locus of the apolipoprotein(a) [apo(a)] gene. The apo(a) gene encodes multiple repeats of a sequence exhibiting up to 85% DNA sequence homology with plasminogen kringle IV (K.IV), a lysine binding domain. In our search for sequence polymorphisms in the K.IV coding domain, we identified a polymorphism predicting a Thr→Pro substitution located at amino acid position 12 of kringle IV type 8 of apo(a). The functional and clinical significance of this polymorphism was analysed in a case-control study and by comparing the in vitro lysine binding characteristics of the two Lp(a) subtypes. The case-control study (involving 153 subjects having symptomatic atherosclerosis and 153 age and gender matched normolipidemic controls) revealed an overall allele frequency for the Thr12-→Pro substitution in kringle IV type 8 of 14% and a negative association between presence of the Pro12-subtype and symptomatic atherosclerosis (p <0.03). The in vitro lysine binding studies, using Lp(a) isolated from subjects homozygous for either Thr12 or Pro12 in K.IV type 8, revealed comparable lysine-Sepharose binding fractions for the two subtypes. The binding affinity (Kd) for immobilised plasmin degraded des- AA-fibrin (DesafibTM-X) was also comparable for the two subtypes, however a decreased maximal attainable binding (Bmax) for immobilised desafibTM-X was observed for the Pro12-subtype Lp(a).

Background. The present study explores the influence of the host’s age and gender upon the inflammatory infiltrate. We aimed to quantify the inflammatory infiltrate caused by cysticercosis, which is related to aging, in the heart and in the encephalon.Methods. 75 autopsy protocols with cysticercosis diagnosis from department of pathology at a university hospital from 1970 to 2008 were reviewed. Two groups were formed: elderly with cysticercosis and nonelderly with cysticercosis. We used KS-300 (Kontron-Zeiss) software for morphometric analysis of the inflammation.Results.The elderly had an average of3.1±2.5cysticerci, whereas the non-elderly had2.7±3.8parasites. The non-elderly group with cysticercosis had significantly more inflammation, both cardiac and encephalic, than the elderly group. The elderly females with cysticercosis had more cardiac and encephalic inflammation.Conclusions. In this study, we showed that the non-elderly had significantly more cardiac and encephalic inflammation than the elderly, and that such inflammatory infiltrate decreases with age and depends upon the evolutionary stage of the cysticercus. Furthermore, there are differences concerning gender in the intensity of the inflammatory response due to cysticerci in the heart and brain parenchyma during senescence. Even during this period, women continue to have a more intense response to the parasitosis.

Tonsils are believed to be the initial site of the John Cunningham virus (JCV) infection. The long-term effect of childhood tonsillectomy on JCV status in multiple sclerosis (MS) patients has not been investigated. In this retrospective case–control study, we analyzed data of 144 JCV seropositive cases and 82 JCV seronegative controls from three outpatient MS clinics in the United States. Early tonsillectomy (before the age of 8) was reported among 8 (5.56%) JCV seropositive subjects and 19 (23.17%) controls. Early tonsillectomy was associated with JCV negative status (adjusted odds ratio = 5.39, 95% confidence interval = 2.13–13.62, p < 0.001) independent of age and gender.

Objectives. Routine measles-mumps-rubella (MMR) vaccine use has greatly decreased the incidence of mumps in the U.S. However, a resurgence of mumps occurred in 2006. We investigated the large outbreak at a university and assessed risk factors for disease. Methods. We described the outbreak and conducted a case-control study. We interviewed case students (identified from student health service and health department records) and control students (selected from a randomly ordered administrative list) and assessed their vaccination status. We compared case students with ≥2 MMR doses and control students with ≥2 MMR doses in univariate and multivariate analyses. Results. The mean age of the 174 case students was 20.9 years; 65% were women. Ninety-seven case students and 147 control students were enrolled in the study. Two-dose MMR coverage was 99% among case and control students with complete records. Only 33% of case students reported exposure to someone with mumps. Case students were more likely than control students to be aged 18 to 19 years (vs. aged 22 years, adjusted odds ratio [AOR] = 5.55; 95% confidence interval [CI] 2.09, 14.74), to report exposure to mumps (AOR=2.31, 95% CI 1.13, 4.73), and to have worked/volunteered on campus (AOR=2.91, 95% CI 1.33, 6.33). Also, women in dormitories had increased odds of mumps compared with men in dormitories. Conclusion. High two-dose MMR coverage was not sufficient to prevent the outbreak. Further study is needed to better understand the effects of dormitory residency and gender on mumps transmission. Clinicians should be vigilant for mumps in young adults presenting with parotitis regardless of immunization history.

Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) is the most important and widely used technology which is mainly used to analyze the protein molecular weight. Fibrin zymography based on the SDS-PAGE is the best method for qualitative analysis of unknown plasminogen activators (PAs), especially for the analysis of molecular weight. In electrophoresis technique, molecular weight marker is the most important factor. However, it is difficult to detect protein molecular weight markers in fibrin zymography. In this study, some important factors, such as concentrations of fibrinogen and plasminogen, are discussed. Our results provide an efficient and convenient method which can clearly exhibit the dark blue bands of protein molecular weight (MW) markers and the transparent bands of PAs against the light blue background on one gel at the same time, and show high sensitivity.
