Inhibitory effects of intestinal electrical stimulation on food intake, weight loss and gastric emptying in rats

The aim was to investigate the effects of intestinal electrical stimulation (IES) on food intake, body weight, and gastric emptying in rats. An experiment on food intake and weight change was performed in 22 rats on a control diet and 10 diet-induced obese (DIO) rats for 4 wk with IES or sham IES. The effect of IES on gastric emptying was performed in another 20 rats in the control group. We found that 1) in control rats, 4-wk IES resulted in a reduction of 18.2% in the total amount of food intake compared with sham-IES ( P = 0.02); the rats treated with IES had a weight change of −1 ± 7.8g ( P = 0.03), which was equivalent to a weight loss of 6.2% due to IES when adjusted for normal growing. 2) Acute IES delayed gastric emptying by 20% in the control rats ( P < 0.01). 3) In the DIO rats, 1-wk IES with the same parameters as those used in the control rats resulted in a significant reduction in the total amount of food intake (126.6 ± 6.3 g vs. 116.9 ± 3.2 g, P < 0.01). More reduction in food intake was noted, and a significant weight change was also observed when stimulation energy was increased. 4) No adverse events were observed in any of the experiments. In conclusion, IES delays gastric emptying, reduces food intake, and decreases weight gain in control growing rats. These data suggest that it is worthy to explore therapeutic potentials of IES for obesity.

The aim of this study was to investigate the effects and mechanisms of intestinal electrical stimulation (IES) on gastric tone, antral and pyloric contractions, and gastric emptying in dogs. Female hound dogs were equipped with a duodenal or gastric cannula, and one pair of serosal electrodes was implanted in the small intestine. The study consisted of five different experiments. Liquid gastric emptying was assessed by collection of chyme from the duodenal cannula in a number of sessions with and without IES and with and without N-nitro-l-arginine (l-NNA). Postprandial antral and pyloric contractions were measured with and without IES and in the absence and presence of l-NNA or phentolamine by placement of a manometric catheter into the antrum and pylorus via the duodenal cannula. Gastric tone was assessed by measurement of gastric volume at a constant pressure. Gastric emptying was substantially and significantly delayed by IES or l-NNA compared with the control session. IES-induced delay of gastric emptying became normal with addition of l-NNA. IES reduced gastric tone, which was blocked by l-NNA. IES also inhibited antral contractions (frequency and amplitude), and this inhibitory effect was not blocked by l-NNA but was blocked by phentolamine. IES alone did not affect pyloric tone or resistance, but IES + l-NNA decreased pyloric tone. In conclusion, IES reduces gastric tone via the nitrergic pathway, inhibits antral contractions via the adrenergic pathway, does not affect pyloric tone, and delays liquid gastric emptying. IES-induced delay of gastric emptying is attributed to its inhibitory effects on gastric motility.

Acupuncture may modulate the imbalance of the autonomic nervous system. It is well known that restraint stress delays gastric emptying via inhibiting parasympathetic activity and/or stimulating sympathetic activity in rats. We have previously shown that electroacupuncture (EA) improves delayed gastric emptying induced by restraint stress in rats. To investigate whether the beneficial effect of EA on delayed gastric emptying is associated with its modulatory effects on autonomic nervous activity, we utilized spectral analysis of heart rate variability (HRV). In rats, the power in the low frequency (LF; 0.04–1.0 Hz) and high frequency (HF; 1.0–3 Hz) band of HRV represent sympathetic and parasympathetic activities, respectively. Electrocardiography (ECG)-electrodes were implanted on the subcutaneous tissues of the back. One week after the surgery, ECG was recorded before, during and after the restraint stress loading in a conscious state. EA (10 Hz) was applied at bilateral acupuncture points [ST-36 (lower leg) or BL-21 (back)] during restraint stress loading. In response to restraint stress, heart rate and LF component were increased, suggesting the increased activity of sympathetic tone. EA at ST-36 significantly reduced the elevated heart rate and LF, compared to that of control group. EA at ST-36 also significantly increased HF component after finishing the stress loading. In contrast, EA at BL-21 had no significant effect on the heart rate, LF and HF. It is suggested that EA at ST-36 stimulates parasympathetic activity and inhibits sympathetic activity under the restraint stress in rats.

Weight loss resulting from decreased caloric intake raises levels of the orexigenic hormone, ghrelin. Because ingested nutrients suppress ghrelin, increased ghrelin levels in hypophagic weight loss may result from decreased inhibitory input by ingested food, rather than from lost weight. We assessed whether ghrelin levels increase in response to exercise-induced weight loss without decreased caloric intake. We randomized 173 sedentary, overweight, postmenopausal women to an aerobic exercise intervention or stretching control group. At baseline, 3 months, and 12 months, we measured body weight and composition, food intake, cardiopulmonary fitness (maximal oxygen consumption), leptin, insulin, and ghrelin. Complete data were available for 168 women (97%) at 12 months. Exercisers lost 1.4 ± 0.4 kg (P &lt; 0.05 compared with baseline; P = 0.01 compared with stretchers) and manifested a significant, progressive increase in ghrelin levels, whereas neither measure changed among stretchers. Ghrelin increased 18% in exercisers who lost more than 3 kg (P &lt; 0.001). There was no change in caloric intake in either group and no effect on ghrelin of exercise per se independent of its impact on body weight. In summary, ghrelin levels increase with weight loss achieved without reduced food intake, consistent with a role for ghrelin in the adaptive response constraining weight loss and, thus, in long-term body weight regulation.

SUMMARYIndividually-penned adult Merino wethers were fed for 16 weeks on a basal diet of ground, pelleted oat straw or Phalaris straw. Urea was supplied either in a salt-urea block containing 30% urea, or as crystalline urea incorporated into the pellets. A commercial mineral supplement was given during the first 8 weeks, but this was replaced with a laboratory-prepared mineral mix during the second 8 weeks.Urea supplementation significantly reduced live-weight loss. Over the 16 weeks control sheep lost a mean of 14·5 kg, and supplemented sheep 8·7 kg body weight. Most of this response to urea occurred during the second half of the experimental period.Food intakes of all sheep increased over the first 2 weeks and then fell. In the unsupplemented groups, levels of intake then remained fairly low, whereas in the sheep fed urea food intakes rose over the final 6 weeks of the experiment. There was a significant linear regression of live-weight change on food intake only with the urea-fed sheep. Intakes of urea-N from the blocks were extremely variable between individual sheep and between periods within sheep; in most cases, however, they appeared to be adequate, as this type of supplementation gave results similar to those obtained by mixing urea throughout the roughage.There was no significant effect of urea on total wool growth during the experimental period, although wool growth in the supplemented shepp showed a rise towards the end of the experiment. Wool growth in any 4-weekly period was closely related to food intake and live-weight change during that period.

Rapid degradation of glucagon-like peptide-1 (GLP-1) by dipeptidyl peptidase-4 suggests that endogenous GLP-1 may act locally before being degraded. Signaling via the vagus nerve was investigated in 20 truncally vagotomized subjects with pyloroplasty and 10 matched healthy controls. Subjects received GLP-1 (7-36 amide) or saline infusions during and after a standardized liquid mixed meal and a subsequent ad libitum meal. Despite no effect on appetite sensations, GLP-1 significantly reduced ad libitum food intake in the control group but had no effect in the vagotomized group. Gastric emptying was accelerated in vagotomized subjects and was decreased by GLP-1 in controls but not in vagotomized subjects. Postprandial glucose levels were reduced by the same percentage by GLP-1 in both groups. Peak postprandial GLP-1 levels were approximately fivefold higher in the vagotomized subjects. Insulin secretion was unaffected by exogenous GLP-1 in vagotomized subjects but was suppressed in controls. GLP-1 significantly reduced glucagon secretion in both groups, but levels were approximately twofold higher and were nonsuppressible in the early phase of the meal in vagotomized subjects. Our results demonstrate that vagotomy with pyloroplasty impairs the effects of exogenous GLP-1 on food intake, gastric emptying, and insulin and glucagon secretion, suggesting that intact vagal innervation may be important for GLP-1's actions.

The aim of this study was to investigate the plasma and tissue levels of nesfatin-1 and irisin hormones, which were discovered in recent years and are associated with endocrine and metabolic functions, in kumiss-supplemented mice. Sixteen BALB/C male mice were divided into two groups as control and kumiss groups. During the experiment, the kumiss was added to the drinking water of mice at a ratio of 1:1 to obtain a daily 2×108 cfu/mL bacterial colony, and was given once a day orally for 20 weeks. The weights and food intake of the animals were monitored during the experiment. The nesfatin-1 and irisin levels in plasma and tissue samples were determined using ELISA kits. Kumiss supplementation reduced the live weight for 2-12 weeks (P<0.05). However, no significant difference was observed after the 12th week. The feed consumption of the kumiss group was lower at the beginning and the 10th week, and at the end, compared to the control group (P<0.05). The plasma levels of nesfatin-1 and irisin (P<0.001) decreased while the liver levels increased (P<0.05 and P<0.001, respectively). The results indicate that plasma and liver levels of nesfatin-1 and irisin are regulated by diet and are effective in weight loss and food intake.
