In vitro experiment was carried out to see the effect of saw dusts of Pinus kesiya, Shorea robusta, and Callicarpa arborea on Trichoderma harzianum, isolate TH-13 mass production, along with its biotization effect on Capsicum chinensis Jacq. “Bhut Jolokia.” Early mycelium initiation (2 days) occurred in S. robusta followed by P. kesiya and C. arborea (3.5 days). The sporulation was observed earlier in S. robusta (100% after 6 days) than P. kesiya (33.4% after 8 days) and C. arborea (16.7% after 9 days) but no sporulation was observed in control. The complete sporulation was also earlier in S. robusta (100% after 10 days) than P. kesiya (33.4% after 15 days) and C. arborea (16.4% after 18 days). Higher conidial yield 86×106 was also in S. robusta than P. kesiya 70×106 and C. arborea 45×106, respectively. The increase in height (60–70 cm), number of leaves (600–650), and yield of chili (120–150 fruits) were also more in inoculated C. chinensis seedlings than control. It is concluded that S. robusta saw dust is the best substrate for mass production of compost fungal activator and can be used in nursery practices for quality stock production of various crops/plantations.

The experiment was conducted to investigate the combined effect of different plant growth regulators and charcoal supplementation in MS medium on growth and development of plantlets regenerated from protocorm like bodies (PLBs) of hybrid orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations with charcoal supplementation was studied. The result revealed that the use of different growth regulators had significant effect on different parameters studied. The maximum weight of PLBs (5.123 g) was obtained from the combination of BAP + IBA at 1.0 mg/l each. The highest shoot height (3.239 cm) and maximum number of rooted plantlets (4.473) was obtained from 1.0 mg/l each of BAP + NAA combination. The maximum number of leaves (3.490) and the maximum length of leaves (1.946 cm) were obtained from 1.0 mg/l each of BAP + IBA and the highest leaf width (1.166 cm) was obtained from 0.5 mg/l BAP +1.0 mg/l IBA combination. The highest root length was obtained from 0.5 mg/l each of BAP + IAA and the maximum number of regenerated plantlets (20) was obtained from 0.5 mg/l IAA + 1.0 mg/l IBA combination. However, the maximum fresh weight of single shoot (0.219 g) and the maximum number of roots per plantlet (6.300) was obtained from 1.0 mg/I each of IAA + IBA combination. The present study was conducted to investigate the effect of growth regulator NAA on in vitro shoot proliferation, rooting, and plantlet establishment. Among the different concentrations of NAA, the best increase in shoot weight (0.25 g) and shoot number (8.83) were observed from 0.1 mg/I NAA. The highest shoot length (2.60 cm), number of leaves (4.83), number of roots (5.15), and root length (2.67 cm) were obtained with 0.2 mg/I NAA at 60 DAT. Key Words: Dendrobium orchid, NAA, MS media. Abstract Taro Colocasia esculenta is a staple food in most countries of the South Pacific region. However, the recent changes in the climatic conditions and the associated factors became a threat to growing taro in the region. Rising sea level and frequent dry and wet conditions had led to increased soil salinity which is a limiting factor in producing taro in these atoll islands. The aim of this research was to screen selected taro varieties based on their ability to tolerate salinity conditions in Samoa. Effects of three salinity levels (0.2, 3% and 5%), and growth hormones Benzylaminopurine (BAP) and Thidiazuron (TDZ) were examined on growth and development of in vitro grown taro varieties. Plant height, number of leaves, and number of roots produced were compared in the selected varieties. Results suggest that all varieties had callus induction and regeneration in MSO medium. Addition of BAP induced better growth and the plants produced higher number of leaves, while TDZ improved roots proliferation in C. esculenta. Solomon variety performed best in high salinity conditions. Samoa-1 grew taller and had more leaves and roots compared to Samoa-2 variety. Thus, we suggest Solomon and Samoa-1 could perform better in high-salinity conditions.

This study was conducted at the plant tissue plant of the Biotechnology Research Center for the purpose of determining the effect ofadding different concentrations of Nacl on the growth and development of the citrus rootstocks (Narang and Cleopramtradrin). Citrus seeds were cultured on MS media supplemented with different concentrations ofNaCL100,80,60,40,20,0 mM. The result showed that citrus rootstocks growth negatively infected by salinity while plant length and number of laves decreased by increasing of salinityfor booth rootstocks .plant length and number of leaves were significantly higher in treatment 0 and 20 mMNaCL compared with other treatments, also the results showed that the plants continued to grow well until 60 mM concentration..

This article presents the results of experimental studies of the effect of biostimulants Agrinos 1 and Agrinos 2, containing strains of microorganisms Azotobacter vinelandii and Clostridium pasteurinum, etc., as well as a complex of nutrients (macro-, microelements, protein, amino acids, chitin, chitosan) on the activation of metabolic processes and reduction of stress effects on strawberry plants obtained by in vitro method. The effectiveness of the reaction of biostimulants on the adaptive processes of growth and development of microplants of garden strawberry varieties Alba, Kemiya, Clery has been studied. In the course of the research, a positive effect of biological products on the growth of the vegetative mass of plants was established: an increase in the height of the plants compared to the control by 20-25%, the number of leaves by 25-28% and an increase in the root system of plants by 30-40%. In the Clery variety, the use of the biostimulants promoted the formation of the runners. In addition, the introduction of biostimulants into the soil contributed to the improvement of the quality indicators of the soil. The content of nitrate nitrogen in the soil increased by 18%.

ABSTRACT Micropropagation of small fruits such as blackberry has been employed due to the need to obtain plants with high phytosanitary quality. Bioreactor technology has been used to improve efficiency in seedling production. Thus, the objective of this work was to evaluate the best culture medium volume and sucrose concentration for blackberry micropropagation in a temporary immersion bioreactor. In vitro blackberry shoots were segmented containing two buds and an internode (1.0 cm) and placed into MS medium supplemented with inositol (0.1 g L-1), BAP (1 mg L-1) and sucrose (10, 20, 30 or 40 g L-1) at different medium volumes (150, 175 and 200 mL). The total length, number of leaves, number of shoots, and number of hyperhydric shoots were evaluated 56 days after start of the project. For blackberry development and propagation in a bioreactor system, the best results were shown at a medium volume of 175 ml and a sucrose concentration of 20 g L-1.

This study aimed to characterize the halotolerant capability, in vitro, of selected actinomycetes strains and to evaluate their competence in promoting halo stress tolerance in durum wheat in a greenhouse experiment. Fourteen isolates were tested for phosphate solubilization, indole acetic acid, hydrocyanic acid, and ammonia production under different salt concentrations (i.e., 0, 0.25, 0.5, 0.75, 1, 1.25, and 1.5 M NaCl). The presence of 1-aminocyclopropane-1-carboxylate deaminase activity was also investigated. Salinity tolerance was evaluated in durum wheat through plant growth and development parameters: shoot and root length, dry and ash-free dry weight, and the total chlorophyll content, as well as proline accumulation. In vitro assays have shown that the strains can solubilize inorganic phosphate and produce indole acetic acid, hydrocyanic acid, and ammonia under different salt concentrations. Most of the strains (86%) had 1-aminocyclopropane-1-carboxylate deaminase activity, with significant amounts of α-ketobutyric acid. In the greenhouse experiment, inoculation with actinomycetes strains improved the morpho-biochemical parameters of durum wheat plants, which also recorded significantly higher content of chlorophylls and proline than those uninoculated, both under normal and stressed conditions. Our results suggest that inoculation of halotolerant actinomycetes can mitigate the negative effects of salt stress and allow normal growth and development of durum wheat plants.
