The seeds ofPsoralea corylifoliaL. (Fabaceae) are a commonly used medicinal herb in eastern Asia with many beneficial effects in clinical therapies. In this study, a simple, sensitive, precise, and specific reverse phase high-performance liquid chromatography (HPLC) method was established for quantification of 9 flavonoids inP. corylifolia, including isobavachin, neobavaisoflavone, bavachin, corylin, bavachalcone, bavachinin, isobavachalcone, corylifol A, and 4′-O-methylbavachalcone. Based on this method, a quantitative analysis of multicomponents by single marker (QAMS) was carried out, and the relative correction factors (RCFs) were calculated for determining the contents of other flavonoids. The accuracy of QAMS method was verified by comparing with the results of external standard method, as well as the feasibility and adaptability of the method applied on quality control ofP. corylifolia. The 9 compounds were baseline separated in 60 min with a good linearity of regression coefficient(R2)over 0.9991. The accuracies of QAMS were between 92.89% and 109.5%. The RSD values offin different injection volume were between 2.3% and 3.6%. The results obtained from QAMS suggested that it was a convenient and accurate method to determine multicomponents especially when some authentic standard substances were unavailable. It can be used to control the quality ofP. corylifolia.

A valid and encyclopaedic evaluation method for assessing the quality of Sanhuang Gypsum Soup (SGS) has been set up based on analysis of high-performance liquid chromatography (HPLC) fingerprint combined with the quantitative analysis of multicomponents by single marker (QAMS) method, hierarchical cluster analysis (HCA), and similarity analysis. 20 peaks of the common model were obtained and used for the similarity analysis and HCA analysis. Berberine was selected as an internal reference, and the relative correction factors of mangiferin, geniposide, liquiritin, epiberberine, coptisine, baicalin, palmatine, harpagosid, wogonoside, cinnamic acid, cinnamic aldehyde, baicalein, glycyrrhizic acid, and wogonin were established. The accuracy of quantitative analysis of multicomponents by the single-marker method was verified by comparing the contents of the fourteen components calculated by the external standard method with those of the quantitative analysis of multicomponents by the single-marker method. No significant difference was found in the quantitative results of the established quantitative analysis of multicomponents by a single-marker method and an external standard method. In summary, these methods were applied to evaluate the quality of SGS successfully. As a result, these evaluation methods have great potential to be widely used in the quality control of traditional Chinese medicines (TCM).

Abstract Capilliposide B (CPS-B) and Capilliposide C (CPS-C), as the key components in Lysimachia capillipes Hemsl., increasingly aroused the interest and research concern of many researchers due to the good bioactivities. Nowadays, the reference standards of CPS-B and CPS-C yield were very limited. Due to the deficit of reference standards, the determination could be difficult to carry out, and the quality control and evaluation would be restrained afterwards. To solve this urgent problem, a quantitative analysis of multi-components by single-marker (QAMS) method was proposed and established based on high-performance liquid-chromatography tandem evaporative light-scattering detector. In this QAMS method, the content of the two bioactive components could be calculated by buddlejasaponin IV, which is applied as an external standard and readily obtained. And the methodological experiments were evaluated and indicated accuracy, stability and feasibility of this QAMS method. Therefore, in this study, this built method would properly meet the requirement of determination of CPS-B, CPS-C and quality control of the L. capillipes Hemsl. plant.

Fermented Cordyceps sinensis, the succedaneum of Cordyceps sinensis which is extracted and separated from Cordyceps sinensis by artificial fermentation, is commonly used in eastern Asia in clinical treatments due to its health benefit. In this paper, a new strategy for differentiating and comprehensively evaluating the quality of products of fermented Cordyceps sinensis has been established, based on high-performance liquid chromatography (HPLC) fingerprint analysis combined with similar analysis (SA), hierarchical cluster analysis (HCA), and the quantitative analysis of multicomponents by single marker (QAMS). Ten common peaks were collected and analysed using SA, HCA, and QAMS. These methods indicated that 30 fermented Cordyceps sinensis samples could be categorized into two groups by HCA. Five peaks were identified as uracil, uridine, adenine, guanosine, and adenosine, and according to the results from the diode array detector, which can be used to confirm peak purity, the purities of these compounds were greater than 990. Adenosine was chosen as the internal reference substance. The relative correction factors (RCF) between adenosine and the other four nucleosides were calculated and investigated using the QAMS method. Meanwhile, the accuracy of the QAMS method was confirmed by comparing the results of that method with those of an external standard method with cosines of the angles between the groups. No significant difference between the two methods was observed. In conclusion, the method established herein was efficient, successful in identifying the products of fermented Cordyceps sinensis, and scientifically valid to be applicable in the systematic quality control of fermented Cordyceps sinensis products.

This paper establishes the HPLC method of determining three nitrophenol isomers in municipal sewage, surface water and tap water. After the sample is filtered and distilled, conduct the monitoring analysis on the distillate with HPLC-DAD and quantify it in external standard method. In the municipal sewage treated in bioanalysis method one type of target object o-nitrophenol is found, whose content is 0.11mg·L-1, no target object is found in all the other samples. This method has the adventages of high separation efficiency and high analysis rate. The experiment result shows that the linear correlation coefficient is above 0.983, the recovery rate lies between 78.6% and 105.2%, the minimum detection limit is 0.05mg·L-1(S/N=3.0).

Objective. To develop a quantitative analysis of multicomponents by single-marker (QAMS) method for the simultaneous determination of polar active components inFructus Corni.Methods. Loganin was selected as the internal reference, and the relative correction factors (RCFs) of gallic acid, 5-hydroxymethyl-2-furfural, morroniside, sweroside, cornin, 7α-O-methylmorroniside, 7β-O-methylmorroniside, 7α-O-ethylmorroniside, 7β-O-ethylmorroniside, and cornuside were established. The contents of multicomponents were then calculated based on their RCFs, respectively. Contents of the 11 components were also calculated by external standard method and compared with those of the QAMS method.Results.The contents of the 11 components in 21 crude and 10 processedFructus Corniproducts were measured. No significant difference was found in the quantitative results of the QAMS and external standard methods.Conclusion. QAMS could serve as an accurate and convenient method in determining the polar and active components inFructus Corniand its processed products.

Objective. To establish a quantitative analysis of multicomponents by single marker (QAMS) method for the simultaneous determination of 4 active components such as protocatechuic acid, catechin, quercetin, and luteolin in Plantago depressa. Method. 4 active components in Plantago depressa were studied. Quercetin was used as an internal reference to establish the relative correction factors among protocatechuic acid, catechin, and luteolin and calculate the contents of each component; the results were compared with those measured by the external standard method. Results. 4 components showed a good linear relationship in their respective concentration ranges (r > 0.9995). The relative correction factors (fs/k) of protocatechuic acid, catechin, and luteolin were 1.1992, 0.8613, and 1.6069, respectively. The method had good durability. The contents of protocatechuic acid, catechin, and luteolin calculated by QAMS were not significantly different from those measured by the external standard method. Conclusion. QAMS can be used to determine the content of 4 components in Plantago depressa at the same time, and the method is simple, accurate, and can be used for quality control.

In this paper, a valid evaluation method for the quality control of Danggui Kushen pills (DKP) has been established based on quantitative analysis of multicomponents by single marker (QAMS). Gallic acid, matrine, oxymatrine, catechin, ferulic acid, and rutin were selected as the indexes for quality evaluation of DKP. The analysis was achieved on an Agilent ZORBAX SB-C18 column (250  mm × 4.6  mm, 5 μm) via gradient elution. Gallic acid was used as internal standard to determine the relative correction factors (RCF) between gallic acid and other five constituents in DKP. The contents of those components were calculated at the same time. The accuracy of QAMS method was verified by comparing the contents of six components calculated by external standard (ES) method with those of the QAMS method. It turned out that there was no significant difference between the quantitative results of QAMS method and external standard method. The proposed QAMS method was proved to be accurate and feasible according to methodological experiments, which provided an accurate, efficient, and economical approach for quality evaluation of DKP.

ABSTRACT The issue of food safety has acquired increased importance, and fraud is a major concern for the food industry. Among different types of food adulteration, there is the sale of frozen-thawed smoked salmon product as fresh, which not only decreases the quality of products but also misleads consumers and may involve associated health risks. In response to this problem, we tested the performance of histology to identify smoked salmon as fresh or frozen-thawed as a valid analytical method, so food business operators and official controllers can reliably and correctly classify the storage state of the product. Three groups of samples were prepared: group A (n = 36), fresh samples; group B (n = 36), frozen at −18°C for 30 days; and group C (n = 36), stored at −3°C for 30 days after packaging. Two histopathologists examined all samples in blind evaluations and classified them as fresh or frozen-thawed. Sensitivity, specificity, and interrater agreement were calculated. Results show high performance with the test: 80.6% sensitivity (95% confidence intervals [CI]: 64 to 91.8%); 95.6% specificity (95% CI: 89.1 to 98.8%); and Cohen's kappa was 0.81 (95% CI: 0.64 to 0.98%). Histology is a reliable and highly accurate method to differentiate fresh from frozen-thawed smoked salmon and could be used by the industry and official controllers to verify the labeling of the commercial product. Introduction. Recently, mass media published a number of articles doubting health promoting effect of manuka honey (MH). On the other hand, many researchers emphasize beneficial effect of honey on human digestive tract, respiratory system and skin. However, the honeys tested for their health promoting effects came from certified sources and met the applicable harvesting, storage and packing standards between the production and testing. Aim. The purpose of the study was to compare quality features of manuka honey (MH) imported to and sold in, Poland: methylglyoxal (MGO), 5-hydroxymethylfurfural (HMF) and diastase number (DN) declared for product lots by producers and distributors vs. the actual figures. Material and methods. The study tested 45 samples of MH coming from 9 producers, acquired from different local suppliers (importers and distributors), marketed in Poland in 2014-2015. The contents of MGO were measured using the high-performance liquid chromatography (HPLC) method. The contents of HMF were measured using high-performance liquid chromatography with spectrophotometric detection (HPLC-UV) in accordance with the regulation by the Polish Minister of Agriculture. DN were determined using the photometric method acc. To the regulation by the Polish Minister of Agriculture. Results. A review of the test results revealed a relationship between the declared and the actual contents of MGO. For the MH with MGO > 500, the actual values were different more frequently. For the MH with higher declared MGO contents (< 500), commercial quality of the products was worse (too high HMF and too low DN). Conclusions. The MH with MGO under 500 mg/kg can be recognized as satisfactory in terms of MGO contents and commercial quality.

Pharbitidis Semen is a traditional Chinese medicine(TCM) with a long history for treatment of edema and fullness, fecal and urinary retention, phlegm and retained fluid, and abdominal pain due to parasitic infestation. Since Pharbitidis Semen is distributed throughout the country, the quality of the medicine from different origins may be varied. Moreover, the reported method could not control the quality comprehensively. In this article, a fingerprint of Pharbitidis Semen has been established based on a high-performance liquid chromatography (HPLC) method. In addition, the contents of the 2 main effective components were determined simultaneously. The reference HPLC fingerprint was obtained according to the chromatograms of test samples. The similarity values were calculated by the Similarity Evaluation System for Chromatographic Fingerprint of TCM (2004 A edition). Cluster analysis of 10 batches of samples was performed using statistical software (SPSS 20.0). The HPLC fingerprints of 10 batches of Pharbitidis Semen showed 25 well-resolved common peaks in each chromatogram. Two of these peaks were assigned to protocatechuic aldehyde and caffeic acid. As a result, HPLC fingerprint similarities of 10 batches of samples were more than 0.99. Pharbitidis Semen from different habitats could be divided into 3 or 2 groups. The results of cluster analysis showed that samples classified into 1 group were associated with their habitats and breeds. At the same time, quantification results showed that the contents of protocatechuic aldehyde and caffeic acid were in the range of 0.026-0.088 and 0.019-0.053 mg/g−1 respectively. HPLC fingerprint combined with multicomponent quantification and data analysis techniques can be an efficient and useful method for monitoring the quality of Pharbitidis Semen. This study also provides a practical strategy for overall quality evaluation and control of traditional Chinese medicines.
