Valorisation of Argan oil requires the precise identification of different provenances markers. The concentration of tocopherol is regarded as one of the essential parameters that certifies the quality and purity of Argan oil. In this study, 39 Argan samples from six different geographical origins (Safi, Essaouira, Agadir, Taroudant, Tiznit, and Sidi Ifni) from the central west of Morocco were collected and extracted using cold pressing. The total tocopherol amount was found to range from 783.23 to 1,271.68 mg/kg. Generally, γ-tocopherol has the highest concentration in Argan oil. It should also be noted that the geographical origin was found to have a strong effect on the amounts of all tocopherol homologues studied. Principal component analysis of tocopherol concentrations highlighted a significant difference between the different provenances. The content of tocopherol has also been found to be strongly influenced by the distance from the coast and altitude, whereas no significant effect was found regarding other ecological parameters. The prediction ability of the LDA models was 87.2%. The highest correct classification was revealed in coastal provenances (100%), and the lowest values were from the continental ones (71.4%). These results provide the basis for determining the geographical origins of Argan oil production with well-defined characteristics to increase the product’s value and the income of local populations. In addition, this study provides a very promising basis for developing Argan varieties with a high content of tocopherol homologues, as well as contributing to the traceability and protection of Argan oil’s geographical indication.

This study aimed to construct objective and accurate geographical discriminant models for tea leaves based on multielement concentrations in combination with chemometrics tools. Forty mineral elements in 87 tea samples from three growing regions in Guizhou Province (China), namely Meitan and Fenggang (MTFG), Anshun (AS) and Leishan (LS) were analyzed. Chemometrics evaluations were conducted using a one-way analysis of variance (ANOVA), principal component analysis (PCA), linear discriminant analysis (LDA), and orthogonal partial least squares discriminant analysis (OPLS-DA). The results showed that the concentrations of the 28 elements were significantly different among the three regions (p < 0.05). The correct classification rates for the 87 tea samples were 98.9% for LDA and 100% for OPLS-DA. The variable importance in the projection (VIP) values ranged between 1.01–1.73 for 11 elements (Sb, Pb, K, As, S, Bi, U, P, Ca, Na, and Cr), which can be used as important indicators for geographical origin identification of tea samples. In conclusion, multielement analysis coupled with chemometrics can be useful for geographical origin identification of tea leaves.

Objective To extract information that affect sport performance, the technique of urine metabolism and quantitative difference(QD)analysis were combined in search of the characteristic metabolites of the rowing athletes. Methods Morning urine were collected in three consecutive weeks, main peaks of the 1H NMR spectrum which have significant difference between the medalists and non-medalists were selected from thousands of one dimensional NMR hydrogen spectrum of urine. Pattern recognition method based on metabonomics combined with QD so that the metabolites which could reflect the competitive level of the elite athletes could be selected from the main components. Results 1.The optimal principal component of rowing athletes were principal component 1 and 5(PC1&PC5). 2. Results of 7-fold cross validation showed that the PLS-DA model were stable, reliable and has good prediction ability. Results of the repeatability experiment showed that sample test accuracy were above 85%. 3. N-methylnicotinamide was obtained by multi-criteria assessment methods as the characteristic metabolite. 4. Substance concentration related to aerobic and anaerobic metabolism were different in urine of the rowing athletes. Conclusions The athletes' urine contains the information of sport performance. Metabonomics combined with QD analysis could be widely applied in the evaluation of rowing athletes' competitive ability. The role of nicotinic acid in regulating energy metabolism and protecting human body might be a necessary condition with which athletes could tolerance high strength training and competition.

Abstract Hepatocellular carcinoma (HCC) is the most prevalent cancer in the liver. The majority of ingested tryptophan is processed in the liver through the kynurenine pathway, the endpoint of which is de novo NAD+ biosynthesis. Dysregulation of tryptophan-kynurenine metabolism and NAD+ synthesis may promote mitochondrial malfunction, tumor reprogramming, and carcinogenesis. Using a publicly available gene expression dataset from liver hepatocellular carcinoma (LIHC) samples available through The Cancer Genome Atlas (TCGA; n = 371), we employed Principal Component Analysis (PCA), hierarchical clustering, gene-pattern expression profiling, and survival analysis to cluster patients and determine overall survival. Our analysis of genes encoding kynurenine pathway enzymes determined that patients with high QPRT expression had a poor prognosis with decreased median survival, with no effect on the maximum survival. There is a significant difference in the survival between patients with high QPRT expression relative to patients with high HAAO/AFMID expression (HR = 1.2, [95% CI 0.5-1.8] P = 0.0181, Gehan-Breslow-Wilcoxon Test). Patients with high QPRT expression have higher survival rates compared with low QPRT expression (HR = 1.4, [95% CI 0.9-2.2] P = 0.0344, Gehan-Breslow-Wilcoxon Test). To test the consequences of kynurenine-pathway inhibition in mitochondrial function and morphology we use 4-Cl-3HAA, an irreversible HAAO inhibitor, and observed a small increase in mitochondrial fragmentation in HepG2 cells after 24 hours of treatment. We conclude that kynurenine metabolism may be useful as a biomarker to predict patient prognosis among HCC patients. In ongoing work, we are testing QPRT inhibitors in cell culture as a potential adjuvant for chemotherapies.

In this work, fatty-acid profiles, including trans fatty acids, in combination with chemometric tools, were applied as a determinant of purity (i.e., adulteration) and provenance (i.e., geographical origin) of cosmetic grade argan oil collected from different regions of Morocco in 2017. The fatty acid profiles obtained by gas chromatography (GC) showed that oleic acid (C18:1) is the most abundant fatty acid, followed by linoleic acid (C18:2) and palmitic acid (C16:0). The content of trans-oleic and trans-linoleic isomers was between 0.02% and 0.03%, while trans-linolenic isomers were between 0.06% and 0.09%. Discriminant analysis (DA) and orthogonal projection to latent structure—discriminant analysis (OPLS-DA) were performed to discriminate between argan oils from Essaouira, Taroudant, Tiznit, Chtouka-Aït Baha and Sidi Ifni. The correct classification rate was highest for argan oil from the Chtouka-Aït Baha province (90.0%) and the lowest for oils from the Sidi Ifni province (14.3%), with an overall correct classification rate of 51.6%. Pairwise comparison using OPLS-DA could predictably differentiate (≥0.92) between the geographical regions with the levels of stearic (C18:0) and arachidic (C20:0) fatty acids accounting for most of the variance. This study shows the feasibility of implementing authenticity criteria for argan oils by including limit values for trans-fatty acids and the ability to discern provenance using fatty acid profiling.

A total of 779 samples of edible nuts (melon seeds, watermelon seeds, pumpkin seeds, and cantaloupe seeds) from Southern Punjab (Pakistan), were collected during the summer and the winter seasons. The natural occurrence of aflatoxins (AFs) and vitamin E (tocopherols) levels were investigated using HPLC. The results have shown that 180 (43.4%) of samples from the winter season and 122 (33.4%) samples from the summer season were found positive for AFs. Elevated average levels of total AFs (20.9 ± 3.10 μg/kg, dry weight) were observed in watermelon seeds without shell, and the lowest average amount (15.9 ± 3.60 μg/kg) were documented in melon seeds without shell samples from the winter season. An elevated average amount of total AFs 17.3 ± 1.50 μg/kg was found in pumpkin seeds available without a shell. The results have documented a significant difference in total AFs levels in edible seeds available with shells versus without shells (α = 0.05 & 0.01). The highest dietary intake of 6.30 μg/kg/day was found in female individuals from consuming pumpkin seeds (without shell) in the winter season. A value of 3.00 μg/kg/day was found in pumpkin seed without shell in the summer season in female individuals. The highest total tocopherol levels were 22.2 ± 7.70 ng/100 g in pumpkin seeds samples from the winter season and 14.5 ± 5.50 mg/100 g in melon seed samples from the summer season. The variation of total tocopherol levels in edible seeds among the winter and summer seasons showed a significant difference (p ≤ 0.0054), except watermelon seeds samples with non-significant differences (p ≥ 0.183).

Leaves are plant structures that express important traits of the environment where they live. Leaf description has allowed identification of plant species as well as investigation of abiotic factors effects on their development, such as gases, light, temperature, and herbivory. This study described populations of Dalbergia ecastaphyllum through leaf geometric morphometrics in Brazil. We evaluated 200 leaves from four populations. The principal component analysis (PCA) showed that the first four principal components were responsible for 97.81% of variation. The non-parametric multivariate analysis of variance (NPMANOVA) indicated significant difference between samples (p = 0.0001). The Mentel test showed no correlation between geographical distances and shape. The canonical variate analysis (CVA) indicated that the first two variables were responsible for 96.77 % of total variation, while the cross-validation test showed an average of 83.33%. D. ecastaphyllum leaves are elliptical and ovate.

Since the nineties of the previous century, qualified geographic names have been covered by a harmonised EU-wide protection system whose main feature is the recognition of exclusive rights. Such rights are generally parallel to those deriving from the registration of a brand. Accordingly, they are not solely protected by the measures provided in the rules on unfair competition but also by a EU protection system based on granting the group of traders that had sought and obtained recognition of the protected designation of origin or geographical indication a monopoly over the use of a given geographic name and the possibility to seek remedy against any unlawful use of it. In this sense, the information provided generally benefits the market as well as merchants and consumers.

Free amino acids in cacao beans are important precursors to the aroma and flavor of chocolate. In this research, we used inferential and explanatory statistical techniques to verify the effect of different edaphic crop conditions on the free amino acid profile of PH-16 dry cacao beans. The decreasing order of free amino acids in PH-16 dry cacao beans is leucine, phenylalanine, glutamic acid, alanine, asparagine, tyrosine, gamma-aminobutyric acid, valine, isoleucine, glutamine, lysine, aspartic acid, serine, tryptophan, threonine, glycine. With the exception of lysine, no other free amino acid showed a significant difference between means of different edaphic conditions under the ANOVA F-test. The hydrophobic free amino acids provided the largest contribution to the explained variance with 58.01% of the first dimension of the principal component analysis. Glutamic acid stands out in the second dimension with 13.09%. Due to the stability of the biochemical profile of free amino acids in this clonal variety, it is recommended that cacao producers consider the genotype as the primary source of variation in the quality of cacao beans and ultimately the chocolate to be produced.

AbstractSiraitia grosvenorii (LHG) is widely used as a medicinal and edible material around the world. The objective of this study was to develop an effective method for the authentication of the geographical origin of LHG in its main producing area Guangxi, China, which is identified as Chinese Protected Designation of Origin product, against other producing regions in China. The content of 14 elements (K, Na, Ca, P, Mg, Al, B, Ba, Cu, Fe, Mn, Ni, Zn, and Sr) of 114 LHG samples was determined by inductively coupled plasma optical emission spectrometry. Multivariate analysis was then performed to classify the geographical origin of LHG samples. The contents of multielement display an obvious trend of clustering according to the geographical origin of LHG samples based on radar plot and principal component analysis. Finally, three supervised statistical techniques, including linear discriminant analysis (LDA), k-nearest neighbours (k-NN), and support vector machine (SVM), were applied to develop classification models. Finally, 40 unknown LHG samples were used to evaluate the predictive ability of model and discrimination rate of 100%, 97.5% and 100% were obtained for LDA, k-NN, and SVM, respectively. This study indicated that it is feasible to attribute unknown LHG samples to its geographical origin based on its multielement content coupled with chemometric techniques.

Abstract. In southern Brazil there are several problems regarding the hematophageal activity of black flies, and the information on the distribution of species are still insufficient. In this study, our main goals were to recognize Simuliidae species and to analyze abiotic factors regarding the distribution of immature stages (larva and pupa) in streams from the Ijuí River basin, Northwest of Rio Grande do Sul, Brazil. We sampled simulids specimens in 27 study sites, covering three regions (high, intermediate and low altitude). Concomitantly, we recorded abiotic factors in the studied sites, such as: temperature, electrical conductivity, dissolved oxygen, atmospheric pressure, pH, altitude and stream width. For data analysis, we performed Kruskal-Wallis, ANOVA, Principal Component Analysis (PCA), Analysis of Indicator Species and Canonical Correspondence (CCA). A total of 10 species of black Flies was recorded. Simulium pertinax Kollar, Simulium incrustatum Lutz and Simulium orbitale Lutz were the most frequent. There was no significant difference in richness and density of the species between the different regions studied, except for Simulium jujuyense Paterson & Shannon. The sampled environments were segregated according to the location within the basin, and only S. jujuyense was considered an indicator species of the upper region. These results can be explained by the homogeneous conditions of the Ijuí River basin, which have suffered anthropogenic pressure mainly due to agricultural activities, resulting in losses in local aquatic biodiversity. Also, it may indicate that the control must be conducted considering the basin as a whole, and not only as isolated stretches of the streams.Diversidade e distribuição espacial de borrachudos (Diptera: Simuliidae) na bacia hidrográfica do rio Ijuí, Rio Grande do Sul, BrasilResumo. No sul do Brasil existem problemas com atividade hematofágica de borrachudos e as informações referentes à distribuição das espécies ainda são insuficientes. O presente estudo visa reconhecer as espécies de Simuliidae e analisar fatores abióticos quanto a distribuição dos estágios imaturos (larva e pupa) em riachos da Bacia do Rio Ijuí, no Noroeste do Rio Grande do Sul, Brasil. Foram coletados simulídeos em 27 pontos de estudo, cobrindo três regiões da bacia (alta, intermediária e baixa altitude). Concomitantemente, foi feito o registro de fatores abióticos nos trechos estudados, tais como: temperatura, condutividade elétrica, oxigênio dissolvido, pressão atmosférica, pH, altitude e largura do riacho. Foram realizadas análises de Kruskal-Wallis ou ANOVA, Análise de Componentes Principais (PCA), Análise de Espécie Indicadora e de Correspondência Canônica (CCA). Foi registrado um total de 10 espécies de Simuliidae. Simulium pertinax Kollar, Simulium incrustatum Lutz e Simulium orbitale Lutz foram as mais frequentes. Não ocorreu diferença na riqueza e na densidade das espécies entre as diferentes regiões estudadas, exceto para Simulium jujuyense Paterson & Shannon. Os ambientes nos pontos de amostragem foram segregados de acordo com a sua localização na bacia, e apenas S. jujuyense foi considerada uma espécie indicadora da região alta. Tais resultados podem ser explicados pelas condições homogêneas da Bacia do Rio Ijuí, que vêm sofrendo pressão antropogênica principalmente devido às atividades agrícolas, resultando em perdas na biodiversidade aquática local. Também, indicam que o controle deve ser realizado considerando a bacia como um todo e não apenas em trechos isolados dos riachos.
