Objective: The aim of the present study was to evaluate anti-inflammatory activity of methanolic extract of Terminalia coriacea. Materials and Methods: A methanolic extract of T. coriacea leaves was studied in albino Wistar rats with carrageenan-induced paw edema, an acute model, and cotton pellet-induced granuloma, a chronic model, at 3 oral test doses (125, 250, and 500 mg/kg). Aspirin 100 mg/kg was used as a positive control. Paw volume and wet and dry weights of cotton pellets were determined. The data were analyzed by one-way ANOVA followed by Dunnett’s multiple comparison test. Results: The test extract at doses of 125 and 250 mg/kg decreased paw volume and wet and dry weights of cotton pellets. The highest test dose (500 mg/kg) displayed a response comparable to that of the standard drug (p < 0.01) on paw volume. The extract produced similar (p < 0.05) decrease in wet weight of the cotton pellet at 125 and 250 mg/kg, whereas the effect of 500 mg/kg of the extract was comparable to that of aspirin 100 mg/kg (p < 0.01). The extract of T. coriacea at 500 mg/kg induced the most significant (p < 0.01) effect on wet weight of granulomatous tissue. Conclusion: The methanolic extract of T. coriacea leaves successfully decreased paw edema as well as dry and wet weights of granulomatous tissue in both acute and chronic inflammatory models thus confirming the anti- edematogenic, antitransudative, and antiproliferative properties of T. coriacea.

Abstract. Belay R, Makonnen E. 2018. Anti-inflammatory activities of ethanol leaves extract and solvent fractions of Zehneria scabra (Cucurbitaceae) in rodents. Biofarmasi J Nat Prod Biochem 18: 42-56. Zehneria scarba (L.f.) Sond is one of the medicinal plants used in folkloric medicine of Ethiopia for years to treat various inflammatory disorders. The present study was aimed to validate the anti-inflammatory activity of crude 70% ethanol leaves extract (70EE) against a sub-acute model and further evaluate the solvent fractions (AF, BF, and CF) in an acute (carrageenan-induced paw edema), sub-acute (formaldehyde induced arthritis) and chronic (cotton pellet induced granuloma) inflammatory models. The 70EE was first prepared by maceration, and the fractions were obtained by sequential partitioning with chloroform and n-butanol from the aqueous suspension of crude 70EE. The test groups, then, received 100, 200 and 400 mg/kg of the crude 70EE or the fractions (AF, BF, and CF) at the same dose levels, whereas positive controls received aspirin (200mg/kg) or dexamethasone (0.5mg/kg) and negative controls received vehicle (2% tween 80 or distilled water, 10 mL/kg). All tested doses of the crude 70EE showed significant inhibition of formaldehyde induced arthritis at the 10th day of treatment, on which the 400mg/kg dose showed the maximum anti-arthritic effect (%A = 60.5; p < 0.001). In the carrageenan-induced paw edema, all the three fractions showed a statistically significant effect, in fact, with different onset and magnitude. In this model, the AF was found to be the most active fraction, and the 400mg/kg dose demonstrated the maximum effect (%A = 76.25; p < 0.001) at 5h post-induction, which is much better than the effect of aspirin at the dose employed. The overall order of efficacy in inhibiting the exudative component of carrageenan-induced paw edema was found to be AF> BF> CF. The AF was also found to be the most active fraction in inhibiting the exudative component of chronic inflammation in the cotton pellet induced granuloma model, where the maximum effect (%A = 43.10, p < 0.001) was exhibited by a dose of 400mg/kg. The AF was also the most active fraction in inhibiting formaldehyde induced arthritis, in which the BF and CF relatively showed a comparable effect throughout day 4-10. On the contrary, in the cotton pellet induced granuloma model, the CF was found to be the most active fraction in inhibiting the proliferative and granulomatous component of chronic inflammation, and the overall order of effectiveness was found to be CF> AF> BF. Besides, 400mg/kg of CF demonstrated the maximum inhibition of granuloma formation (%A = 55.52; P < 0.001). The phytochemical analysis revealed the differential distribution of secondary metabolites into the three fractions, which either singly or in concert appeared to be responsible for the observed effects. The data obtained from the present study collectively indicate that the extract and fractions of leaves of Z.scabra possessed a significant anti-inflammatory activity, upholding the folkloric use of the plant.

Introduction: Guazuma ulmifolia (Sterculiaceae) is commonly used in folk medicine as a single drug remedy to treat various ailments like; skin diseases, headache, wounds, ulcers, asthma, kidney disease and malaria. However, the use of the herb for treatment of pain and inflammation has not been scientifically investigated. Objective: This study of 80% methanolic crude extract of the leaves of Guazuma ulmifolia was evaluated for its analgesic and anti-inflammatory properties using established animal models. Material and methods: Wister albino rats were randomly divided into four groups of six rats per group. The control group was orally given 2 ml/kg (p.o.) of distilled water. The positive control received standard drug (Diclofenac 10 mg//kg, Indomethacin 20 mg/kg). The rest of the groups were treated with 80% methanolic extract of Guazuma ulmifolia at doses of 250 and 500 mg/kg. Animals were then subjected to tests using hot plate and tail-clip for analgesic activity, for anti-inflammatory activity carrageenan-induced paw edema test and histamine-induced paw edema test was used. Result and Discussion: Guazuma ulmifolia extract showed a dose-dependent significant reduction of pain in analgesia models (p<0.001) with 500 mg/kg dose producing the highest reduction. The extract significantly reduced carrageenan-induced inflammation in a dose-independent manner, in which the highest reduction of inflammation was observed at 500 mg/kg. Conclusion: The data collectively indicate that 80% methanolic extract of Guazuma ulmifolia leaves has potential analgesic and anti-inflammatory activities.

Background: Inflammation is a complex reaction to various injurious agents such as infections, trauma, foreign bodies, tissue necrosis, physical and chemical agents, that consists of vascular responses, migration and activation of leukocytes and systemic reactions. Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) are currently the most commonly prescribed drugs for treating pain and inflammation. The traditional NSAIDs usually cause various adverse effects on long term use. Gmelina arborea Linn (Gambhari) belongs to family Verbenaceae. The roots, leaves, flowers, fruits and bark are used for treating different ailments as anthelmintic, analgesic, anti-pyretic, antidiabetic, antimicrobial, diuretic and other common disorders.Methods: The study was carried out by using inflammatory models in wistar rats. The anti-inflammatory activity of G. arborea was compared with standard drug aspirin. The study parameters for acute inflammation was assessment of reduction in inflammation & the percentage inhibition of the paw edema. The parameter for the sub-acute inflammation was percentage inhibition of the dry granuloma weight.Results: The low and high dose of G. arborea root extract significantly showed the anti-inflammatory activity when compared to control group. The high dose of G. arborea extract showed comparable results in parameters like reducing inflammation, percentage inhibition of paw edema and dry granuloma weight in acute carrageenan paw edema and sub-acute inflammation cotton pellet granuloma models with standard aspirin treated group.Conclusions: Since G. arborea root extract was having all the qualities required for anti-inflammatory drug. However, no clear inference can be drawn at this stage and hence we consider the work for further extensive research.

The present study was intended to evaluate Anti-inflammatory activity ofC. rottleriextracts (Hydroalcoholic, Methanol, Ethyl acetate and Hexane). The Anti-inflammatory activity ofC.rottleriextracts at doses of 125mg/kg, 250mg/kg and 500mg/kg using carrageenan induced rat paw edema model compared with standard drug (Indomethacin). The selected plant extracts significantly inhibited paw edema along with the standard drug Indomethacin. Of all extracts, methanol extract produced significant effect on reduction of increased paw thickness, hydro alcoholic and ethyl acetate extracts produced moderate percentage inhibition and hexane extract produced low level of percentage inhibition in reducing paw edema on carrageenan induced rats. In all extracts, methanol extract at a dose of 500mg/kg showed more percentage inhibition i.e . 53.47±2.19. From the results obtained during the study it is concluded thatC. rottlerihaving the bioactive molecule responsible for Anti-inflammatory activity by individually or by combination of different bio-active compounds present in it. Further is necessary for isolation and characterization of bioactive molecules which are responsible for the selected plant biological activities.

Objective: Objective of the study was to synthesize and evaluate a series of novel compounds, ethyl 2-(2-cyano-3-(substituted phenyl)acrylamido)- 4,5-dimethylthiophene-3-carboxylates, for in vitro antioxidant and in vivo anti-inflammatory activities.Methods: Ethyl 2-(2-cyano-3-(substituted phenyl)acrylamido)-4,5-dimethylthiophene-3-carboxylates were synthesized by knoevenagel condensation of active methylene group of ethyl 2-(2-cyanoacetamido)-4,5-dimethylthiophene-3-carboxylate with substituted benzaldehydes. The synthesized compounds were evaluated for their in vitro antioxidant properties in three different models, viz., reduction of 1,1-diphenyl-2-pycrylhydrazyl free radical, scavenging of nitric oxide free radical, and ferric ion-induced lipid peroxidation using rat brain homogenate. Few selected compounds with good antioxidant properties were pharmacologically evaluated for anti-inflammatory activity by carrageenan-induced rat paw edema model.Results: Clean and efficient synthetic procedure was used for the preparation of series of compounds. The structures of synthesized compounds were confirmed by infrared, 1H nuclear magnetic resonance and mass spectra. The antioxidant activity data revealed that the compounds of ethyl 2-(2-cyano-3-(substituted phenyl)acrylamido)-4,5-dimethylthiophene-3-carboxylate containing phenolic substitution showed greater antioxidant activity. Hence, the active compounds were evaluated for anti-inflammatory activity and found to possess good activity. The percentage inhibition of rat paw edema obtained for the evaluated compounds was in the range of 70.2-83.1, comparable to the standard drug diclofenac (85.0%).Conclusion: The use of inexpensive, eco-friendly and readily available reagents, easy work-up and high purity of products makes the procedure a convenient and robust method for the synthesis of title compounds. The compounds of ethyl 2-(2-cyano-3-(substituted phenyl)acrylamido)-4,5- dimethylthiophene-3-carboxylate containing phenolic substitution showed greater antioxidant and anti-inflammatory activities.

Nonsteroidal anti-inflammatory drugs (NSAIDs) have been widely used for the treatment of inflammation. However, despite their effectiveness, most NSAIDs cause various side effects that negatively affect the management of inflammation and, in part, pain. Thus, there is a need to search for new anti-inflammatory agents with few, or no, side effects. Natural products of plant, animal, or microorganism origin have been good sources of new bioactive compounds. The present study was carried out to evaluate the acute and chronic anti-inflammatory activities of the essential oil of the rhizomes of Zingiber zerumbet (Zingiberaceae) using the carrageenan-induced paw edema and cotton pellet-induced granuloma tests, respectively. The effect of the essential oil on inflammatory- and noninflammatory-mediated pain was also assessed using the formalin test. Essential oil of Z. zerumbet, at doses of 30, 100, and 300 mg/kg, was administered intraperitoneally to rats. The substance exhibited significant anti-inflammatory activity both in acute and chronic animal models. The essential oil also inhibited inflammatory- and noninflammatory-mediated pain when assessed using the formalin test. In conclusion, the essential oil of Z. zerumbet possessed anti-inflammatory activity, in addition to its antinociceptive activity, which may explain its traditional uses to treat inflammatory-related ailments.

Objective: The object of the study is to design, synthesize and biological evaluation of isoniazid derived 1,2,4-triazoles compounds.Methods: Isoniazid based 1,2,4-triazoles derivatives have been synthesized by reaction of Isoniazid with carbon disulfide in basic medium (KOH) to form Potassium dithiocarbazinate salt and reaction with hydrazine hydrate converted into 4-amino-5-(pyridin-4-yl)-4H-1,2,4-triazole-3-thiol. These compounds were reacted with seven different benzaldehyde to form 4-[(substituted phenyl)-methylene]-amino-5-(pyridine-4-yl)-4H-1,2,4-triazol-3-thiol (4). The final compounds were synthesized by reaction with four different acetanilide to form 4-[substituted phenyl)-methylene]-amino-3-(N-substitutedcarboxamidomethylthio)-5-(pyridine-4-yl)-4H-1,2,4-triazoles derivatives. All these compounds were characterized by IR, 1H-NMR, [13]C-NMR and elemental analysis. The antimicrobial activity was determined by the cup plate method. Acute anti-inflammatory activity determined by using carrageenan-induced rat paw edema model.Results: Series PJ-A4, PJ-A7 and PJ-A13 showed more than 90% of the zone of inhibition against both Gram positive and Gram negative organisms. The antifungal study suggested that among synthesized compounds series PJ-A4, A7, A9, A11 and A13 showed more than 90% of zone of inhibition, A2, A10 and A12 shows more than 80% of the zone of inhibition. Anti-inflammatory study data indicate that compounds PJ-A4, PJ-A8, PJ-A9 and PJ-A13 produced 70 to 76% of paw edema inhibition compare to standard drug Ibuprofen which showed 83.3% after 5 h. Conclusion: Results suggested that the isoniazid based 1,2,4-triazole derivatives have significant antibacterial, antifungal and anti-inflammatory activity.

Background: In recent years there has been growing interest in therapeutic use of natural products, especially those derived from plants. P. granatum is very common dietary ingredient in many parts of India and has remarkable biological and medicinal properties.Methods: In the present study, the methanolic extract of fruit peels of Punica granatum Linn. (MEPG) were investigated for anti-inflammatory activity by simple, reliable, less toxic and less time consuming HRBC membrane stabilization method. The presentation of hypo tonicity induced HRBC membrane lysis was taken a measure of anti-inflammatory activity. Their activities were compared with standard drug diclofenac.Results: The results of the study demonstrated that P. granatum contains active constituents, which possess anti-inflammatory activity which is probably related to the inhibition of prostaglandin synthesis.Conclusions: It is concluded that methanolic extract of P. granatum fruit peel possesses significant anti-inflammatory activity and this is a possible rationale for its folkloric use as an anti-inflammatory agent.

Series of novel Schiff bases of Isatin the  equimolar amines and 5-Dicrboxymethyl (R=COO2Me) substituted isatins (1 mmol of each) were added to 96% w/w ethanol (20 mL) containing 8 drops of glacial acetic acid. The mixture was heated under reflux for 5 h and then cooled to room temperature. The resulting solid was collected by filtration, washed with cold ethanol and dried in open air. The derivatives thus prepared had sufficient analytical purity. anticonvulsant activity performed by method as Animals were weighed and numbered. Mice were divided into 7 groups of six animals each. Group 1 served as control which was treated with vehicle (2% v/v Tween 80), group 2 was treated with standard drug phenytoin (25 mg/kg, i.p.) and groups 3– 7 were treated with newly synthesized oxadiazole derivatives (25 mg/kg, i. p.). One hour after injection, the animals were subjected to electro shock through ear electrodes of 80 mA for 0.2 sec by electroconvulsiometer AND ANTI-inflammatory activity measured by Weigh the animals and number them. Mark the animals with picric acid for individual animal identification. Divide rats into 5 groups of 6 rats each. Note the initial paw volume of each rat by dipping just beyond tibio-tarsal junction by mercury displacement method. The pharmacological screening of the synthesized compounds showed anti convulsant activity ranging from 56.2 % to 76.3 % inhibition of epileptic seizures in mice, where as the standard drug Phenytoin showed 83.95 % inhibition of epileptic seizures in mice. The compound iiih4 from each group was found to be nearly potent to Phenytoin which is used as standard drug. Anti-inflammatory activity ranging from 31.09 to 63.11 % inhibition of rat paw edema volume after 3 hours, whereas the standard drug Indomethacin showed 62.06 % inhibition of rat paw edema volume after 4 hours.  The compound iiih3 was found to be nearly more potent then indomethacin which is used as standard drug Keywords: Isatin; Schiff bases; Anti convulsant activity; Anti-inflammatory activity; Isatin.

Abstract: Rubiadin is identified as a bioactive anthraquinone that exists in some quinone rich plants. The current research was carried out to evaluate the potential anti-inflammatory impact of Rubiadin in acute and chronic inflammation test models in rodents. The anti-inflammatory activity of Rubiadin was examined in cotton pellet-induced granuloma and carrageenan-induced edema as chronic and acute inflammation models in rats. TNF-α level and histopathological changes were assessed using sampled foot tissue of rat in the acute model. Also, the IL-1β level was assessed in the chronic model. One-way ANOVA (post hoc Tukey’s) analysis was used for comparing the groups. Rubiadin (0.5 mg/kg, i.p.) induced a significant reduction in TNF α level and the paw edema compared to the control group in carrageenan test. Also, it was observed that the anti-inflammatory activity of Rubiadin (0.5 mg/kg, i.p.) is comparable to mefenamic acid (30 mg/kg, i.p.) as the standard drug. Rubiadin was effective in granuloma induced by cotton pellet concerning the granuloma and transudate formation amount. Rubiadin’s anti-inflammatory effects were associated with a significant IL-1β decrease in this model. The results suggest that Rubiadin as a natural compound can possess significant peripheral anti-inflammatory impacts.
