The epidemiological characteristics of bovine leptospirosis in animals and herds in Mato Grosso do Sul were investigated to determine parameters such as disease frequency and the serovars reactant in beef cattle herds. A total of 4,629 beef cattle herds were examined against 33 Leptospira spp. serovars. The serum samples were submitted to the microscopic agglutination test (MAT) for the serological diagnosis of leptospirosis. The MAT results showed that 3,814 (82.39%) of the 4,629 animals evaluated were seropositive for the bacterium, with serological reactions mainly to serogroup Sejroe, serovar Wolffi (36.49%). The observed high frequency of reactive animals demonstrates the relevance of the infection. Therefore, general and specific measures should be implemented to contain and/or prevent infection of the animals in the studied region.

A Inseminação artificial em tempo fixo (IATF) está se difundindo no Brasil, pois contribui para o aumento da produtividade. O objetivo neste trabalho foi avaliar o uso do GnRH em protocolos de IATF de vacas criadas no pantanal Sul-mato-grossense. Participaram da pesquisa 531 vacas paridas da raça Nelore, que foram avaliadas de acordo com o escore de condição corporal (ECC), submetidas a três tipos de protocolos de IATF (lote 1, 2 e 3). Após 40 dias da inseminação artificial foi realizado diagnóstico de gestação. As matrizes que não receberam GnRH no lote 1 (estro aparente), lote 2 (estro parcial) e lote 3 (sem apresentação de estro) apresentaram, respectivamente 55,4%, 19,2% e 25,9% de prenhez, enquanto as que receberam o fármaco apresentaram 48,6%, 37,2% e 38,8%. Relacionado ao ECC, pode-se observar que a maior taxa de prenhez ocorreu em animais com ECC 3. Primíparas apresentaram melhor taxa de prenhez quando receberam GnRH independente do ECC. Concluiu-se que o uso de GnRH mostrou-se eficiente para melhoria da taxa de prenhez principalmente em primíparas, assim como nos animais que não apresentaram estro ou apresentaram estro parcial, e que matrizes com ECC 3 apresentaram melhores taxas de prenhez que vacas com escore inferior. Palavras-chave: bovino de corte; inseminação artificial em tempo fixo; produtividade; GnRH; Nelore.   Evaluation of cow reproductive performance in the region of Mato Grosso do Sul swamp submitted to TAI with GnRH application   ABSTRACT: The technique of timed artificial insemination (TAI) is spreading in Brazil, as it contributes to the increase of productivity. The objective of this work was to evaluate the use of GnRH in TAI protocols of cows raised in the swamp of Mato Grosso do Sul. 531 Nelore breed calved cows participate in the survey, which were evaluated according to the body condition score (BCS), and then performed three types of TAI protocols (batch 1, 2 and 3). After 40 days of the insemination, a pregnancy diagnosis was made. The matrices that did not receive GnRH in batch 1 (apparent oestrus), batch 2 (partial oestrus) and batch 3 (no oestrus presentation) presented respectively 55.4%, 19.2% and 25.9% of pregnancy, while those receiving the drug showed 48.6%, 37.2% and 38.8%. Related to BCS, it can be observed that the highest pregnancy rate occurred in animals with BCS 3. Primiparous had a better pregnancy rate when receiving GnRH regardless of BCS. It was concluded that the use of GnRH was efficient to improve the pregnancy rate mainly in primiparous as well as animals that showed no oestrus or partial oestrus, and matrices with BCS 3 had better pregnancy rates than cows with lower scores. ABSTRACT Evaluation of the comfort and animal welfare parameters enables determining the best environmental conditions for livestock creation. The present study was aimed to determine the thermal comfort index for beef cattle using interpolation techniques for the State of Mato Grosso do Sul during extreme summer and winter seasons for a period of 10 years. The analysis was performed on the climatic variables data provided by the National Institute of Meteorology, Brazil. The maximum (THImax) and minimum (THImin) temperature and humidity indices (THIs) of 28 weather stations distributed across the state were calculated, and the thematic maps of the THIs (maximum and minimum) calculated for the summer and winter seasons were prepared. The results revealed a state of emergency in the two largest animal-producing cities in the state; namely, Corumbá and Ribas do Rio Pardo, while two other cities, Chapadão do Sul and Costa Rica emerged as favorable regions for the production of beef cattle. The Spline method used in the present study, therefore, proved to be a suitable tool for analyzing a small number of weather stations distributed over a large territorial area.

Abstract Neospora caninum is an obligate intracellular protozoan with canids (Canis domesticus, Canis lupus dingo, Canis latrans, Canis lupus) as its definitive hosts. The objective of this study was to detect anti-N. caninum antibodies in pregnant women seen at referral center for prenatal screening in the state of state Mato Grosso do Sul, Brazil. A total of 188 serum samples from pregnant women provided by the Instituto de Pesquisa, Ensino e Diagnósticos da APAE de Campo Grande (IPED/ APAE) were subjected to IFA test and western blot analysis. The samples were divided into three groups: 23/99 samples from the seropositive group for toxoplasmosis were positive for anti-N. caninum IgG antibodies, and 9/99 positive for IgM; in the HIV group, 7/33 were positive for IgG; and in the HIV+toxoplasmosis group, 13/56 were positive for IgG and two positive for IgM. The seropositivity for IgG was assessed by western blot by testing 43 IFA test positive samples using rNcSRS2 (Nc-p43) as antigen. The serological results of the present study suggest that exposure of these pregnant women to the parasite N. caninum and presence of IgM antibodies are indicative of recent infection. Further studies are needed to establish the possibility of active infection.

ABSTRACT: This study aimed to evaluate exposure of wild small mammals to spotted fever group (SFG) rickettsiae in Mato Grosso do Sul State, central-western Brazil. Serum samples of 68 small mammals were analyzed by indirect immunofluorescence assay (IFA) against six Rickettsia species from Brazil. Overall, 37.5% (9/24) marsupials and 6.8% (3/44) small rodents were seroreactive to at least one of the Rickettsia species, with end point titres ranging from 64 to 512. These results suggested that wild small mammals were infected by SFG rickettsiae, and could participate in the ecology of rickettsiae in Mato Grosso do Sul, Brazil.

Leptospirosis is a zoonotic disease of global importance, especially in tropical countries. The current Leptospira spp. seroprevalence in cattle from central and northern Madagascar is unknown. Thus, the aim of this study was to determine the seroprevalence resulting from infections with pathogenic Leptospira spp. in zebu cattle from these areas. Serum samples from 194 animals were tested by microscopic agglutination test (MAT) using a panel of 12 serovars as antigens. Samples with a titer of ≥1:100 were considered positive. The overall seroprevalence was 59.3% (95% CI; 52.0–66.2%) with titers ranging from 1:100 to 1:1600. Among the seropositive animals, the most frequent antibody reactions were against serovar L. Tarassovi (serogroup L. Tarassovi) with 40.2% (33.3–47.5%), followed by L. Hardjo (L. Sejroe) with 13.9% (9.5–19.8%), L. Grippotyphosa (L. Grippotyphosa) with 9.8% (6.2–15.1%), L. Pomona (L. Pomona) with 7.7% (4.5–12.7%) and L. Autumnalis (L. Autumnalis) with 5.2% (2.6–9.5%). Less than 5% of the samples reacted positively against the remaining serovars. These results indicate a very high exposure of Malagasy cattle to Leptospira spp. which, consequently, poses a definite risk for people working with cattle acquiring this zoonotic infection.

Background: Leptospirosis is a re-emergent contagious infectious disease, caused by pathogenic leptospires that are transmitted by the urine of infected animals or bacteria-contaminated water and mud. In tropical and subtropical countries it presents high prevalence due to the temperature and humidity conditions that favor the maintenance of the agent in the environment. This disease can affect several species, in Brazilian cattle is an endemic disease, and studies have shown a high occurrence of Leptospira spp. infection in beef and dairy herds. Domestic cats as well as other species of the Family Felidae seem to be resistant to leptospirosis. However, it has been demonstrated under experimental conditions that cats may become infected by ingestion of infected rodents and contaminated water. The present study investigated the occurrence of Leptospira spp. infection in cattle, feral cats and rodents of the Fernando de Noronha Island.Materials, Methods & Results: Fernando de Noronha Island is located 360 km far from Recife and Natal, capitals of the states of Pernambuco and Rio Grande do Norte, Brazil, respectively. It has an area of approximately 18.4 km2 and constitutes the submerged part of a volcanic edifice currently inactive, which base rests 4,000 m deep in the Atlantic Ocean. Blood samples were collected from all the cattle raised in the Island (n = 88), 200 feral cats and 150 rodents, and the sera were screened by MAT (Serogroups: Australis; Autumnalis; Ballum; Bataviae; Canicola; Cynopteri; Djasiman; Grippotyphosa; Hebdomadis; Icterohaemorrhagiae; Icterohaemorrhagiae; Panama; Pomona; Pyrogenes; Sejroe e Tarassovi) for detection of anti-Leptospira spp. antibodies. Initially all sera were screened at 1:100 dilution and those with 50% or more agglutination were titrated at two-fold geometric dilutions. The serum titer was defined as the reciprocal of the highest positive dilution. The plane coordinates obtained by Global Position System (GPS) were used for developing a spatial map of the Fernando de Noronha Island. The geo-referenced data were plotted in the ArcGIS 10.1 software. Approximately 22% (20/88) and 12% (19/150) of the cattle and rodents were serologically reactive against Leptospira spp. antigens, respectively. The antibody titers of cattle ranged from 100 to 800 as shown. All the rodents screened were reactive against only one serovar and their antibody titers ranged from 100 to 3200. None of the serum samples from cats was reactive against the serovars tested. The serogroup Icterohaemorrhagiae predominated among the seropositive cattle, being found in 100% of the reactive samples. In rodents, the serogroups Icterohaemorrhagiae, Djasiman and Australis were responsible for 73.7% (14/19), 21.0% (4/19) and 5.2% (1/19) of the infections, respectively.Discussion: We believe that rodents and cattle play an important role in the dissemination of this disease, thus, it is necessary adopting prophylactic measures aimed at leptospirosis in the study area, in view of the human cases of leptospirosis reported and confirmed in the Island. These results are unprecedented in an insular environment in Brazil. Strategies aimed at better sanitary management of the cattle herds as well as population control of rodents must be implemented in the Fernando de Noronha Island to secure a more sustainable animal production and minimize the risks to public health.

ABSTRACT: The working equid population in Corumbá, Southern Pantanal, is very large and has a crucial role in the main economic activity of the State of Mato Grosso do Sul, the beef cattle industry. The aim of the present study was to estimate the prevalence of equine infectious anaemia (EIA) in working equids of ranches in the municipality of Corumbá, by the official agar gel immunodiffusion (AGID) test, and evaluate the adoption of the Programme for the Prevention and Control of Equine Infectious Anaemia proposed by Embrapa Pantanal and official entities in the 1990s. From September to November 2009, forty ranches distributed through the area of the municipality were visited, and serum samples were obtained from 721 equines and 232 mules. According to previous publications and the present data, it was concluded that the prevalence of EIA in this population has increased from 18.17% to 38.60%, which represents at this time approximately 13,000 infected animals. There was no significant difference between the apparent prevalence of equines and mules. It was also verified that the control programme was not known by the greater part of the interviewed ranch owners, managers and foremen and, in their perception, EIA is not a primary threat to address. Among the studied variables, the serologic testing practice significantly reduced the risk for the presence of EIA seropositivity, as well as the separation of riding equipment and segregation of seropositives.

RESUMO: Foi realizado um estudo epidemiológico da leptospirose em fêmeas acima de 24 meses, provenientes de 246 rebanhos, e 2.766 animais amostrados aleatoriamente nos nove municípios que compõem a região do Pantanal de Mato Grosso do Sul, bem como identificados os fatores de risco associados à doença. As amostras de sangue foram coletadas no período de setembro a novembro de 2009 e examinadas pelo teste de aglutinação microscópica ante uma coleção de 24 antígenos vivos de Leptospira spp., representantes dos sorovares Australis, Bratislava, Autumnalis, Butembo, Castellonis, Batavie, Canicola, Whitcombi, Cynopteri, Grippotyphosa, Hebdomadis, Copenhageni, Icterohaemorrhagiae, Javanica, Panamá, Pomona, Pyrogenes, Hardjo, Wolffi, Shermani, Tarassovi, Sentot, Andamana e Patoc. Adicionalmente, representantes de doze estirpes de leptospiras isoladas no Brasil foram adicionados à coleção de antígenos do teste de soroaglutinação microscópica (SAM). A prevalência aparente foi de 66% e a prevalência real de animais infectados, de 79,80%, com intervalo de confiança (IC) de 95% (78,3-81,3) e 241 rebanhos apresentando pelo menos um animal reagente. Os sorovares mais prováveis foram o Hardjo seguido pelo Wolffi. Os resultados demonstram que a leptospirose bovina continua presente no Pantanal, com alta prevalência tanto em rebanhos quanto em indivíduos, sendo os principais fatores de risco para a doença o tipo de exploração e a raça.

Background: Leptospirosis has a worldwide distribution with high social and economic impact. It is caused by a variety of Leptospira spp. serovars, particularly in tropical regions. Various species of animals can be affected by Leptospira spp. including cattle, horses, sheep, goats and swine, and can act as a reservoir for human infection. In Brazil, sheep leptospirosis has been diagnosed in a variety of States, however, little is known about sheep flocks of Mato Grosso. Considering the marked growth of sheep industry in Mato Grosso State in past recent years, this study aimed to evaluate the frequency of serologically positive sheep for Leptospira spp. serovars in the State of Mato Grosso (Brazil).Materials, Methods & Results: Blood samples of 346 sheep, which belonged to 16 sheep flocks of the medium northern region of Mato Grosso was obtained by jugular vein puncture. Serum was obtained by blood centrifugation and stored at -20°C until the test to be performed. The diagnosis was performed using the microscopic agglutination test. Serovars Australis, Autumnalis, Bataviae, Bolívia, Castelonis, Celledoni, Grippotyphosa, Hardjo, Hardjobovis, Hebdomadis, Icterohaemorrhagiae, Javanica, Lagoa, Norma, Panama, Pomona, Sejroe, Shermani, Szwajizak, and Wolffi were tested. Reactions were considered positive with minimum titration of 1:100. Considering all serovars, 54.9% of sheep were positive, and the serovar more frequently detected was Celledoni (22.8%), followed by Javanica (19.4%), Castellonis (16.5%), and Norma (15.3%), with predominance of titration of 100 for these serovars, while only one sheep had a titration of 1:800 detected for serovar Hardjo. All flocks (100%) were positive (at least one positive animal), with the frequency of animals positive to Leptospira spp. varying of 31.3% to 79.2% in flocks.Discussion: Infection for Leptospira spp. in humans has a large social and sanitary impact. Under an animal health perspective, beyond sanitary impact, leptospirosis have a large economic impact and can act as important reservoirs for Leptospira spp. for human infection, especially in tropical conditions, which favor the persistence of agent in the environment. The occurrence of sheep leptospirosis has been diagnosed in several States of Brazil as Rio Grande do Sul, Paraíba, Rio de Janeiro, São Paulo and Maranhão, with frequency of herds positive for Leptospira spp. varying of 5.4% to 47.4%, however, the frequency of positive sheep in this study was higher than these previous reports in other Brazilian States. The most frequent serovar in sheep flocks have a great importance for prophylactic control. However, there were identified as main frequent serovar Celledoni and Javanica, which are considered unusual in sheep, and indicates a peculiar profile of ovine serovars for the region. Considering de high frequency of positive farms (100%) in Mato Grosso and the lack of studies about the occurrence of leptospirosis in this region, our results suggest the occurrence of risk factors that can favor the dispersion and survival of agent. Contact of the sheep evaluated in this study with other animal species such as cattle, dogs, and horses that can serve as a source for sheep infection or may become contaminated from sheep. Then, the profile of sheep infection by Leptospira spp. in the State of Mato Grosso has a peculiar profile of serovars, with high frequency of infection of animals and herds.

Leptospirosis is an important socioeconomic disease in humans, as well as in domestic and wild animals, being caused by Leptospira spp. Bovine animals are considered reservoirs of this disease, because they intermittently disseminate the bacteria into the environment through their urine. In this way, the cattle an important source of Leptospira infection. The objective of this study was to detect Leptospira spp. antibodies and DNA in bovine females from two refrigerated slaughterhouses in the microregion of Umuarama, Paraná, Brazil. In particular, blood and urine samples from 52 crossbred bovine females older than 36 months from the two slaughterhouses were used. The microscopic agglutination test (MAT) was used to detect leptospiral antibodies, and the polymerase chain reaction (PCR) and subsequent sequencing were used to detect Leptospira DNA. The MAT yielded 22 (42.3%) serum samples considered reagent, while the nested PCR test resulted in one amplified sample (1.9%) of 289 bp. This single sample was then amplified again using primers for the SecY gene (549 bp). Sequencing of this gene characterized the bacteria as L. borgpetersenii that were similar to the serovar Hardjo of the genotype Hardjobovis. This is the first molecular confirmation of Hardjobovis-like L. borgpetersenii in the urine of crossbred bovine females older than 36 months from slaughterhouses in the microregion of Umuarama. This study’s results show that it is important to combine serological and molecular diagnosis in the detection of Leptospira spp. Therefore, both methods were used to improve our understanding of the epidemiology of this disease in bovine animals from the microregion of Umuarama. In addition, the analysis informed the subsequent adoption of preventive measures and educational One Health actions to prevent economic losses related to the herd, as well as social losses related to workers and the environment.
