Xylaria nigripes ( XN) is a medicinal fungus that was used traditionally as a diuretic, nerve tonic, and for treating insomnia and trauma. In this study, we elucidated possible mechanisms of neuroprotective effects of XN mycelia extracts. XN mycelia were produced by fermentation. Hot water extract and 70% ethanol extract of XN mycelia were evaluated on hydrogen peroxide (H2O2)-induced apoptosis in PC12, a rat pheochromocytoma cell line. Both XN extracts effectively protected PC12 cells against H2O2-induced cell damage by inhibiting release of lactate dehydrogenase, decreasing DNA damage, restoring mitochondrial membrane potential, and arresting abnormal apoptosis through upregulation of Bcl-2 and downregulation of Bax and caspase 3. Compared to water extract, ethanol extract showed not only greater neuroprotective effects but also a higher antioxidant activity by scavenging DPPH radicals, inhibiting lipid peroxidation, and reducing power. High phenolic content and antioxidant activity may provide the neuroprotective properties of XN ethanol extract.

Ganoderma lucidum (Leyss.:Fr.) Karst is one of the medicinal mushrooms, which possesses enviable antioxidant properties. Objective of this investigation was to evaluate antioxidant activity, reducing power, scavenging abilities on 1.1-diphenyl-2-picrylhydrazyl (DPPH) radicals and chelating effects on ferrous ions of hot water extracts obtained from carpophore and spores of this mushroom. Hot water extract from carpophore (Gl-I) showed high antioxidant activity of 85.7 ? 0.7%, at 10 mg/ml, while antioxidant activity of hot water extract from spores (Gl-Is) was 9.2 ? 0.3% at 10 mg/ml. Reducing power of Gl-I reached a plateau of 3.4 ? 0.1 at 20 mg/ml, and 0.3 ? 0.0 at 20 mg/ml for Gl-Is. At 10 mg/ml, scavenging ability on DPPH radicals of Gl-I increased to 96.8 ? 2.5%, whereas Gl-Is scavenged DPPH radicals by 69.6 ? 2.5% at 10 mg/ml. Gl-I chelated 81.6 ? 3.6 % of ferrous ions at 20 mg/ml, while the chelating effect of Gl-Is was 73.8 ? 1.7%. The antioxidative activities of hot water extracts from carpophore and spores of the mushroom G. lucidum were concentration dependent and increased with an increase in the concentration.

The objectives of this study were to determine phytochemical compositions, chemiluminescence antioxidant activities, and neuroprotective effects on PC12 cells for water, methanol, and 95% ethanol extracts of the air-dried fruit ofTerminalia chebulaRetzius. The water extract afforded the greatest yield, and total phenolic and tannin content. The methanol extract yielded the greatest total triterpenoid content. Based on four chemiluminescence antioxidant assays, the three extracts showed various degrees of antioxidant activity. The methanol extract showed good antioxidant activity based on the horseradish peroxidase-luminol-hydrogen peroxide (H2O2) assay. The water extract appeared to have good antioxidant activities in cupric sulfate-Phen-Vc-H2O2and luminol-H2O2assays. Pyrogallol-luminol assay showed the 95% ethanol extract to have good antioxidant activity. The methanol and water extracts presented neuroprotective activities on H2O2-induced PC12 cell death at 0.5–5.0 μg/mL. Further investigations are necessary to verify these activitiesin vivo.

(1) Background: Coreopsis lanceolata L. is a perennial plant of the family Asteraceae, and its flower is known to contain flavonoids with various bioactivities. We evaluated the effect of Coreopsis lanceolata L. flower (CLF) extracts on H2O2-induced oxidative stress (OS) in neuronal cells and mouse neurons. (2) Methods: The flowering part of CL was used as CLF1 (70% ethanol extract) and CLF2 (water extract), and 10 types of phenolic compounds were quantified using high-performance liquid chromatography. To evaluate the neuroprotective effects of CLF, the antioxidant activities of the extracts were measured, and the expression levels of antioxidant enzymes and proteins related to OS-induced apoptosis in neuronal cells and mouse neurons treated with the extracts were investigated. (3) Results: In the in vitro study, CLF ameliorated H2O2-induced oxidative stress and induced the expression of antioxidant enzymes in PC12 cells. Furthermore, CLF1 enhanced the expression of the Bcl-xL protein but reduced the expression of Bax and the cleavage of caspase-3. In the same manner, CLF1 showed neuroprotective effects against OS in vivo. Pretreatment with CLF1 (200 mg/kg) increased the Bcl-2 protein and decreased Bax compared with the 1-methyl-4-phenylpyridinium ion (MPP+)-treated C57BL/6 mice model group. Our results suggest that the protective effects of CLF1 on MPP+-induced apoptosis may be due to its anti-apoptotic activity, through regulating the expression of the Bcl-2 family. (4) Conclusions: CLF1 exerts neuroprotective effects against OS-induced apoptosis in PC12 cells in a Parkinson’s disease model mouse. This effect may be attributable to the upregulation of Bcl-2 protein expression, downregulation of Bax expression, and inhibition of caspase-3 activation. These data indicate that CLF may provide therapeutic value for the treatment of progressive neurodegenerative diseases.

The standard extracts of Hypericum perforatum L. (SEHP), a well-known medicinal plant, are used for the treatment of depression, exhibited upgrading and significant protective effects on the trauma of PC12 cells induced by 200 μM H 2 O 2 in a dose-dependent manner within 24-hour treatment. Cell viability was assessed by the MTT method, and in situ cellular hydrogen peroxide ( H 2 O 2)-induced oxidative stress was examined by measurement of reactive oxygen species (ROS) formation using CDCFH procedures. Intra- and extra-cellular ROS levels decreased significantly to 71.9% and 50.0% of the control at a moderate concentration of 20 μg/ml, respectively, suggesting that SEHP could easily enter the cells and play important roles in reducing ROS levels. Our results were proved by detection of DNA fragmentation and inspection of cell morphology of PC12 cells. SEHP can obviously block DNA fragmentation and prevent the cells from shrinking and turning round of H 2 O 2-induced apoptosis in PC12 cells at concentrations of 10~100 μg/ml. This data suggests SEHP may be a candidate for application in neurodegenerative diseases such as Alzheimer's disease or Parkinson's disease.

Aims: The leafy stem and fruit of P.murex have been reported to be used in folk medicine to treat male reproductive system ailments. This study was undertaken to assess the antioxidant potential of the aqueous extracts of P.murex leafy stem and fruit. Methodology: Extracts were prepared by macerating the powder in water. Total phenolics amount were determined by Folin-Ciocalteu reagent, flavonoids were quantified by aluminum chloride method and total tannin content was estimated by hexacyanoferric method. The in vitro antioxidant activity of the extracts were assessed through 2,2´-diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, hydrogen peroxide (H2O2) inhibition assay and lipid peroxidation assay. Results: Total phenolic compounds, flavonoids and tannins content were respectively equal to 48.91± 1.67 µg/mg Gallic Acid Equivalents (GAE); 56.01± 0.90 µg/mg Rutin Equivalents (RUE); 16.92± 1.22 µg/mg Tannic Acid Equivalents (TAE) for the leafy stem extract while they were equal to 26.26± 0.54 µg/mg GAE; 47.88± 2.39 µg/mg RUE; 7.94± 0.31 µg/mg TAE for the fruit. The leafy stem extract exhibited a more pronounced DPPH scavenging activity, reducing power, hydrogen peroxide scavenging activity and lipid peroxidation activity than the fruit extract. Conclusion: The antioxidant activity of the leafy stem aqueous extract was significantly more potent than that of the fruit extract. Further studies will find out the extracts pharmacological efficacy and innocuity.

Terminalia catappa L. is one of the herbal plants that contain flavonoids that play a role in wound healing including to bleeding after tooth extraction. The aim of our study was to identify the chemical compounds contained in Terminalia catappa L. leaves. This type of research was experimental. The sample in this study was the leaves of Terminalia catappa number 3-6 from the base, at a tree height of 6 meters picked as much as 6 kilograms. Data analysis qualitatively and quantitatively. The highest content in ethanol extract of Terminalia catappa L of saponins and alkaloids were fractioned with distilled water and the lowest content were fractioned with hexane. Meanwhile distilled water is also used to fractioned the highest content of tannins and phenol. While ethyl acetate was used to fractioned the lowest content of tannins and the highest content of flavonoid. The lowest flavonoid in distilled water extract was fractioned using hexane. For the lowest content of phenol was fractioned using hexane. The results show the highest content of saponins is 3,787.80 mg/100g, the lowest is 166.67 mg/100g. The highest content of Alkaloids is 1,798.57 mg/100g, and the lowest is 576.80 mg/100g. The highest content of tannins is 53,140.72 mg/100g, the lowest is 8,391,803 mg/100g. The highest content of flavonoids is 2,5964.14 mg/100g, the lowest flavonoid is 462.84 mg/100g. The highest content of phenol 29,968.05 mg/100g, the lowest is 225.46 mg/100g. The highest antioxidant activity with AAI (Antioxidant Activity Index) obtains value of 0.36. This shows the moderate antioxidant ability. Terminalia catappa L. contain saponins, alkaloids, tannins, flavonoids, and triterpenoids. The active compound of Terminalia catappa L will generally be produced optimally if a polar solvent is used.

Antioxidant compounds play a significant role in preventing and scavenging free-radicals by reducing oxidative stress and providing protection to humans against degenerative diseases and infections. Obviously, antioxidant molecules of plant origin are pivotal to combat the oxidative harm in cells. Present work intended to evaluate the antioxidant capacity of the fruits of Spondias pinnata. Methanol extracts of the fruits at two stages of maturity were prepared and investigated by various antioxidants analyses such as total antioxidant activity, reducing power and radical scavenging assays (DPPH, nitric oxide and hydrogen peroxide). Its total phenol, flavonoid and tannin contents were also determined. Spondias pinnata fruit extracts exhibited effective antioxidant activity and its IC50 values of the unripe fruits were 65, 66, 72.23, 83.25, 66.75 ?g/ml and ripe fruits were 124.24, 92.50, 97.66, 144.10, 72.25 ?g/ml, for total antioxidant activity, reducing power, DPPH radical scavenging, nitric oxide radical scavenging and hydrogen peroxide scavenging assays respectively. The extracts, especially unripe fruit extract had good amounts of total phenolic and flavonoid contents which might contribute the antioxidant activities considerably. It is evident from the study that the fruits of S. pinnata possessed potent antioxidant activity and it can be considered as a good dietary choice among the underutilized fruits as well as common fruits. Therefore, wild mango may be a good choice of antioxidants of plant origin for dietary and pharmaceutical uses.
