The objectives of this study were to determine phytochemical compositions, chemiluminescence antioxidant activities, and neuroprotective effects on PC12 cells for water, methanol, and 95% ethanol extracts of the air-dried fruit ofTerminalia chebulaRetzius. The water extract afforded the greatest yield, and total phenolic and tannin content. The methanol extract yielded the greatest total triterpenoid content. Based on four chemiluminescence antioxidant assays, the three extracts showed various degrees of antioxidant activity. The methanol extract showed good antioxidant activity based on the horseradish peroxidase-luminol-hydrogen peroxide (H2O2) assay. The water extract appeared to have good antioxidant activities in cupric sulfate-Phen-Vc-H2O2and luminol-H2O2assays. Pyrogallol-luminol assay showed the 95% ethanol extract to have good antioxidant activity. The methanol and water extracts presented neuroprotective activities on H2O2-induced PC12 cell death at 0.5–5.0 μg/mL. Further investigations are necessary to verify these activitiesin vivo.

In this study, we investigated the phenolic compounds in hop strobile extracts and evaluated their antioxidant property using DPPH and ABTS assay. The total phenolic compound (TPC) and total flavonoid compound (TFC) estimated in two different solvent extracts considerably varied depending on the extraction solvent. The most abundant phenolic compound in hop strobile was humulones (α-acid) with levels ranging from 50.44 to 193.25 µg/g. El Dorado accession revealed higher antioxidant activity in ethanol extracts (DPPH: IC50 124.3 µg/mL; ABTS: IC50 95.4 µg/mL) when compared with that of the other accessions. Correlations between DPPH (IC50) scavenging TFC in ethanol extract (TFC_E, −0.941), and TPC_E (−0.901), and between ABTS (IC50) scavenging TFC_E (−0.853), and TPC_E (−0.826), were statistically significant at p < 0.01 level, whereas no significant correlation was observed between antioxidant activities, TPC and TFC in water extract. This study is the first to report that variations in the level of phenolic contents and antioxidant activity of various hop cultivars depended on the type of extraction solvent used and the cultivation regions. These results could provide valuable information on developing hop products.

AbstractSample particle size is an important parameter in the solid–liquid extraction system of natural products for obtaining their bioactive compounds. This study evaluates the effect of sample particle size on the phytochemical composition and antioxidant activity of brown macroalgae Sargassum cristaefolium. The crude ethanol extract was extracted from dried powders of S.cristeafolium with various particle sizes (> 4000 µm, > 250 µm, > 125 µm, > 45 µm, and < 45 µm). The ethanolic extracts of S.cristaefolium were analysed for Total Phenolic Content (TPC), Total Flavonoid Content (TFC), phenolic compound concentration and antioxidant activities. The extract yield and phytochemical composition were more abundant in smaller particle sizes. Furthermore, the TPC (14.19 ± 2.08 mg GAE/g extract to 43.27 ± 2.56 mg GAE/g extract) and TFC (9.6 ± 1.8 mg QE/g extract to 70.27 ± 3.59 mg QE/g extract) values also significantly increased as particle sizes decreased. In addition, phenolic compounds epicatechin (EC), epicatechin gallate (ECG), epigallocatechin (EGC), and Epigallocatechin gallate (EGCG) concentration were frequently increased in samples of smaller particle sizes based on two-way ANOVA and Tukey’s multiple comparison analysis. These results correlate with the significantly stronger antioxidant activity in samples with smaller particle sizes. The smallest particle size (< 45 µm) demonstrated the strongest antioxidant activity based on DPPH, ABTS, hydroxyl assay and FRAP. In addition, ramp function graph evaluates the desired particle size for maximum phytochemical composition and antioxidant activity is 44 µm. In conclusion, current results show the importance of particle size reduction of macroalgae samples to increase the effectivity of its biological activity.

Three Selaginella species, S. ornata, S. plana, and S. willdenowii, from Java Island, Indonesia, have been known to have antioxidant properties; however, in vivo antioxidant activities of these species have not been reported. This research aimed to evaluate the in vivo antioxidant activity of ethanolic extract of three Selaginella species. The 70% ethanol extract of three Selaginella species at four different doses was administered to mice one day before being treated with oxidative stress. The liver tissue of mice treated with or without oxidative stress was analyzed their lipid peroxidation by measuring MDA concentration and Superoxide Dismutase (SOD) activities. The results showed that there were variations in antioxidant activity among the three Selaginella species. In general, the dose of 0.3 g extract kg-1 BW has been able to reduce lipid peroxidation and increase SOD activity. The administration of S. ornata extract to the mice at 1.2 g extract kg-1 BW reduced the MDA concentration to the lowest level, but the same dose of two other Selaginella extracts caused toxic effects in mice. The antioxidant activities of S. ornata and S. plana were better than that of S. willdenowii extract, and among those species, S. ornata has the best antioxidant activity.

The plants of the genus Phlomis are native to Turkey, North Africa, Europe and Asia.  Phlomis bovei De Noé (Lamiaceae) is a rare Algerian endemic plant, commonly known as Kayat El Adjarah. The objective of this study is to quantify the polyphenol content and to evaluate the antioxidant activities of the ethanolic extract (EthE) of Phlomis bovei De Noé. The plant extract was prepared by macerating 100 g of ground material in 1000 ml of 85 % ethanol for 72 h, then the filtrate was evaporated using a rotary evaporator at a temperature of 45C and the filtrate was dried. The total content in polyphenols was determined using Folin Ciocalteu method. Flavonoid content in extracts was determined using Aluminum trichloride assay and the total content of tannins was determined according to Bate Smith method. The antioxidant activity was investigated in vitro by the DPPH (2, 2-diphenyl-1-picryl-hydrazyl), ABTS and iron chelating assays. The results indicate that the ethanol extract was rich in polyphenols and has an important capacity of scavenging the DPPH and ABTS free radical with an IC50 of 0.05 and 0.018 mg/ml, respectively. However, in the ferrous ions chelating ability test, PBEE extract showed moderate chelating activity (IC50 = 1.59 ± 0.05 mg/ml). To conclude, the obtained results may contribute to add possible scientific data to use Phlomis bovei industrially as herbal medicine and as supplementary sources for natural antioxidant drugs in food. Objective:: Euphorbia is a plant from the Euphorbiaceae family and it has traditionally been used to treat gonorrheal, respiratory tract infection, skin infections and malaria. The aim of present study was to evaluate of antibacterial, antifungal and antioxidant activities of latex and extracts of Euphorbia monostyla. Materials and Methods:: antimicrobial activity was studied by Disc-well diffusion and MIC-MBC methods, also its antioxidant activity was evaluated by DPPH and FRAP methods. Total phenolic content was evaluated by Folin-Ciocalteu reagent. Results:: It is displayed as a result of work that solvent polarity will play a key role to increase phenolic solubility. Methanol extract gave the highest percentage yield and ethyl acetate resulted to the lowest percentage yield. The antimicrobial test showed that whole extracts had no beneficial uses on microorganisms in compare of antibiotics types. Analysis of antioxidant assays revealed that aqueous and methanol extracts had highest antioxidant properties. Conclusion:: whole extracts had any beneficial uses on microorganisms in compare of antibiotics types but aqueous extract of E. monostyla had moderate antioxidant activity and may contain valuable active compounds. This work is the first study about E. monostyla and further study is suggested to isolate its active compounds.

SummaryIntroductıon: Instead of synthetic antioxidants, using of natural products with antioxidant activity is demanded.Objectıve: The aim of this study is to evaluate the antioxidant activity and total phenolic and flavonoid contents of theTrachystemon orientalis(borage) obtained from Ordu.Methods: The water and ethanol extracts of the borage plant were investigated by using severalin vitroantioxidant activity tests such as total antioxidant capacity, O2•−,•OH, DPPH and ABTS radicals scavenging activities, hydrogen peroxide scavenging activity, FRAP, Fe2+chelating and reducing power assays in order to make effective comparisons.Results: Water extract of theT. orientalishad the more phenolic (90 mg GA/g extract) and flavonoid (56.88 mg CT/g extract) contents than ethanol extract. Furthermore, both of the extracts showed strong antioxidant and radical scavenging activities determined by differentin vitromodels. The highest total antioxidant capacity as ascorbic acid equivalents of 244.50 mg/g extract was also calculated for water extract. Generally, a relationship between total phenolic content and antioxidant activity was established. It is also an important finding for an edible food source that the phenolic and flavonoid content and antioxidative activities for water extract was higher than the ethanol extract.Conclusıon:T. orientaliscould be used as a food ingredient instead of synthetic antioxidants and all results will contribute to the recent increase in investigations on using natural products in many areas such as food, pharmacy, alternative medicine and natural therapy.

To evaluate possibility as a skin whitening agent of Sorghum bicolor (S. bicolor), its antioxidant activity and anti-melanogenic effect on 3-isobutyl-1-methylxanthine (IBMX)-induced melanogenesis in B16/F10 melanoma cells were investigated. The result of total phenolic contents (TPC) indicated that 60% ethanol extract of S. bicolor (ESB) has the highest contents than other ethanol extracts. Antioxidant activity was evaluated using the 2,2’-azino-bis-(3-ethylbenzothiazolin-6-sulfonic acid) diammonium salt (ABTS)/1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activities and malondialdehyde (MDA) inhibitory effect. These results showed ESB has significant antioxidant activities. Inhibitory effect against tyrosinase was also assessed using L-tyrosine (IC50 value = 89.25 μg/mL) and 3,4-dihydroxy-L-phenylalanine (L-DOPA) as substrates. In addition, ESB treatment effectively inhibited melanin production in IBMX-induced B16/F10 melanoma cells. To confirm the mechanism on anti-melanogenic effect of ESB, we examined melanogenesis-related proteins. ESB downregulated melanogenesis by decreasing expression of microphthalmia-associated transcription factor (MITF), tyrosinase and tyrosinase-related protein (TRP)-1. Finally, 9-hydroxyoctadecadienoic acid (9-HODE), 1,3-O-dicaffeoylglycerol and tricin as the main compounds of ESB were analyzed using the ultra-performance liquid chromatography-ion mobility separation-quadrupole time of flight/tandem mass spectrometry (UPLC-IMS-QTOF/MS2). These findings suggest that ESB may have physiological potential to be used skin whitening material.

This study was designed to examine the in vitro antioxidant DPPH free radical-scavenging activity, and the total phenolic and flavonoid content of ethanol and methanol extracts of Salvia amplexicaulis Lam. in the whole plant and different parts, leaves, stems and flowers. The largest amounts of extract yield were obtained from the flowers, 14.14% and 12.00 % (w/w) in the ethanol of methanol extracts, respectively. The ethanol extract of leaves (16.07 ?g/ml) and methanol extract of the whole plant (21.28 ?g/ml) showed the highest activity against the DPPH radical. The ethanol extract of the leaves was the richest in phenols (222.40 mg GAE/g) and flavonoids (49.81 mg QE/g), whereas the methanol extract of the whole plant contained the highest amount of phenolics (180.89 mg GAE/g) and flavonoids (38.15 mg QE/g). A very strong linear correlation between antioxidant activity and the phenolic content of the extracts was established. The obtained results suggest that S. amplexicaulis could be regarded as an important source of natural antioxidants.

Xylaria nigripes ( XN) is a medicinal fungus that was used traditionally as a diuretic, nerve tonic, and for treating insomnia and trauma. In this study, we elucidated possible mechanisms of neuroprotective effects of XN mycelia extracts. XN mycelia were produced by fermentation. Hot water extract and 70% ethanol extract of XN mycelia were evaluated on hydrogen peroxide (H2O2)-induced apoptosis in PC12, a rat pheochromocytoma cell line. Both XN extracts effectively protected PC12 cells against H2O2-induced cell damage by inhibiting release of lactate dehydrogenase, decreasing DNA damage, restoring mitochondrial membrane potential, and arresting abnormal apoptosis through upregulation of Bcl-2 and downregulation of Bax and caspase 3. Compared to water extract, ethanol extract showed not only greater neuroprotective effects but also a higher antioxidant activity by scavenging DPPH radicals, inhibiting lipid peroxidation, and reducing power. High phenolic content and antioxidant activity may provide the neuroprotective properties of XN ethanol extract.

Objective: The present investigation focuses on the use of Cardiospermum halicacabum L. in their phytochemical and biological activities. Methods: In this study, in vivo stem and in vitro callus ethanolic extracts of C. halicacabum were tested for their phytochemical attributes by qualitative method, Fourier transform infrared (FTIR), antioxidant, antibacterial, and bioactive compound properties. The bactericidal activity of the in vivo stem and in vitro callus extract has been evaluated in both Gram+ve and Gram-ve microorganisms using the disk diffusion method. Results: The highest frequency (78%) of well developed, dark green organogenic callus was induced from stem explant on Murashige and Skoog (MS) medium supplemented with 0.7 mg/l 2,4-Dichlorophenoxyacetic acid (2, 4-D) and 0.5 mg/l benzyl adenine (BA). The results of FTIR spectra confirmed the presence of functional groups in wild stem and in vitro callus extract of C. halicacabum with various peaks. The total phenolic content in ethanolic extract of in vivo plant and in vitro callus was 80.46 mg gallic acid equivalent (GAE)/g dry weight and 76.4 mg GAE/g dry weight, respectively. The highest percentage of tannins was measured at 78.03 in wild stem ethanol extracts followed by 75.22 in callus extract. The antioxidant activity of 2,2-diphenyl-2- picrylhydrazyl (DPPH) ethanol extract was found to be 206.54 μg/ml. IC50 values of the stem extracts of C. halicacabum are 306 μg/ml and 286 μg/ml in callus extract, respectively. Antibacterial activity of the ethanol extract was higher for Staphylococcus aureus (S. aureus) with a 17 mm zone of inhibition. Conclusion: The present investigation recommended that the callus ethanolic extract function as a good source of biologically active compounds and natural antioxidants.
