Objectives: Autologous Platelet Rich Plasma (PRP) therapy, is considered to be a promising solution in accelerating the healing process of injured skeletal muscle tissue. In addition to the release of growth factors, PRP also promotes concentrated anti-inflammatory signals, including interleukins. However, the impact of the intramuscular administration of the PRP on hematologic and biochemical responses has not been fully elucidated in exercise induced muscle damage. Methods: Twelve healthy moderately active male volunteers, without previous experience with eccentric/concentric elbow flexors exercise, participated in this study. They were divided into two groups: control group (CONTROL, n=6) and platelet rich plasma administration group (PRP, n=6) group. To induce muscle damage, subjects in both groups performed concentric/eccentric contractions with load of (80 % 1RM) maximal voluntary contraction of the elbow flexors until point of exhaustion of the non-dominant arm. The non-dominant arms of the PRP group were treated with autologous PRP (Regen ACR-C, Regen Lab, Switzerland) post-24h exercise induced damage (DOMS). Subsequently, 4 ml PRP samples was injected using a 20-gauge needle into the region of the biceps brachii of the non-dominant arm under sterile aseptic conditions. Venous blood samples were collected pre-, and 4 days post-exercise, and analyzed for complete blood counts, serum ferritin, iron, iron binding capacity (IBC), creatinine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT) as markers of muscle damage and inflammation. Results: We found that the baseline levels of iron, ferritin, IBC, CK, LDH, AST and ALT were similar in control and PRP groups. However, 24 h following exercise induced muscle damage a significant increase in these parameters was observed in both groups. Interestingly, PRP administration decreased plasma iron levels compared to the control group but this was only achieved on the second day of post-exercise induced muscle damage. In addition, the plasma IBC levels increased in PRP group from day 2 to 4 post exercise compared to control group. PRP administration had no effect on plasma ferritin, CK, LDH, AST, and LDH levels. Conclusion: Acute exhaustive exercise increased muscle damage markers, including plasma iron, IBC and ferritin levels, indicating metabolic stress due to exercise induced muscle damage. PRP administration decreased the iron levels post-exercise and may have a role to play in the recovery of exercise induced muscle damage.

Background: Compression garment utilization is very popular among runners despite a lack of consensus in the literature regarding a beneficial impact. The aim of the study was to investigate the impact of training and competing with compression garments on exercise-induced muscle damage and performance in ultramarathon runners. Hypothesis: Compression garments will reduce the severity of exercise-induced muscle damage and improve performance in long-distance runners compared with control conditions. Study Design: Prospective, randomized controlled trial. Level of Evidence: Level 1. Methods: The study was conducted in healthy, uninjured endurance runners (n = 41) participating in a 56-km ultramarathon. The experimental group (n = 20; 14 males, 6 females) trained for 6 weeks and participated in the race wearing below-knee compression garments while the control group (n = 21; 15 males, 6 females) did not. Participants were tested on 4 occasions for various markers of exercise-induced muscle damage and running performance. Results: Ankle circumference measurements increased significantly less ( P = 0.01, Cohen d = 0.9) in the experimental group from immediately after until 2 days post-race compared with the control group. No further statistically significant changes were detected over time in midcalf circumferences, muscle architecture, or race performance. Selected pain ratings were statistically significant and worse in the experimental group. Conclusion: There are limited indications of a beneficial impact of compression garments with improvements in ankle circumference measurements. No ergogenic impact was detected. Clinical Relevance: There is limited evidence to support the continued utilization of commercially available below-knee compression garments during running for the purpose of muscle recovery or as a performance aid.

This study investigated whether maximal voluntary isometric contractions (MVC-ISO) would attenuate the magnitude of eccentric exercise-induced muscle damage. Young untrained men were placed into one of the two experimental groups or one control group (n = 13 per group). Subjects in the experimental groups performed either two or 10 MVC-ISO of the elbow flexors at a long muscle length (20° flexion) 2 days prior to 30 maximal isokinetic eccentric contractions of the elbow flexors. Subjects in the control group performed the eccentric contractions without MVC-ISO. No significant changes in maximal voluntary concentric contraction peak torque, peak torque angle, range of motion, upper arm circumference, plasma creatine kinase (CK) activity and myoglobin concentration, muscle soreness, and ultrasound echo intensity were evident after MVC-ISO. Changes in the variables following eccentric contractions were smaller (P < 0.05) for the 2 MVC-ISO group (e.g., peak torque loss at 5 days after exercise, 23% ± 3%; peak CK activity, 1964 ± 452 IU·L–1; peak muscle soreness, 46 ± 4 mm) or the 10 MVC-ISO group (13% ± 3%, 877 ± 198 IU·L–1, 30 ± 4 mm) compared with the control (34% ± 4%, 6192 ± 1747 IU·L–1, 66 ± 5 mm). The 10 MVC-ISO group showed smaller (P < 0.05) changes in all variables following eccentric contractions compared with the 2 MVC-ISO group. Therefore, two MVC-ISO conferred potent protective effects against muscle damage, whereas greater protective effect was induced by 10 MVC-ISO, which can be used as a strategy to minimize muscle damage.

Tart cherry juice (TC) and pomegranate juice (POM) have been demonstrated to reduce symptoms of exercise-induced muscle damage (EIMD), but their effectiveness has not been compared. This randomized, double-blind, parallel study compared the effects of TC and POM on markers of EIMD. Thirty-six non-resistance trained men (age 24.0 (Interquartile Range (IQR) 22.0, 33.0) years, body mass index (BMI) 25.6 ± 4.0 kg·m−2) were randomly allocated to consume 2 × 250 mL of: TC, POM, or an energy-matched fruit-flavored placebo drink twice daily for nine days. On day 5, participants undertook eccentric exercise of the elbow flexors of their non-dominant arm. Pre-exercise, immediately post-exercise, and at 24 h, 48 h, 72 h and 96 h post-exercise, maximal isometric voluntary contraction (MIVC), delayed onset muscle soreness (DOMS), creatine kinase (CK), and range of motion (ROM) were measured. The exercise protocol induced significant decreases in MIVC (p < 0.001; max decrease of 26.8%, 24 h post-exercise) and ROM (p = 0.001; max decrease of 6.8%, 72 h post-exercise) and significant increases in CK (p = 0.03; max increase 1385 U·L−1, 96 h post-exercise) and DOMS (p < 0.001; max increase of 26.9 mm, 48 h post-exercise). However, there were no statistically significant differences between treatment groups (main effect of group p > 0.05 or group x time interaction p > 0.05). These data suggest that in non-resistance trained men, neither TC nor POM enhance recovery from high-force eccentric exercise of the elbow flexors.

Previous necropsy studies have shown that up to 60% of sled dogs competing in endurance sled dog races have areas of muscle damage. Human athletes and trainers have maintained a strong belief, based mostly on personal experiences, that massage can be effective for pre-race preparation and expedited recovery between consecutive events. We proposed that massage could lessen the degree of creatine phosphokinase (CPK) release when utilised prior to endurance activities. A randomised crossover study was performed using minimally conditioned sled dogs from a sled dog racing kennel. Each dog in the treatment group received a 14 min massage immediately prior to controlled endurance exercise. The pre-exercise CPK activity was 64±11 IU/l (mean ± standard deviation). The difference between pre-exercise CPK and post-exercise CPK for both groups was expressed as delta CPK. The delta CPK for the massage group was 62±22 IU/l and 74±55 IU/l for the control group. There was no significant change in concentration of CPK between pre- and post-exercise in the treatment and control group (P=0.2282). The lack of a significant difference between the control and massage group suggests that pre-exercise massage may not reduce the severity of exercise-induced muscle damage.

Hormone therapy (HT) is a potential treatment to relieve symptoms of menopause and prevent the onset of disease such as osteoporosis in postmenopausal women. We evaluated changes in markers of exercise-induced skeletal muscle damage and inflammation [serum creatine kinase (CK), serum lactate dehydrogenase (LDH), and skeletal muscle mRNA expression of IL-6, IL-8, IL-15, and TNF-α] in postmenopausal women after a high-intensity resistance exercise bout. Fourteen postmenopausal women were divided into two groups: women not using HT (control; n = 6, 59 ± 4 yr, 63 ± 17 kg) and women using traditional HT (HT; n = 8, 59 ± 4 yr, 89 ± 24 kg). Both groups performed 10 sets of 10 maximal eccentric repetitions of single-leg extension on the Cybex dynamometer at 60°/s with 20-s rest periods between sets. Muscle biopsies of the vastus lateralis were obtained from the exercised leg at baseline and 4 h after the exercise bout. Gene expression was determined by RT-PCR for IL-6, IL-8, IL-15, and TNF-α. Blood draws were performed at baseline and 3 days after exercise to measure CK and LDH. Independent t-tests were performed to test group differences (control vs. HT). A probability level of P ≤ 0.05 was used to determine statistical significance. We observed significantly greater changes in mRNA expression of IL-6, IL-8, IL-15, and TNF-α ( P ≤ 0.01) in the control group compared with the HT group after the exercise bout. CK and LDH levels were significantly greater after exercise ( P ≤ 0.01) in the control group. Postmenopausal women not using HT experienced greater muscle damage after maximal eccentric exercise, indicating a possible protective effect of HT against exercise-induced skeletal muscle damage.

Objectives. To estimate the impact of ellagic acid (EA) towards healing tooth socket in diabetic animals, after tooth extraction.Methods. Twenty-fourSprague Dawleymale rats weighing 250–300 g were selected for this study. All animals were intraperitoneally injected with 45 mg/kg (b.w.) of freshly prepared streptozotocin (STZ), to induce diabetic mellitus. Then, the animals were anesthetized, and the upper left central incisor was extracted and the whole extracted sockets were filled with Rosuvastatin (RSV). The rats were separated into three groups, comprising 8 rats each. The first group was considered as normal control group and orally treated with normal saline. The second group was regarded as diabetic control group and orally treated with normal saline, whereas the third group comprised diabetic rats, administrated with EA (50 mg/kg) orally. The maxilla tissue stained by eosin and hematoxylin (H&E) was used for histological examinations and immunohistochemical technique. Fibroblast growth factor (FGF-2) and alkaline phosphatase (ALP) were used to evaluate the healing process in the extracted tooth socket by immunohistochemistry test.Results. The reactions of immunohistochemistry for FGF-2 and ALP presented stronger expression, predominantly in EA treated diabetic rat, than the untreated diabetic rat.Conclusion. These findings suggest that the administration of EA combined with RSV may have accelerated the healing process of the tooth socket of diabetic rats, after tooth extraction.

Abstract Objectives Food intake and exercise are considered modulators of the immune system. Specifically, intermittent fasting protocols have been demonstrated to reduce inflammation and alter cytokine responses. The objective of the current study was to determine if a form of intermittent fasting known as time-restricted feeding (TRF) would alter immune parameters in response to exercise. Methods 8-week-old C57BL/6 male mice were divided into three groups based on feeding schedule; group one had access to food ad libitum (Control) and groups two and three had access to food in a time restricted manner. Access was allowed for six hours per day either immediately after running (TRF-imm) or six hours after running (TRF-del). Mice ran on a treadmill for 1 hour, 5 days per week for eight weeks. Diet consisted of 21% protein, 16% fat and 64% carbohydrate. Weight, glucose and ketone levels, and immune populations were analyzed. Systemic IL-6 and TNF-α levels were measured before and after running. In a subpopulation, cytokine response to lipopolysaccharides (LPS) was also monitored. Results All mice gained weight during the eight-week intervention, but TRF-imm gained significantly less weight than Control (P = 0.02). No differences were detected in glucose levels. The ketone body β-hydroxybutyrate (BHB) was significantly higher at week eight in TRF groups (P ≤ 0.03) but running induced BHB in all groups to approximately 1 mM. Running reduced the blood lymphocytes levels (P &lt; 0.05), with a concomitant increase of granulocytes (P &lt; 0.05) in all groups. There was a small increase in monocytes only in the Control group (P = 0.017). No differences were detected in splenic immune populations, including CD4 and CD8 T cells, and CD11b + cells. Both IL-6 and TNF-α levels were low in all groups before exercise; however, post exercise IL-6 was increased, but not to the same extend in all groups. The IL-6 response was blunted in the TRF groups. The reduced levels of IL-6 was not due to loss of immune function, as both IL-6 and TNF-α were readily induced by exposure of mice to LPS. Conclusions Time-restricted feeding protocols did not induce differences in immune cell composition in blood or spleen but resulted in attenuated exercise-induced IL-6 levels.
