The study aimed to investigate and characterize the maternal effects of feeding Saccharomyces cerevisiae var. boularddii (Scb) to sows from late-gestation through lactation on progeny cortisol, immune status, and stress responsiveness from birth to 14 days post-weaning. Eighty-four piglets were born to sows fed control (CON) or probiotic (PRO) boluses twice daily for 59 days. Blood samples were obtained at birth and 24 h later to assess prenatal effects; 7, 14, and 21 day-of-age to assess potential developmental effects; and at 24 h, 7, and 14 days post-weaning to assess the effects of weaning stress on immune and cortisol responses. Pigs born to PRO sows had less robust cortisol response and enhanced immune parameters at birth and 24 h later, indicating less stress. In response to weaning, pigs born to and nursed by PRO sows displayed unique cortisol and immune profiles than CON pigs. These results indicate that feeding sows Scb probiotics during late gestation reduces stress responsiveness to farrowing stress while increasing immune cell populations. Pigs nursed by PRO sows had a more robust initial cortisol response and enhanced neutrophil function and B-cell lymphocyte proliferation in response to weaning stress. These data imply it may be possible to maternally alter immune and stress responses in utero and during suckling in the short-term and up to 14 days post-weaning. However, more research is needed to optimize this strategy.

The aim of our study was to evaluate the influence of asymptomatic infection and the occurrence of symptomatic COVID-19 on specific biochemical, renal, and immune parameters—renalase, neutrophil gelatinase-associated lipocalin (NGAL) cystatin C (CysC), and creatinine—and their weekly fluctuations during a one-month observation period in COVID-19 patients admitted to hospital. The study involved 86 individuals: 30 patients with diagnosed COVID-19, 28 people with asymptomatic infection confirmed with IgG antibodies—the IG(+) group—and 28 individuals without any (IgG, IgE) anti-SARS-CoV-2 antibodies—the IG(−) group. In the COVID-19 group, blood was drawn four times: (1) on day 0/1 after admission to hospital (C1 group), (2) 7 days later (C7 group), (3) 14 days later (C14 group), and (4) 28 days later (C28 group). In the IG(−) and IG(+) groups, blood was drawn once. There were no significant differences in creatinine, Cys C, and uric acid between any of the analyzed groups. NGAL levels were significantly higher in IG(+) and at all time-points in the COVID-19 groups than in controls. A similar observation was made for renalase at the C7, C14, and C28 time-points. Plasma renalase, NGAL, and CysC are unrelated to kidney function in non-critically ill COVID-19 patients and those with asymptomatic infection. Renalase and NGAL are most likely related to the activation of the immune system rather than kidney function. Asymptomatic SARS-CoV-2 infection causes a rise in plasma NGAL levels similar to those observed in symptomatic COVID-19 patients. Therefore, more attention should be paid to tracking and monitoring the health of these people.

The penis is the primary site of HIV acquisition in heterosexual men. Elevated penile inflammatory cytokines increase sexual acquisition risk, and topically applied cytokines enhance foreskin HIV susceptibility in an explant model. However, the impact of penile-vaginal sex on these immune parameters is undefined. Heterosexual couples were recruited to the Sex, Couples and Science (SECS) Study, with the collection of penile swabs, semen, cervico-vaginal secretions, and blood after a period of abstinence, and repeated sampling up to 72 hours after either condomless (n = 30) or condom-protected (n = 8) penile-vaginal sex. Soluble immune parameters were quantified by multiplex immunoassay. Co-primary immune endpoints were penile levels of IL-8 and MIG, cytokines previously linked to penile HIV acquisition. One hour after sex there were dramatic increases in penile IL-8 and MIG levels, regardless of condom use, with a gradual return to baseline by 72 hours; similar patterns were observed for other chemoattractant chemokines. Penile cytokine changes were similar in circumcised and uncircumcised men, and repeated measures ANOVA and ANCOVA models demonstrated that the degree of change after condomless sex was explained by cytokine levels in their partners’ cervico-vaginal secretions. This may have important implications for the biology of penile HIV acquisition.

In the current study, Melissa officinalis L. essential oil (MOEO), a novel sedative and anaesthetic, was employed in transport water to obtain a lower stress effect and higher survival rate for live marine fish. The effect of MOEO and various types of anaesthetics, administered at a low temperature on gill morphology, liver function and immunological parameters of living sea bass (Lateolabrax maculatus) subjected to transport stress, was evaluated to optimize the anaesthetic and sedative concentrations during live sea bass transport. Light microscopy and scanning electron microscopy of sea bass, subjected to simulated live transport for 72 h, demonstrated that the changes in the morphological characteristics of gill tissue treated with 40 mg/L MOEO (A3 group) were minimal in comparison to those observed in untreated sea bass. The results of pyruvate kinase (PK), phosphofructokinase (PFK), hexokinase (HK), hepatic glycogen (Gly), superoxide dismutase (SOD), lipid peroxides (MDA) and Caspase-3 assays indicated that the glycolysis rate, energy consumption, lipid peroxidation and hepatocyte apoptosis were the lowest in the A3 group. The values of the two immune parameters, lysozyme (LZM) and fish immunoglobulin M (IgM), indicated the strongest immunity ability in the A3 group. After 12 h recovery, sea bass treated with 30 mg/L MS-222 (B group) displayed a 100% survival rate, sea bass treated with 20 mg/L (A2 group) and 40 mg/L (A3 group) MOEO displayed a 96% survival rate, sea bass treated with 20 mg/L eugenol (C group) had a 94% survival rate, and untreated sea bass (CK group) had a 50% survival rate. Therefore, the addition MOEO to the transport water had anaesthetic and sedative effects similar to MS-222 and eugenol. The results confirmed that the addition of MOEO to the transport water could reduce tissue damage, energy metabolism, and the oxidative stress response in sea bass during transport.

Abstract The objective was to determine temporal changes in hematological and immune parameters in response to naturally occurring bovine respiratory disease (BRD) in commercially managed stocker calves. Forty newly weaned beef steers purchased from auction markets were housed at a commercial stocker operation in Crossville, TN. Blood samples, rectal temperature, and thoracic ultrasonography (TUS; 1: normal to 3: severe) were collected on days 0, 7, 14, and 21. Castration status (FC: freshly castrated; PC: previously castrated) was determined on arrival based on presence of a fresh castration site at the scrotum. Calves received antibiotics for BRD based on clinical severity scoring (CSS; 0: moribund, 4: moribund) and rectal temperature. Complete blood counts (CBC) were performed. Calves were categorized based on the number of treatments (NumTrt) received (0x, 1x, and 2x). Temporal variations in CBC and immune parameters were analyzed using mixed model repeated measure ANOVA (Proc GLIMMIX; SAS 9.4). Variation of CBCs and immune parameters based on TUS was determined using mixed model ANOVA. There was a NumTrt by day interaction effect on the responses of white blood cells (WBC) (P = 0.04) and Haptoglobin (HPT) (P = 0.04). On d 21, WBC were greater in the 2x NumTrt group than other groups, but there were no differences in WBC between NumTrt levels on other days. Haptoglobin was greater in the 2x group on d 14 and 21 than 0x or 1x. Red blood cells (RBC) (P = 0.02) and WBC (P = 0.04) differed between FC and PC group, and lower RBC and WBC were observed in the FC group. A castration status × day effect for mean corpuscular volume (MCV; P = 0.04) was observed where FC group had higher MCV at d 14 and 21 than the PC group. Tumor necrosis factor-α differed based on NumTrt (P = 0.03) and higher concentrations were found in 2x group. We observed a day effect for IL-1β (P = 0.009) and TNF-α (P = 0.001). Significant effect of TUS on HPT at d 14 (P = 0.0004) and d 21 (P = 0.002) was observed. Combining HPT and platelet explained 15% of the variability in treatment status on a given day. Whereas HPT and hemoglobin explained 10% of the variability in lung consolidation status. Although hematological and immunological parameters varied largely in our study, the potential of combining HPT with hematological variables should be studied further. Results from this study would help in understanding temporal changes in CBC and immune parameters in newly received stocker cattle.

The biological response to chemical pollutants reflects the acid–base status of an aquatic ecosystem. The gercacinid, Cardiosoma armatum (75±0.1 g) was exposed to acidified waters to evaluate the effects on its immune parameters. The crabs were exposed to pH 4.0, 5.0, 6.0 and 7.8 (control) for 28 days. The hematological parameters of control crabs and crabs exposed to varied doses of acidified water indicated a marked reduction. Significant (p<0.05) higher alkaline phosphatase and albumen were obtained in pH 4.0, 5.0 and 6.0 compared to control; other values were mostly similar to control. The highest superoxide dismutase (SOD) (252.61±0.06 min/mg pro) was recorded in control group, while highest CAT activity (2.08±0.16 min/mg protein) was recorded in crabs exposed to pH 4 treatment. Furthermore, the control group's SOD activity was significantly higher than the exposed groups. With a lower pH, the quantities of malondialdehyde increased substantially and were significantly different from the control group. While these findings demonstrate that changes in pH have limited impact on energy use, decreasing immune system conditions show that C. armatum is susceptible to pH variations and may be influenced in aquaculture, where a pH drop is more prominent.

Aim: The aim of this study was to assess the levels of some hematological, coagulation and immune parameters among male oil refinery workers in Port Harcourt, Nigeria. Study Design: This study is a cross-sectional study. Place and Duration of Study: Rivers State University Teaching Hospital (RSUTH), Port Harcourt, Shell Petroleum Development Company of Nigeria Limited and Modular Oil Refinery, Rivers State, between January 2021 and September 2021. Methodology: A total of one hundred (100) subjects (50 oil and gas workers as test subjects and 50 non-oil and gas workers as control subjects), were enrolled in the study. The convenient sampling technique was employed in the study. Venous blood samples were collected from all subjects and tested for Prothrombin Time (PT), Activated Partial Thromboplastin Time (APTT), Fibrinogen concentration, Full Blood Count (FBC) (Haemoglobin (Hb), Packed Cell Volume (PCV), Total White Blood Cell (WBC) count, Lymphocyte, Neutrophils, monocytes and Platelet Count), CD4 cell count, CD3 cell count and CD8 cell count. Data generated were analyzed statistically using Graph-Pad Prism, Version 8.0.2. Results: There was statistically significant decrease (P<.001) in Prothrombin time (PT) (11.55±0.73s), International Normalized Ratio (INR) (0.85±0.05), Fibrinogen (202.4±27.4mg/dl), Platelet count (185.6±37.1(103/μl)) and Neutrophils (46.6±6.4%) in oil refinery workers exposed to gas flare, while there was a statistical significant increase in APTT (31.8±4.15s), Hb (13.7±1.0g/dl), PCV (41.1±3.2%), Monocytes (8.4±3.0%) in subjects exposed to gas flare over control subjects with Prothrombin time(12.23±0.82s), INR(0.90±0.06), Fibrinogen(252.0±57.0mg/dl), platelet count(213.3±49.5(103/μl)) and Neutrophils(52.6±11.7%). Other parameters showed no statistical significant difference at P<.05 in both test and control subjects. Comparison of the mean ± standard deviation of the studied parameters in test subjects based on age using Analysis of Variance showed no statistically significant difference in all parameters at P<.05. Also, Comparison of the mean ± standard deviation of the studied parameters in test subjects based on duration of Exposure using Analysis of Variance showed a significant decrease in CD8 cells as the years of exposure increase (2-5years exposure = 865±319, 6-10years exposure = 579±288, 11-20 years exposure = 591± 286, F- Value = 3.869, P- Value = 0.0278). Conclusion: In conclusion, based on the findings, some haematogical, coagulation and immune parameters increased while others decreased in male oil refinery workers and duration of exposure can also be considered as a risk factor and age was considered not a risk factor as to cause any aberrations in the studied parameters.

BackgroundColorectal cancers (CRCs) with microsatellite instability-high (MSI-H) are hypermutated tumors and are generally regarded as immunogenic. However, their heterogeneous immune responses and underlying molecular characteristics remain largely unexplained.MethodsWe conducted a retrospective analysis of 73 primary MSI-H CRC tissues to characterize heterogeneous immune subgroups. Based on combined tumor-infiltrating lymphocyte (TIL) immunoscore and tertiary lymphoid structure (TLS) activity, MSI-H CRCs were classified into immune-high, immune-intermediate, and immune-low subgroups. Of these, the immune-high and immune-low subgroups were further analyzed using whole-exome and transcriptome sequencing.ResultsWe found considerable variations in immune parameters between MSI-H CRCs, and immune subgrouping of MSI-H CRCs was performed accordingly. The TIL densities and TLS activities of immune-low MSI-H CRCs were comparable to those of an immune-low or immune-intermediate subgroup of microsatellite-stable CRCs. There were remarkable differences between immune-high and immune-low MSI-H CRCs, including their pathological features (medullary vs mucinous), genomic alterations (tyrosine kinase fusions vs KRAS mutations), and activated signaling pathways (immune-related vs Wnt and Notch signaling), whereas no significant differences were found in tumor mutational burden (TMB) and neoantigen load. The immune-low MSI-H CRCs were subdivided by the consensus molecular subtype (CMS1 vs CMS3) with different gene expression signatures (mesenchymal/stem-like vs epithelial/goblet-like), suggesting distinct immune evasion mechanisms. Angiogenesis and CD200 were identified as potential therapeutic targets in immune-low CMS1 and CMS3 MSI-H CRCs, respectively.ConclusionsMSI-H CRCs are immunologically heterogeneous, regardless of TMB. The unusual immune-low MSI-H CRCs are characterized by mucinous histology, KRAS mutations, and Wnt/Notch activation, and can be further divided into distinct gene expression subtypes, including CMS4-like CMS1 and CMS3. Our data provide novel insights into precise immunotherapeutic strategies for subtypes of MSI-H tumors.
