AbstractLawsonia intracellularis is endemic to swine herds worldwide, however much is still unknown regarding its impact on intestinal function. Thus, this study aimed to characterize the impact of L. intracellularis on digestive function, and how vaccination mitigates these impacts. Thirty-six L. intracellularis negative barrows were assigned to treatment groups (n  =  12/trt): (1) nonvaccinated, L. intracellularis negative (NC); (2) nonvaccinated, L intracellularis challenged (PC); and (3) L. intracellularis challenged, vaccinated (Enterisol® Ileitis, Boehringer Ingelheim) 7 weeks pre-challenge (VAC). On days post-inoculation (dpi) 0 PC and VAC pigs were inoculated with L. intracellularis. From dpi 19–21 fecal samples were collected for apparent total tract digestibility (ATTD) and at dpi 21, pigs were euthanized for sample collection. Post-inoculation, ADG was reduced in PC pigs compared with NC (41%, P  <  0.001) and VAC (25%, P  <  0.001) pigs. Ileal gross lesion severity was greater in PC pigs compared with NC (P  =  0.003) and VAC (P  =  0.018) pigs. Dry matter, organic matter, nitrogen, and energy ATTD were reduced in PC pigs compared with NC pigs (P  ≤  0.001 for all). RNAscope in situ hybridization revealed abolition of sucrase-isomaltase transcript in the ileum of PC pigs compared with NC and VAC pigs (P  <  0.01). Conversely, abundance of stem cell signaling markers Wnt3, Hes1, and p27Kip1 were increased in PC pigs compared with NC pigs (P  ≤  0.085). Taken together, these data demonstrate that reduced digestibility during L. intracellularis challenge is partially driven by abolition of digestive machinery in lesioned tissue. Further, vaccination mitigated several of these effects, likely from lower bacterial burden and reduced disease severity.

Thus, this study aimed to characterize the impact of vaccination on growth performance and disease severity during experimental challenge. Thirty-six L. intracellularis negative barrows were assigned to treatment groups as follows (n = 12/trt): 1) nonvaccinated, L. intracellularis negative (NC); 2) nonvaccinated, L intracellularis challenged (PC); and 3) L. intracellularis challenged, vaccinated (Enterisol® Ileitis, Boehringer Ingelheim) at 1 week postweaning (VAC). On days post inoculation (dpi) 0 (7 weeks post-weaned) PC and VAC pigs were inoculated with L. intracellularis. Individual feed disappearance and BW were recorded weekly and all pigs were euthanized at dpi 21. Post-inoculation, ADG was reduced in PC (P &lt; 0.001) and VAC (P = 0.001) pigs compared with NC pigs. Average daily gain was additionally reduced in PC pigs compared with VAC pigs (P &lt; 0.001). Similarly, ADFI was reduced in PC (P &lt; 0.001) and VAC (P = 0.029) pigs compared with NC pigs, and further reduced in PC pigs compared with VAC pigs (P = 0.032). Overall G:F was reduced in PC pigs compared with NC (P &lt; 0.001) and VAC (P = 0.015) pigs, which did not differ. For antibody response and fecal shedding, NC pigs remained negative. Antibody responses and fecal shedding were lower in VAC pigs compared with PC pigs (P &lt; 0.05). Ileal gross lesion severity was greater in PC pigs compared with VAC (P = 0.018) and NC (P = 0.003) pigs, which did not differ. Similarly, ileal lesion length was greater in PC pigs compared with NC (P = 0.007) and VAC (P = 0.045) pigs. Microscopic lesion severity was greater in PC pigs compared with both PC and VAC pigs (P &lt; 0.05). Taken together, these data characterize the impact L. intracellularis has on growth performance of pigs and highlight the importance of vaccination to prevent disease associated performance losses.

Abstract Swine dysentery (SD) induced by Brachyspira hyodysentariae (Bhyo) causes colitis and mucohemorrhagic diarrhea in grow-finish pigs, however little is known about the physiological changes that occur to the gastrointestinal tract during Bhyo infection. Thus, the objective of this study was to evaluate the impact of a Bhyo challenge on intestinal function and integrity of pigs fed two divergent diets. A total of 36 Bhyo negative gilts (24.3 ± 3.6 kg BW) were selected and assigned to one of three treatment groups (n=12 pigs/trt): 1) Bhyo negative, 20% DDGS diet (CON), 2) Bhyo challenged, 20% DDGS diet (DDGS), and 3) Bhyo challenged, 10% DDGS, 5% beet pulp and 5% resistant potato starch diet (RS). Pigs were fed diets 21 days prior to challenge and on days post inoculation (dpi) 0 and 1, pigs were inoculated with Bhyo or sham. Fecal samples were collected for ATTD and pigs were euthanized for colon collection within 72 hours of initial observation of clinical SD, or at the end of the study (dpi 10-16). Tissues were assessed for ex vivo measures of intestinal integrity and mitochondrial function. The challenge resulted in high morbidity, with 88% of DDGS and RS pigs developing clinical SD. Colon transepithelial resistance was increased in DDGS pigs compared with CON and RS pigs (P=0.005), and colon macromolecule permeability was reduced in both DDGS and RS pigs compared with CON pigs (P=0.006), likely due to mucoid discharge. Colonic mitochondrial oxygen consumption was not impacted by treatment (P &gt;0.10). Further, ATTD of DM, OM, N, and GE were reduced in DDGS pigs compared with CON pigs (P&lt; 0.001), whilst nutrient digestibility was not reduced in RS pigs. Taken together, these data show Bhyo does not appear to reduce ex vivo colonic integrity. Further, the RS diet may reduce severity of a Bhyo challenge.

Loose dry sow housing became mandatory in the European Union from January 2013 onwards. One of the major causes of injuries to sows’ hooves and associated lameness is fighting on concrete/slatted flooring at grouping. Previous studies observed that in sows submitted to feed restriction, feed supplemented with dietary fiber reduced the time spent in the standing position and increased the time spent in the lying position. Therefore, we investigated the effect of increased levels of dietary fiber (7.2-7.5% crude fiber/kg dry matter) on the severity of hoof lesions of group housed sows in three Greek swine herds. The feet of 596 sows were initially examined for lesions upon their entry to the lactation facilities. Lesions scored included hoof hyperkeratinization, erosions or cracks and toe and dew claws overgrowths. When exiting the farrowing facilities they were offered recipes with increased dietary fiber throughout one or two subsequent gestations. Thirty-eight percent were re-examined for feet lesions during the first and sixty-two percent during the second lactation after initial examination. The proportion of sows with at least one lesion on any foot, at first scoring, was more than 95% in all herds. The most frequently and severely affected sites were the heel and the elongated toes and dew claws. The increased dietary fiber had no effect on lesion severity on any of the hoof sites considered.

A study was undertaken to compare the feeding of a ground-pelleted concentrate mixture with a textured one. Parameters studied were in situ rumen degradability characteristics of the concentrates, dry matter intake and yield of milk and milk components. A concentrate mixture consisting of barley, corn, canola meal and a vitamin-mineral premix was manufactured in two forms. In the one form, all ingredients were ground and pelleted while in the second, only the canola meal and premix were pelleted while the corn and barley were steam rolled prior to mixing. Initially the degradability characteristics and digestive tract disappearance of the two concentrates were determined in an in situ trial using two non-lactating Holstein cows fitted with rumen and duodenal cannulae. Nylon bags containing 2–3 g of material were prepared in quadruplicate (two replicates per cow per incubation time) and incubated in reverse order 72, 48, 24, 12, 8, 4, 2 and 0 h for each concentrate. Total intestinal disappearance was determined using the mobile nylon bag procedure. The results of the in situ trial indicated that the degradable fraction b of the dry matter was greater for the textured compared with the pelleted form (34.7 vs. 31.2%) while effective degradability of the dry matter was higher for the pelleted form (74.5 vs. 68.0%). Total tract disappearance of the dry matter and protein fractions were not influenced by form of concentrate but rumen disappearance of crude protein was higher for the pelleted form (70.4 vs. 64.2%). The feeding trial employed 14 lactating Holstein cows fed a forage mixture consisting of corn silage and grass silage. Milk and milk protein yields and protein content of the milk were higher, while milk fat content was lower, for cows fed the pelleted compared with those fed the textured concentrate. Rumen pH (6.79 vs. 6.58) was lower for cows fed the pelleted compared with the textured concentrate. There was, however, no effect of form of concentrate on concentrations of volatile fatty acids in the rumen fluid. Form of concentrate fed did not influence dry matter intake but in terms of net energy (Mcal NEL) the diet containing the pelleted concentrate tended to have a higher energy value than the diet containing the textured concentrate (1.76 vs. 1.66 Mcal NEL kg DMI−1). The marked difference in the ratio of milk fat to milk protein, 1.14 vs. 1.29 for pelleted and textured concentrates, respectively, coupled with the impact on rumen degradability characteristics indicated that minor differences in processing can have an impact on product yield from the milk and the efficiency of utilization of the diet. Abstract This study aimed to (1) evaluate the effects of flint maize processing methods on the estimation of the readily soluble fraction (a), the potentially degradable fraction (b) and the rate of degradation of b (c) for dry matter (DM), organic matter (OM) and starch in the rumen; and (2) verify whether two different applications of in situ technique can be used to estimate in vivo DM, OM and starch digestibilities. Five ruminally cannulated Nellore bulls (265 ± 18.2 kg; 8 ± 1.0 mo) were distributed in a 5 × 5 Latin square. Three experimental diets were composed of 0.30 whole-plant maize silage, 0.10 supplement and 0.60 of one of the following processing methods: dry ground maize grain (DMG); high-moisture maize (HMM); reconstituted maize grain silage (RMG). Two additional diets were composed of 0.10 supplement, 0.80 snaplage and 0.10 stalklage (SNAP-80); or 0.10 supplement and 0.90 snaplage (SNAP-90). Digestibilities were estimated using in vivo procedure or predicted from in situ technique using a single 24 h incubation point or an equation proposed in previous literature. Diets based on ensiled grains presented greater (P < 0.05) fraction a and c and lower (P < 0.05) fraction b of DM, OM and starch compared to DMG. Both alternative use of in situ technique accurately estimated (P > 0.05) in vivo DM, OM and starch digestibilities. The results suggest that ensilage process may increase the availability of nutrients. The two different applications of in situ technique showed precision and accuracy to estimate in vivo digestibility.

AbstractThe objective of this study was to determine whether feeding tannin-containing hays to heifers and mature beef cows influences enteric methane (CH4) emissions and nitrogen (N) excretion relative to feeding traditional legume and grass hays. Fifteen mature beef cows (Exp. 1) and 9 yearling heifers (Exp. 2) were each randomly assigned to treatment groups in an incomplete bock design with 2 periods and 6 types of hays with 3 hays fed each period (n = 5 cows and 3 heifers per treatment). Groups were fed tannin-containing [birdsfoot trefoil (BFT), sainfoin (SAN), small burnet (SML)] or non-tannin-containing [alfalfa (ALF), cicer milkvetch (CMV), meadow bromegrass (MB)] hays. Each period consisted of 14 d of adjustment followed by 5 d of sample collection. Nine cows and 9 heifers were selected for the measurement of enteric CH4 emissions (sulfur hexafluoride tracer gas technique), and excretion of feces and urine, while dry matter intake (DMI) was measured for all animals. The concentration of condensed tannins in SAN and BFT was 2.5 ± 0.50% and 0.6 ± 0.09% of dry matter (DM), respectively, while SML contained hydrolyzable tannins (4.5 ± 0.55% of DM). Cows and heifers fed tannin-containing hays excreted less urinary urea N (g/d; P &lt; 0.001) and showed lower concentrations of blood urea N (mg/dL; P &lt; 0.001) than animals fed ALF or CMV, indicating that tannins led to a shift in route of N excretion from urine to feces. Additionally, cows fed either BFT or CMV showed the greatest percentage of retained N (P &lt; 0.001). Enteric CH4 yield (g/kg of DMI) from heifers (P = 0.089) was greatest for MB, while daily CH4 production (g/d) from heifers (P = 0.054) was least for SML. However, digestibility of crude protein was reduced for cows (P &lt; 0.001) and heifers (P &lt; 0.001) consuming SML. The results suggest that tannin-containing hays have the potential to reduce urinary urea N excretion, increase N retention, and reduce enteric CH4 emissions from beef cattle. The non-bloating tannin-free legume CMV may also reduce environmental impacts relative to ALF and MB hays by reducing N excretion in urine and increasing N retention.

Abstract Objectives were to evaluate the effects of cattle breed, Holstein or Angus, and forage inclusion on total tract digestibility and ruminal pH in cattle fed a whole shelled corn (WSC) based diet. Six Holstein and 6 Angus steers, were assigned to a 2 × 3 factorial arrangement of treatments. Factors included breed, Holstein or Angus, and forage inclusion at 0, 8, or 16% forage (dry matter (DM) basis). Steers were fed in a replicated 3 × 3 Latin square, split-plot design. Each period consisted of 14 d diet adaptation followed by 7 d of sample collection. Data were analyzed using the MIXED procedures in SAS (v9.4 SAS Inst. Inc., Cary, NC). Repeated measures were used to analyze changes in ruminal pH over time. There was no interaction of breed × diet (P ≥ 0.19) on dry matter intake (DMI) or digestibility; however, Holstein steers had greater (P = 0.03) DMI than Angus steers. Despite the impact of breed on intake, there was no effect (P ≥ 0.33) of breed on diet digestibility. Digestibility of DM increased (linear; P &lt; 0.01) as forage was removed from the diet, but there were no differences (P ≥ 0.32) in NDF and starch digestibility. However, due to the change in diet, NDF intake digested on a grams per day basis increased (P ≤ 0.01) and starch intake digested (g/d) decreased (P = 0.01) as forage inclusion increased. There was a tendency for breed × diet interaction (P = 0.08) on ruminal pH. Holstein steers fed 8% or 16% forage had greater ruminal pH than Holstein steers fed 0% forage; but, ruminal pH of Angus steers was not altered by diet.

This study aims to determine theeffectiveness of the implementation of UAS which is carried out online using the Google Form applicationmedia at SMP MA'ARIF NU CIMANGGU. This research uses the quantitative survey method whichdescribes the online UAS implementation activities at SMP MA'ARIF NU CIMANGGU during thepandemic. The object consists of 35 student respondents. The data sample collection is done using aquestionnaire / questionnaire containing questions related to the implementation of UAS online using theGoogle Form application at SMP MA ' ARIF NU CIMANGGU. Based on the results of this study, theimplementation of online UAS runs effectively f and good, it can be seen from the results of satisfactorystudent scores. The application used is of course the Googke Form as the main media, and Whatsapp as amedium for interaction between teachers and students. The obstacles experienced are regarding badconnections, limited quota, schedules that frequently change, as well as difficulties in understandingsubject matter.Keywords: Covid-19,impact,effectiveness
