On the determination of the dorso-ventral polarity in the amphibian embryo: suppression by lactate of the formation of Ruffini's flask-cells

Cells isolated from the vegetal hemisphere of the blastula of Ambystoma mexicanum differentiate spontaneously into fibroblast-like cells. Similar cells may be formed from animal cells, provided they are induced either by vegetal cells or by Li+. We have found that lactate and various inhibitors of RNA synthesis suppress the spontaneous cell differentiation. The effect of lactate differs from that of the other agents in so far as lactate must be present before the second day of culture to suppress the outgrowth of flbroblasts on the third day; the other inhibitors are active also when added on the second day. An explanation of this difference may possibly be found in the fact that lactate suppresses incorporation of RNA but only by 40%. The effect on the differentiation of various substances supposed to interfere with the metabolism of lactate was established. The results obtained were suggestive, but not conclusive. It is concluded that the effect of anaerobiosis may be explained as a lactate inhibition. The amounts of lactate under aerobic conditions are so slight that it is unlikely, but not impossible, that lactate is directly involved in the control of differentiation.

The western North American dioecious species Rumex paucifolius is shown to be a tetraploid with 2n = 28 chromosomes. It is the third tetraploid known within the subgenus Acetosa, and the first polyploid dioecious taxon of that group, the others having either 2n = 14 ♂, 15 ♀ (R. Acetosa and relatives), or 2n = 8 ♂, 9 ♀ (R. hastatulus). The sex chromosomes of R. paucifolius are of the XX:XY type, the male sex being heterogametic. The X is a large chromosome, while the Y is the smallest chromosome of the complement. The mechanism of sex determination of R. paucifolius follows the Melandrium–Acetosella scheme with strongly epistatic male determinants in the Y–chromosome. Other dioecious Acetosae follow the Drosophila–Acetosa scheme of sex determination with a balance between the number of X and autosome complements, the Y being sexually inert. It is concluded from the observed cytogenetical and morphological differences that R. paucifolius should constitute a section of its own, Paucifoliae, which should be placed as far as possible from the section Acetosa, though within the same subgenus. The other American dioecious endemic, R. hastatulus, is placed in a subsection of the section Acetosa.

At present there are three methods for obtaining values of the stacking fault energy y of face-centred cubic (f.c.c.) materials by direct observation of dislocationstacking fault configurations in the electron microscope. These are based on measurements of extended three-fold dislocation nodes (e.g. Whelan 1958; Brown and ThOlen 1964), faulted dipole configurations (e.g. Haussermann and Wilkens 1966; Steeds 1967), and triangular Frank dislocation loops and stacking fault tetrahedral (e.g. Silcox and Hirsch 1959; Loretto, Clarebrough, and Segall 1965). The main advantages of the third method over the other two are that it is applicable to materials of a very wide range of stacking fault energy and involves only simple length measurements of defects that are easily recognized. However, it has suffered from the disadvantage that the values of y deduced from these measurements relied on an incomplete theory. The present authors have reconsidered this problem and, subject to the limitations of isotropic linear elasticity, have taken into account the major variables that may affect the values of y. It is the purpose of this note to present the results of this theory in a form in which values of y may easily be obtained from measurements of Frank dislocation loops and stacking fault tetrahedral without the resources of a large digital computer.

Somites are mesodermal structures which appear transiently in vertebrates in the course of their development. Cells situated ventromedially in a somite differentiate into the sclerotome, which gives rise to cartilage, while the other part of the somite differentiates into dermomyotome which gives rise to muscle and dermis. The sclerotome is further divided into a rostral half, where neural crest cells settle and motor nerves grow, and a caudal half. To find out when these axes are determined and how they rule later development, especially the morphogenesis of cartilage derived from the somites, we transplanted the newly formed three caudal somites of 2.5-day-old quail embryos into chick embryos of about the same age, with reversal of some axes. The results were summarized as follows. (1) When transplantation reversed only the dorsoventral axis, one day after the operation the two caudal somites gave rise to normal dermomyotomes and sclerotomes, while the most rostral somite gave rise to a sclerotome abnormally situated just beneath ectoderm. These results suggest that the dorsoventral axis was not determined when the somites were formed, but began to be determined about three hours after their formation. (2) When the transplantation reversed only the rostrocaudal axis, two days after the operation the rudiments of dorsal root ganglia were formed at the caudal (originally rostral) halves of the transplanted sclerotomes. The rostrocaudal axis of the somites had therefore been determined when the somites were formed. (3) When the transplantation reversed both the dorsoventral and the rostrocaudal axes, two days after the operation, sclerotomes derived from the prospective dermomyotomal region of the somites were shown to keep their original rostrocaudal axis, judging from the position of the rudiments of ganglia. Combined with results 1 and 2, this suggested that the fate of the sclerotomal cells along the rostrocaudal axis was determined previously and independently of the determination of somite cell differentiation into dermomyotome and sclerotome. (4) In the 9.5-day-old chimeric embryos with rostrocaudally reversed somites, the morphology of vertebrae and ribs derived from the explanted somites were reversed along the rostrocaudal axis. The morphology of cartilage derived from the somites was shown to be determined intrinsically in the somites by the time these were formed from the segmental plate. The rostrocaudal pattern of the vertebral column is therefore controlled by factors intrinsic to the somitic mesoderm, and not by interactions between this mesoderm and the notochord and/or neural tube, arising after segmentation.

Abstract A procedure is presented for the simultaneous determination of phenylephrine HCl (PE), chlorpheniramine maleate (CM), and methscopolamine nitrate in commercial tablets or capsules by liquid chromatography (LC) with 2 UV absorbance detectors in series. Reference and sample solutions are prepared in methanol. LC separations are performed on a 7.5 cm Novapak silica column. The mobile phase is prepared by mixing 930 mL methanol with 70 mL of a 0.5% aqueous solution of 1-pentanesulfonic acid, sodium salt. The injection volume is 20 L; the flow rate is approximately 1 mL/min. Retention times are approximately 1.5 min for PE, 3 min for CM, and 6 min for methscopolamine nitrate. One detector determines the first 2 compounds at 265 nm, but the third compound does not produce a detectable peak. The other detector set at 210 nm generates peaks for all 3 compounds, but only methscopolamine is within the recorder range; the other 2 compounds are exceedingly off scale. If it is not feasible or desirable to arrange 2 UV absorbance detectors in series, separate determinations can be made, one for the first 2 compounds and the other for the third component of the mixture. Two commercial samples of tablets and 2 commercial samples of capsules were analyzed by the proposed method. Recovery studies were also conducted with amounts of the 3 compounds ranging from 80 to 120% of the quantities present in the sample solutions.

In this paper, a new formula has been presented for accurate calculation of the third virial coefficient with Kihara potential. The obtained formula allows us to the determination of thermodynamic properties of imperfect gases. The validity of the formula has been tested by application to some gases C2H6, C3H8, C(CH3)4, and n – C5H12. The obtained results have been compared with the other studies. These comparisons show that the formula developed in this study is in good agreement with the data available in the literature.

Domains in GaAs can exist which are related to one another by the inversion symmetry, i.e., the sites of gallium and arsenic in one domain are interchanged in the other domain. The boundary between these two different domains is known as an antiphase boundary [1], In the terminology used to describe grain boundaries, the grains on either side of this boundary can be regarded as being Σ=1-related. For the {110} interface plane, in particular, there are equal numbers of GaGa and As-As anti-site bonds across the interface. The equilibrium distance between two atoms of the same kind crossing the boundary is expected to be different from the length of normal GaAs bonds in the bulk. Therefore, the relative position of each grain on either side of an APB may be translated such that the boundary can have a lower energy situation. This translation does not affect the perfect Σ=1 coincidence site relationship. Such a lattice translation is expected for all high-angle grain boundaries as a way of relaxation of the boundary structure.

IntroductionA simple and accurate technique to determine the Burgers vector of a dislocation has become feasible with the advent of HVEM. The conventional image vanishing technique(1) using Bragg conditions with the diffraction vector perpendicular to the Burgers vector suffers from various drawbacks; The dislocation image appears even when the g.b = 0 criterion is satisfied, if the edge component of the dislocation is large. On the other hand, the image disappears for certain high order diffractions even when g.b ≠ 0. Furthermore, the determination of the magnitude of the Burgers vector is not easy with the criterion. Recent image simulation technique is free from the ambiguities but require too many parameters for the computation. The weak-beam “fringe counting” technique investigated in the present study is immune from the problems. Even the magnitude of the Burgers vector is determined from the number of the terminating thickness fringes at the exit of the dislocation in wedge shaped foil surfaces.

Summary Aim of the study was to find out wether there is a common stop of growth of mandibular bone, so that no individual determination of the optimal time for surgery in patients with asymmetric mandibular bone growth is needed. As there are no epiphyseal plates in the mandibular bone, stop of growth cannot be determined on X-ray films. Methods: Bone scans of 731 patients [687 patients (324 male, 363 female) under 39 y for exact determination of end of growth and 44 (21 male, 23 female) patients over 40 y for evaluation of nongrowth dependant differences in tracer uptake] were reviewed for the study. All the patients were examined 3 hours after injection of 99mTc-DPD. Tracer uptake was measured by region of interest technique in different points of the mandibular bone and in several epiphyseal plates of extremities. Results: Tracer uptake in different epiphyseal plates of the extremities shows strong variation with age and good correlation with reported data of bone growth and closure of the epiphyseal plates. The relative maximum of bone activity is smaller in mandibular bone than in epiphyseal plates, which show well defined peaks, ending at 15-18 years in females and at 18-21 years in males. In contrast, mandibular bone shows no well defined end of growing but a gradually reduction of bone activity which remains higher than bone activity in epiphyseal plates over several years. Conclusion: No well defined end of growth of mandibular bone exists. The optimal age for surgery of asymmetric mandibular bone growth is not before the middle of the third decade of life, bone scans performed earlier for determination of bone growth can be omitted. Bone scans performed at the middle of the third decade of life help to optimize the time of surgical intervention.

SummaryTwo quantitative procedures for autoprothrombin C are described. In one of these purified prothrombin is used as a substrate, and the activity of autoprothrombin C can be measured even if thrombin is in the preparation. In this procedure a reaction mixture is used wherein the thrombin titer which develops in 20 minutes is proportional to the autoprothrombin C in the reaction mixture. A unit is defined as the amount which will generate 70 units of thrombin in the standardized reaction mixture. In the other method thrombin interferes with the result, because a standard bovine plasma sample is recalcified and the clotting time is noted. Autoprothrombin C shortens the clotting time, and the extent of this is a quantitative measure of autoprothrombin C activity.

SummaryAn assay for rapid factor XIII activity measurement has been developed based on the determination of the ammonium released during fibrin stabilization. Factor XIII was activated by thrombin and calcium. Ammonium was measured by an ammonium-sensitive electrode. It was demonstrated that the assay procedure yields accurate and precise results and that factor XIII-catalyzed fibrin stabilization can be measured kinetically. The amount of ammonium released during the first 90 min of fibrin stabilization was found to be 7.8 ± 0.5 moles per mole fibrinogen, which is in agreement with the findings of other authors. In 15 normal subjects and in 15 patients suffering from diseases with suspected factor XIII deficiency there was a satisfactory correlation between the results obtained by the “ammonium-release-method”, Bohn’s method, and the immunological assay (r1 = 0.65; r2= 0.70; p<0.01). In 3 of 5 patients with paraproteinemias the values of factor XIII activity determined by the ammonium-release method were markedly lower than those estimated by the other methods. It could be shown that inhibitor mechanisms were responsible for these discrepancies.
