1. Amino acids are actively reabsorbed by the antennal gland of Carcinus in all concentrations of sea water, but there is never complete recovery of these molecules from the urine; even in animals in 100% sea water amino acids are eliminated in the urine. 2. The urine concentration of amino acids is related to (i) the blood amino acid concentration and (ii) the rate of urine production. An increase in either or both of these factors, for example, when animals are in dilute sea water, raises the U/B ratio of amino acids. 3. The rate of reabsorption of amino acids increases as the amino acid load on the antennal gland is raised. When reabsorption is maximal, the rate of transport is about seven times greater than at normal concentrations of amino acids in blood and urine when the animal is in 100% sea water. 4. Increases in blood amino acid concentrations when crabs are placed directly into dilute sea water are taken to be the result of changes in free amino acids of muscle which occur under these conditions. This intracellular regulation in dilute media is initiated when the blood sodium concentration is approximately 400 mM/l.

1. U/B ratios for inulin and sorbitol have been measured in 100%, 75% and 50% sea water. 2. When Carcinus is in 100% sea water, a U/B ratio of 1 for both sorbitol and inulin is reached and maintained after 40 and 115 hr. respectively. A reason for this difference is suggested. 3. In dilute media the rate of increase of U/B ratio for both molecules is more rapid than in 100% sea water. For instance, in 50% sea water the inulin U/B ratio is 1 after 50-60 hr. Sorbitol ratios are generally less than 1 in dilute sea water and a possible explanation of this is given. 4. In any particular concentration of sea water blood and urine concentrations of injected solutes fall at approximately the same rate once the maximum urine concentration has been reached. 5. It is concluded that primary urine production in Carcinus is by a filtration mechanism and that reabsorption of water from the urine does not normally occur.

1. The space measured by inulin distribution, the ‘inulin volume’, has been determined, and represents approximately 20% of the body weight in crabs ranging in size from 20.0 to 57.2 g. 2. After the injection of labelled inulin into crabs, the increase in activity of the medium is equal to the fall in blood inulin in all dilutions of sea water. Clearance of inulin from the blood is due only to urine production, and therefore the molecule can be used for quantitative investigations of antennal gland function. 3. Urine production in various concentrations of sea water has been determined by measuring the clearance of inulin from the blood and the rates at which the tracer appeared in the external media. By these methods the mean rate of urine production in 100% sea water was estimated to be 4.4% body weight per day. In dilute sea water the rate of urine production increases; for example, in 50% sea water the urine flow is four times greater than in normal sea water.

1. The mechanism of sodium balance in Carcinus maenas has been investigated. 2. Measurements of sodium outflux showed no evidence of a decrease in surface permeability to sodium in dilute sea water. 3. The rate of urine production in normal sea water was 3.6% body weight per day and the sodium loss through the urine was insignificant compared with the total sodium loss. In 40% sea water the urine rate was increased to 30% body weight per day and the loss in the urine accounted for 20% of the total loss. 4. Measurements of sodium influx and calculation of the active component showed that the active uptake mechanism was fully saturated at all external concentrations in which the animals could survive. 5. Regulation of the blood sodium concentration is effected largely by the activation of the sodium uptake mechanism. This prevents the blood concentration falling below a critical level as long as the external concentration itself is not too low.

1. Glucose is actively reabsorbed by the antennal gland. 2. In 100% sea water there is a blood threshold concentration of approximately 150 mg.%. This does not indicate the maximum rate of glucose reabsorption. 3. Below the threshold, reabsorption is not always complete, but may be if carbohydrate sources are limited or absent. 4. The threshold level depends on the rate at which fluid passes through the antennal gland, an increase in urine production rate being correlated with a reduction in the glucose threshold. 5. Phloridzin inhibits reabsorption and glucose U/B ratios approach unity, indicating that glucose is filtered passively into the antennal gland.

1. The results of some chemical analyses for inorganic and organic constituents of the seminal plasma of Balanus balanus are presented. 2. The inorganic ions show a cation deficit of 178 m-equiv./l., which is probably made up by free amino-acids. 3. Cystine is a prominent amino acid present. 4. Potassium and calcium are present in excess of their quantities in sea water. 5. Reducing sugars, compared with the amount found in sea-urchin spermatozoa, are found in moderate quantities, 1 mg./ml. 6. Phosphorus of all kinds is present in only small quantities (total of o.14 mg./ml.). 7. Some phosphatases are present. 8. There are 21 /µg./ml. of ascorbic acid; the function of this is discussed in relation to its possible contribution to the protective action against the poisoning of -SH groups by thiol-reactive agents.

1. Non-protein and protein nitrogen fractions of the isopod Sphaeroma rugicauda were measured in animals adapted to 100 and 2% sea water. 2. The non-protein nitrogen component was reduced in animals acclimatized to the lower salinity. 3. Free amino acids accounted for 88 and 74% respectively of the non-protein nitrogen in the two salinities. 4. In 2% sea water taurine, proline, glycine, alanine and glutamic acid showed the greatest decreases in concentration compared to the levels measured in animals adapted to 100% sea water. 5. The decrease in total free amino acids of animals acclimatized to 100% sea water and transferred to 2% sea water was measured. 6. The total free amino acid concentration is reduced to the 2% sea water level within 12 hr. after transfer. 7. Free amino acid, haemolymph sodium and total body sodium levels after transfer to 2% sea water were compared. 8. The asymmetry between the fall in haemolymph sodium concentration and the decrease in total body sodium under these conditions is thought to be due to a water shift from the haemolymph into the tissues. 9. It is suggested that the osmotic pressure of the cells falls at a slower rate than that of the haemolymph.

SummaryAn in vitro investigation has been made of the relative roles of the gut and tegument in the absorption of the neutral amino acids L-alanine and L-leucine by a marine fish-gill parasite, Diclidophora merlangi. The use of ligatures to preclude oral ingestion of trace-labelled medium has proved inadequate, invariably damaging the tegument, as revealed by stereoscan electron microscopy, and resulting in artifactual levels of absorption. Three alternative procedures have given consistently reliable data on the route of entry of low molecular weight substrates. (1) Ultrastructural examination of worms previously incubated in electron-dense cationic tracers has shown that, in vitro, there is no oral intake of sea water. (2) The suspending of worms in trace-labelled medium with the mouth out of the medium and comparing amino acid uptake with that of worms totally immersed in medium has revealed no statistically significant difference in the absorption levels. (3) Application of section (freeze-dried) auto-radiography to detect diffusible isotope has demonstrated directly transtegumental absorption of a neutral amino acid. It is concluded from these experiments that Diclidophora has a tegumental transport system for absorbing certain neutral amino acids, and whilst, clearly, the worm is sanguinivorous and digests blood in a well-developed gut, it may also be capable of supplementing this diet with low molecular weight organic nutrient absorbed directly from sea water via the tegument.

Amino acids, both free and combined as protein, and some other nitrogenous constituents of Calanus finmarchicus have been examined. Seventy-six per cent of the total nitrogen in C. finmarchicus was present in protein amino acids, 14% in the free amino acid fraction, 6% in trimethylamine oxide, and 1·5% in betaine. These findings are discussed in relation to previous work on nitrogenous constituents of Calanus.The free amino acid fraction of Calanus is compared with that of higher Crustacea and it is suggested that this fraction may be important in the adaptation of the animal to dilute sea water.Amino acid nitrogen made up 40% of the total nitrogen of particulate material from Clyde sea water. This finding is compared with other recent analyses of the phytoplanktonic nitrogen and total particulate nitrogen of inshore waters.

Abstract The optical isomers of amino acids can be easily separated by gas chromatography using capillary columns coated with the chiral polysiloxane peptide, Chirasil-Val. Quantitative trace amino acid analysis in complex mixtures such as biological fluids, sea water, or protein hydrolysates can be achieved by enantiomer labeling: The D-amino acid enantiomers, which do not occur naturally, are added to the sample prior to analysis as internal standards. Because the D-enantiomers show the same physical and chemical properties as the natural L-enantiomers, they are ideal standard references. In routine analysis, the derivatization is achieved with a new automated derivatization robot. The D-standard serves as overall internal standard for the whole analytical procedure from sample enrichment to derivatization, chromatography, and response of the detector.

Pembenihan ikan kerapu dihadapkan pada rendahnya sintasan, terutama fase awal hidupnya. Rendahnya sintasan diduga berhubungan dengan belum berkembangnya organ pencernaan larva dan tidak tersedianya pakan yang siap diserap tubuh. Pemberian asam amino bebas seperti taurin dan glutamin melalui rotifer diharapkan dapat meningkatan ketersediaan nutrien yang dapat diserap dengan cepat oleh larva, sehingga sintasan larva tinggi dan dapat tumbuh lebih cepat. Penelitian bertujuan mengukur pengaruh glutamin atau taurin terhadap sintasan dan pertumbuhan larva kerapu bebek Cromileptes altivelis. Larva umur 1 hari (10 ekor.L-1) dipelihara dalam bak fiberglass volume 500 L yang diisi air laut sampai 330 L. Larva diberi pakan rotifer diperkaya 0,5 g glutamin, 0,5 g taurin, atau tanpa glutamin dan taurin. Larva dipelihara selama 16 hari dengan pemberian pakan 2 kali  per hari. Satu jam setelah pemberian pakan dilakukan pengamatan terhadap jumlah rotifer dalam saluran pencernaan larva ikan. Hasil penelitian menunjukkan jumlah rotifer dalam saluran pencernaan larva meningkat seiring dengan bertambahnya umur. Namun, jumlah rata-rata rotifer yang dikonsumsi larva pada perlakuan 0,5 g glutamin atau 0,5 g taurin lebih banyak dibanding perlakuan tanpa taurin dan glutamin. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g glutamin atau 0,5 g taurin memiliki sintasan yang tidak berbeda (P>0,05), masing-masing 19,2±2,25%, dan 24,5±1,20%, dan secara signifikan lebih banyak dibanding larva di perlakuan tanpa glutamin dan taurin yang nilainya 13,0±1,70%. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g taurin secara signifikan lebih panjang (5,5±0,18 mm) dari ikan di perlakuan 0,5 g glutamin (5,3±0,12 mm). Demikian juga, larva di perlakuan glutamin lebih panjang dari pelakuan tanpa glutamin dan taurin (5,0±0,16 mm). Pengkayaan rotifer menggunakan asam amino bebas taurin atau glutamin dapat meningkatkan sintasan dan pertumbuhan larva kerapu bebek. Namun, taurin lebih baik dibanding glutamin karena memberi efek ke pertumbuhan ikan yang lebih cepat. Kata kunci: taurin, glutamin, kerapu bebek, Cromileptes altivelis. The hatchery production of grouper faced to the problem of low surival rate, especially during the first feeding period. During this period, the digestive organ of larvae was underdeveloped, and the nutrient in live food was not readily absorbed by larvae. Feeding on free amino acids, such as taurine or glutamine, enriched rotifer would be enhanced the nutrient readily absorbed thereby resulting to the improvement of growth and survival rate of larvae. A duplicate experiment was conducted to evaluate the effect of rotifer enriched with free amino acid of glutamine or taurine, on the growth and survival rate of larval grouper Cromileptes altivelis. One day old larvae were cultured in 500 l fiberglass containing 330 l of sea water with a density of 10 ind.L-1. Larvae were fed on rotifer enriched with either 0.5 g glutamine, 0.5 g taurine, or without glutamine nor taurine. Larvae were fed on rotifer twice a day at 08.00 and 14.00, and cultured for 16 days. One h after feeding, rotifer content in the gut of larvae were observed. Results showed that larvae fed on rotifer enriched with 0.5 g taurine or 0.5 g glutamie had higher rotifer content in the gut than that in the other group. Larvae fed on rotifer enriched with 0.5 g glutamine or 0.5 g taurine had the same survival rate (P>0.05), 19.2±2.25% and 24.5±1.20%, respectively; these values were significantly higher than that in the other group which was 13.0±1.70%.  The total length of larvae fed on rotifer enriched with 0.5 g taurine (5.5±0.18 mm) significantly was the biggest compared than others; while those fed on rotifer enriched with 0.5 g glutamine (5.3±0.12 mm) was bigger than fish fed on rotifer without glutamine nor taurine (5.0±0.16 mm). Thus, it can be concluded that survival rate and growth of larvae were improved when feeding on rotifer enriched with taurine or glutamine. Nevertheless, taurine was more efective than glutamine due to its effect resulted the highest growth performance of larval grouper. Keywords: taurine, glutamine, grouper, Cromileptes altivelis
