Amino acids and some other nitrogenous compounds in Calanus finmarchicus

Amino acids, both free and combined as protein, and some other nitrogenous constituents of Calanus finmarchicus have been examined. Seventy-six per cent of the total nitrogen in C. finmarchicus was present in protein amino acids, 14% in the free amino acid fraction, 6% in trimethylamine oxide, and 1·5% in betaine. These findings are discussed in relation to previous work on nitrogenous constituents of Calanus.The free amino acid fraction of Calanus is compared with that of higher Crustacea and it is suggested that this fraction may be important in the adaptation of the animal to dilute sea water.Amino acid nitrogen made up 40% of the total nitrogen of particulate material from Clyde sea water. This finding is compared with other recent analyses of the phytoplanktonic nitrogen and total particulate nitrogen of inshore waters.

1. Non-protein and protein nitrogen fractions of the isopod Sphaeroma rugicauda were measured in animals adapted to 100 and 2% sea water. 2. The non-protein nitrogen component was reduced in animals acclimatized to the lower salinity. 3. Free amino acids accounted for 88 and 74% respectively of the non-protein nitrogen in the two salinities. 4. In 2% sea water taurine, proline, glycine, alanine and glutamic acid showed the greatest decreases in concentration compared to the levels measured in animals adapted to 100% sea water. 5. The decrease in total free amino acids of animals acclimatized to 100% sea water and transferred to 2% sea water was measured. 6. The total free amino acid concentration is reduced to the 2% sea water level within 12 hr. after transfer. 7. Free amino acid, haemolymph sodium and total body sodium levels after transfer to 2% sea water were compared. 8. The asymmetry between the fall in haemolymph sodium concentration and the decrease in total body sodium under these conditions is thought to be due to a water shift from the haemolymph into the tissues. 9. It is suggested that the osmotic pressure of the cells falls at a slower rate than that of the haemolymph.

L-Methionine-1-14C was fed through the roots to 6-day-old pea seedlings. After 10 h over 80% of the absorbed radioactivity was recovered in the free amino acid fraction, 8% in the extracted proteins, 5% as carbon dioxide, and 2.2% in the organic acid fractions. Analysis of the amino acids revealed that only 24% of the total activity was recovered as methionine, 40% as homoserine, 10% as isoleucine, and smaller but significant amounts of aspartate, O-acetylhomoserine, and cystathionine. Dilution values strongly imply that aspartate is not an intermediate in the extensive conversion of methionine to homoserine; rather a more direct pathway is involved.

The nitrogen compounds of the faeces of dairy cows on purified, protein-free feed with urea and small amounts of ammonium salts as the sole source of nitrogen (0-feed). on low-protein feed in which part of the protein was replaced with urea (ULP-feed) and on normal protein-containing feed (NorP-feed) were studied. The total nitrogen contents of the dry matter of the faeces were 2.5 ± 0.7 % on 0-feed, 2.4 ± 0.4 % on ULP-feed and 2.3 ± 0.6 % on NorP-feed. The proportion of urea- and ammonium nitrogen of the total nitrogen was 3.2 ± 2.4, 4.5 ± 2.4 and 3.7 ± 1.8 % respectively, and the proportion of nitrogen soluble in 70 % ethanol 10.9 ± 5.7, 9.9 ± 2.4 and 10.4 ± 3.8 % of the total nitrogen; the proportion of nitrogen in the free amino acid fraction was 4.1 ±2.9 on 0-feed, 1.6 ± 1.4 (P < 0.05) on ULP-feed and 3.2 ± 2.9 % on NorP-feed. The nitrogen insoluble in 70 % ethanol as a proportion of the total nitrogen was 82.3 ± 7.2 on 0-feed, 85.4 ± 5.0 on ULP-feed and 82.3 ± 7.2 % on NorP-feed. Of the total nitrogen of this insoluble fraction, the following proportions were released as amino acids by acid hydrolysis: 62.15 ± 8.5 % on 0-feed, 61.6 ± 10.7 % on ULP-feed and 59.0 ± 13.9% on NorP-feed. (α, Ɛ-diaminopimelic acid was 1.7 ± 0.6, 0.7 ± 0.2 (P < 0.05) and 1.2 ± 0,4 mol % respectively of the total amino acids of the hydrolyzate. The soluble nitrogen fraction was separated into free amino acid and »peptide» fractions, and the amino acid composition of both was determined, the latter after acid hydrolysis

Pembenihan ikan kerapu dihadapkan pada rendahnya sintasan, terutama fase awal hidupnya. Rendahnya sintasan diduga berhubungan dengan belum berkembangnya organ pencernaan larva dan tidak tersedianya pakan yang siap diserap tubuh. Pemberian asam amino bebas seperti taurin dan glutamin melalui rotifer diharapkan dapat meningkatan ketersediaan nutrien yang dapat diserap dengan cepat oleh larva, sehingga sintasan larva tinggi dan dapat tumbuh lebih cepat. Penelitian bertujuan mengukur pengaruh glutamin atau taurin terhadap sintasan dan pertumbuhan larva kerapu bebek Cromileptes altivelis. Larva umur 1 hari (10 ekor.L-1) dipelihara dalam bak fiberglass volume 500 L yang diisi air laut sampai 330 L. Larva diberi pakan rotifer diperkaya 0,5 g glutamin, 0,5 g taurin, atau tanpa glutamin dan taurin. Larva dipelihara selama 16 hari dengan pemberian pakan 2 kali  per hari. Satu jam setelah pemberian pakan dilakukan pengamatan terhadap jumlah rotifer dalam saluran pencernaan larva ikan. Hasil penelitian menunjukkan jumlah rotifer dalam saluran pencernaan larva meningkat seiring dengan bertambahnya umur. Namun, jumlah rata-rata rotifer yang dikonsumsi larva pada perlakuan 0,5 g glutamin atau 0,5 g taurin lebih banyak dibanding perlakuan tanpa taurin dan glutamin. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g glutamin atau 0,5 g taurin memiliki sintasan yang tidak berbeda (P>0,05), masing-masing 19,2±2,25%, dan 24,5±1,20%, dan secara signifikan lebih banyak dibanding larva di perlakuan tanpa glutamin dan taurin yang nilainya 13,0±1,70%. Larva yang diberi rotifer hasil pengkayaan dengan 0,5 g taurin secara signifikan lebih panjang (5,5±0,18 mm) dari ikan di perlakuan 0,5 g glutamin (5,3±0,12 mm). Demikian juga, larva di perlakuan glutamin lebih panjang dari pelakuan tanpa glutamin dan taurin (5,0±0,16 mm). Pengkayaan rotifer menggunakan asam amino bebas taurin atau glutamin dapat meningkatkan sintasan dan pertumbuhan larva kerapu bebek. Namun, taurin lebih baik dibanding glutamin karena memberi efek ke pertumbuhan ikan yang lebih cepat. Kata kunci: taurin, glutamin, kerapu bebek, Cromileptes altivelis. The hatchery production of grouper faced to the problem of low surival rate, especially during the first feeding period. During this period, the digestive organ of larvae was underdeveloped, and the nutrient in live food was not readily absorbed by larvae. Feeding on free amino acids, such as taurine or glutamine, enriched rotifer would be enhanced the nutrient readily absorbed thereby resulting to the improvement of growth and survival rate of larvae. A duplicate experiment was conducted to evaluate the effect of rotifer enriched with free amino acid of glutamine or taurine, on the growth and survival rate of larval grouper Cromileptes altivelis. One day old larvae were cultured in 500 l fiberglass containing 330 l of sea water with a density of 10 ind.L-1. Larvae were fed on rotifer enriched with either 0.5 g glutamine, 0.5 g taurine, or without glutamine nor taurine. Larvae were fed on rotifer twice a day at 08.00 and 14.00, and cultured for 16 days. One h after feeding, rotifer content in the gut of larvae were observed. Results showed that larvae fed on rotifer enriched with 0.5 g taurine or 0.5 g glutamie had higher rotifer content in the gut than that in the other group. Larvae fed on rotifer enriched with 0.5 g glutamine or 0.5 g taurine had the same survival rate (P>0.05), 19.2±2.25% and 24.5±1.20%, respectively; these values were significantly higher than that in the other group which was 13.0±1.70%.  The total length of larvae fed on rotifer enriched with 0.5 g taurine (5.5±0.18 mm) significantly was the biggest compared than others; while those fed on rotifer enriched with 0.5 g glutamine (5.3±0.12 mm) was bigger than fish fed on rotifer without glutamine nor taurine (5.0±0.16 mm). Thus, it can be concluded that survival rate and growth of larvae were improved when feeding on rotifer enriched with taurine or glutamine. Nevertheless, taurine was more efective than glutamine due to its effect resulted the highest growth performance of larval grouper. Keywords: taurine, glutamine, grouper, Cromileptes altivelis

The effect of storage at subzero temperatures (-5°C, -12°C, and -20°C) on hake ( Merluccius merluccius L.) muscle free-amino acid fraction was evaluated. A significant increase in free aspartic acid, serine, threonine, arginine, β-alanine, tyrosine, methionine, valine, phenylalanine, isoleucine, leucine, and lysine was found at -5°C, whereas at -12°C, a significant decrease in free glutamic acid, glycine, methyl-histidine, β-alanine, taurine, alanine, and leucine was the most noticeable. No changes in the free amino acid fraction were observed at -20°C. Activity of different kind of enzymes, aminopeptidases, aminoacid deaminases, and decarboxylases might be involved in the changes observed at -5°C and -12°C.

The leaves and flowers of several genetic and cytoplasmic male-sterile lines were examined for free and protein amino acid content. Fewer amino acids were observed in the leaves in the free amino acid fraction than in the protein fraction. In both samples leaves of all lines had a similar number and quantity of amino acids. Amino acid content in the protein fraction was lower in young buds than mature flowers. Differences in amino acids between normal and sterile flowers were more quantitative than qualitative.

Studies with acetate-2-14C showed that leaves of light-grown seedlings of the mutant were less efficient in the synthesis of protein particularly at the 6-day stage. The synthesis of leucine from labeled acetate was also lower in the mutant at this stage. Metabolism of leucine-U-14C confirmed the lower rate of protein synthesis in light by the mutant at the 6-day stage whereas, in the dark, protein synthesis in the two lines was similar. There was essentially no difference in the use of labeled glycine by the two lines. Free and protein amino acids accounted for over 75% of the label. Glycine was rapidly converted to serine, which contained over 65% of the label in the free amino acid fraction but less than 50% of the label in the protein fraction. Evidence for the existence of two separate pools of serine was obtained. Only 3 to 5% of the label from glycine was detected in chlorophyll.Administration of δ-aminolevulinic acid increased chlorophyll synthesis in the mutant in the same proportion as it did in the normal. It was, therefore, concluded that inadequate production of δ-aminolevulinic acid or its precursors was not responsible for the virescent character of the mutant. Rather it seems that the mutation caused a partial inhibition of synthesis of chlorophyll holochrome protein which accounted for the slower accumulation of chlorophyll in the mutant.

Aim and Objective:: To determine the mechanisms present in the etiopathogenesis of nasal polyposis. It is not clear whether amino acids contribute in a causal way to the development of the disease. Therefore, the aim of this study was to determine the plasma-free amino acid profile in patients with nasal polyposis and to compare the results with a healthy control group. Materials and Methods:: This was a prospective controlled study that took place in the Otolaryngology Department at the Harran University Faculty of Medicine between April 2017 and April 2018. Plasmafree amino acid profile levels were studied in serum samples taken from a patient group and a healthy control group. Patients who were diagnosed with bilateral diffuse nasal polyposis and were scheduled for surgical interventions were included in this study. Individuals whose age, gender, and body mass index values were compatible with that of the patient group and who did not have any health problems were included in the control group. All the participants whose levels of plasma-free amino acid were thought to be affected by one or more of the following factors were excluded from the study: smoking and alcohol use, allergic rhinitis presence, the presence of acute or chronic sinusitis, a history of endoscopic sinus surgery, unilateral nasal masses, a history of chronic drug use, systemic or topical steroid use in the last three months for any reason, and liver, kidney, hematological, cardiovascular, metabolic, neurological, or psychiatric disorders or malignancies. However, the gamma-aminobutyric acid (gaba: ng = 0.16 (0.10 – 0.24); cg = 0.21 (0.19 – 0.29); p = 0.010) plasma concentration was significantly lower in the nasal polyposis group than in the healthy control group. Conclusion: In this study, plasma levels of 15 free amino acids were significantly higher in the nasal polyposis group than in the healthy control group. A plasma level of 1 free amino acid was found to be significantly lower in the nasal polyposis group compared to the healthy control group. Therefore, it is important to determine the possibility of using the information obtained to prevent the recurrence of the condition and to develop effective treatment strategies. This study may be a milestone for studies of this subject. However, this study needs to be confirmed by further studies conducted in a larger series.

AbstractWild soybean (Glycine soja) is a valuable genetic resource for soybean improvement. Seed composition profiles provide beneficial information for the effective conservation and utilization of wild soybeans. Therefore, this study aimed to assess the variation in free amino acid abundance in the seeds of wild soybean germplasm collected in Japan. The free amino acid content in the seeds from 316 accessions of wild soybean ranged from 0.965 to 5.987 mg/g seed dry weight (DW), representing a 6.2-fold difference. Three amino acids had the highest coefficient of variation (CV): asparagine (1.15), histidine (0.95) and glutamine (0.94). Arginine (0.775 mg/g DW) was the predominant amino acid in wild soybean seeds, whereas the least abundant seed amino acid was glutamine (0.008 mg/g DW). A correlation network revealed significant positive relationships among most amino acids. Wild soybean seeds from different regions of origin had significantly different levels of several amino acids. In addition, a significant correlation between latitude and longitude of the collection sites and the total free amino acid content of seeds was observed. Our study reports diverse phenotypic data on the free amino acid content in seeds of wild soybean resources collected from throughout Japan. This information will be useful in conservation programmes for Japanese wild soybean and for the selection of accessions with favourable characteristics in future legume crop improvement efforts.
