Aedes mosquitoes were found to lay their eggs in the cryptic breeding sites. Eliminating cryptic and open breeding sites is essential in reducing dengue virus transmission. However, it is often challenging for health officers to assess these breeding sites which are usually missed during larval surveillance. The autodissemination approach may produce a better outcome by manipulating female mosquitoes to disperse insecticide to other Aedes spp. mosquito habitats. Thus, the present study aims to evaluate the effectiveness of the pyriproxyfen autodissemination technique using Mosquito Home System against the population of mosquitoes. This study was conducted in Bandar Baru Bangi, Selangor, Malaysia. The Mosquito Home System was deployed to control Aedes spp. populations at treatment sites using before-after-control-impact (BACI) design. The presence of pyriproxyfen distribution was confirmed using the WHO larval bioassay which resulted in 10-35% larvae mortalities. Autodissemination of pyriproxyfen significantly reduced the population size of mosquito eggs (p<0.05), larvae (p<0.05), and ovitrap index (p<0.05) at the treatment areas compared to the control areas. Moreover, rainfall was correlated positively against ovitrap index (r = 0.247), larvae (r = 0.420), and eggs (r = 0.422). The study provides promising results for controlling Aedes spp. populations and also highlights the potentials of this technique as an alternative in vector control programmes. However, further studies on larger scale field trials are warranted.

Dengue virus (DENV), a member of the Flavivirus genus of the Flaviviridae family, can cause dengue fever (DF) and more serious diseases and thus imposes a heavy burden worldwide. As the main vector of DENV, mosquitoes are a serious hazard. After infection, they induce a complex host–pathogen interaction mechanism. Our goal is to further study the interaction mechanism of viruses in homologous, sensitive, and repeatable C6/36 cell vectors. Transcriptome sequencing (RNA-Seq) technology was applied to the host transcript profiles of C6/36 cells infected with DENV2. Then, bioinformatics analysis was used to identify significant differentially expressed genes and the associated biological processes. Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was performed to verify the sequencing data. A total of 1239 DEGs were found by transcriptional analysis of Aedes albopictus C6/36 cells that were infected and uninfected with dengue virus, among which 1133 were upregulated and 106 were downregulated. Further bioinformatics analysis showed that the upregulated DEGs were significantly enriched in signaling pathways such as the MAPK, Hippo, FoxO, Wnt, mTOR, and Notch; metabolic pathways and cellular physiological processes such as autophagy, endocytosis, and apoptosis. Downregulated DEGs were mainly enriched in DNA replication, pyrimidine metabolism, and repair pathways, including BER, NER, and MMR. The qRT-PCR results showed that the concordance between the RNA-Seq and RT-qPCR data was very high (92.3%). The results of this study provide more information about DENV2 infection of C6/36 cells at the transcriptome level, laying a foundation for further research on mosquito vector–virus interactions. These data provide candidate antiviral genes that can be used for further functional verification in the future.

Abstract: Dengue Hemorrhagic Fever (DHF) is one of the most serious health problems in Indonesia which often causes outbreaks with numerous deaths. The disease is transmitted byAedes sp.females. Generally, dengue virus transmission occurs horizontally from human carriers, and the dengue viruses are passed on bytheir vectors through blood sucking activity. After propagation in the mosquito, the viruses are transmitted to human recipients. In addition, there is a vertical transmission (transovarial) of dengue virusesin the ova of Aedes sp.females. The viruses propagate in the ova that undergo  metamorphosis to become larvae, pupae, and imagoes. The transovarial transmission of dengue virusesin its vectors in endemic areas could be a causative key which is responsible for the phenomenon of increasing cases of DHF. Any effort to prevent and control DHF requires a thorough understanding about virDen transmission, including this transovarial transmission in Aedes spfemales. Transmisi virus dengue umumnya terjadi secara horizontal, yaitu dari manusia pembawa virus dengue ke nyamuk vektor Aedes sp. melalui aktivitasnya mengisap darahSetelah mengalami propagasi  dalam  tubuh nyamuk, virus dengue ditularkan ke  manusia penerima. Selain itu, transmisi virus dapat terjadi secara vertikal (transovarial) yaitu virus dengue dalam tubuh nyamuk vektorAedes sp. betinake ovum, kemudian berpropagasi dalam ovum, larva, pupa, dan imago. Transmisi transovarial virus dengueke vektornya di daerah endemik bisa menjadi kunci penyebab yang bertanggung jawab terhadap fenomena peningkatan kasus deman berdarah dengue. Upaya pencegahan dan penanggulangan DBD memerlukan pengetahuan yang matang tentang adanya infeksi transovarial virDen pada nyamuk Aedes sp. Kata kunci : DBD, transmisi transovarial,  Aedes sp.

Dengue Hemorrhagic Fever (DHF) is a disease caused by Dengue virus. In 2016, the number of DHF patients in areas of Health services of Rowosari was 247 cases, IR 289.6/100.000 populations and the highest number of the patients was from Meteseh Village. The most usual mechanism happened in the vertical transmission, transovarial infection, from adult mosquito to the egg. The study was conducted to describe of entomological status of House Index (HI), Container Index (CI), Breteau Index (BI), House index (HI), Ovitrap Index (OI) of larvae as well as transovarial infection in Aedes sp. The research method was descriptive method by conducting test to the entomology status of DHF incidence. Based on the survey in Meteseh sub-village (XVI), a result found that average House Index 8.99%, Container Index 2.88%, Breteau Index 16.40%, HI 8,99 Ovitrap Index 35.46% with the egg density at 1.023 eggs. The identified mosquitoes were 30, and from the identification result of Aedes sp., two Aedes species were found, 28 Aedes aegypti,and only 2 Aedes albopictus. The transovarial infection in Aedes sp. using ELISA method was found 6 mosquito samples which were positive of dengue virus, with the detail in neighborhood 01 (1 sample), neighborhood 02 (2 samples), neighborhood 03 (3 samples). DHF in Meteseh Sub village in Semarang City was probably happened because correlated with transovarial infection.

Background: The higher incidence of dengue fever in Pakistan demands additional efforts in order to limit the disease. Despite active public health campaigns, low public awareness is one of the factors facilitating dengue virus transmission. For effective preventive measures, the assessment of the knowledge gap and then taking appropriate steps to fill the gap is required.The objective of this study is to assess knowledge about dengue fever prevention among people visiting Benazir Bhutto Hospital, Rawalpindi.Methods: The descriptive cross-sectional study of 6 months duration was conducted with 280 participants selected via nonprobability convenience sampling. After informed consent, an interview was conducted based on a questionnaire that assessed socio-demographic parameters and knowledge about dengue virus transmission and prevention. Data were analyzed through SPSS v. 22. The study was approved by the Ethical Review Board (ERB) of Rawalpindi Medical University and Allied hospitals.Results: Out of 280 respondents, 54.6% were males and 45.4% females and the mean age was 35.0 ± 13.1 years. The respondents having high knowledge scores were 66(23.6%) while those having moderate and low scores were 159 (56.8%) and 55 (19.6%) respectively. Educated respondents (p=0.03) and urban residents (p=0.05) had higher knowledge scores.Conclusion: The majority of the participants know about dengue fever. However, only one out of every four respondents has good knowledge scores for dengue fever prevention.
