CMS (cytoplasmic male sterile) line is one of the most important hybrid parents in hybrid rice production technology using Three-Line systems. Test-crosses for selection of candidate local maintainer lines were conducted using 24 rice varieties and two CMS-WA (wild abortive) lines IR78374A/B and 1A/B. The F1 of the two combinations, namely IR78374A/P519 and 1A/MR243 had less than 10% spikelet fertility (6.4 and 7.2%, respectively), indicated that P519 and MR243 are partially maintainers and could be used for the development of new locally adapted CMS lines. As a results, two CMS lines, namely 0025A/B and 0047A/B were generated after 5 to 6 successive backcrossing of IR78374A with P519 and 1A with MR243. The CMS 0025A/B and 0047A/B lines had maturation age which is comparable to other commonly grown inbred varieties. The percentage of stigma exertion rate for CMS 0025A/B and 0047A/B were 17.9 and 21.3%, respectively, lower than that of IR78374A/B (43.2%). Both showed poor out-crossing rate. CMS 0025A/B and 0047A/B are classified as long and slender grain. They had intermediate amylose content. Their gel consistency was hard, comparable to that of MR263 (medium).  CMS 0025A/B and 0047A/B lines were also moderately resistant to foliar blast disease and brown planthopper infestation.

Thirty-six rice genotypes were evaluated for thirteen different quality parameters along with yield/plant to assess genetic estimates of the traits and the extent of genetic diversity among the varieties. Analysis of variance was conducted to determine GCV, PCV, heritability and GA of the genotypes with respect to all characters. Significant variation was observed in many traits among the genotypes offering scope for selection. Correlation analysis determined the nature of relationship among these characters. UPGMA studies revealed six major clusters and cluster I and II were the largest with maximum number of genotypes. The study identified that the varieties namely, Black Gora, Kalinga-2, Dudheswar, ARC 10086, IR-36, IR-64 and Nipponbare possessed good quality traits and high yield performance. The current study indicated that developing rice varieties for consumer acceptance with good grain quality traits along with high yield will be very useful in rice breeding and in selection of parents for hybridization to combine both high yield and improved quality traits.

Since introduction of rice into Uganda in 1904, improvement of the irrigated and rain-fed lowland types was undertaken to address a number of production and quality constraints in three consecutive and overlapping phases. The initial phase was achieved through evaluation of introduction, selection of promising lines and subsequent release of the selected lines for production by the farmers. In the second phase, genetic potential of traits and characteristics of interest were analyzed and used to guide selection of suitable parents for hybridization and the third phase employed genotyping approach in screening and selection of the parental lines and the segregating populations to enhance the breeding efficiency for the traits of importance. Simultaneously, the key production constraints addressed included resistance to rice yellow mottle virus (RYMV), rice blast, bacterial leaf blight and narrow leaf spot diseases as well as submergence tolerance and cold tolerance. The quality traits considered for the improvement alongside the grain yield parameters were the grain aroma, amylose content, shape and size. These interventions have resulted into release and wide adoption of seven rice varieties in Uganda besides several breeding lines which have informally diffused into different major rice production agro-ecology. Subsequently, it can be concluded that a substantially strong and functional breeding platform for rice in Uganda has been established.

Selection of rice varieties and application of amendments are effective measures to ensure food safety. Here we report that in the non-Cd area, the grain quality of all rice varieties met the Chinese National Grain Safety Standards (CNGSS). In the high-Cd area, rice varieties showed significant different bioaccumulation of Cd with lower rice yields than those in non-Cd area with the average decrease of 31.1%. There was a negative correlation between grain Cd content and yields. A total of 19 rice varieties were selected as low Cd accumulating rice varieties and their Cd content met CNGSS in the low-Cd area. Six of them met CNGSS in the high-Cd area. The application of amendments significantly reduced Cd content in rice grains by 1.0–84.7% with an average of 52.6% and 13 of varieties met CNGSS. The amendments reduced available Cd content in soils by 1.1–75.8% but had no significant effects on rice yields. Therefore, the current study implied that proper agronomic management with selection of rice varieties and soil amendments was essential in controlling Cd accumulation in rice grains.

We have succeeded in selecting four higher yield mutants from five gamma-ray irradiated high-yielding Japanese rice varieties using a novel approach. A total of 464 M2 plants which had heavier total panicle weights per plant were first selected from 9801 irradiated M2 plants. Their higher yields were confirmed by yield trials conducted for three years with a six to ten-pairwise replicated plot design. FukuhibikiH6 and FukuhibikiH8 were selected from an irradiated high-yielding variety Fukuhibiki and showed 1.2% to 22.5% higher yield than their original significantly. YamadawaraH3 was selected from an irradiated high-yielding variety Yamadawara and its yield advantages were 2.7% to 3.9%. However, there was no difference in the genotypes of the 96 SNP (single nucleotide polymorphism) markers between the higher yield mutants and their respective original varieties. The differences in the measured phenotypical traits between each mutant and its original variety were not constant and the actual differences were marginal. Therefore, the higher yields of the selected mutants were likely to have been caused by physiological traits rather than phenotypical traits. The selection method used in this study is an application of the directed evolution method which has long been commonly used in the substantial improvements of microorganisms and their proteins.

The availability of molecular marker systems allowed estimating the relationships among various taxa. This study was aimed at assessing the genetic diversity among ten aromatic rice (Oryza sativa L.) pools from Bangladesh by means of randomly amplified polymorphic DNA (RAPD) markers. These varieties were evaluated for polymorphisms after amplification with 10 decamer primers. A total of 60 RAPD fragments were generated among the assessed varieties with a polymorphism percentage of 80. Cluster analysis by the unweighted pair group method of arithmetic means (UPGMA) showed that these 10 varieties could be placed into two groups with a similarity ranging from 65 to 86% depicting adjacent association between Rajbhog and Kalijira‐12, whereas Maloti belongs to a separate group maintaining maximum distance from rest of the varieties. The analysis revealed that the intervarietal genetic relationship of several varieties is related to their center of origin. As expected, most of the varieties have a narrow genetic base. The present results could be used for the selection of possible parents to generate a mapping population and utilized by the breeders for assessing the genetic diversity of rice genotypes.Plant Tissue Cult. & Biotech. 27(2): 217-225, 2017 (December)

Hygromycin is an efficient selective agent in transformation studies of wide ranges of crop. In this study, different concentrations of hygromycin were used to observe the effect on untransformed calli death, percent germination and seedling growth of three rice varieties (Oryza sativa L.) viz BRRI dhan29, MR219 and Taipei309. Hygromycin killed the untransformed calli and inhibited the germination of tested varieties in a concentration dependent manner. Among the tested varieties, the lowest and the highest calli death was observed in MR219 and Taipei309 respectively in all the concentrations of hygromycin. Whereas, the highest and the lowest percent germination were observed in MR219 and Taipei309 respectively. The minimal inhibitory concentration (MIC) for selection of calli were calculated as 42, 40 and 47 mg/L hygromycin for BRRI dhan29, MR219 and Taipei309 respectively. During germination, 35, 62 and 32 mg/L hygromycin were suitable for the selection of BRRI dhan29, MR219 and Taipei 309 respectively. Shoot and root growth reduction after germination was increased with the increased concentration of hygromycin. Besides, root growth was more sensitive to hygromycin than the shoot. These results suggest that hygromycin increases calli death, decreases percent germination, and shoot and root growth in all varieties with the increasing rate of hygromycin. But these characteristics vary with different degrees in different genotypes as well as different stages.Bangladesh Rice j. 2014, 18(1&2): 38-43
