Phenolic compounds such as kaempferol (1), quercetin (2), and methyl caffeate (3) have been isolated from the ethyl acetate extract of Kalanchoe prolifera (Crassulaceae). The Flavanone compound with anti diabetic activity was isolated from ethyl acetate extract of Artocarpus communis leaves using column chromatography techniques. The structure of the flavanone compound was elucidated on the basic of spectroscopic evidence and comparison to published values. This compound, 8-geranyl-4,5,7-trihydroxyflavone, showed strong anti diabetic activity on α-glucosidase inhibition assay with IC50 18.120 µg mL-1.   Keywords: Artocarpus communis, 8-geranyl-4,5,7-trihydroxyflavone, anti diabetic activity

The research characterization of cytotoxic fraction against P-388 leukemia murine cells from the extract honje (Etlingera elatior) seed have been reported. This research lead to isolated and characterization of cytotoxic compounds against P-388 leukemia murine cells from the extract E. elantior seed. The extract of E. elantior seed was maserated by methanol, n-hexane, and ethyl acetate, respectively and estimated their cytotoxic activity against P-388 leukemia murine cell with 3- (4, 5-dimetiltiazol-2-yl) -2,5-difeniltetrazolium bromide (MTT) assay guided toxicity test against of shrimp Artemia salina Leach. Brine shirmp Lethality Test (BSLT) method. The active extracts will be separated by fractionation using column chromatography, radial chromatography, and for analyzing the purity of isolate will estimate by HPLC. The chemical structure of pure isolate will be identified by spectroscopies data UV Vis, FTIR, NMR and MS. The ethyl acetate extract from honje seed have cytotoxic activity by leukemia P-388 cell  with IC50 19.21 µg/mL. The compound toxic as cytotoxicagainst P-388 leukemia murine cells is flavonoid compouds their is resveratrol, lapachol, apigenin, methylated chrysin, 6,2’-dihydroxyflavanone, 3-hydroxy-3,4’-dymethoxyflavone and 4’-hydroxy-5,7-dimethoxyflavanone.DOI: http://dx.doi.org/10.15408/jkv.v0i0.3640

Agricultural wastes cause a serious environmental problem in Egypt. Utilization of these wastes by an environmentally friendly way is a very important issue. The objective of this study was to utilize rice straw into high-value products. Antioxidant activity, total soluble phenols, and flavonoids were evaluated in ethyl acetate extract prepared from rice straw after alkaline hydrolysis. Total phenols were found to be 221.6 mg gallic acid equivalents (GAE), while total flavonoids were 4.9 mg rutin equivalents (RE). The extract exhibited strong antioxidant activity measured by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method and its IC50 value was 0.4 mg/ml. Furthermore, the ethyl acetate extract possessed high antioxidant activity assayed by β-carotene bleaching method and its value of lipid peroxidation inhibition (LPI) was 75.4%. It also exhibited high reducing power and its IC50 value was equal to 0.06 mg/ml. HPLC analysis indicated that this extract contained seven phenolic acids including; protocatechuic, caffeic, syringic, p-coumaric, ferulic, rosmarinic and cinnamic, in addition to two flavonoids; qurecetin and kaempferol. Results indicated that ferulic and p-coumaric acids were the major soluble phenolic acids in rice straw, and their concentrations were 3.9 and 2.9 mg/g DW. It can be summarized that the strong antioxidant activity of ethyl acetate extract, prepared from rice straw, was highly correlated with its high level of phenolic compounds.

Chisocheton balansae C.DC., is one of the Meliaceae family plants which is the endemic plants from Soputan Mountain, North Sulawesi, Indonesia. This study was aimed to determine the chemical structure of flavan-3-ol compounds from ethyl acetate extract of C. balansae C.DC stembark. Dried powder of C. balansae C.DC stem bark was extracted consecutively with n-hexane, ethyl acetate, and methanol solvents. Four flavan-3-ol compounds, named catechin (1), epicatechin (2), epigallocatechin-3-O-gallate (3), and epicatechin-3-O-gallate (4) were successfully isolated from ethyl acetate extract. The chemical structure of these isolates was determined by spectroscopic methods (1H-NMR, 13C-NMR, DEPT, and 2D-NMR) and comparison with previous reported spectral data. These compounds are first time reported from this plant.

Various species of Kalanchoe plant has been widely used for traditional medicine and also as an ornamental plant. This research is a continuing search for secondary metabolites from Kalanchoe plants in Indonesia. The fresh leaves of Kalanchoe pinnata (6 kg) was extracted at room temperature with methanol to obtain a concentrated extract. The concentrated extract of methanol was further partitioned successively with n-hexane and ethyl acetate. Yellow solid of pure isolates from ethyl acetate extract was separated by various chromatographic techniques. The chemical structure of isolates was determined by spectroscopic analysis of UV, IR , MS, 1H-NMR, 13C-NMR data and a comparison wih those previously reported on literature and identified as a flavonoid compound 3,3’,4’,5,7 pentahydroxyiflavone also known as kuersetin which belong to the flavonol class.

Water lily (Nymphaeaceae antares) contains a vast array of phenolic compounds with important natural antioxidative properties that could inhibit the adverse effects of the reactive oxygen species produced in living things. These substances are complex in composition which requires a suitable extraction medium to exploit the phenolic compounds. This study was designed to determine the antioxidative properties in petal and stamen of water lily extracted using different extraction solvents namely ethyl acetate and 60% methanol. Antioxidant properties of the samples were determined using 2,2-diphenyl -1-picrylhydrazyl (DPPH) radical scavenging activity, ferric thiocyanate (FTC) method, thiobarbituric acid (TBA) test, total phenolic content (TPC), total flavonoid content (TFC) and total tannin content (TTC) along with individual flavonoids content using High Performance Liquid Chromatography (HPLC). The yield of extract was found to be the highest in petals extracted with 60% methanol (44.55±1.63%) while stamen extracted with ethyl acetate yielded the lowest (24.50±1.09%). In the DPPH inhibition method, petal extracted with ethyl acetate obtained the highest scavenging activity (89.15±3.62%). FTC analysis revealed that ethyl acetate extract of petal experienced the highest percentage inhibition (76.70±6.48%) while in TBA analysis ethyl acetate extract of stamen exhibited the highest percentage inhibition (70.78±1.39%). Flavonoids such as quercetin, kaempferol, rutin, gallic acid, catechin, epigallocatechin, p-coumaric acid and myricetin were found to be present in ethyl acetate and 60% methanol extracts of water lilies stamen and petal. The information from this study indicates that water lily petals and stamens may be integrated as a good source of nutrients in the food industry, the pharmaceutical industry. If the advantages of water lily are not used and integrated into everyday life for health benefits, it will be a massive waste of natural resources.

Aim and Objective: Origanum acutidens (Hand.–Mazz.) Ietsw. is an endemic and perennial plant grown mainly in East Anatolia. Recently, natural plant products have attracted interest due to their safety and therapeutic effects. Therefore, the aim of this study was to investigate phytochemical contents and biological effects of Origanum acutidens. Materials and Methods: The aerial parts of O. acutidens were extracted with water, ethyl acetate, nbutanol, and methanol/chloroform solvents. Phenolic compounds and other constituents of the extracts were analyzed by HPLC/TOF-MS. The Ethyl Acetate extract (EA) was fractionated by flash chromatography. The extracts and fractions were investigated for their antiproliferative activities on human cervical adenocarcinoma (HeLa) cell line by using BrdU ELISA assay. Antioxidant activities of the extracts and fractions were evaluated by complementary test systems, namely determination of total phenolic contents, metal chelating ability and DPPH radical scavenging assay. Results: Among the extracts, Ethyl Acetate extract (EA) exhibited the highest antiproliferative activity (IC50 = 15.71 ± 0.04 µg/mL) on HeLa cells. It was therefore fractionated by flash chromatography to obtain 10 fractions which were investigated for their phenolic compounds and bioactivities. Rosmarinic acid was determined as the major component of EA and its fractions. EA exhibited higher antiproliferative activity against HeLa cell line than its fractions and 5-fluorouracil (5-FU) at the concentration of 100 µg/mL. EA and its fractions (F10, F6, F4, F7, F3, and F2) displayed higher radical scavenging activity compared to Butylated Hydroxytoluene (BHT). These effects may be attributed to the presence of rosmarinic acid in EA and its active fractions. Conclusion: This study demonstrated that O. acutidens is an essential natural source of polyphenols and a potent natural antioxidant and antiproliferative agent for food and pharmaceutical industries.

Chemical composition of a phenolic complex of the Siberian spruce bark (Picea obovata Ledeb.) and a structure their oligomeric phenolic compounds are studied.The samples of the spruce bark have been collected in the vicinity of Fr. Baikal in September 2015. Extractive substances were recovered from the crushed bark by extraction with ethyl acetate at the boiling point of the solvent for 4 hours. The resinous substances were extracted from the obtained ethyl acetate extracts by hexane by the infusion method for 3 days. The bark samples, after extraction with ethyl acetate, were dried to remove solvent residues, then the water-soluble substances were extracted by extraction with distilled water at 90 °C for 4 hours.The yields of the extractive substances extracted with ethyl acetate and water from the spruce bark were 3,4 and 6,9% in absolutely dry bark (a.d.b.), respectively. The content of phenolic compounds in the ethyl acetate extract of the spruce bark have been 42,4% relative. The ethyl acetate extract of a spruce bark, pretreated with hexane, was fractionated on a silica gel column using mixture chloroform-acetone as the eluent with an increase in the fraction of the latter (from 0 to 100%). The content of monomeric and oligo-, polymeric phenolic compounds in the extract was determined.Based on the data of 1Н and 13C NMR spectroscopy, it was established that the flavonoid dihydroquercetin and stilbene glycosides – isorhapontin and astringin are the main monomeric phenolic extractive substances of Siberian spruce bark. It is shown that the structure of oligomeric phenolic compounds of the spruce bark includes structural fragments of stilbene glycosides, mainly monomeric blocks with the structure of isorhapontin and astringin.

In our course of screening for secondary metabolite derived from marine bacterial, we isolate the antimicrobial compound collysmicin A from the ethyl acetate extract of Streptomyces sp Q-629K. Separation of this compound was carried out by silica gel open column chromatography. Purification of an active compound was done using HPLC C18 with acetonitril-water system. Determination of chemical structure was done by 1H, 13C NMR and LC-MS analysis. Collysmicin A was contained in fraction 3, fraction 7.2 and fraction 8.7. The antimicrobial assayed from purified compound Fr.8.7 gave diameter inhibition approximately 13 mm against S. aureus and 12 mm against B. subtilis .   Keywords: antimicrobial, collismycin A and marine Streptomyces sp

There is no research on the chemical constituents investigation and bioactivity evaluation of Buchanania lucida Blume (Anacardiaceae). From the stems of Buchanania lucida collected at Dong Nai province, the ethyl acetate and n-butanol extracts were prepared. Four phenolic compounds: protocatechuic methyl ester (1), aloe-emodin (2), naringenin (3), and glucosyringic acid (4) were isolated from these extracts. The chemical structure of these compounds were elucidated by 1D and 2D NMR spectra and comparison with published data. These compounds were first reported in Buchanania lucida.
