A comparison of cytoplasmic and chemically-induced male sterility systems for hybrid performance in wheat ( Triticum aestivum L.)

Cytoplasmic ( Triticum timopheevii -based) and chemically induced (CH9832- based) male sterility systems were compared for hybrid performance in wheat. A total of 40 genotypes including 10 CMS-based hybrids, 10 CHA-based hybrids, 10 B lines and 10 R lines were included in the experiment. analysis of variance revealed significant differences between the genotypes for all the characters studied. Differences between the hybrids and their parents were significant for all the characters. There were also significant differences between the hybrids resulting from the two systems of sterility. This study of 10 comparable hybrids showed, on average, midparent heterosis of 30.2 and 7.3% for CMS- and CHA-derived hybrids, respectively. Generally, the CMS-based hybrids were superior to their CHA-based equivalents for grain yield performance. In spite of the incomplete fertility the higher grain yield in CMS-based hybrids was attributed to the profuse tillering and high thousand-grain weight. On the other hand, the CHA-based hybrids had lower yield performance due to the lower thousand-grain weight and tillering. The lower grain weight and tillering in these hybrids compared with their CMS-based equivalents might be due to the toxic effect of the CHA, which was carried over to the hybrid and affected vigour. As a whole the CMS system was found to be better than the CHA system (based on the particular CHA used in this study) for hybrid performance in wheat.

Mutagenesis is one of the powerful genetic strategies for crop improvement programmes. A chemically induced mutated genetic resource for detecting novel variations by Targeting Induced Local Lesions IN Genomes (TILLING) has been developed in recently released bread wheat (Triticum aestivum) cultivar DPW621-50. A total of 3,478 M2 plants were evaluated for plant height, number of tillers/plant, thousand grain weight, number of seeds/spike and grain yield/plant. A large variation was observed for all the traits. The highest frequency (52.2%) of lines had similar height between 91-100 cm to the non-mutagenized DPW 621-50 control followed by 28.9% of lines with height between 81-90 cm. A large variation was observed in number of tillers/plant which ranged from 1-35 tillers/plant. The highest frequency (32.09%) lines had 31-40 seeds/spike followed by 29.84% lines with 41-50 seeds/spike. Few lines (0.35%) had more than 70 seeds/spike with normal seed size as their thousand grain weight ranged between 34.82-43.82g. Chlorophyll deficient, grassy type and sterile mutants were also observed. This population may serve as new genetic resource for functional genomics studies and novel variants for different traits in elite germplasm can be made available to the plant breeders for wheat improvement.

The experiments were laid out to understand the mechanisms causing yield limitations imposed by post-anthesis water and nitrogen deficiencies in plants with modified source-sink ratios. Two soil-water regimes were allotted to the main plots. At anthesis, three levels of N were applied: none, 25% and 50% of total the N supply. Spike-halving caused reduction in grain yield at both water regimes and all N supply levels, showing that the reduction in grain number can not be compensated by a higher individual grain weight. Sink reduction by trimming 50% of the spikelets reduced grain number per ear by 38.5% and increased individual grain weight by 12.0%, which shows the plasticity in grain weight and grain set of wheat if sufficient assimilates are available. Additional nitrogen supply at anthesis had no significant effect on the total aboveground biomass, but increased grain yield through more allocation of dry matter to grains. Our findings suggest that for rainfed wheat with optimum N supply and supplemental irrigation, wheat growers should choose cultivars with a high grain number per ear and manage the crop to increase grain number per unit of land (sink capacity).

Spring wheat (Triticum aestivum L.) varieties Gaurab, Kanchan, Balaka, Sonora, Protiva, Pavon, and Anza were used as parent materials to estimate heterosis in a set of 7 × 7 diallel crosses. The diallel trial was carried out for seven parental material and their 21 F2 progenies under two contrasting cultural conditions for different yield and yield contributing characters. Cultural conditions I is provided by the BARI recommended doses of fertilizer and irrigation, and 2 have no fertilizer but two irrigations once at crown root initiation stage and twice at panicle initiation stage. Heterosis was measured as i) Relative heterosis and ii) Heterobeltiosis. The result of relative heterosis revealed cross Sonora × Anza exhibited superior performance for grain yield/plot in environment-i. Desirable negative heterosis was observed in cross Balaka × Anza in environment-I and Pavon x Anza in environment-2 for days to 50% heading character. For days to maturity, desirable negative heterosis was found in cross Pavon x Anza in both cultural environments. Estimate of heterobeltiosis for different yield contributing characters showed that cross Sonora x Anza exhibited highest heterosis for grain yield/plant in environment-1 and Kanchan x Balaka in environment-2. Cross Pavon x Anza exhibited superior relative heterosis and heterobeltiosis for 100-grain weight in both cultural environments. Analysis of genetic diversity and variability is an initial step for crop development in a plant breeding program. Thirty foxtail millet genotypes were evaluated for their ten agro-morphological traits viz. grain yield, days to heading, days to maturity, plant height, leaf length, leaf width, number of nodes per tiller, panicle length, thousand-grain weight, and leaf blast during April to August season of 2018 at Kabre, Dolakha, Nepal. Clustering was done based on variability observed among the genotypes for all the ten traits by the average linkage method. The genotypes were grouped into six clusters. The shortest inter cluster distance between Cluster I and Cluster II was 17.63 and the longest was between Cluster IV and Cluster VI with 47.55. Cluster I had the maximum leaf blast score with the minimum leaf length. The Cluster II had the maximum grain yield and days to 50% heading and the number of nodes per tiller. The Cluster III was characterized by the maximum leaf length and leaf blast value. The genotypes grouped into the Cluster IV were characterized by the lowest value of days to 50% heading and 80% days to maturity and those of Cluster V had the maximum thousand-grain weight. The genotypes categorized into Cluster VI had the maximum plant height, leaf length, and panicle length. Cluster II and Cluster V were good because of their agro-morphological traits. The presence of a high level of diversity among the genotypes indicated their suitability for selection in crop breeding programs. Correlation analysis of genotypes having the maximum panicle length leaf area and higher plant height and shorter maturity period are pre-requisite for improvement in grain yield.

Eleven genotypes of bread wheat were crossed with three testers in the rabi season of 2018-19 inline x tester mating fashion to obtain 33 F1 hybrids, which were evaluated with parents and two standard checks of wheat viz., UP 2855 and HD 2967. The experiment was laid out in randomized block design with three replications at Norman E. Borlaug Crop Research Centre, G.B. Pant University of Agriculture and Technology, Pantnagar in the rabi season 2019-20. Observations were recorded on various qualitative and quantitative characters including grain yield. The results obtained revealed significant differences among all the genotypes, indicating the presence of aconsiderable amount of genetic variability. The cross combinations SOKOLL x HI 1621, followed by QBP 12-11 x HI 1621, NAC/TH.AC//3*PVN/3/MIRLO/BUC/4/ 2*PASTOR/5/… x HD 3237 were observed to be the better heterotic F1s in terms of grain yield. The hybrid QLD 75 x HI 1621 was identified as the best heterotic combination for grain weight per spike and harvest index. Whereas, for grains per spike F1, NW 6036 x HI 1621, for 1000 grain weight F1, SOKOLL x HD3237, for spike length F1, NAC/TH.AC//3*PVN/3/ MIRLO/BUC/4/2*PASTOR/5/… x HD 3237, for peduncle length F1, VORB/4/D67.2/PARANA 66.270 x HI 1621 and for days to 75% heading and days to maturity F1, PRL/2*PASTOR*2//FH6-1-7/3/KINGBIRD#1//… x PBW 725 were observed to be the better heterotic combinations. The crosses with higher heterotic values may provide an opportunity for isolation of desirable purelines in advanced generations.

Effect of drought stress on cold season wheat varieties investigated at Jolgeh-rokh station of Torbat Heydarieh agricultural and resources research center during 2012 and 2013. The experiment contained 18 inbred lines and promised varieties studied in optimal and limited irrigation levels. Two separate complete randomized block designs with three replications were carried out. At limited irrigation level plants did not irrigated at dough stage and physiological maturity stage. Measured treats was: plant height, spike length, peduncle length, grains/spike, grain weight, thousand grain weight, spike weight, harvest index, grain and biological yield.  Results showed that Cold wheat 14 was the best variety in both irrigation levels. Grain yield significantly correlated with grain yield and biological yield at optimal condition. There was a positive correlation between biological yield peduncle weight and harvest index with grain yield at drought condition. Stepwise regression analysis showed that yield variation control by thousand grain weight, grains/spike and spike weight at optimal irrigation condition. Peduncle length was the only variable enters to regression model at drought condition. Peduncle length and weight had the highest indirect effect on yield as shown by path analysis. Factor analysis indicated that three factors accounted for about 80 percent of the total variation among characters. Investigating the drought susceptibility indices showed that mean productivity index and stress susceptibility index was the best in selecting tolerate varieties.

SummaryThree spring barley varieties, Proctor, Vada and Mosanne, were inoculated with Rhynchosporium secalis(Oudem.) J. J. Davies at Growth Stages 3, 7, 10·1 and 10·5 (Large, 1954). The treatments included single inoculation at each growth stage and multiple inoculations at two, three and four of the above growth stages. There was no effect of inoculation oil the number of fertile tillers. Thousand-grain weight was reduced in all seven inoculation treatments in Mosanne but only the quadruple inoculation adversely affected the other two varieties. Numbers of grains of both Mosanne and Vada were reduced by treatments which included an inoculation at ear emergence but Proctor was affected only by the quadruple inoculation. In terms of disease assessment Mosanne was very susceptible and Proctor moderately resistant but Vada, with symptom scores only slightly better than Mosanne, exhibited a degree of tolerance which was reflected by the grain weight results in which small increases were recorded. This tolerance mechanism, possibly acting through compensation in this variety, is compared with compensation reported in wheat.

An experiment on combining ability was carried out with 21 crosses produced from 7×7 diallel cross without reciprocal for grain yield and yield contributing characters in maize. Analysis of variance for combining ability showed that mean square (MS) due to GCA & SCA were highly significant for all characters except GCA in plant height, cob length and 1000 grain weight and SCA in maturity and row/cob indicated that all but mentioned traits were governed by both additive and non-additive gene action. Variances due to GCA were higher for all characters except thousand grain weight revealed that the predominance of additive gene action for all characters except thousand grain weight. Parent CML 487 and Ki 21 were the best general combiner for yield and most of the yield contributing characters. Parent BMZ 57 & BMZ 15 were the best general combiner for dwarf & earliness in plant. Among all the crosses CML 473 × Ki 21, CML 487 × Ki 21 and CML 429 × BIL 182 exhibited significant positive SCA effect for grain yield. The cross CML 429 × BIL 182 may be considered as the best cross with recorded significant mean value and desired SCA for traits like 1000 grain weight, yield (t/ha), days to 50% pollen shedding, days to 50% silking, plant height, ear height and days to maturity. Four year trials were undertaken to study the milling and baking rye quality, the hybrid variety Picasso and population varieties Dankowskie nowe and Selgo having been examined coming from three different locations of the Czech Republic. The variety significantly (<i>P</i> < 0.01) influenced the specific weight, grain size, amylograph maximum, and grain yield. The year of harvest significantly (<i>P</i> < 0.01) influenced the thousand grain weight, maltose content, protein content, amylograph maximum, and grain yield. The location significantly (<i>P</i> < 0.01) influenced the thousand grain weight, protein content, amylograph maximum, and grain yield. The highest pentosan content (average of the four years and three locations) was achieved by the hybrid variety Picasso (8.04%), which had the highest Falling number (232 s) as well as amylograph maximum (597 AU). The location Hradec nad Svitavou proved to be the best (8.02% pentosans) while the year 2005 (8.34% pentosans) was the most positive. A positive correlation (<i>P</i> < 0.05) was found between the pentosan content and the Falling number.

Chemically induced male sterility (CIMS) systems in wheat are among the male sterility types used for hybrid wheat (Triticum aestivum L.) production in China. Some studies suggested that male sterile line Xi'nong 1376-CIMS induced by chemical hybridizing agents (CHA) may suffer from oxidative stress as its cyanide-resistant respiration is lower than that of Xi'nong1376. To elucidate the metabolic mechanism of reactive oxygen species (ROS) in the CIMS anthers, the metabolism changes in the production and scavenging of ROS and gene expression related to ROS-scavenging enzymes were investigated in the anther of Xi'nong 1376-CIMS and Xi'nong1376.Anthers of Xi'nong 1376-CIMS had higher contents of [Formula: see text] and H2O2 than those of 1376, which corresponds to expression level of the NADPH oxidase (NOX) gene, and has higher contents of malondialdehyde compared with 1376. Simultaneously, there were lower activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascrodate peroxidase (APX) in scavenging ROS in the anthers of the Xi'nong 1376-CIMS line than in Xi'nong1376. Meanwhile, the expressions of SOD, POD, CAT and APX genes in 1376 were always higher at different levels than those in the Xi'nong 1376-CIMS line except for POD in stage 1. Therefore, it is possible that the sterility in Xi'nong 1376-CIMS is related to the abortion of microspores induced by chronic oxidative stress caused by an abnormal increase in ROS.
