INCREASED SEBUM SECRETION IN ADULT FEMALE RATS AFTER NEONATAL TREATMENT WITH TESTOSTERONE PROPIONATE

SUMMARY Testosterone propionate (TP) was administered to female rats at 3 and 6 days of age. When adult the sebum secretion rate of these rats was significantly increased, although the response to TP was normal. Neonatal androgen treatment had no effect on the preputial gland weight of adult female rats. On the other hand ovarian weight was reduced. After ovariectomy of normal adult rats there was an increase in sebum secretion to a level comparable to that found in adult rats given TP as neonates. By contrast there was no change in sebum secretion after ovariectomy of adult rats given TP as neonates and the rate of sebum secretion in these rats was comparable to that in ovariectomized normal adults. It is suggested that after neonatal androgen treatment the normal inhibition of the sebaceous glands by the ovaries was abolished and consequently the rate of secretion was increased. Thus the endocrine state in early life may have a lasting effect on sebaceous gland activity in the adult.

SUMMARY The effect of varying doses of testosterone propionate (TP) on the sebum secretion and preputial gland weight of castrated and hypophysectomized—castrated rats was studied. The changes in preputial gland weight closely paralleled those of sebum secretion. Small doses of TP (0·05 mg) stimulated sebum secretion in hypophysectomized-castrated rats, but not in the castrated rats. Higher doses of TP increased sebum secretion in both the hypophysectomized—castrates and the castrated rats. While 0·5 mg TP completely restored sebum secretion to normal levels in castrated rats but not in the hypophysectomized—castrated rats, the actual increase in sebum secretion produced by this dose of TP was similar in both groups. It is concluded that at physiological doses the action of TP on the sebaceous glands is independent of the pituitary. However, the presence of the pituitary is required for normal sebaceous gland activity.

SUMMARY Adult male rats which had been castrated prepuberally secreted less sebum than intact rats. When hypophysectomy followed castration there was a further decrease in sebum secretion. Treatment of the castrated and hypophysectomized castrated rats with testosterone propionate produced a large and comparable increase in the sebum secretion of both groups. Testosterone propionate also caused a marked increase in the preputial gland weight of the castrated rats, although a much smaller response occurred after hypophysectomy. The effect of hypophysectomy on the response of the sebaceous and preputial glands to testosterone is discussed.

SUMMARY The effects of α-MSH and testosterone propionate on sebum secretion, sebaceous gland volume, dermal lipogenesis, and preputial gland weight and lipogenesis were examined in hypophysectomized rats. Hypophysectomy reduced sebum secretion, sebaceous and preputial gland size, and dermal and preputial gland lipogenesis. The greatest effects were seen on the biosynthesis of wax esters and squalene. Testosterone propionate (TP) increased sebum secretion, sebaceous gland volume and preputial gland weight and lipogenic activity, but had no significant effect on the pattern of lipid labelling. α-MSH had no effect on sebaceous or preputial gland size, but increased sebum secretion and dermal lipogenesis, especially wax ester biosynthesis. When given together TP and α-MSH had a synergistic effect on sebum secretion and on dermal and preputial gland lipogenesis, and the pattern of lipid labelling was shifted towards normal. TP and α-MSH also showed synergism in increasing preputial gland weight, but together they had no greater effect on sebaceous gland volume than that achieved with TP alone. These results suggest that TP and α-MSH have different actions on the sebaceous glands with α-MSH acting predominantly on lipogenesis and TP on cellular proliferation and turnover leading to an increase in gland size. Preputial glands differ from cutaneous sebaceous glands in their response to α-MSH and androgen which could be a reflection of their more specialized function.

ABSTRACT The influence of early androgen treatment, light deprivation (by blinding), pinealectomy and superior cervical ganglionectomy on the reproductive system of female rats was tested. Early postnatal treatment of rats with testosterone propionate caused adult rats to exhibit the characteristic signs of androgen sterilization; these included polyfollicular ovaries, normal-sized uteri and persistent vaginal cornification. If early androgentreated rats also were blinded the ovaries were smaller in size and contained fewer follicles, the uteri were greatly reduced in size and the incidence of vaginal oestrus was decreased by approximately 50% If in addition to blinding, androgen-sterilized animals were subjected to either removal of the pineal gland or superior cervical ganglia, the reproductive organs and the vaginal smears were indistinguishable from those of testosterone-treated rats with eyes. These data indicate that the inhibitory influence of blinding on the pituitary-ovarian axis was mediated through the sympathetic nervous system and the pineal gland. The restraining influence of light deprivation on the growth of the reproductive organs was not permanent as illustrated by the fact that if these animals were kept to 120 days of age the ovaries and uteri grew to the same level as those of pinealectomized control rats.

SUMMARY An anti-androgenic steroid, 17α-methyl-B-nortestosterone was shown to reduce sebum production in spayed female rats treated simultaneously with testosterone. The sebum production was assessed by measuring the increase in the amount of ether-extractable hair fat over a period of 8 days immediately after washing the rats. Testosterone increased the secretion of sebum mainly by stimulating mitosis in the sebaceous glands and it was shown that the anti-androgenic steroid acted at this site, i.e. by suppressing mitosis. These effects were in contrast to the action of oestradiol which also depressed sebaceous activity, but appeared to do so by interfering with synthesis in the sebaceous cells.

ABSTRACT In vitro experiments carried out with uterus preparations of ovariectomized adult rats indicate the presence in this tissue of a 20β-hydroxysteroid-oxidoreductase which catalyzes the conversion of 20β-hydroxy-4-pregnen-3-one to progesterone. Since a hepatic 20β-hydroxysteroid-oxidoreductase is absent in adult female rats, the myometrial enzyme can be responsible for the biological activity of 20β-hydroxy-4-pregnen-3-one in these animals. Besides progesterone five metabolites were isolated and identified after incubation of [4-14C]20β-hydroxy-4-pregnen-3-one with uterine tissue: 20β-hydroxy-5α-pregnan-3-one, 20β-hydroxy-5β-pregnan-3-one, 5α-pregnane-3α,20β-diol, 4-pregnene-3α,20β-diol and 4-pregnene-3β,20β-diol. The conversion of 20β-hydroxy-4-pregnen-3-one to progesterone permits us to regard all five steroids isolated as progesterone metabolites in the rat uterus. 20β-hydroxy-5β-pregnan-3-one is the first C21-metabolite with a 5β(H)-configuration isolated in the rat uterus, which indicates the presence of 5β-reductase in this tissue.

Background: Puberty is a developmental transition in which an estrogenic surge occurs, mediating the release of xenoestrogens, like aluminium. Aluminium’s effect on anxiety in rodents at the different developmental stages is inconsistent. Aims: This study aimed at investigating the effect of the metalloestrogenic property of aluminium on anxiety-like behavioral changes in prepubertal and young adult female rats. Objective: Considering this aim, our objective was to evaluate the anxiety-like behavior by the elevated plus maze in prepubertal and young adult female rats with or without acute exposure to aluminium. Methods: To address this property of aluminium, 5mg/Kg body weight (Al-5) and 10 mg/Kg body weight (Al-10) of aluminium was administered intraperitoneally to female rats at two developmental stages, prepubertal (PP; n = 8 for each dose) and young adult (YA; n = 6 for each dose) for two weeks. Post-treatment, three days behavioral assessment of the rats was done employing elevated plus maze. Results: Reduced escape latency was seen in Al-5, Al-10 pre-pubertal rats, and Al-5 young-adult rats on day 3. A significant reduction in open arm time was seen in the Al-5 young-adult rats. Aluminium treatment in the pre-pubertal rats reduced their head dipping and grooming. Reduced sniffing, head dipping, and stretch-attended posture in the treated young-adult female rats showed that they had impaired risk-taking tendency. Conclusion: Differential effect on the anxiety-like behavior in the pre-pubertal and young-adult female rats might be due to the metalloestrogenic property of aluminium, acting differently on the two age groups.

SUMMARY Adult rats of both sexes were either gonadectomized or hypophysectomized and gonadectomized. Three to eight weeks later they were treated for 14 consecutive days with oil or with 75 or 200 μg testosterone propionate (TP) per 100 g body weight. The animals were killed and for each sex the gonadectomized animals were compared with the hypophysectomized-gonadectomized animals as far as their NADPH- and NADH-dependent 3α-hydroxysteroid dehydrogenases (3α-HSD) in renal microsomes, transcortin levels in serum and five organ weights relative to total body weight were concerned. For two of the latter, i.e. the relative kidney and prostatic weights, no significant differences were found. Transcortin levels, relative adrenal weights and renal NADPH-dependent 3α-HSD activities were higher in oil-treated gonadectomized animals than in oil-treated hypophysectomized-gonadectomized animals. The opposite was found for the relative weights of uterus and seminal vesicles and renal NADH-dependent 3α-HSD activities. These differences between gonadectomized and hypophysectomized-gonadectomized animals disappeared after TP treatment as far as transcortin levels were concerned but remained for the five other parameters. After gonadectomy sexual differences subsisted for all parameters studied. But whereas intact male rats had higher NADH-dependent 3α-HSD activities than female rats the opposite was found after gonadectomy. After gonadectomy plus hypophysectomy the between sex differences disappeared as far as transcortin levels were concerned but remained in the other parameters studied.

The recovery time course of muscle atrophied by immobilization was followed after removal of hindlimb casts from adult female rats. Increases of only 9% in body weight, 4% in gastrocnemius weight, and 10% in soleus weight occurred in controls during the 78-day duration of the experiment. There were no increases in the amounts of total protein or of citrate synthase activities in gastrocnemius or soleus during the first 3 days after removal of hindlimb casts; thereafter, there were increases in these paramters. Citrate synthase activities per mg of gastrocnemius protein were significantly higher at the 16th and 50th day of recovery. No significant differences for citrate synthase activity per mg of soleus occurred during recovery. Until the 50th day of recovery, no significant differences for total protein in soleus and for total protein and wet weight of gastrocnemius were observed between control and recovery values. However, the wet weight of the soleus returned rapidly during recovery and was not significantly different from control during recovery.

One-year-old adult female rats were fasted for 12 or 36 h followed by a 30-min infusion of insulin. The responses of the fractional rate of protein synthesis (Ks) in the individual muscles (measured in vivo) to fasting were small and mostly nonsignificant. After 12 h of fasting, only the epitrochlearis muscle (ET) showed a significant decrease in Ks, and, even after 36 h of fasting, a significant decrease in Ks was seen in only ET, extensor digitorum longus, and tensor fasciae latae (TFL). After the 36-h fast, infusion of insulin restored the fed Ks in all muscles except TFL. The fiber-type composition of the individual muscles appeared to influence the muscles' responsiveness to the fasting, since the highly glycolytic TFL was the most sensitive (particularly after 36 h of fasting), whereas the highly oxidative adductor longus and soleus muscles were unaffected by either fasting or insulin. In a second experiment, refeeding of fasted adult rats also had little effect on Ks, consistent with the low sensitivity to fasting shown by the first experiment. The parallel results in the two experiments confirmed that the low responsiveness to fasting and insulin infusion in these adult rats was not a result of failure to absorb in “fed” animals or insufficient levels of insulin during insulin infusions. In contrast, a third experiment showed that muscle protein synthesis in the gastrocnemius muscle from young adult (5-mo-old) female rats was significantly reduced after only 12 h of fasting.
