Lactogenic actions of different growth hormone preparations in pregnant and lactating rats

Abstract We studied the capacity of different GH preparations, natural human (h)GH, recombinant hGH (rhGH), rat (r)GH, ovine (o)GH, bovine (b)GH and porcine (p)GH, and ovine prolactin (oPRL), to stimulate lactogenesis in ovario-hysterectomized pregnant rats or intact lactating rats treated with bromocriptine (BC). Ovariohysterectomy (OVX-HYS) performed at 0800 h on day 19 of pregnancy induced lactogenesis, i.e. increases in mammary casein and lactose and positive response to the oxytocin test, 28 h later. Lactogenesis was prevented by treatment with BC (1·5 mg/kg) immediately after surgery (OVX-HYS-BC). The hormones were given at doses of 0·25 or 0·5 mg/rat (except rhGH given only at 0·5 mg/rat) at 1200 and 2000 h on day 19. Casein was increased by both doses of oPRL and hGH, rhGH and 0·25 mg oGH, and lactose by both doses of oPRL, rhGH and 0·25 mg rGH. The other GH preparations had no effect. The oxytocin test demonstrated the presence of milk in the mammary tissues of the OVX-HYS rats and in the OVX-HYS-BC plus oPRL (0·25 and 0·5 mg) or rhGH-treated groups. Injection of BC to pregnant rats at 2000 h on day 20 and at 0800 h on day 21 decreased litter growth on the first 4 days postpartum. Two-thirds of the litters resumed growth after day 4, indicating the recuperation of milk production, while the rest never recuperated. Serum prolactin in BC-treated rats was reduced until day 4 postpartum. On day 6 the rats which had recuperated had normal values, while those which had still not recuperated had lower values. BC-treated rats were injected s.c. with 0·25 mg each of oPRL, hGH, rGH, oGH, bGH or pGH, or 0·25 or 0·5 mg rhGH/rat, immediately postpartum and 12, 24 and 36 h later. hGH and 0·5 mg rhGH induced levels of milk production similar to controls except on day 3. oPRL and rhGH (0·25 mg), induced a partial reversion of the effect of BC. rGH and oGH had a slight effect on days 1 and 2 and all the litters resumed growth on day 7. In contrast, pGH and bGH were inactive. The affinity of hGH for the prolactin receptor, measured as displacement of 125I-labelled oPRL binding to crude liver membranes, was comparable with that of oPRL. While rhGH was ten times less active than oPRL, rPRL was 100 times lower and all the other GH preparations had at least 104 times lower capacity to displace 125I-labelled oPRL. These results indicate that both natural and recombinant hGHs are potent inductors of milk synthesis in pregnant or lactating rats, most probably due to their actions at the level of the prolactin receptor. rGH and oGH have a partial action, while pGH and bGH seem to be inactive. The actions of non-human GHs may be explained by their somatogenic properties exclusively, and indicate that GH may play a role in the optimization of milk production during lactation and an accessory role in the induction of lactogenesis in pregnant rats. ABSTRACT The changes in oestrogen, progesterone and prolactin receptor levels in target organs, and the macroscopic and microscopic modifications of uterus, ovary, adrenal and pituitary gland induced by long-term administration of high doses of medroxyprogesterone acetate (MPA) were investigated in female rats. Medroxyprogesterone acetate was injected i.m. for 30 days at daily doses of 7·5, 15 and 75 mg/kg. Oestrogen and/or progesterone-binding capacities were remarkably reduced at all doses of MPA used both in the uterus and pituitary gland. Furthermore, MPA caused a very evident reduction in the weight of pituitary glands, ovaries, adrenals and uterus. In all MPA-treated rats corpora lutea were absent from the ovaries, whereas the adrenals showed a significant reduction in the thickness of the cortex. In accordance with this, there was no evidence of ACTH-producing cells in the pituitary glands. Prolactin-producing cells were also absent, while GH-producing cells were present. Serum prolactin levels were significantly reduced at all doses of MPA used. A dramatic reduction of prolactin receptor concentrations was observed in the liver and the ovaries of MPA-treated rats. The results suggest that MPA acts as an antioestrogenic drug both by reducing the number of oestrogen receptors in target tissues and by changing the structure (and perhaps the function) of those organs (pituitary glands, ovaries and adrenals) which are, directly or indirectly, a source of oestrogens. The decreased synthesis of prolactin and the reduction of the number of prolactin receptors (which, on the contrary, are both increased by oestrogens) might be considered as additional antioestrogenic effects of MPA. PURPOSE: Determine the effects of the MSG (monosodium glutamate) in the offspring of pregnant rats through the comparison of the weight, NAL (nasal-anal length) and IL (Index of Lee) at birth and with 21 days of life. METHODS: Pregnant Wistar rats and their offspring were divided into 3 groups: GC, G10 and G20. Each of the groups received 0%, 10% and 20% of MSG, respectively from coupling until the end of the weaning period. RESULTS: Neither weight nor NAL were different among the groups at birth. The group G20 at birth had an IL lower than the group GC (p<0,05) and with 21 days of life presented weight and NAL lower than the groups G10 and this lower than the GC (p<0,01). Otherwise the G20 at 21 days of life had the IL similar to the other two groups. The weight profit percentage from birth to the 21st day of life was lower in the G20 regarding the other two groups (p<0,01). The G20 had a NAL increase percentage from birth to the 21st day of life lower than the G10 and this lower than the GC (p<0,01). CONCLUSIONS: MSG presented a dose-dependent relation in the variables weight and NAL. It caused a decrease in the growth pattern as well as in the weight gain pattern until the 21st day of life. The IL of the group 20% had an increased in relation to the control group after 3 weeks of follow up.

The present research objective was to evaluate the performance and blood parameters of lactating cows fed with a diet of two tannin sources, based on sorghum (condensed tannin)  with increasing concentrations of tannic acid (hydrolyzable tannin’s). Increasing levels of tannic acid in a sorghum-based diet for five Holstein/Zebu crossbred lactating cows were subject to a 5 × 5 Latin square experimental design. To assess the effect on cow intake, digestibility, milk production, and blood parameters. All cows received 9.87 kg/DM of corn silage as roughage and 6.38 kg of concentrate consisting of 2.58 kg/DM of ground sorghum 0.87 kg/DM of cornmeal 1.32 kg/DM of soybean bran 0.44 kg/DM of wheat bran 0.2 kg/ DM of urea and 0.18 kg/ DM of mineral mixture. Diet 1 (control) contained low-tannin sorghum and the other diets contained high-tannin sorghum. The levels of tannic acid added to the diets were established based on the quantity of condensed tannin in high-tannin sorghum. Thus, diets 2, 3, 4 and 5 were supplemented with 1.5g (13.%DM), 79.5g (2.6%DM), 157.5g (3.9%DM) and 235.5g (5.2%DM) of tannic acid, totalling 0.078, 0.156, 0.234 and 0.321 total tannin kg/day respectively, to assess the effect on microbial protein synthesis. The diets contained 35 kg of corn silage (roughage) and 6.40 kg of concentrate. The natural and dry matter intake did not significantly differ between groups, except for ether extract (EE) intake that significantly differed. The DM apparent digestibility, crude protein, EE, neutral detergent fiber, total carbohydrate, and non-fibrous carbohydrate did not differ (P > 0.05) with the increase in dietary tannin supplementation. The GOT levels increased linearly. The blood glucose, triglyceride, and cholesterol (total, LDL and HDL) levels did not significantly differ. Hemoglobin showed a significant difference. No significant difference in urea, creatinine and uric acid occurred. Diets using two tannins sources supplementation caused no decrease in the dietary intake or digestibility in the animals. The GOT level changed significantly, showing linear behavior, however below the toxicity level, without any change in the other blood parameters. Milk production decreased with the increase in dietary tannin supplementation.

The objective of the present study was to evaluate the effect of the grade of crossbreeding (Lacaune x Manchega) and environmental factors on milk production in a commercial flock in Spain. A total of 5769 milk production records of sheep with different degrees of purity of the Lacaune breed crossed with Manchega were used as follows: 100% Lacaune (n = 2960), 7/8 Lacaune (n = 502), 13/16 Lacaune (n = 306), 3/4 (n = 1288), 5/8 Lacaune (n = 441) and 1/2 Lacaune: Manchega (n = 272). Additional available information included the number of parity (1 to 8), litter size (single or multiple), and the season of the year of lambing (spring, summer, autumn and winter). A mixed model was used to evaluate the level of crossbreeding and environmental factors on milk production. The 100% Lacaune sheep presented the highest milk production with respect to the F1 Lacaune x Manchega sheep (p < 0.01), showing that as the degree of gene absorption increases with the Manchega breed, it presents lower milk yield. The 100%, 13/16, and 3/4 Lacaune genotypes had the highest milk yields with respect to the 1/2 Lacaune/Manchega breed (p < 0.001). The Lacaune registered on average 181.1 L in a period adjusted to 160 days of lactation (1.13 L/ day). Likewise, the parity number, litter size, and season of lambing effects showed significant differences (p < 0.01). It was concluded that 13/16 and 3/4 Lacaune/Manchega ewes presented the highest milk yields with respect to the other crosses.

1. Rates of lipolysis were measured at different concentrations of glucagon in adipocytes prepared from parametrial adipose tissue of fed or starved rats in different reproductive states. All experiments were performed in the presence of a high concentration of adenosine deaminase (1 unit/ml). 2. Maximal rates of lipolysis (elicited by 25 nM-glucagon in each instance) were higher in adipocytes from peak-lactating rats than those from pregnant animals in both the fed and starved states. 3. Of adipocytes from fed animals, those from peak-lactating rats were the most sensitive to glucagon, whereas those from late-pregnant and early-lactating rats were 1-2 orders of magnitude less sensitive. 4. Adipocytes from 24 h-starved rats showed a much smaller stimulation of lipolysis by glucagon, making the assessment of sensitivity difficult. Therefore, rates of lipolysis were also measured in the presence of a maximally anti-lipolytic dose of insulin. The presence of insulin did not alter the relative sensitivities to glucagon of adipocytes from fed animals in different reproductive states, although all dose-response curves were shifted to the right. When lipolysis in adipocytes from starved animals was measured in the presence of insulin, it became evident that starvation for 24 h markedly increased the sensitivity of adipocytes from late-pregnant rats to glucagon, but did not affect that of cells from animals in the other reproductive states. 5. It is concluded that the large changes in sensitivity to glucagon that occurred during the reproductive cycle may enable the modulation of adipose-tissue lipolysis in vivo to satisfy the different metabolic requirements of the animal in the transition from pregnancy to peak lactation.

Recently we have shown that prostaglandin-induced luteolysis in pregnant rats involves resistance to prolactin-receptor signalling through the JAK2/STAT5 pathway.1 In the present study, we investigate whether PGF2alpha acts similarly to inhibit GH signalling in the ovine corpus luteum. The oestrous cycle of ewes was synchronised using cloprostenol and CIDR-G devices with oestrus detected by testosterone treated wethers with raddles. Twelve days after the first recorded oestrous mark, ewes were given an intramuscular injection of either saline or cloprostenol (125 µg), followed 1 h later with an intravenous injection (jugular vein) of either vehicle or 1.5 mg recombinant bovine GH (rbGH, Monsanto). After a further 15 min ewes were killed by pentobarbitone overdose and the corpus luteum removed. Tyrosine phosphorylation of STAT5 (STAT-P) in the corpus luteum was determined by immunoprecipitation and Western blot (n = 4 ewes/treatment). STAT5-P levels were relatively low in all ewes that were not treated with rbGH. Treatment with rbGH significantly (P < 0.01) increased STAT5-P in the corpus luteum of ewes pretreated with saline, compared to both control groups. However the STAT5-P response to rbGH was significantly (P < 0.01) reduced by the pretreatment with cloprostenol, although the response remained significantly (P < 0.05) higher than both control groups. In summary we have shown that (1) as expected, the GH-receptor signals through STAT5 in the ovine corpus luteum and (2) cloprostenol induces resistance to this GH-receptor signalling pathway. A study was conducted to analyse the effect of various factors on cumulative monthly milk yields in Frieswal cows at Military Dairy Farm, Meerut, over a period of eight years (2006-2013). A total of 441 part lactation records of cows born to 30 Frieswal sires were analyzed using the SAS (2002) software. The least squares’ model developed for the cumulative 30, 60, 90, 120, 150, 180, 210, 240, 270 and 300 days yields accounted for 13.5, 13.5, 12.8, 12.0, 11.5, 11.6, 11.3, 10.3, 13.7 and 13.7 per cent of the total variation, respectively. The overall least squares’ averages for the cumulative 30, 60, 90, 120, 150, 180, 210, 240, 270 and 300 days of first lactation yields were estimated as 313.29, 660.83, 998.19, 1321.58, 1627.06, 1916.78, 2201.26, 2445.01, 2741.04 and 2972.64 kg, respectively. The effects of sire, season and period of calving were found to be statistically non-significant on any of the cumulative monthly part lactation yields while the regression of monthly part yields on AFC was found to be highly significant (P<0.01). Based on the results it was concluded that the AFC was the most significant factor which alters the cumulative part lactation yields in Frieswal cattle while the other factors like sire of cows, season and period of calving did not alter the part yields significantly. The cows calved during rainy season produce comparatively higher yield followed by summer and winter calvers. It may also be concluded that the genetic selection programme implemented in the Frieswal project resulted in increasing the milk production over the years of study.
