Veiled Chameleons (Chamaeleo calyptratus) are an increasingly popular model organism, and we have maintained a colony at the Stowers Institute since 2010. Due to the poikilothermic nature of these animals, achieving the correct macro and microenvironments plays an important role in successful long term colony maintenance. Despite being bred in captivity for many generations, relatively little husbandry information has been published regarding housing in a research environment. Recognizing that husbandry conditions and environmental parameters can impact research results we provide information about the housing, husbandry, maintenance, nutrition, and health for our colony of Veiled Chameleons. This information will be applicable to any chameleon housed in the facility in 2021.

The starlet sea anemone (Nematostella vectensis) is an emerging model organism, and we have maintained a colony at the Stowers Institute since 2007. Nematostella are known as a simple sea anemone, related to other cnidarians such as jellyfish and corals. Native to estuarine environments across the Atlantic coast of North America, from Novia Scotia to Florida, they encounter a variety of environmental conditions (e.g., temperature, salinity). Acknowledging that husbandry conditions and environmental parameters can impact research results we provide information about the housing, nutrition, maintenance, and health for our colony of Nematostella. This information will be applicable to any Nematostella housed in the facility in 2021.

Planaria are an emerging model organism, and we have maintained a colony of asexual and sexual worms at the Stowers Institute for Medical Research since 2010. Their small size allows us to maintain a large census across several systems requiring a small footprint. The planaria systems at the Stowers Institute allow for standardization of husbandry and environmental parameters. Acknowledging that husbandry conditions and environmental parameters can impact research results we provide information about the housing, nutrition, maintenance, and health for our Planaria colonies. This information will be applicable to any Planaria housed in the facility in 2021.

The utilization of an automated multitest laboratory as a data acquisition center and of a computer for trie data processing and analysis permits large scale preventive medical research previously not feasible. Normal test values are easily generated for the particular population studied. Long-term epidemiological research on large numbers of persons becomes practical. It is our belief that the advent of automation and computers has introduced a new era of preventive medicine.

Ipilimumab (IPI) provides a ten-year overall survival in almost 20 % of selected patients participated in several phase II-III trials. However, the expanded access program (EAP) looks more like routine practice than like clinical trials& This is why the results of such application could be different. Here we present the long-term follow-up data of single center EAP. Ninety-six patients with disseminated melanoma progressing after at least one lines of drug therapy were included at the N.N. Petrov National Medical Research Center of Oncology. Sixty-seven (70 %) patients had stage IV M1c, 35 patients (36 %) had elevated LDH before initiating IPI therapy. All patients received IPI 3 mg / kg IV every 3 weeks for a maximum of 4 cycles. Totally, 320 cycles (mean - 3.3 per patient) were conducted. Grade 3-4 immuno-mediated adverse events (imAE) observed in 18 (19 %) patients. Three patients died of adverse events, possibly associated with ongoing therapy. The median time to progression was 3 (95 % CI, 2.4 to 3.5) mo., the median overall survival was 13 (95 % CI, 8.3 to17.6) mo. Previous immunotherapy with dendritic cell vaccines decreased the risk of death by 48 % (Log-rank p = 0.049). The wild type BRAF status increased three-year overall survival from 29 to 68 % (p = 0.042). Our data confirms long-term safety and efficacy of IPI in patients with pretreated disseminated melanoma in the close to real practice setting.

Abstract Various information technology (IT) infrastructures for biobanking, networks of biobanks and biomaterial management are described in the literature. As pre-analytical variables play a major role in the downstream interpretation of clinical as well as research results, their documentation is essential. A description for mainly automated documentation of the complete life-cycle of each biospecimen is lacking so far. Here, the example taken is from the University Medical Center Göttingen (UMG), where the workflow of liquid biomaterials is standardized between the central laboratory and the central biobank. The workflow of liquid biomaterials from sample withdrawal to long-term storage in a biobank was analyzed. Essential data such as time and temperature for processing and freezing can be automatically collected. The proposed solution involves only one major interface between the main IT systems of the laboratory and the biobank. It is key to talk to all the involved stakeholders to ensure a functional and accepted solution. Although IT components differ widely between clinics, the proposed way of documenting the complete life-cycle of each biospecimen can be transferred to other university medical centers. The complete documentation of the life-cycle of each biospecimen ensures a good interpretability of downstream routine as well as research results.

Multiple sclerosis (MS) is an autoimmune disorder that affects ~2.5 million people globally. Women of reproductive age are highly susceptible to this disease. This study aims to explore the association between MS and pregnancy. Articles related to the topic under investigation were identified; the search terms included “pregnancy”, “multiple sclerosis”, “MS”, and “women”. Only articles published between 2010 and 2020 were included in the review. This review shows that researchers have attempted to explore the link between pregnancy and MS, and the results from previous studies indicate that pregnancy reduces the risk of MS relapse. However, evidence suggesting that pregnancy can affect the long-term progression of MS is lacking. The research results also indicate that MS does not increase the risk of maternal and fetal complications. MS remains a serious autoimmune disorder that affects many women worldwide. The data gathered during this review indicate that a significant correlation exists between pregnancy and MS relapse rates. The findings presented in this review can aid in the management of MS during pregnancy. Furthermore, these research results provide vital insights that caregivers can use to monitor patients with MS during pregnancy.

Few, if any, of children's behavioural or cognitive characteristics assessed in the first years of life demonstrate stability until later childhood; early characteristics have so far failed to show an association with future psychopathology. This longitudinal study, from 4–8 months to 4.7 years old, focused on stability and change of extreme temperamental traits in groups of infants subselected from a large birth cohort. Persistent extreme temperament at four and eight months old did not increase stability of temperament to four years of age, relative to other children in the whole population. Sizeable change occurred, and the environmental parameters associated with negative temperamental change did not seem to be the same as those related to positive change. Boys with extreme scores were more stable, while girls appeared more prone to positive change. It is hypothesised that the direction of temperamental change in the first years could be more meaningful for long-term prediction of disorders than any one assessment of temperament taken at any one year.

Abstract The article is a review of completed research on developed and implemented innovative and technologically advanced technical systems. According to the Industry 4.0 concept they can have a significant impact on the efficiency of production processes and product development. In this perspective, the key aspect seems to be maintenance management of technical systems, realized both in the operate phase as well as during service and repair works. There were discussed research results of authors and developed application solutions supporting decision-making processes, in terms of three main periods of realization of exploitation processes: short, medium and long-term.

Abstract. This article describes an innovative, complex and affordable monitoring system designed for joint observation of environmental parameters, rock block dilatations and temperature distribution inside the rock mass with a newly designed 3-meter borehole temperature sensor. Global radiation balance data are provided by pyranometers. The system introduces a novel approach for internal rock mass temperature measurement, which is crucial for the assessment of the changes in the stress field inside the rock slope influencing its stability. The innovative approach uses an almost identical monitoring system at different sites allowing easy setup, modularity and comparison of results. The components of the monitoring system are cheap, off-the-shelf and easy to replace. Using this newly designed system, we are currently monitoring three different sites, where the potential rock fall may endanger society assets below. The first results show differences between instrumented sites, although data time-series are relatively short. Temperature run inside the rock mass differs for each site significantly. This is very likely caused by different aspects of the rock slopes and different rock types. By further monitoring and data processing, using advanced modelling approaches, we expect to explain the differences among the sites, the influence of rock type, aspect and environmental variables on the long-term slope stability.
