Aims: Ulcerative colitis is a disease of the bowel that occurs in all ages and affects both males and females. This research study was designed to investigate the effect of age and sex on the healing of colitis in rats. Methodology: Twenty - eight rats were randomly distributed into four groups of seven animals per group; adult male rats, mid age male rats, adult female rats and mid age female rats. Mid age and adult Wistar rats were 7- 8weeks and 14 weeks old respectively. Colitis was induced through a single intra-colonic instillation of 7% acetic acid (1mL/100g body weight) and allowed to heal for 14 days. Blood samples were obtained for analysis. Colon samples were also obtained for histomorphological study and biochemical assays (Myeloperoxidase activities, Superoxide dismutase, Glutathione, Catalase and Malondialdehyde) levels. Results: There was no significant difference in Malondialdehyde concentration, catalase, Superoxide dismutase, Myeloperoxidase, Platelet Distribution Width, Platelet Count, Basophil cell numbers, Eosinophil cell numbers, platelet cells, Mean Platelet Volume , Mean Cell Volume and white blood cells across the groups. The Glutathione concentration in mid age male rats was significantly increased when compared with adult male rats. The haemoglobin , Lymphocytes and Mean Cell Haemoglobin levels were increased while neutrophils and monocyte levels were decreased in the younger female rats. The histomorphological study revealed poorly preserved surface epithelia layer of the colon in adult male rats while mid age male and female rats showed moderately preserved surface epithelia layer, adult female rats showed normal surface epithelia layer. Conclusion: Mid age rats heal faster than adult rats while in terms of sex, female rats tends to heal faster than male rats.

AbstractDue to the ethnopharmacological use of Campsiandra laurifolia (Fabaceae), popularly known as Acapurana, to treat wounds and ulcers, associated with the lack of alternative treatments for intestinal inflammations such as ulcerative colitis (UC), the present work sought to characterize its phytochemical and antioxidant activities, and to evaluate remedial action in experimental colitis with acetic acid. Phytochemical analyzes were performed through qualitative and quantitative colorimetric tests of the main secondary metabolites. In the colitis model, 24 male Wistar rats aged ± 60 days old were used, divided into 4 groups: Control (CO) control + aqueous extract of C. laurifolia 50 mg/kg (CO + A50); Colitis (CL); and Colitis + aqueous extract of C. laurifolia 50 mg/kg (CL + A50). Measurement of sphincter anal pressure and histological tests of the large intestine, lipoperoxidation (LPO), enzyme activity of superoxide dismutase (SOD), and levels of glutathione (GSH) were performed. For statistical analysis, the oxidative stress (OS) results were expressed as means ± standard error, adopting a significance level of p < 0.05. The screening indicated the presence of flavonoids, saponins and tannins in the extract, with high levels of phenolic compounds and tannins, and was related to high antioxidant capacity. In the histological analysis, the CL group presented loss of the crypts, edema and inflammatory infiltrate. The use of C. laurifolia extract restructured the crypts, decreased edema and increased sphincter anal pressure, with a decrease in LPO, SOD, and an increase in GSH. It is suggested that the use of C. laurifolia extract reduces OS due to its antioxidant power conferred by the phenolic compounds present in the extract.

Abstract Objectives Vanadium has been reported to possess relevant therapeutic properties such as anti-diabetic and anti-tumoral. This study aimed at determining the effects of vanadium on experimentally induced colitis in rats. Methods Forty-five male Wistar rats (103 ± 3.90 g, n=15) were used for this study and were divided into three groups. Group 1 (Untreated control) had nothing added to their drinking, while groups 2 and 3 received sodium metavanadate at a dose of 50 and 200 mg/L respectively in their drinking water for 10 weeks. Colitis was thereafter induced by intra colonic administration of 1.50 mL of 6% acetic acid. Animals were sacrificed on day 0 (pre-induction), three- and seven-days post induction. Blood samples were collected for haematological variables and the distal 8 cm of the colon was collected for macroscopic, histological and biochemical (malondialdehyde-MDA, superoxide dismutase-SOD, catalase-CAT, glutathione peroxidase- GPx and nitrite concentration- NO) assessment. Results Low dose vanadium proved beneficial in ameliorating acetic acid-induced colitis by improving both histopathological and haematological changes. Gross observation showed a faster healing rate in vanadium treated groups (50 and 200 mg/L) compared with untreated control at day 3 (40 and 26.20 vs. 2.50%) and day 7 (80 and 66.70 vs. 42%) respectively. Vanadium also appears to exert its beneficial effects on acetic acid-induced colitis via up regulation of antioxidant enzymes (SOD, CAT, GPx) and NO while decreasing the over production of MDA. Conclusions Vanadium at small concentration functions as an essential trace element and may be able to promote healing process during ulcerative colitis.

The aim of the current study was to evaluate the therapeutics potential of eluxadoline (ELX) loaded solid lipid nanoparticles (SLNs) in ulcerative colitis. ELX loaded SLNs were prepared using three different lipids according to the solvent emulsification technique. The optimization of prepared SLNs (F1-F3) were carried out based on size, PDI, zeta potential, percent drug entrapment (%EE), and loading (%DL). The lipid (stearic acid) based SLNs (F2) was optimized with particle size (266.0 ± 6.4 nm), PDI (0.217 ± 0.04), zeta potential (31.2 ± 5.19 mV), EE (65.0 ± 4.8%), and DL (4.60 ± 0.8%). The optimized SLNs (F2) was further evaluated by DSC, FTIR, SEM, in vitro release, and stability studies, which confirmed the successful encapsulation of ELX in SLNs. The efficacy of optimized SLNs (F2) in comparison to the pure ELX drug was assessed in acetic acid induced colitis rat models. It was observed that the delivery of ELX by SLNs alleviated the induced acetic acid colitis significantly. Thus, ELX loaded SLNs delivery to the colon has a significant potential to be developed for the treatment of ulcerative colitis.

Abstract Background: Mucosal inflammation and ulcer play important roles in the pathogenesis of ulcerative colitis. As as traditional Chinese medicine compound composed of Periplaneta americana and Taraxacum mongolicum, Ento-PB is always prescribed for the treatment of ulcer and inflammatory diseases. As for the significant role of P. americana in terms of promoting mucosal healing, the compatibility of the anti-inflammatory drug T. mongolicum may enable Ento-PB to simultaneously play anti-inflammatory and promote mucosal healing effects on the treatment of UC. Therefore, this study aimed to evaluate the therapeutic potential and possible mechanism of Ento-PB for UC by establishing an acetic acid-induced colitis model in dogs.Methods: Preliminary identification to the chemical components of compound Ento-PB was carried out through high performance liquid chromatography. A cross-bred dogs model of acetic acid-induced ulcerative colitis was established to evaluate the efficacy of compound Ento-PB. The expression levels of inflammatory cytokines C-reactive protein (CRP), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-10 (IL-10) in plasma were measured by carrying out enzyme-linked immunosorbent assay (ELISA).Results: With the extension of treatment time, Ento-PB could effectively improve clinical symptoms of UC cross-bred dogs. Colonoscopy displayed that mucosal redness, swelling and congestion decreased gradually, and obviously repaired after mucosal injury. The intestinal texture was gradually clear, and the colonoscopy score gradually reduced. Histopathological examination revealed that the structure of colon was restored significantly, the infiltration of inflammatory cells was reduced, and the histological score was remarkably reduced. At the same time, the results of dynamic monitoring of inflammatory cytokines in plasma proved that Ento-PB can gradually down-regulate the activity of CRP, iNOS and COX-2, reduce the expression levels of inflammatory cytokines TNF-α and IL-1β, and gradually restore anti-inflammatory and the expression level of cytokine IL-10.Conclusions: Ento-PB reduces the level of pro-inflammatory cytokines in a dose- and time-dependent manner and inflammation, improves colon tissue lesions and the repair of intestinal mucosa after injury, and effectively increases acetic acid-induced colon inflammation in UC cross-bred dogs.
