The aim of the study was to elucidate the role and significance of oxytocin in reparative histogenesis of tissues of various genesis using the example of periodontal tissues and extrapulmonary airways.Material and methods. Periodontitis and tracheobronchitis were simulated on sexually mature white outbred male rats. Periodontitis was simulated by traumatic injury to the periodontium, followed by the wound infection with Staphylococcus aureus. Introduction of oxytocin as an additional tool was used in the treatment of periodontitis. Experimental tracheobronchitis was simulated in rats by intratracheal infection with S. aureus (strains with and without antilactoferrin activity). Paraventricular large-cell nuclei of the hypothalamus were additionally destroyed in the simulated experimental tracheobronchitis (in order to reduce the production of endogenous oxytocin). The resulting material (the periodontium, alveolar processes, trachea and extrapulmonary bronchi) was examined using histological methods of investigation.Results. Analysis of histological sections demonstrated that when oxytocin was used in the complex therapy of experimental periodontitis, the proliferation and cyto-differentiation of fibroblasts, osteoblasts and cementoblasts was intensified, followed by their activation; this resulted in the optimization of reparative histogenesis in the structures of the periodontium and alveolar ridge. In animals with destruction of the paraventricular nuclei of the hypothalamus (against the background of an oxytocin deficiency in the body), a decrease in the reparative potential of the tracheal and bronchial mucosa was observed. Moreover, the severity of the inflammatory process in the mucous membrane of the trachea and bronchi with the introduction of staphylococci with antilactoferrin activity was significantly higher than that of animals that were administered staphylococci without antilactoferrin activity.Conclusion. The results obtained indicate the optimizing and correcting effect of the hypothalamic nonapeptide oxytocin on the reparative regeneration of the studied periodontal and the trachea and bronchi structures.

The salts of heavy metals negatively affect the health of the population and cause the development of the pathology of individual organs and systems of the body. Adrenal hormones are one of the key places in regulating and maintaining the basic functions of the body. The aim of the study — the study of morphological rearrangements of the structural components of the adrenal cortex of sexually mature male rats under conditions of prolonged exposure to the combination of heavy metal salts remains relevant and insufficiently studied. The experiment was conducted on 12 animals weighing 250–300 g, aged 7–8 months. Animals of the experimental group used for 90 days the usual drinking water, saturated with a combination of heavy metal salts. Commonly used histological methods of research were used. Long-term admission into the body of sexually-mature rats of a combination of heavy metal salts leads to morphological rearrangements of the stroma, vascular bed, morphology and secretory activity of cells of the adrenal gland, mainly zona fasciculata and zona reticularis. Development in spongy sacs of lipid vacuolar and granular degeneration indicates a weakening of steroidogenesis, which negatively affects the development of compensatory and adaptive processes in the body and the general adaptation syndrome in response to the action of the damaging agent. So, adaptive-compensatory processes in the adrenal cortex with signs of reduced functional activity, characteristic for the initial stages of depletion of the general adaptive syndrome. Prospects for further development are based on conducting studies of adrenal rats in conditions of reparative changes after long periods of influence of the combination of heavy metal salts on the body.

Subject. In the modern world, the prevalence of cancer is growing every year, in this regard, this pathology is one of the most pressing problems in medicine. There is no doubt that malignant neoplasms have a complex interaction with the body. The General mechanisms of occurrence and progression of the tumor process are intensively studied, and to date, a huge amount of material on molecular and clinical Oncology has been accumulated, it is used in related disciplines. The most important sections of dentistry are the diagnosis and treatment of diseases of the mucous membrane. Often, clinical manifestations in the oral cavity are one of the first symptoms indicating a General somatic disease. This article covers the study of the dental status of patients with different localizations of cancer. Objective ― to study the incidence of dental diseases of patients with cancer. Methodology. To conduct a study medical examination of patients with different localizations of cancer was held to determine dental: the definition of DMF-Index, PMA-index, Parodontal Index, and the condition of the mucous membrane of the oral cavity and periodontal tissues. Cancer patients receiving chemotherapy and radiation therapy, patients who refused to participate in the survey were excluded from the survey. Results. The study included 526 patients. The predominance of certain types of dental diseases among patients with a particular cancer pathology was revealed. Conclusions. Our study showed that all patients had decompensated forms of caries. The most common dental diseases combined with cancer are hyperplasia filiform papillae (K14.3), the "geographic" language (K14.1). Periodontal disease occurred in 69,5 % of cases among cancer patients.

Introduction. Inflammatory periodontal disease is one of the most urgent problems of dentistry, which has a social significance due to the high prevalence, pronounced changes in the tissues of the periodontal disease and the body of the patient as a whole, and the defeat of young people.The aim of the study – to investigate the functional state of the nitrogen (II) oxide system in rats with periodontitis without concomitant pathology and against the background of hyper- and hypothyroidism.Research Methods. The study was carried out on 48 white non-linear male rats. The total activity of NO-synthase (NOS) was determined colorimetrically by the number of formed nitrates and nitrites in the incubation medium. The total content of nitrates and nitrites (NOx) was determined by the Gris method.Results and Discussion. Experimental periodontitis is accompanied by increased general activity of NO-synthase in periodontal tissue homogenate by 2.2 times vs control. NOx content in the serum of animals with periodontitis increased by 46.2% and in the periodontal tissue homogenate – by 74.7% compared with the control. In rats with periodontitis against hyperthyroidism, NOS activity increased by 3.9 times relative to the control group of animals and by 75.9% exceeded the rate of rats with periodontitis without concomitant pathology. In rats with periodontitis, against the background of hypothyroidism, the activity of NOS was 29.6% higher than that of rats with periodontitis without concomitant pathology and 2.9-fold of control.Conclusions. Experimental periodontitis is accompanied by a marked increase in the intensity of nitroxidergic processes both in the homogenate of periodontal tissues and in the blood. The imbalance of thyroid hormones increases the synthesis of nitrogen (II) oxide in the experimental periodontitis, especially expressed in hyperthyroidism.

We present the effect of stressogenic factors on the morphological parameters of spermatozoa. Objective. We studied the morpho-physiological features of spermatozoa from male albino rats under experimental stress. Materials and methods. A experiment was carried out using sexually mature outbred white rats. Combined stress was performed to an experimental group: after three days of starvation, an emotional stress of “free swimming in a cage” technique was provided. Under a microscope, the spermatozoa were counted per volume of epididymal suspension, the activity was evaluated, and the morphology was studied. Results. A study of the epididymal suspension of male rats under starvation and “emotional” stress showed a decrease in the total number of spermatozoa and sperm motility indices, and an increase in the abnormal gamete forms compared to the control. Conclusion. The combined effect of even short-term stressogenic factors leads to significant changes in the spermogram, allowing the quantitative and qualitative seminal fluid parameters to be a convincing criterion of maladaptation processes in the body under the stress.

Today special attention to the influence of environmental factors on the condition of the dental-maxillary system and periodontal tissues in children is paid. Scientists have proved that environmentally unfavorable conditions also change the immunological reactivity of the body, form violations of the immune status, and changes in the local immunity in the oral cavity stimulate the development of pathogenic microflora. Our research is devoted to the study of changes in periodontal tissues in children living in ecologically favorable conditions and in ecologically polluted Precarpathian regions, a comparative characterization of changes in periodontium, a treatment plan for chronic catarrhal gingivitis, supplemented with chlorhexidine dentin and quercetin. According to the results of many researches, 0.12% chlorhexidine solution provides a pronounced antimicrobial effect on pathogenic microflora; and under the action of quercetin there is a stabilization of the antioxidant system, improvement of peripheral blood circulation, improvement of rheological properties of the patient's blood. We examined 60 patients with chronic catarrhal gingivitis living in environmentally stable conditions (Ivano-Frankivsk) (1 group), 15 people with intact periodontium and 60 children with chronic catarrhal gingivitis living in contaminated Precarpathian regions (Burshtyn) (group 2). All patients were treated with a set of therapeutic measures, which are recommended for the conventional treatment of patients with chronic catarrhal gingivitis. In groups 1a and 2a, patients were treated with chlorhexidine, calendula and chamomile infusion and 1% mefenamine paste. In groups 1b and 2b we applied a quercetin gel on the mucous membrane of the patient’s gums under an individual dental-gingival cap. For the purpose of comparative characterization of the two groups, an index assessment of the condition of periodontal tissues, study of rheographic parameters of periodontal tissues and radiographic characteristics of jaw bone tissue were performed. The results of the research showed a significant difference between clinical and laboratory parameters in patients with chronic catarrhal gingivitis living in environmentally friendly conditions compared with persons living permanently in ecologically polluted Precarpathian regions (p <0.05 ). In people living in ecologically polluted areas there is a pronounced cyanosis of the gums and mild diffuse cyanosis of the oral mucosa, high rates of bleeding gums. Scientists believe that the main mechanisms of pathogenetic effects on periodontal tissues are transmitted infectious and toxic diseases, trophic and metabolic disorders, changes in neurohumoral regulation, vitamin imbalance in the body, immune disorders, hypoxia, oxidative stress, disorders of plastic processes in any tissues of the body, the mucous membrane of the oral cavity. We proved that the use of the proposed treatment regimen for chronic catarrhal gingivitis (groups 1b and 2b) shows a significantly higher effectiveness over the treatment of basic therapy in groups 1a and 2a. This treatment stimulates metabolic processes in periodontal tissues, reduces cyanosis, edema of periodontal tissues, eliminates inflammatory phenomena in periodontal tissues. The results show that the use of quercetin and 0.12% chlorhexidine solution in the complex treatment of patients with chronic catarrhal gingivitis provides a significant improvement in clinical parameters and laboratory characteristics of periodontal tissues in children living in different environmental conditions.

В условиях непрерывного воздействия неблагоприятных факторов окружающей и производственной среды на человека особую актуальность приобретает изучение механизмов, поддерживающих гомеостаз организма. Длительное поступление фторидов в организм приводит к формированию хронической фтористой интоксикации, патогенез которой вызывает многочисленные споры и дискуссии. До сих пор недостаточно внимания уделяется изучению висцеральной патологии, обусловленной нарушениями иммунного статуса в условиях воздействия на организм соединений фтора. Практически отсутствуют исследования по изучению иммунной реактивности, определяющей морфофункциональный характер ответной реакции печени на ранних стадиях развития фтористой интоксикации. Цель работы - изучение действий патофизиологических механизмов иммунной реактивности печени при субхроническом действии на организм соединений фтора. Методика. Опыты проведены на 210 лабораторных крысах-самцах массой 180-220 г., разделенных на 2 группы: контрольную (n=80) и группу животных с субхроническим действием фторида натрия (n=130). Экспериментальные животные в течение 12 нед имели свободный доступ к водному раствору фторида натрия (концентрация 10 мг/л, что составляет суточную дозу фтора 1,2 мг/кг массы тела). Для изучения иммунологических и биохимических показателей забирали кровь из хвостовой вены через 1, 3, 6, 9, 12 нед от начала эксперимента. Для оценки состояния гуморального звена иммунитета определяли уровень сывороточных иммуноглобулинов (IgA, IgG, IgM) иммуноферментным анализом с помощью наборов реактивов ЗАО «Вектор-Бест» (Новосибирск). Уровень сывороточных цитокинов: TNF-α, IL-1β, 2, 4, 6, 10 определяли на анализаторе Multiskan EX методом иммуноферментного анализа с использованием наборов «Вектор Бест» (Новосибирск). Подсчет общего количества лейкоцитов произведен классическим способом в камере Горяева, анализ лейкоцитарной формулы - в окрашенных мазках периферической крови. Метаболические изменения оценивали по активности ферментов в ткани печени: щелочной фосфатазы (ЩФ), аланин- и аспартатаминотрансфераз (АЛТ, АСТ), лактатдегидрогеназы (ЛДГ), гаммаглутамилтранспептидазы (γ-ГТ). Активность ферментов определяли унифицированными методами с помощью наборов реактивов ЗАО «Вектор-Бест» (Новосибирск) на фотометре PM-750 (Германия). Гистологические исследования печени осуществляли после декапитации крыс, проводимой под эфирным наркозом. Результаты. Показано, что субхроническое воздействие фторида натрия сопровождается формированием внутриклеточных и внутрисосудистых повреждений печени. Активация медиаторов воспаления и развитие иммунологических нарушений в динамике эксперимента способствуют формированию системной воспалительной реакции, которая приводит к появлению стойких морфологических нарушений в печени и изменению активности ферментов основных метаболических путей. Заключение. Полученные результаты могут быть использованы при разработке и проведении профилактических мероприятий в условиях воздействия на организм высоких концентраций фтора с последовательным применением детоксикационной, иммуномодуляторной и органопротекторной коррекции. Studying mechanisms, which maintain the body homeostasis, is particularly important in the conditions of continuous impact of adverse environmental and manufacturing factors. Long-term exposure to fluorides leads to chronic fluoric intoxication, the pathogenesis of which is a subject of multiple controversy and discussions. Not enough attention is still paid to elucidating the visceral pathology associated with fluorine-induced immune disorders. There are virtually no studies of immune reactions that define the morphofunctional nature of the liver response to early stages of fluoric intoxication. Aim. To study pathophysiological mechanisms of hepatic immune reactivity in subchronic exposure of the body to fluorine compounds. Methods. Experiments were performed on 210 male rats weighing 180-220 g. The animals were divided into two groups: 1) control (n=80) and 2) subchronic exposure to sodium fluoride (n=130). The rats had free access to a 10 mg/l aqueous solution of sodium fluoride (daily dose, 1.2 mg/kg body weight) for 12 weeks. Blood was withdrawn from the caudal vein at 1, 3, 6, 9, and 12 weeks of the experiment for immunological and biochemical tests. Histological study of the liver was performed after decapitation of rats under ether anesthesia. Results. The subchronic exposure to sodium fluoride was associated with intracellular and intravascular damage of the liver. Activation of inflammatory mediators and development of immunological disorders during the experiment contributed to a systemic inflammatory reaction, which resulted in persistent morphological injuries of the liver and changes in enzyme activities in major metabolic pathways. Conclusion. The study results can be used for development and implementation of preventive measures against the effects of high fluorine concentrations, which would include a successive use of detoxification, immunomodulation and organ protection.

Hypothyroidism frequently causes cardiopulmonary dysfunction, such as heart failure and respiratory and metabolic deficiencies. This study investigated the effects of Chinese herbal formula B307 on thyroidectomy-induced cardiopulmonary exercise dysfunction in rats. Twenty male rats were equally divided into four groups: negative control with sham treatment, positive control with oral B307 treatment only, thyroidectomy treatment only, and thyroidectomy with B307 posttreatment groups. The feeding dose of B307 was 50 mg/kg per day for 14 days. We examined and then compared the thyroid-stimulating hormone (TSH), free triiodothyronine (T3), free thyroxine (T4), and reactive oxygen species (ROS) from the blood of these four groups. Also, we compared the body weight, neck subcutaneous blood flow, cardiac ejection function, cardiopulmonary exercise function of oxygen consumption (VO2), carbon dioxide production (VCO2), and respiratory quotient (RQ = VCO2/VO2) among the four groups. Our results indicated that thyroidectomized rats had significantly decreased body weight, neck subcutaneous blood flow, cardiac ejection function, serum T3 and T4, and VO2 and VCO2, but had significantly increased ROS and TSH levels and RQ values compared with sham rats (P<0.01–0.05). In addition, thyroidectomized rats receiving oral B307 treatment had significantly increased body weight, neck subcutaneous blood flow, cardiac ejection function, and VO2, but significantly decreased ROS and TSH levels and VCO2 and RQ values compared with thyroidectomized rats (P<0.01–0.05). We suggest that the B307 could be a protective and beneficial alternative treatment for thyroidectomy-induced cardiopulmonary exercise dysfunction.

Sexual performance in adult male rats is highly sensitive to prenatal stress which can affect the functionality of the reproductive system and various brain structures involved in modulating sexual behavior. The immunomodulatory effect of mouse IgG on reproductive maturity in male offspring after LPS exposure in vivo and in vitro was studied. Prenatal IgG injection (20 µg / mouse) had a positive impact on the puberty of male mice whose mothers were exposed to LPS (100 µg / kg) on the 12th day of pregnancy. The number of Sertoli cells were increased, whereas the body weight and the number of symplastic spermatids were decreased in offspring as compared to LPS-treated animals. Besides, IgG had a positive effect on altered hormone levels: reduced estradiol level on the 5th and 14th postnatal days and increased testosterone level on the 30th postnatal day in blood that led to an increased number of mounting attempts in sexually mature males. The cAMP-dependent pathway may be involved in the regulation of the LPS-induced inflammation. IgG reduced the increased level of cAMP in mouse peritoneal macrophages activated by LPS in vitro. IgG is able to modulate inflammation processes, but its exposure time is important.

Long-term consequences of impaired perinatal development are very significant. They appear during the neonatal period and in the first years of life, and persist during ontogenesis. There is little data on the impact of any prenatal factors on the sensitivity of a sexually mature organism to medications. The aim of the study is to assess the impact of early life stress on the development of individual antidepressant sensitivity. Materials and Methods. The authors conducted the experiments on sexually mature outbred male rats. To simulate the early life stress, a standard protocol was used. From the 2nd to 15th days of the postnatal period the pup rats were separated from their mother for 3 hours and kept in an incubator. The open-field test, Porsolt test and Sucrose consumption test were used to determine rat’s anxiety level as well as motor, orientation and exploratory activity at puberty. Then, for 14 days, the rats were intragastrically administered with a fluoxetine solution (10 mg/kg/daily), followed by their full examination. Statistical analysis of results was performed using the Mann-Whitney U-test to compare unrelated groups and Wilcoxon's test to compare related groups. Results. Fluoxetine did not have a pronounced antidepressant effect in animals that survived the early life stress. Such animals demonstrated passive floating during the Porsolt test, without any changes in immobility time. When testing in an open field, a sharp increase in the number of freezing behavior was observed, which was an indicator of an increased anxiety level in animals. Conclusion. The results obtained indicate that the long-term effects of neonatal stress may be associated with a change in antidepressant sensitivity or an increase in development of unwanted adverse reactions. Отдаленные последствия нарушения перинатального развития весьма значительны и не только проявляются в период новорожденности и в первые годы жизни, но и сохраняются в период онтогенеза. Данные о влиянии каких-либо пренатальных факторов на чувствительность половозрелого организма к действию лекарственных веществ в доступной литературе представлены незначительно. Цель исследования – оценить роль стресса раннего периода жизни в формировании индивидуальной чувствительности к действию антидепрессантов. Материалы и методы. Эксперименты проведены на половозрелых беспородных крысах-самцах. Для моделирования стресса раннего периода жизни использовали стандартный протокол, подразумевающий отделение детенышей от матери со 2-го по 15-й дни постнатального периода на 3 ч в условиях инкубатора. В половозрелом возрасте проводили оценку уровня тревожности, двигательной и ориентировочно-исследовательской активности крыс в условиях теста открытого поля, теста Порсолта и теста «Потребление раствора сахарозы». Затем в течение 14 дней крысам внутрижелудочно вводили раствор флуоксетина (10 мг/кг/сут), после чего обследование повторяли в том же объеме. Статистический анализ результатов исследования проводили с использованием U-критерия Манна–Уитни для сравнения несвязанных групп и критерия Вилкоксона для сравнения связанных групп. Результаты. У животных, переживших стресс раннего периода жизни, флуоксетин не оказывал выраженного антидепрессантного действия. У данных животных в тесте Порсолта преобладало пассивное плавание, без изменения длительности иммобильности. При тестировании в открытом поле наблюдалось резкое повышение числа актов фризинга, что является показателем повышенного уровня тревожности у животных. Выводы. Полученные результаты свидетельствуют о том, что отдаленные последствия неонатального стресса могут быть связанны с изменением чувствительности к действию антидепрессантов или повышением риска развития нежелательных побочных реакций.
