Relevance. The purpose of this work is to create new discrete competitive tomato genotypes based on modern biotechnological approaches with increased resistance to the tobacco mosaic virus, economically valuable traits and adapted to the soil and climatic conditions of cultivation in the southern regions of the Russian Federation.Methods. At the first stage of the study, SSR markers taken from literature sources were tested [1;2] and PCR conditions were optimized for the identification of target Tm genes (Tm2a, Tm22, providing resistance to tobacco mosaic virus at the tissue level) in the breeding material available in the department of vegetable and potato growing.Results. Two informative molecular markers Tms 37 and UMD 2060 were selected, which reveal the allelic difference between resistant and susceptible samples. Hybridization of FMS tomato lines with samples having the desired genes of interest in the genotype for obtaining tomato lines resistant to TMV was carried out. Seeds of paternal forms - donors of target genes were obtained for their further use in breeding work. The scientific novelty lies in the study of the discrete genetic material of Solanumlycopersicum, used in the breeding process, and preserving its functions in a new genetic environment.

The article presents stages of development and testing new rice variety Nautilus with blast resistance genes Pi-b and Pi-z. Basing on the use of DNA-marking methods, breeding material carrying the target genes was developed. The obtained material was studied in the links of breeding process in the field. Based on the results of field testing, 6 lines were identified, of which VNIIR 6016 turned out to be the best in terms of a complex of economically valuable traits. In 2016, this sample was transferred to the State variety testing under the name Nautilus. In 2019, a new rice variety Nautilus was included in the register of breeding achievements and approved for use in the Russian Federation in Krasnodar region. The study of variety Nautilus in 2019-2020 in an ecological test confirmed its high resistance to blast, in comparison with other released varieties. It took 10 - 12 years to develop the variety.

Preliminary results reported last year on the ion etching of tobacco mosaic virus indicated that the diameter of the virus decreased more rapidly at 10KV than at 5KV, perhaps reaching a constant value before disappearing completely.In order to follow the effects of ion etching on TMV more quantitatively we have designed and built a second apparatus (Fig. 1), which incorporates monitoring devices for measuring ion current and vacuum as well as accelerating voltage. In addition, the beam diameter has been increased to approximately 1 cm., so that ten electron microscope grids can be exposed to the beam simultaneously.

Tobacco mosaic virus (TMV) infection has been studied in several investigations of Nicotiana tabacum leaf tissue. Earlier studies have suggested that TMV infection does not have precise infective selectivity vs. specific types of tissues. Also, such tissue conditions as vein banding, vein clearing, liquification and suberization may result from causes other than direct TMV infection. At the present time, it is thought that the plasmodesmata, ectodesmata and perhaps the plasmodesmata of the basal septum may represent the actual or more precise sites of TMV infection.TMV infection has been implicated in elevated levels of oxidative metabolism; also, TMV infection may have a major role in host resistance vs. concentration levels of phenolic-type enzymes. Therefore, enzymes such as polyphenol oxidase, peroxidase and phenylalamine ammonia-lyase may show an increase in activity in response to TMV infection. It has been reported that TMV infection may cause a decrease in o-dihydric phenols (chlorogenic acid) in some tissues.

Superoxide overproduction is known to occur in multiple disease states requiring critical care yet non-invasive detection of superoxide in deep tissue remains a challenge. Herein, we report a metal-free magnetic resonance imaging (MRI) and electron paramagnetic resonance (EPR) active contrast agent prepared by “click conjugating” paramagnetic organic radical contrast agents (ORCAs) to the surface of tobacco mosaic virus (TMV). While ORCAs are known to be reduced <i>in vivo</i> to an MRI/EPR silent state, their oxidation is facilitated specifically by reactive oxygen species—in particular superoxide—and are largely unaffected by peroxides and molecular oxygen. Unfortunately, single molecule ORCAs typically offer weak MRI contrast. In contrast, our data confirm that the macromolecular ORCA-TMV conjugates show marked enhancement for <i>T<sub>1</sub></i> contrast at low field (<3.0 T), and <i>T<sub>2</sub></i> contrast at high field (9.4 T). Additionally, we demonstrated that the unique topology of TMV allows for “quenchless fluorescent” bimodal probe for concurrent fluorescence and MRI/EPR imaging, which was made possible by exploiting the unique inner and outer surface of the TMV nanoparticle. <a>Finally, we show TMV-ORCAs do not respond to normal cellular respiration, minimizing the likelihood for background, yet still respond to enzymatically produced superoxide in complicated biological fluids like serum.</a>

The article notes that the growing popularity of healthy lifestyles contributes to the increase in consumption of fruits and berries. At the same time, the analysis of the dynamics of the gardening industry for 2000-2018 shows that the problem of providing fresh fruits and berries to the population remains one of the most important. Based on actual data, linear and logarithmic models of time series of key industry indicators for the period 2010 – 2019 have been compiled. Calculations showed that in the Russian Federation as a whole the trend of reduction of sown areas of perennial fruit plantations will continue with growth of yield and gross fees. In this regard, the issues of distribution and introduction of gardens of intensive type are updated. An overview of the views of domestic scientists-gardeners on the concept of “intensive garden” is given. It has been established that the distribution of intensive gardens is possible only if there are favorable natural and climatic conditions and a developed scientific and production base of nursery management. The current state and problems of gardening in one of the leading regions – the Republic of Dagestan - are considered. A significant technological lag of region in the further development of intensive horticulture has been identified. Various directions of intensification process in horticulture as the main and necessary condition of growth of efficient and sustainable production are summarized.
