Correlation Analysis between MyoG Gene Polymorphism and Carcass Characteristics of Egg quails

Quail is extensively reared in China for characteristic of quick growth, small fodder consumption, early sexual maturity, high egg output and short production period. In this study, the SNP markers in 5’ regulation regions of cytogenin gene (MyoG) among China yellow quail, Beijing white quail and Korean quail were detected by PCR-SSCP method. Moreover, correlations of MyoG 5’ regulation regions with carcass characteristics of quail were analyzed. Results demonstrated thatin egg quail group, six genotypes were detected in Locus A in the control region of MyoG 5’, including AA, BB, CC, AB, AC and BC. The highest frequencies of CC in China yellow quail, Beijing white quail and Korean quail were 0.323, 0.366 and 0.444, respectively. A total of 9 genotypes were detected in locus B in egg quail group, which were AA, BB, CC, DD, EE, AB, AC, AD and AE. AA showed the highest frequency in China yellow quail, Beijing white quail and Korean quail, which were 0.493, 0.385 and 0.406, respectively. There was significant correlation between locus A and leg muscle rate of egg quail (plessthan0.05). Locus B presented significant correlations with carcass net weight, leg muscle weight, dressing percentage, whole net carcass rate and heart rate (plessthan0.05). Loci A and B in the control region of MyoG 5’ can be used as the molecular marker of carcass characteristics of egg quails during marker assisted selection.

Abstract:Electrical stimulation-induced leg muscle contractions provide a useful model for examining the role of leg muscle neural afferents during low-intensity exercise in persons with spinal cord-injury and their able-bodied cohorts. Eight persons with paraplegia (SCI) and 8 non-disabled subjects (CONTROL) performed passive knee flexion/extension (PAS), electrical stimulation-induced knee flexion/extension (ES) and voluntary knee flexion/extension (VOL) on an isokinetic dynamometer. In CONTROLS, exercise heart rate was significantly increased during ES (94 ± 6 bpm) and VOL (85 ± 4 bpm) over PAS (69 ± 4 bpm), but no changes were observed in SCI individuals. Stroke volume was significantly augmented in SCI during ES (59 ± 5 ml) compared to PAS (46 ± 4 ml). The results of this study suggest that, in able-bodied humans, Group III and IV leg muscle afferents contribute to increased cardiac output during exercise primarily via augmented heart rate. In contrast, SCI achieve raised cardiac output during ES leg exercise via increased venous return in the absence of any change in heart rate.

Abstract Background Maize is one of the most important field crops in the world. Most of the key agronomic traits, including yield traits and plant architecture traits, are quantitative. Fine mapping of genes/ quantitative trait loci (QTL) influencing a key trait is essential for marker-assisted selection (MAS) in maize breeding. However, the SNP markers with high density and high polymorphism are lacking, especially kompetitive allele specific PCR (KASP) SNP markers that can be used for automatic genotyping. To date, a large volume of sequencing data has been produced by the next generation sequencing technology, which provides a good pool of SNP loci for development of SNP markers. In this study, we carried out a multi-step screening method to identify kompetitive allele specific PCR (KASP) SNP markers based on the RNA-Seq data sets of 368 maize inbred lines. Results A total of 2,948,985 SNPs were identified in the high-throughput RNA-Seq data sets with the average density of 1.4 SNP/kb. Of these, 71,311 KASP SNP markers (the average density of 34 KASP SNP/Mb) were developed based on the strict criteria: unique genomic region, bi-allelic, polymorphism information content (PIC) value ≥0.4, and conserved primer sequences, and were mapped on 16,161 genes. These 16,161 genes were annotated to 52 gene ontology (GO) terms, including most of primary and secondary metabolic pathways. Subsequently, the 50 KASP SNP markers with the PIC values ranging from 0.14 to 0.5 in 368 RNA-Seq data sets and with polymorphism between the maize inbred lines 1212 and B73 in in silico analysis were selected to experimentally validate the accuracy and polymorphism of SNPs, resulted in 46 SNPs (92.00%) showed polymorphism between the maize inbred lines 1212 and B73. Moreover, these 46 polymorphic SNPs were utilized to genotype the other 20 maize inbred lines, with all 46 SNPs showing polymorphism in the 20 maize inbred lines, and the PIC value of each SNP was 0.11 to 0.50 with an average of 0.35. The results suggested that the KASP SNP markers developed in this study were accurate and polymorphic. Conclusions These high-density polymorphic KASP SNP markers will be a valuable resource for map-based cloning of QTL/genes and marker-assisted selection in maize. Furthermore, the method used to develop SNP markers in maize can also be applied in other species.

Analyzing marker-assisted breeding is an important method utilized in modern molecular breeding. Recent studies have determined that a large number of molecular markers appear to explain the impact of “lost heritability” on human height. Therefore, it is necessary to locate molecular marker sites in poultry and investigate the possible molecular mechanisms governing their effects. In this study, we found a 104-bp insertion/deletion polymorphism in the 5′UTR of the ADPRHL1 gene through resequencing. In cross-designed F2 resource groups, the indel was significantly associated with weight at 0, 2, 4, 6, and 10 weeks and a number of other traits [carcass weight (CW), semi-evisceration weight (SEW), evisceration weight (EW), claw weight (CLW), wings weight (DWW), gizzard weight (GW), pancreas weight (PW), chest muscle weight (CMW), leg weight (LW), leg muscle weight (LMW), shedding Weight (SW), liver rate (LR), and leg muscle rate (LMR)] (P < 0.05). In brief, the insertion-insertion (II) genotype was significantly associated with the greatest growth traits and meat quality traits, whereas the values associated with the insertion-deletion (ID) genotype were the lowest in the F2 reciprocal cross chickens. The mutation sites were genotyped in 4,526 individuals from 12 different chicken breeds and cross-designed F2 resource groups. The II genotype is the most important genotype in commercial broilers, and the I allele frequency observed in these breeds is relatively high. Deletion mutations tend to be fixed in commercial broilers. However, there is still considerable great potential for breeding in dual-purpose chickens and commercial laying hens. A luciferase reporter assay showed that the II genotype of the ADPRHL1 gene possessed 2.49-fold higher promoter activity than the DD genotype (P < 0.05). We hypothesized that this indel might affect the transcriptional activity of ADPRHL1, thereby affecting the growth traits of chickens. These findings may help to elucidate the function of the ADPRHL1 gene and facilitate enhanced reproduction in the chicken industry.

The effects of salbutamol administration to pregnant sows on post-natal growth and carcass characteristics of the progeny were investigated. Salbutamol (4 mg/kg diet) was fed to sows during the first (0 to 38 days, TI), second (39 to 78 days, T2) or third (79 days to term, T3) trimester of pregnancy. At birth, maternal administration of salbutamol had no effect on body weight, dressing percentage or weights of liver, heart, semimembranosus and semitendinosus muscle in proportion to body weight in the progeny. No significant effects of maternal treatment with salbutamol on body weight, dressing percentage, semitendinosus muscle weight in proportion to body weight and loin eye area were observed in the progeny at 21 days. P2 fat depth at 21 days was greater (P < 0-05) in TI than in controls. The percentage of type I fibres in the deep portion of semitendinosus muscle was higher (P < 0.05) in treatment groups as compared with controls: 27.4, 42.8, 36.9 and 36.0% in control, TI, T2 and T3 respectively. At slaughter (90 kg), carcass and meat parameters including length, P2 fat depth, meat colour and drip loss were not affected by maternal administration of salbutamol. However, loin eye areas were larger in T1 than in controls. Larger loin eye areas, coupled with numerically (but not statistically) significant heavier carcasses and lower backfats, resulted in greater estimates of carcass protein mass in TI pigs as compared with controls (P = 0.06).

AbstractSalbutamol (albuterol) is a β-adrenergic agonist marketed as a racemic (50: 50) mixture of R- and S-enantiomers (rac-salbutamol). Since only R-salbutamol is pharmacologically active and S-salbutamol has a longer half-life in humans, we examined R-salbutamol as a performance enhancer and repartitioning agent in domestic chickens. The effects of feeding diets containing R-salbutamol (5, 10 and 15 mg/kg diet) and rac-salbutamol (10 mg/kg diet) from day 21 to 42 post hatching on growth performance, carcass characteristics and tissue concentrations of R- and S-salbutamol in male and female broilers were compared with a control diet. R-salbutamol in the diet lowered the weight gains in both sexes, but the magnitude of reduction was greater in males as indicated by a significant R-salbutamol ✕ gender interaction. R-salbutamol also lowered food intake and improved food conversion ratios in both sexes. The relative weights of breast muscle and leg muscle were significantly increased and the relative weight of the fat pad was significantly decreased in birds of both sexes given diets containing R-salbutamol. Carcass protein content increased and carcass fat content decreased but the differences were not statistically significant. A significant dose-response effect was observed for tissue concentrations of R-salbutamol in all tissues, except the fat. Performance and carcass parameters in chickens given the 5 mg/kg R-salbutamol diet were similar to those given the 10 mg/kg rac-salbutamol diet, but tissue concentrations of R-salbutamol were lower. Chickens given the 10 mg/ kg rac-salbutamol diet had higher tissue concentrations of salbutamol than chickens given the 10 mg/kg R-salbutamol diet and higher concentrations of S-salbutamol than R-salbutamol in liver and leg muscle. Overall, these results demonstrate that R-salbutamol is an effective repartitioning agent in broiler chickens.
