​Evaluation of Homologous Platelet Rich Plasma (PRP) Drop and Leucocyte Platelet Rich Fibrin (L-PRF) Membrane for Corneal Ulcer Healing in Dogs

Background: To evaluate use of homologous platelet rich plasma (PRP) drop and leucocyte-platelet rich fibrin (L-PRF) clot/membrane as regenerative therapy for healing of corneal ulcer in dogs and its subsequent comparison with conventional therapy. Methods: In this randomized study, 30 dogs were divided into three groups of 10 animals each. It was ensured each group had grade I, II and III ulcers. Group I received medicinal treatment with antimicrobial and non-steroidal anti-inflammatory eye drop. In Group II PRP drops and in Group III L-PRF membrane was used to evaluate corneal ulcer healing. Homologous platelet concentrates were prepared in laboratory and used for assessment of healing on various grades of corneal ulcer on basis on reduction in size and depth of ulcer, presence of stromal infiltration, scleral involvement, corneal opacity and vascularization. Result: Post treatment, the size of corneal ulcer started to reduce gradually from day 7 onwards. Significant (p≤0.05) reduction was observed from day 10 in group I after instillation of drops Moxifloxacin and Flurbiprofen and in group II after instillation of PRP. It was measured as 5.35 ± 0.63 and 5.20 ± 0.98 mm at day 0, which reduced significantly (p≤0.05) to 3.90 ± 0.50 and 3.10 ± 0.98 mm at day 10 in group I and group II respectively. In group III there was significant reduction in size of ulcer from day 0 to day 7 measuring 7.80 ± 1.05 and 0.20 ± 0.11mm respectively. Thereafter, from day 10 onwards complete sealing of ulcer was observed in all the cases. Post treatment there was no significant reduction in corneal opacity from day 0 to day 30 in group I dogs and corneal opacity persisted specially in grade II and grade III ulcers. There was significant reduction (p≤0.05) in opacity from 2.90 ± 0.35 to 2.10 ± 0.38 and 3.00 ± 0.37 to 2.40 ± 0.34 in group II and group III dogs from day 0 to day 10 respectively. In group I, II and III there was significant (p≤0.05) reduction in vascularization from 1.50±0.30 to 1.20±0.13, 1.30±0.37 to 0.60±0.22 and 1.40±0.31 to 0.40±0.31 from day 0 to day 15 respectively. There was a significant reduction in vascularization on day 15 in group II and group III compared to group I animals.

ABSTRACT Objective: To evaluate the efficacy of coronally advanced flap (CAF) procedure under microsurgical approach for the management of Miller's Class I and II gingival recession defects with the use of either platelet-rich fibrin (PRF) or amnion membrane (AM) in comparison to CAF alone. Materials and Methods: A total of 45 sites with Miller's Class I or II gingival recession defect were randomly distributed for: Experimental Group I (CAF with PRF) sites (n = 15) which were treated with the microsurgical approach using CAF along with PRF; experimental Group II (CAF with AM) sites (n = 15) were treated with the microsurgical approach using CAF along with AM; control Group III (CAF alone) sites (n = 15) were treated with the microsurgical approach using CAF alone. Vertical gingival recession (VGR), horizontal gingival recession (HGR), gingival thickness (GT) (using transgingival probing [TGP] and ultrasonography [USG]) and patients’ response and acceptance were documented at baseline, 3 months and 6 months after surgical interventions. Results: CAF alone and in combination with PRF or AM, were effective techniques for root coverage with average VGR values of 1.47 ± 0.92 mm (56%), 0.67 ± 1.23 mm (36%) and 0.60 ± 1.06 mm (33%) in Group I (CAF with PRF), Group II (CAF with AM), and Group III (CAF alone), respectively. Complete coverage (100%) was obtained in 33.3% sites of Group I (CAF with PRF), 26.6% sites of Group II (CAF with AM) and 13.3% in Group III (CAF alone). Patients’ response and acceptance for surgical treatment modality in terms of patient esthetic score and decrease in hypersensitivity score was highest for Group I (CAF with PRF), whereas patient comfort score was highest for Group II (CAF with AM). At 6 months follow-up, significant increase in GT measurements (using TGP and USG) in Group I (CAF with PRF), whereas, nonsignificant increase for Group II (CAF with AM) and no change or decrease for Group III (CAF alone) as compared to baseline was observed. Conclusion: The present study observed enhancement in root coverage when PRF or AM are used in conjunction with CAF as compared to CAF alone. These results are based on 6-month followup. Therefore, the long-term evaluation may be necessary to appreciate the clinical effect of autologous PRF and AM.

The used of monosodium glutamate (MSG) oftenly found in almost every food that we consumed everyday. It suggested that MSG can increase appetite, can caused intake much foods, and increased the bodyweight. The aim of study is to prove the changes that occur in male mice by induce of MSG. The experimental study used 24 male mice, 30-45 gram, 12-14 weeks divided into 4 groups, ie group-I (negative control/placebo given aquadest 0.2 cc/20gBW mice); group-II, MSG with the dose of 5 mg/gBW mice); group-III, MSG with the dose of 10 mg/gBW mice and group-IV, MSG with the dose of 20 mg/gBW mice. All of these treatments were administered orally for 14 days. Permanent cervical fracture execution was performed at the end of the study. The body weight measured in pre and post treatment. The significant difference of data between different groups was compared by ANOVA followed by Tukey test. The compared of between pre –post treatment used paired test to analyze the changes of bodyweight This study proved that the comparison of mean values ± SEM of body weight between groups were not significantly different; group I (39,83±3,26), group II (39,33±2,04), group III (38,00±1,39) and group IV (38,33±1,76) (p = 0.930) and the changes of bodyweight pre and post treatmen also not significantly different, whereas p > 0.05. The study concluded that no changes in the body weight between groups and pasca induced by MSG. In the future studies, we consider to examining the effects MSG with longer of duration and more variations doses in MSG.

<p><strong>Objective: </strong>The purpose of this study was to perform an experimental quantitative comparison of primary teeth enamel alterations under three commonly prophylaxis surface polishing treatments. <strong>Material and </strong><strong>Methods: </strong>36 healthy primary teeth naturally exfoliated were selected and randomly separated in three groups. Group I was treated with a rotary instrument set at a low speed, rubber cup and a mixture of water and pumice; group II with a rotary instrument set at a low speed, rubber cup and prophylaxis paste Herjos-F (Vigodent S/A Indústria e Comércio, Rio de Janeiro, Brazil); and group III with sodium bicarbonate spray Profi II Ceramic (Dabi Atlante Indústrias Médico Odontológicas Ltda, Ribeirão Preto, Brazil). All procedures were performed by the same operator for ten seconds, and samples were rinsed and stored in distilled water. Pre-and post-treatment surface evaluation was completed using a surface profilometer Mitutoyo SJ400. <strong>Results: </strong>The results of this study were statistically analyzed with the GraphPad PRISM (version 6, 2010). The pumice and water led to significantly rougher surfaces than other groups in Tukey’s test (Group I: 1.22 Ra; Group II 0.38 Ra; Group III: 1.01 Ra). Pre-and post-treatment comparison revealed significantly rougher enamel surface with pumice. <strong>Conclusion: </strong>Based on this study, it can be concluded that there was an increased enamel surface roughness when teeth were treated with pumice and water in comparison to bicarbonate spray and prophylaxis paste.</p><p><strong>Keywords</strong></p><p><strong></strong>Dental enamel; Dental prophylaxis; Jet abrasive system; Enamel roughness; Primary teeth.<strong> <br clear="all" /> </strong></p>

Mutans streptococci (MS) are one of the most virulent cariogenic pathogens in the oral cavity. The effects of three oral hygiene techniques on salivary mutans streptococci levels were evaluated to see which mechanism would be most efficient in reducing salivary MS levels. Sixty patients, all in the permanent dentition, were selected and randomly distributed into three groups. Group I was asked to use a tongue scraper to brush the tongue once daily in the morning after normal tooth brushing routine. Group II was asked to place and let dissolve a Listerine Oral Care StripTM on the tongue once daily after normal oral hygiene routine. Group III was asked to rinse once every morning, for thirty seconds, with a saturated saline solution after routine tooth brushing. The instructions were done for a period of seven days. Baseline, one hour, three day, and seven day saliva samples were obtained and plated on CRT" by Vivadent MS-sensitive agar. All treatments groups show a significant reduction in colony counts from baseline and one or more post treatment periods and at one or more time periods between treatment groups. The most effective treatment in reducing colony counts was seen within Group I "Tongue Scraping" which demonstrated the greatest change from baseline to each of the post treatment periods. The least effective was Group II "Listerine Strip" which showed a statistically insignificant increase in colony count from baseline to 1-Hour and a significant decrease from baseline at the 7-day period only.

The current study was designed to evaluate the effects of autologous platelets rich plasma (PRP) on the articular cartilage repair. The present study was conducted on twenty-four adult male rabbits. Surgically (24) animals have received the same defect (5mm in diameter) which is made on the lateral condyle of the distal end of the femoral bone. The animals were divided randomly into three equal groups. Group (I) remain without treatment as a positive control group, group (II) which treated with a single dose of prepared PRP injected into the intraarticular space immediately after the creation of the of defect, group (III) treated with two doses of PRP (the first dose administered immediately after the surgery and second dose after (15) days of the first dose. Results were evaluated through the Biochemical analysis of the alkaline phosphatase (ALP) and Ca2+ at 15, 30, and 60 days after treatment. The biochemical findings of the current study showed that the total ALP was not significantly differed in animals of a group (II, III), whereas animals of a group (I) showed an elevation of ALP in the last period which indicates the osseous reaction especially in late stages of cartilage repair; however, the other two groups (II, III) didn’t show a great change in ALP levels in all periods of blood sample collection. The Ca+ levels also showed no significant differences among groups, only in the last period in the control positive group, so we concluded that the increase in ALP, and Ca+ levels in control group is an evidence of continues inflammatory process, and increase ability of ossification of the new tissue formed in the site of the lesion.

C57BL male mice (Jackson Lab., Bar Harbor, Maine) weighing about 18 gms were randomly divided into three groups: group I was fed sweetened liquid alcohol diet (modified Schenkl) in which 36% of the calories were derived from alcohol; group II was maintained on a similar diet but alcohol was isocalorically substituted by sucrose; group III was fed regular mouse chow ad lib for five months. Liver and heart tissues were fixed in 2.5% cacodylate buffered glutaraldehyde, post-fixed in 2% osmium tetroxide and embedded in Epon-araldite.

SummaryMost anticardiolipin antibodies (ACA) associated with antiphospholipid syndrome (APS) are directed against epitopes expressed on β2-glycoprotein I (β2GPI). Despite a good correlation between standard ACA assays and those using purified human β2GPI as the sole antigen, some sera from APS patients only react in the latter. This is indicative of heterogeneity in anti-β2GPI antibodies. To characterize their reactivity profiles, human and bovine β2GPI were immobilized on γ-irradiated plates (β2GPI-ELISA), plain polystyrene precoated with increasing cardiolipin concentrations (CL/β2GPI-ELISA), and affinity columns. Fluid-phase inhibition experiments were also carried out with both proteins. Of 56 selected sera, restricted recognition of bovine or human β2GPI occurred respectively in 10/29 IgA-positive and 9/22 IgM-positive samples, and most of the latter (8/9) were missed by the standard ACA assay, as expected from a previous study. Based on species specificity and ACA results, IgG-positive samples (53/56) were categorized into three groups: antibodies reactive to bovine β2GPI only (group I) or to bovine and human β2GPI, group II being ACA-negative, and group III being ACA-positive. The most important group, group III (n = 33) was characterized by (i) binding when β2GPI was immobilized on γ-irradiated polystyrene or cardiolipin at sufficient concentration (regardless of β2GPI density, as assessed using 125I-β2GPI); (ii) and low avidity binding to fluid-phase β2GPI (Kd in the range 10–5 M). In contrast, all six group II samples showed (i) ability to bind human and bovine β2GPI immobilized on non-irradiated plates; (ii) concentration-dependent blockade of binding by cardiolipin, suggesting epitope location in the vicinity of the phospholipid binding site on native β2GPI; (iii) and relative avidities approximately 100-fold higher than in group III. Group I patients were heterogeneous with respect to CL/β2GPI-ELISA and ACA results (6/14 scored negative), possibly reflecting antibody differences in terms of avidity and epitope specificity. Affinity fractionation of 23 sera showed the existence, in individual patients, of various combinations of antibody subsets solely reactive to human or bovine β2GPI, together with cross-species reactive subsets present in all samples with dual reactivity namely groups III and II, although the latter antibodies were poorly purified on either column. Therefore, the mode of presentation of β2GPI greatly influences its recognition by anti-β2GPI antibodies with marked inter-individual heterogeneity, in relation to ACA quantitation and, possibly, disease presentation and pathogenesis.

<p class="p1"><span class="s1"><strong>Background:</strong> The inspiratory oxygen fraction (FiO<sub>2</sub>) is usually set between 60% and 100% during conventional extracorporeal circulation (ECC). However, this strategy causes partial oxygen pressure (PaO<sub>2</sub>) to reach hyperoxemic levels (&gt;180 mmHg). During anesthetic management of cardiothoracic surgery it is important to keep PaO<sub>2</sub> levels between 80-180 mmHg. The aim of this study was to assess whether adjusting FiO<sub>2</sub> levels in accordance with body temperature and body surface area (BSA) during ECC is an effective method for maintaining normoxemic PaO<sub>2</sub> during cardiac surgery.</span></p><p class="p1"><span class="s1"><strong>Methods:</strong> After approval from the Ethics Committee of the University of Acıbadem, informed consent was given from 60 patients. FiO<sub>2</sub> adjustment strategies applied to the patients in the groups were as follows: FiO<sub>2</sub> levels were set as 0.21 × BSA during hypothermia and 0.21 × BSA + 10 during rewarming in Group I; 0.18 × BSA during hypothermia and 0.18 × BSA + 15 during rewarming in Group II; and 0.18 × BSA during hypothermia and variable with body temperature during rewarming in Group III. Arterial blood gas values and hemodynamic parameters were recorded before ECC (T1); at the 10th minute of cross clamp (T2); when the esophageal temperature (OT) reached 34°C (T3); when OT reached 36°C (T4); and just before the cessation of ECC (T5).</span></p><p class="p1"><span class="s1"><strong>Results:</strong> Mean PaO<sub>2</sub> was significantly higher in Group I than in Group II at T2 and T3 (<em>P</em> = .0001 and <em>P</em> = .0001, respectively); in Group I than in Group III at T1 (<em>P</em> = .02); and in Group II than in Group III at T2, T3, and T4 <br /> (<em>P</em> = .0001 for all). </span></p><p class="p1"><span class="s1"><strong>Conclusion: </strong>Adjustment of FiO<sub>2</sub> according to BSA rather than keeping it at a constant level is more appropriate for keeping PaO<sub>2</sub> between safe level limits. However, since oxygen consumption of cells vary with body temperature, it would be appropriate to set FiO<sub>2</sub> levels in concordance with the body temperature in the <br /> rewarming period.</span></p>

The article presents the comparative analysis of the state of the cervix in women with endometrial polyps and micropolyps. Patients and methods. The study involved 130 patients aged 18-35 years: 70 patients with endometrial polyps (group I), 30 patients with micropolyps (group II) and 30 patients of the control group (group III). Results. According to the anamnesis of women in the I group were significantly more frequent diseases of the cervix, which corrected physical surgery methods, in particular cryodestruction. In group II, the representatives of these indicators were similar to healthy. Normal colposcopic picture met significantly less frequently in patients and I, and II group. The differences in the incidence of HPV high oncogenic risk in all groups were not statistically significant. Conclusion. Destructive methods used in the detection of any changes in the cervix are often overly aggressive, form scars and contributing to inflamaciones process. In the chain of events leading to the formation of PE, cervical pathology and its correction can take the basic place. Key words: endometrial polyp, micropolyps, chronic endometritis, uterine cervix, colposcopy.

Изучено влияние кормовой добавки в форме индивидуальных болюсов Метраболь на возможность снижения негативного влияния субклинического кетоза (СКК) и результативность искусственного осеменения высокопродуктивных молочных коров в посттранзитный период. Животные были подобраны по принципу условных аналогов и разделены на 3 группы в зависимости от концентрации β-оксимасляной кислоты в их крови. В I группе (n=18) у коров не было СКК, концентрация кислоты составляла <1,0 ммоль/л, во II группе (n=21) у животных зарегистрирован СКК, концентрация кислоты — 1,0…1,4 ммоль/л. С целью поиска решения по снижению негативного влияния СКК на репродуктивные показатели была сформирована III группа коров, переболевших СКК (n=24) с концентрацией β-оксимасляной кислоты в крови 1,0…1,4 ммоль/л, но с использованием болюсов, применяемых для ускоренного восстановления репродуктивной функции коров после отела. Важной составляющей опыта явилось то, что к моменту гормональной синхронизации у коров всех 3 групп фиксировалось отсутствие СКК (снижение β-оксимасляной кислоты до уровня <1,0 ммоль/л). Для синхронизации полового цикла выбрана схема Пресинх-овсинх. Установлено, что гормональная синхронизация животных, перенесших СКК, в сравнении со здоровыми, не позволяет снизить негативное влияние данного нарушения обмена веществ. Во II группе у животных зафиксированы самые низкие репродуктивные показатели (количество использованных доз; индекс осеменения, сервис-период; время от первого до успешного осеменения; результат первичного осеменения; количество животных, осемененных 3 и более раз). В III группе применение болюсов позволило, в сравнении с животными II группы, не только избежать отрицательного воздействия СКК, но и превзойти по изучаемым параметрам животных из I группы. The effect of the feed additive in the form of individual boluses Metrabol on the possibility of reducing the negative effect of subclinical ketosis (CCK) and the effectiveness of artificial insemination of highly productive dairy cows in the post-transit period was studied. The animals were selected according to the principle of conditional analogs and divided into 3 groups depending on the concentration of β-hydroxybutyric acid in their blood. In group I (n=18), the cows did not have SSC, the acid concentration was <1.0 mmol/L, in group II (n=21), SSC was registered in animals — 1.0...1.4 mmol / L. In order to find a solution to reduce the negative effect of CCB on reproductive indices, a group III of cows was formed who had CCB (n = 24) with a concentration of β-hydroxybutyric acid in their blood of 1.0...1.4 mmol/L, but using boluses, used to accelerate the restoration of the reproductive function of cows after calving. An important component of the experiment was that by the time of hormonal synchronization, cows of all 3 groups had no SSC (decrease in β-hydroxybutyric acid to a level of <1.0 mmol/L). To synchronize the sexual cycle, the Pressinh-ovsinh scheme was chosen. It was found that hormonal synchronization of animals that underwent SSC, in comparison with healthy ones, does not allow reducing the negative effect of this metabolic disorder. In group II, animals had the lowest reproductive indices (number of doses used; insemination index, service period; time from first to successful insemination; result of primary insemination; number of animals inseminated 3 or more times). In group III, the use of boluses made it possible, in comparison with animals of group II, not only to avoid the negative effect of SSC, but also to surpass the animals from group I in terms of the studied parameters.
