Neurometabolic disorders are an important group of diseases that mostly occur in neonates and infants. They are mainly due to the lack or dysfunction of an enzyme or cofactors necessary for a specific biochemical reaction, which leads to a deficiency of essential metabolites in the brain. This, in turn, can cause certain neurometabolic diseases. Disruption of metabolic pathways, and the inhibition at earlier stages, may lead to the storage of reaction intermediates, which are often toxic to the developing brain. Symptoms are caused by the progressive deterioration of mental, motor, and perceptual functions. The authors review the diseases with microcephaly, which may be one of the most visible signs of neurometabolic disorders.

AbstractThe insertion sequence IS26 has long been known to play a major role in the recruitment of antibiotic resistance genes into the mobile resistance gene pool of Gram-negative bacteria and IS26 also plays a major role in their subsequent broad dissemination. Related IS, IS431/257 and IS1216 are important in the same roles in Gram positive bacteria. However, until recently the properties of IS26 movement that could potentially explain this ability had not been explored. A much needed insight has come from our recent demonstration that IS26 uses a novel targeted mechanism that is conservative. The targeted conservative mechanism is much more efficient than the known replicative mechanism, which is now more accurately called copy-in. A recent review “The IS6 family, a clinically important group of insertion sequences including IS26” by Varani, He, Siguier, Ross and Chandler published in Mobile DNA has substantially misrepresented the recent studies on the targeted conservative mechanism and at the same time incorrectly implied that any mechanism established for IS26 can be assumed to apply to a range of IS that are at best very distantly related. A few of the most important issues are examined in this comment. Readers are advised to consult the original literature to check facts before drawing firm conclusions.

AbstractThe IS6 family of insertion sequences is a large but coherent group which was originally named to avoid confusion between a number of identical or nearly identical IS that were identified at about the same time and given different names (IS15D, IS26, IS46, IS140, IS160, IS176). The underlying common mechanistic feature of all IS6 family members which have been investigated is that they appear to transpose by replicative transposition and form pseudo compound transposons with the flanking IS in direct repeat and in which associated genes are simply transferred to the target replicon and lost from the donor.In the accompanying letter Hall raises a number of very serious and wide-ranging criticisms of our recent review article concerning the IS6 family of insertion sequences. She clearly feels that we have undervalued her work and that we question or ignore certain of her in vivo results. This impression is almost certainly the result of the standard of proof we generally apply to mechanistic aspects of transposition where we think it important to identify transposition intermediates including the types of synaptic, strand cleavage and strand transfer complexes involved.

Taking into consideration the gradually widespread use of the internet, children are thought to be an important group both in terms of the time they spend on the internet and the dangers they will be exposed to in the virtual world. Internet addiction is essentially a type of behavioral addiction, characterized by a certain behavioral pattern, which is confronted with both physical, psychological, and social consequences. The research aims to establish a correlation between children's addiction to computer games with the cyber violence that they may be exposed to base on this addiction. For this reason, this study is a systematic review that aims to examine the effects of computer addiction as a subtype of internet addiction, which is a type of behavioral addiction, on children and its relationship with cyber violence. The research significantly contributes to the literature by revealing the effects of COVID-19 on children's addiction to computer games and the correlation of children's addiction to computer games with cyber violence.

The closed-nested honey bees are an important group that has been successfully bred traditionally and in a modern way. The traditional honey beekeeping practices are still favorable by local people living near natural habitats. Many rural areas in Indonesia are well known as producers of honey from the traditional honey collecting and traditional honey beekeeping of the closed-nested honey bees. However, there is limited information on the diversity of the honey bees that had supported the honey productions and their traditional honey beekeeping. This research was to provide an overview of the diversity of the honey bee species that are used in the wild honey collecting and their traditional honey beekeeping in four selected study sites in the islands of Java, Bawean, Kalimantan, and Peleng. We recorded three species of closed-nested native honey bees in the traditional honey collecting and traditional honey beekeeping, namely Apis cerana, A. koschevnikovi, and A. nigrocincta. We observed that traditional beekeeping of A. cerana was carried out in Tasikmalaya and Bawean Island, and that of A. cerana and A. koschevnikovi were carried out in Kayan Hilir. On Peleng Island, people do not do beekeeping but collect honey directly from the forest. Honey collecting and beekeeping practices are related to changes in the seasons of the flowering period in their habitats. The knowledge of the flowering period is needed to know the seasonal movement of honey bees from forest to village and vice versa.

Covid-19 pandemic created a new environment to New Green Deal - flagship initiative of European Commission and crucial element of Ursula von der Leyen political manifesto. Author argues, that faced with the possibility of weakening or postponing ambitious environmental programme because of new challenges the commission, backed by important group pf member states decided to build a direct link between reconstruction after pandemics and greening of the economy. The result was a push forward with decarbonisation commitments  - the stimuli for change should be bound together to strengthen  their interdependence and build a momentum for modernization of EU in both economic and political dimensions. Conditionality of Next Generation Europe financial instrument is one of the links between these two dimensions.

Chromone glycosides comprise an important group of secondary metabolites. They are widely distributed in plants and, to a lesser extent, in fungi and bacteria. Significant biological activities, including antiviral, anti-inflammatory, antitumor, antimicrobial, etc., have been discovered for chromone glycosides, suggesting their potential as drug leads. This review compiles 192 naturally occurring chromone glycosides along with their sources, classification, biological activities, and spectroscopic features. Detailed biosynthetic pathways and chemotaxonomic studies are also described. Extensive spectroscopic features for this class of compounds have been thoroughly discussed, and detailed 13C-NMR data of compounds 1–192, have been added, except for those that have no reported 13C-NMR data.

Pyrazines (1,4-diazirines) are an important group of natural products that have tremendous monetary value in the food and fragrance industries and can exhibit a wide range of biological effects including antineoplastic, antidiabetic and antibiotic activities. As part of a project investigating the secondary metabolites present in understudied and chemically rich Actinomycetes, we isolated a series of six pyrazines from a soil-derived Lentzea sp. GA3-008, four of which are new. Here we describe the structures of lentzeacins A-E (1, 3, 5 and 6) along with two known analogues (2 and 4) and the porphyrin zincphyrin. The structures were determined by NMR spectroscopy and HR-ESI-MS. The suite of compounds present in Lentzea sp. includes 2,5-disubstituted pyrazines (compounds 2, 4, and 6) together with the new 2,6-disubstituted isomers (compounds 1, 3 and 5), a chemical class that is uncommon. We used long-read Nanopore sequencing to assemble a draft genome sequence of Lentzea sp. which revealed the presence of 40 biosynthetic gene clusters. Analysis of classical di-modular and single module non-ribosomal peptide synthase genes, and cyclic dipeptide synthases narrows down the possibilities for the biosynthesis of the pyrazines present in this strain.
