Abstract-Diabetes is a defect in the ability of the body to convert glucose (sugar) to energy. Glucose is the main source of energy in our body. When food is digested it is metabolized into fats, proteins, or carbohydrates. Glucose is then transferred to the blood and is used by the cells for energy production. To investigate the antidiabetic effect ethanolic extracts of flowers of Moringa oleifera against dexamethasone induced insulin resistance in wistar albino rats. To study the antidiabetic effect, flowers ofMoringaoleiferawerecollectedandauthenticated, extracted and investigated for acute toxicity and dexamethasone induced hyperglcemia. The animals treated with EEMOF at a dose of 100mg/kg and 200mg/kg prevented the development ofhyperglycemia,hypercholesteremiaandhypertriglyceridemia in dexamethasone induced insulin resistance models. Oral administration of Moringa Oleifera 100mg/kg and 200mg/kg reduces serum glucose, triglyceride, total cholesterols and LDLconcentration and improve the concentration of HDLin dexamethasone administered rats. The lignin Moringa Oleifera showed significant anti-diabetic effect in rats after oral administration. The present study demonstrated that Moringa Oleifera could be useful in Management of diabetes associated with abnormalities in lipid profiles. Further study need to isolate, identify the active compounds and find out thepossiblemechanismofactions.

Moringa oleifera leaf have been used for treatment of diabetes, in this work we studied pancreatic gene expression. The gene expression was done by PCR machine. The plant was harvested from garden within Madonna University and was identified in the Department of Pharmacognosy of the University by Dr P. Osuagwu. Each of the extracts was analyzed for bioactive components using Gas Chromatography–Mass Spectrometry (GC-MS). Male wistar albino rats (n=40) six weeks old weighing 150-250g were purchased from the animal farm of Madonna University Elele. From this work we observed that, there was significant increase in the expression of ADIPOQ gene in the diabetic rats treated with 300mg/kg of M. oleifera leaf (1.52±0.13), when compared with that of the non-diabetic rats (1.00±0.02). From this study, it could be inferred that M. oleifera leaf powder contains polyphenols that inhibit carbohydrate metabolism, reduced blood glucose level, enhance expression ADIPOQ genes.

Background: Moringa oleifera (M. oleifera) is a commonly used medicinal and nutritive plant. The aqueous leaf extract from this plant contains a high concentration of alkaloids and they are toxic to body organs especially the liver leading to hepatotoxicity after long term exposure. However, if N-Acetyl cysteine (NAC) is administered together with M. oleifera extract, it may have a hepatotoxic protective effect. Objective: To establish the protective effect of N-Acetyl cysteine against M. oleifera aqueous leaf extract-induced hepatotoxicity in the Wistar albino rats. Methods: An experimental laboratory-based study conducted at department of Physiology Makerere University, College of Health Sciences. Three treatment groups of six Wistar albino rats each, were dosed intragastrically once a day for 28 days. Group I; negative control, received 8.05g/kg bwt of M. oleifera extract plus 1ml of normal saline (NS), Group II; test group, received 8.05g/kg bwt of M. oleifera extract plus 50mg/kg of NAC. Group III; positive control, received 750mg/kg bwt of Paracetamol plus 50mg/kg of NAC. On the 14th and 29th day, three animals selected randomly from each group were sacrificed; blood samples were collected, the liver was harvested for histopathological analysis.  Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), bilirubin and total protein levels were determined. Results: There was an increase in serum ALT, AST and ALP levels in the M. oleifera extract plus NS group which was a sign of hepatotoxicity. The M. oleifera extract plus NAC group showed normal serum ALT, AST and ALP levels with no significant changes in the bilirubin (P-value = 0.9089) and total protein levels (P-value = 0.8858). Conclusion: The results have provided evidence that NAC administration with M. oleifera extract effectively prevents the occurrence of M. oleifera leaves xtract- induced hepatotoxicity.
