Effects of the Combined Treatment of Ethanol Extract of Moringa oleifera and Pleurotus ostreatus on Blood Glucose, Amylase and Lipid Profile of Alloxan-induced Diabetic Wistar Albino Rats

Objective: The aim of this study was to investigate the effect of ethanol extracts of stem and leaf of Bacolepis nervosa as antihyperglycemic, anti-hyperlipidemic and antioxidant activity in alloxan-induced diabetic rats. Methods: Diabetes was induced in wistar albino rats by administration of alloxan monohydrate (150 mg/kg). The ethanol extract of B. nervosa leaf and stem at a dose of 150 and 300 mg/kg body weight was administrated at a single dose per day to diabetes-induced rats for a period of 14 d. The effect of ethanol extract of B. nervosa leaf and stem on blood glucose, insulin, urea, creatinine, HbA1C, serum protein, albumin, globulin, serum enzymes, serum lipid profiles, lipid peroxidase (LPO) and antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and reduced glutathione (GSH) were measured in the diabetic rats.Results: The ethanol extract of B. nervosa stem and leaf elicited significant reduction in blood glucose (p<0.001), serum enzymes (SGPT, SGOT, ALP) (p<0.01), lipid parameters (TC, TG, VLDL-LDL, PL) (p<0.01) except HDL-C and significantly increased insulin (p<0.01), HDL-C (p<0.05),, GPx, GSH, SOD and CAT (p<0.05) at the dose of 300 mg/kg when compared with the diabetic-induced control.Conclusion: From the above results, it is concluded that ethanol extracts of B. nervosa leaf and stem possesses significant antihyperglycemic, antihyperlipidemic and antioxidant effect in alloxan induced diabetic rats.

Aim: The role of nutrition in lipid metabolism continues to generate a lot of research interest, especially as dyslipidaemia is implicated in a host of diseases. The work investigated the effect of ethanol extract of  Curcubita pepo leaves on the lipid profile of Wister rats. Methodology: A total of 18 rats divided into three groups of 6 rats each were employed in the investigation. The first group (baseline) was sacrificed after purchase; second group (control) was fed rat chow, and the third group (test) was fed a composite feed containing rat chow and 10% extract of C. pepo leaves.  Results: The lipid profile (total cholesterol, triglycerides, LDL, HDL and VLDL) of the rats was assayed after 18 days of feeding and the result showed no significant difference (P>0.05) between the test group and control for the lipids assayed. However, there was a marked increase in the HDL level of the test group (0.500±0.057 mmol/L) compared to the control (0.268±0.043 mmol/L), it was nonetheless statistically non-significant (P=0.068).  The HDL increasing effect observed may stem from the rich phytochemistry of the leaves, as preliminary phytochemical investigation showed the leaves to contain saponin, tannin, alkaloids, flavonoids and glycosides. Conclusion: We conclude that the leaves of C. pepo may play a medicinal role in maintaining lipid homeostasis.

Moringa oleifera, popularly known as “miracle tree” belongs to the family, Moringaceae. It is a medicinal plant in which the leaves are the most nutritious part, being a significant source of vitamins and protein among others. This study was conceived and designed based on the gaps in the research that has been performed and what is known about the plant. In this study, the effect of Moringa oleifera leaves extract on alloxan induced diabetes in Wistar albino rats was investigated. A total of forty five (45) rats were acclimatized for a period of two weeks, then randomly divided into five (5) groups (1, 2, 3, 4, and 5) of  nine (9) rats each and fed with standard feed and water. Group 1 which is the control was fed with just water and standard feed while Hyperglycemia was induced in groups 2, 3, 4, & 5 intra-peritoneally after an over-night fasting using alloxan at a concentration of 130 mg/kg b.w. and allowed for 48hours which resulted in a high blood glucose level between 300 mg/dl and 600 mg/dl. Group 2 was not given any treatment while Groups 3, 4, & 5 were treated with doses 100 mg/kg b.w., 200 mg/kg bw, and 400 mg/kgbw of Moringa oleifera leaf extract respectively for a period of four weeks. A glucometer was used to check the blood glucose level of the animals before and after treatment. The results of Groups 3, 4, & 5 (172.0±4.75 mg/dl, 142.9±47.25 mg/dl, 70.6±24.46 mg/dl respectively) showed a significant decrease (p< 0.05) in blood glucose level of the induced rats when compared with Group 2 (316±47.17 mg/dl) which was induced only alloxan. It can therefore be concluded that this study has shown that the extract of Moringa oleifera leaves offers an anti-diabetic effect in Wistar albino rats.

Sonchus arvensis L. leaves have been used traditionally to treat various disease conditions. This study is designed to evaluate cardioprotective potential of ethanol extract of S. arvensis leaves on isoproterenol-induced myocardial infarction in Wistar rat. Male Wistar albino rats were divided into three main groups: negative control (saline only), positive control (isoproterenol only), and S. arvensis extract treated groups. S. arvensis extract was administered in three doses; 50, 100, and 200 mg/kg b.w. p.o for 14 days. On day 13 and 14, isoproterenol (85 mg/kg bw) was given intraperitoneally to positive control and extract treated groups. The parameters studied were cardiac biomarker enzymes which were Creatine Kinase (CK), Creatine Kinase-MB (CK-MB), Aspartate Transaminase (AST), Alanine Transaminase (ALT) and Lactate Dehydrogenase (LDH). The results showed that S. arvensis at dose of 50 mg/kg b.w. could significantly (P<0.05) reduce the level of CK, CK-MB, AST, ALT, and LDH in myocardial infarcted rats compared to positive control. The increase of the dose of S. arvensis extract was not followed by an increase of its cardioprotective activity. In conclusion, Sonchus arvensis L. leaves extract at dose of 50 mg/kg b.w. has potential to be developed as cardioprotective drug.
