Garcinia kydia Roxburgh merupakan salah satu spesies dari genus garcinia, yang secara khusus belum banyak diketahui aktivitas senyawa bioaktif, namun pada penelitian sebelumnya terbukti berpotensi sebagai agen penghambat α-glukosidase. Pada penelitian ini, akan dilakukan investigasi aktivitas antioksidan terhadap berbagai fraksi yang diperoleh dari daun G. kydia Roxb. Metode penelitian dilakukan dengan menentukan aktivitas antioksidan melalui analisis in-vitro menggunakan microplete reader dengan metode radical scavenging (Metode DPPH) dan ferric reducing antioxidant power (Metode FRAP). Hasil menunjukkan bahwa fraksi yang paling aktif dari ekstrak etil asetat adalah FEA8 (DPPH) dan FEA7 (FRAP), sedangkan fraksi paling aktif dari ekstrak metanol adalah FMT4 (DPPH) dan FMT6 (FRAP). Aktivitas antioksidan tertinggi di antara empat fraksi adalah FEA7 dan FEA8. Aktivitas fraksi FEA7 dan FEA8 ditentukan oleh nilai EC50, sehingga diperoleh masing-masing 9,73 dan 9,26 μg/mL (DPPH); 10,28 dan 13,94 μg/mL (FRAP). Garcinia kydia Roxb. berpotensi sebagai agen antioksidan dengan mekanisme yang berbeda. Sehingga hasil penelitian terkait terdapatnya potensi antioksidan tersebut dapat memberikan alternatif terapi diabetes yang disertai komplikasi stres oksidatif.

AbstractWe conducted this study to determine and compare the content of phenolic compounds and flavonoids in the argan and olive leaves as well as their antioxidant capacity in aqueous, methanolic, and ethyl acetate extracted fractions. In vitro antioxidant activity was evaluated in comparison with synthetic antioxidants by assessing DPPH• radical scavenging capacity, ferric reducing antioxidant power, scavenging ability by inhibiting the β-carotene/linoleic acid emulsion oxidation, and by the ABTS radical scavenging activity assay. Total phenolic content in argan samples ranged from 221.69 ± 2.07 to 1.32 ± 0.01 mg GAE/g DW and in olive samples from 144.61 ± 0.82 to 1.21 ± 0.02 mg GAE/g DW. Total flavonoids content in argan samples varied from 267.37 ± 1.12 to 25.48 ± 0.02 mg QE/g DW, while in olives from 96.06 ± 0.78 to 10.63 ± 0.05 mg QE/g DW. In vitro antioxidant studies strongly confirmed the antioxidant potency of argan and olive leaves and their richness in secondary metabolites that are effective in free radicals scavenging and metal chelating capacities, indicating their antioxidant power.

Syzygium caryophyllatum L. Alston (Family: Myrtaceae, Sinhala: Heendan) is a red-listed plant that has been used in traditional medicine in Sri Lanka for the treatment of diabetes, but it is yet to be exploited for its potential uses as a functional food or a source of supplements. The present study focused on the evaluation of antidiabetic property of S. caryophyllatum fruits and leaves assessing antioxidant, antiglycation, and antiamylase activities and functional mineral element composition. The crude extracts (CR) of leaves and fruits were fractionated into hexane (Hex) ethyl acetate (EA) and aqueous (AQ) and evaluated for bioactivities along with the crude extracts. The isolated fraction (C3) of Hex fraction of fruit showed significantly high (p<0.05) antiamylase activity with IC50 value 2.27 ± 1.81 μg/mL where the Hex fraction of fruits exhibited the IC50 value as 47.20 ± 0.3 μg/mL which was higher than acarbose (IC50: 87.96 ± 1.43 μg/mL). The EA fraction of leaves showed highest values for DPPH radical scavenging activity, ferric reducing antioxidant power, and oxygen radical absorbance capacity. Significantly high (p<0.05) ABTS radical scavenging activity and iron chelating activity were observed in Hex fraction of fruit. The composition of volatiles in leaf oil was studied with GC-MS, and 58 compounds were identified. Inductively coupled plasma-mass spectrometry data revealed the presence of biologically significant trace elements such as Fe, Zn, Mg, Cu, Se, and Sr in leaves and fruits. It is concluded that the Hex fraction of S. caryophyllatum fruits will be a good source for the formulation of supplements for diabetic management with further evaluation of potency and efficacy.

This study aimed to isolate, prepare and identify the main flavonoids from a standardized Smilax glabra flavonoids extract (SGF) using preparative HPLC, MS, 1H NMR and 13C NMR, determine the contents of these flavonoids using UPLC, then compare their pharmacological activities in vitro. We obtained six flavonoids from SGF: astilbin (18.10%), neoastilbin (11.04%), isoastilbin (5.03%), neoisoastilbin (4.09%), engeletin (2.58%) and (−)-epicatechin (1.77%). The antioxidant activity of six flavonoids were evaluated by determining the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and 2,2′-Azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS+) radical scavenging activity and ferric reducing antioxidant power (FRAP). In addition, the anti-inflammatory activity of six flavonoids were evaluated by determining the production of cytokines (IL-1β, IL-6), nitric oxide (NO) using enzyme linked immunosorbent assay and the NF-κB p65 expression using Western blotting in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The results showed that (−)-epicatechin, astilbin, neoastilbin, isoastilbin and neoisoastilbin had strong antioxidant activities, not only in DPPH and ABTS+ radicals scavenging capacities, but in FRAP system. Furthermore, all the six flavonoids could significantly inhibit the secretion of IL-1β, IL-6, NO (p < 0.01) and the protein expression of NF-κB p-p65 (p < 0.01) in LPS-stimulated RAW264.7 cells. This study preliminarily verified the antioxidant and anti-inflammatory activities of six flavonoids in S. glabra.

Objective: This research is to study the production of natural red pigment by Monascus purpureus TISTR3615 in the submerged and solid-statefermentation system using Pathumthani-1 rice as a carbon source.Methods: The antioxidant activity of the red pigment was evaluated in vitro 2,2-diphenyl-1-picrylhydrazyl (DPPH), ABTS radical scavenging assay,and ferric-reducing antioxidant power (FRAP) assay, including total phenolic compound.Results and Discussion: The maximum of red pigment production was 0.55±0.02/ml (OD 680 nm) after incubation at 30°C for 24 days. Theantioxidant activity based on inhibition DPPH (%), ABTS radical scavenging activity (%), and FRAP activity (mM Fe2+/g) was 97.80±1.51,68.64±0.46, and 0.32±0.021, respectively. The total phenolic content was 164.78±2.82 μg GAE/mg.Conclusion: It was estimated that Monascus pigments, leading to nutraceutical and pharmaceutical applications, cosmetic industry, and foodindustry.

Oxidative stress is the result of the disparity between pro-oxidants and antioxidants in an organism, and it is important in the pathogenesis of several degenerative disorders, such as arthritis, Alzheimer’s, cancer, and cardiovascular diseases. Free radicals can damage biomolecules, such as nucleic acids, lipids, proteins, polyunsaturated fatty acids, and carbohydrates, and the DNA leading to mutations. The use of antioxidants is effective in delaying the oxidation of biomolecules. Antioxidants are complexes found in the food that can retard or deter oxidation by preventing the initiation and propagation of oxidizing chain reactions. Medicinal plants have been used for centuries by man to manage diseases and have a host of antioxidant complexes. Traditionally, Caesalpinia volkensii, Vernonia lasiopus, and Acacia hockii have folkloric remedies against associated oxidative stress-mediated complications. However, the upsurge in its use has not been accompanied by scientific validations to support these claims. In this study, in vitro antioxidant activity of Caesalpinia volkensii, Vernonia lasiopus, and Acacia hockii collected from Embu County (Kenya) were determined by radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical in addition to ferric reducing antioxidant power analyzed against that of L-ascorbic acid as the standard. The obtained results revealed remarkable antioxidant activities of the studied plant extracts as evidenced by the low IC50 and EC50 values. These antioxidant activities could be due to the presence of antioxidants phytochemicals such as flavonoids, phenols, terpenoids, and saponins among others. Therefore, the therapeutic potential of this plant could be due to their antioxidant properties. This study recommends bioassay of the extracts against oxidative stress-related disorders for development of phytomedicine with antioxidant properties.

The best described pharmacological property of flavonoids is their capacity to act as potent antioxidant that has been reported to play an important role in the alleviation of diabetes mellitus. Flavonoids biochemical properties are structure dependent; however, they are yet to be thoroughly understood. Hence, the main aim of this work was to investigate the antioxidant and antidiabetic properties of some structurally related flavonoids to identify key positions responsible, their correlation, and the effect of methylation and acetylation on the same properties. Antioxidant potential was evaluated through dot blot, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, ABTS+radical scavenging, ferric reducing antioxidant power (FRAP), and xanthine oxidase inhibitory (XOI) assays. Antidiabetic effect was investigated throughα-glucosidase and dipeptidyl peptidase-4 (DPP-4) assays. Results showed that the total number and the configuration of hydroxyl groups played an important role in regulating antioxidant and antidiabetic properties in scavenging DPPH radical, ABTS+radical, and FRAP assays and improved bothα-glucosidase and DPP-4 activities. Presence of C-2-C-3 double bond and C-4 ketonic group are two essential structural features in the bioactivity of flavonoids especially for antidiabetic property. Methylation and acetylation of hydroxyl groups were found to diminish thein vitroantioxidant and antidiabetic properties of the flavonoids.

Objective: The intent  of this report  was to investigate the effect of aqueous root extract of Clerodendrum serratum L. for antioxidant activity using divergent models viz. DPPH scavenging assay, Superoxide scavenging assay and Ferric Reducing Antioxidant Power (FRAP) assay.Materials and Methods: The root of C. serratum was extracted using water. The yield of aqueous extract was 10%w/w. The outcome was examined statistically by the regression method.Results and discussions: The IC50 values are 85.43 µg/ml and 107.59 µg/ml for DPPH radical scavenging and Superoxide scavenging assay respectively whereas  FRAP showed significant reducing power activity with increased concentration of sample. The pilot study showed, a significant correlation existed between concentrations of the extract and percentage engrossment of free radicals.Conclusion: The antioxidant property may be corresponding to the polyphenols and flavonoids adjacent in the extract. These results clearly revealed that C. serratum might be effective against diseases analogous with free radical mediated. Keywords Clerodendrum serratum, DPPH, Superoxide, FRAP, Rutin, Antioxidant

Phytochemical investigations of Taxus baccata L. by successive chromatographic methods resulted in the isolation of the lignans lariciresinol (1), taxiresinol (2), 3´-demethylisolariciresinol- 9´-hydroxyisopropylether (3), isolariciresinol (4), and 3-demethylisolariciresinol (5) as well as taxoids. Compounds 1 - 5 were evaluated for their acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and lipoxygenase (LOX) inhibitory activities, which play a role in the pathogenesis of Alzheimer’s disease (AD), by in vitro spectrophotometric methods, while they were also screened for their antioxidant capacity in 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging, ferrous ion-chelating effect, and ferric-reducing antioxidant power (FRAP) at 125, 250, 500, and 1000 μg ml-1. All compounds exhibited a moderate inhibition against both BChE and LOX, whereas they were inactive towards AChE. The compounds displayed a great scavenging activity against DPPH especially at 500 and 1000 μg ml-1. Besides, they were found to exert noteworthy reducing antioxidant power on ferric ions. In particular, the FRAP of compounds 2 (3.552 ± 0.02), 4 (3.021 ± 0.71), and 5 (3.533 ± 0.01) were as high as that of the reference chlorogenic acid (3.618 ± 0.01) at 1000 μg ml-1. None of the compounds exhibited chelating ability against ferrous ions.

The decoction of Tessaria absinthioides is used in traditional medicine of South America as hypocholesterolemic, balsamic, and expectorant; but it is also useful for the prevention of hepatitis, renal insufficiency, and diabetes, and is used as digestive. A lyophilized decoction from the aerial parts of this plant (TLD) collected in San Juan (TLDSJ) and Mendoza (TLDM) provinces (Argentina) and one collection from Antofagasta, Chile (TLDCH) were characterized regarding antioxidant and antibacterial activities, phenolics and flavonoids content, and ultrahigh resolution liquid chromatography Orbitrap MS analysis UHPLC–PDA–OT-MS/MS metabolite profiling. The antioxidant properties were carried out "in vitro" using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and trolox equivalent antioxidant activity (TEAC) methods, ferric-reducing antioxidant power (FRAP), and lipoperoxidation in erythrocytes (LP). The antibacterial activity was evaluated following the Clinical and Laboratory Standards Institute (CLSI) rules. TLDSJ, TLDM, and TLDCH displayed a strong DPPH scavenging activity (EC50 = 42, 41.6, and 43 µg/mL, respectively) and inhibition of lipoperoxidation in erythrocytes (86–88% at 250 µg TLD/mL), while a less effect in the FRAP and TEACantioxidant assays was found. Additionally, the decoctions showed a content of phenolics compounds of 94 mg gallic acid equivalents (GAE)/g, 185 GAE/g, and 64 GAE/g, for TLDSJ, TLDM, and TLDCH samples, respectively. Regarding the flavonoid content, the Chilean sample was highlighted with 19 mg quercetin equivalents (QE)/g. In this work, several phenolic compounds, including sesquiterpenes, flavonoids, and phenolic acids, were rapidly identified in TLDSJ, TLDM, and TLDCH extracts by means UHPLC–PDA–OT-MS/MS for the first time, which gave a first scientific support to consider this medicinal decoction from both countries as a valuable source of metabolites with antioxidant effects, some with outstanding potential to improve human health.

In the current research, the total and free genistein and daidzein contents were determined in two endemic Genista species (G. sandrasica and G. vuralii) by an HPLC method. The highest amount of total genistein and total daidzein was found in G. sandrasica, 0.582 % and 0.113 %, respectively, whereas only the free daidzein content of G. sandrasica was higher than that of G. vuralii. The antioxidant activity of the crude methanol and hydrolyzed extracts of these species was evaluated by three in vitro methods; namely DPPH free radical scavenging, ferrous ionchelating and ferric-reducing antioxidant power (FRAP) tests at 0.25, 0.50, and 1.0 mg ml. The hydrolyzed extracts of both species displayed greater antioxidant activity than the crude methanol extracts in all tests. Total phenol and flavonoid contents in the extracts were determined via the Folin-Ciocalteau and AlCl3 reagents, respectively. G. vuralii was richer in terms of total phenol and flavonoid contents compared to G. sandrasica.
