Nutrien daun kelor (Moringa oleifera) dan jerami kacang hijau (Vigna radiata) sebagai hijauan pakan ternak cukup berkualitas sehingga perlu diproteksi untuk meningkatkan efektivitas penggunaannya di dalam rumen. Daun nangka mengandung total tanin sebesar 7,08%, sehingga potensial digunakansebagai bahan untuk memproteksi bahan pakan sumber protein. Studi in vitro perlu dilakukan untuk mengevaluasi penggunaan tepung daun nangka untuk meningkatkan efektivitas penggunaan hijauan berupa daun kelor dan jerami kacang hijau. Tujuan dari studi yang dilakukan adalah untuk mengetahuipengaruh penambahan tepung daun nangka terhadap nilai biologis daun kelor dan jerami kacang hijau. Evaluasi produksi metana juga dilakukan untuk mengetahui efisiensi proses fermentasi. Perlakuan penelitian adalah: 1) jerami kacang hijau; 2) jerami kacang hijau + 0,7% daun nangka; 3) jerami kacanghijau + 1 ,4% daun nangka; 4) daun kelor; 5) daun kelor + 0,7% daun nangka; 6) daun kelor + 1 ,4% daun nangka. Penelitian ini menggunakan rancangan acak lengkap dengan pengulangan sebanyak tiga kali. Pengamatan dilakukan terhadap produksi gas total (inkubasi ke-0, 2, 4, 6, 8, 10, 12, dan 24 jam),konsentrasi gas metana (%), karakteristik produksi gas dan karakteristik produk fermentasi rumen. Hasil penelitian menunjukkan bahwa penambahan tepung daun nangka tidak berpengaruh terhadap produksi gas total, produksi gas maksimum dan laju degradasi kedua jenis hijauan pakan. Penambahan 0,7 dan1,4% tepung daun nangka dapat menurunkan konsentrasi gas metana daun kelor berturut-turut sebesar 4,93% dan 3,19%. Penambahan tepung daun nangka juga mampu meningkatkan rasio CO2:CH4 substrat jerami kacang hijau masing-masing sebesar 4,42 dan 6,49%. Tepung daun nangka juga tidakmemberikan efek negatif terhadap produk fermentasi rumen (pH, NH3, VFA total, dan degradasi bahan organik).

Background: Protease inhibitors have been isolated from plants and present several biological activities, including immunomod-ulatory action. Objective: This work aimed to evaluate a Moringa oleifera flower trypsin inhibitor (MoFTI) for acute toxicity in mice, hemolytic activity on mice erythrocytes and immunomodulatory effects on mice splenocytes. Methods: The acute toxicity was evaluated using Swiss female mice that received a single dose of the vehicle control or MoFTI (300 mg/kg, i.p.). Behavioral alterations were observed 15–240 min after administration, and survival, weight gain, and water and food consumption were analyzed daily. Organ weights and hematological parameters were analyzed after 14 days. Hemolytic activity of MoFTI was tested using Swiss female mice erythrocytes. Splenocytes obtained from BALB/c mice were cultured in the absence or presence of MoFTI for the evaluation of cell viability and proliferation. Mitochondrial membrane potential (ΔΨm) and reactive oxygen species (ROS) levels were also determined. Furthermore, the culture supernatants were analyzed for the presence of cytokines and nitric oxide (NO). Results: MoFTI did not cause death or any adverse effects on the mice except for abdominal contortions at 15–30 min after administration. MoFTI did not exhibit a significant hemolytic effect. In addition, MoFTI did not induce apoptosis or necrosis in splenocytes and had no effect on cell proliferation. Increases in cytosolic and mitochondrial ROS release, as well as ΔΨm reduction, were observed in MoFTI-treated cells. MoFTI was observed to induce TNF-α, IFN-γ, IL-6, IL-10, and NO release. Conclusion: These results contribute to the ongoing evaluation of the antitumor potential of MoFTI and its effects on other immunological targets.

Background: The need for suitable antibacterial agents effective against Multi-drug resistant Gram-negative bacteria is acknowledged globally. The present study was designed to evaluate the possible antibacterial potential of an extracted compound from edible flowers of Moringa oleifera. Methods: Five different solvents were used for preparing dried flower extracts. The most effective extract was subjected to fractionation and further isolation of the active compound with the highest antibacterial effect was obtained using TLC, Column Chromatography and reverse phase- HPLC. Approaches were made for characterization of the isolated compound using FTIR, NMR and Mass spectrometry. Antibacterial activity was evaluated according to the CLSI guidelines. Results: One fraction of aqueous acetic acid extract of M. oleifera flower was found highly effective and more potent than conventional antibiotics of different classes against Multi-drug resistant Gram-negative bacilli (MDR-GNB) when compared. The phytochemical analysis of the isolated compound revealed the presence of hydrogen-bonded amine and hydroxyl groups attributable to unsaturated amides. Conclusion: The present study provided data indicating a potential for use of the flowers extract of M. oleifera in the fight against infections caused by lethal MDR-GNB. Recommendations: Aqueous acetic acid flower extract of M. oleifera is effective, in-vitro, against Gram-negative bacilli. This finding may open a scope in pharmaceutics for the development of new classes of antibiotics.

Introduction: Diabetes mellitus (DM) has been recognized as the seventh leading cause of global mortality; however, researchers seek alternative means to manage the menace. The current study sought to investigate antioxidant potentials, α-amylase, and α-glucosidase inhibitory activities of ethanolic extract of Moringa oleifera flower in vitro. Methods: Antioxidant properties of the extract were appraised by assessing its inhibition against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl (OH•), and hydrogen peroxide (H2O2) free radicals, as well as ferric reducing antioxidant power (FRAP), the antidiabetic activity was evaluated by α-amylase and α-glucosidase inhibition.Results: In this study, ethanolic extract of M. oleifera flower demonstrated a significant (P < 0.05) inhibition against DPPH free radical (43.57–83.56%) in a concentration-dependent manner, while FRAP (101.76 ± 1.63 mg/100 g), OH• scavenging ability (71.62 ± 0.95 mg/100 g), and H2O2 free radical scavenging capacity (15.33 ± 1.20 mg/100 g) were also observed. In the same manner, ethanolic extract of M. oleifera flower revealed a significant (P < 0.05) inhibition against α-amylase (IC50= 37.63 mg/mL) and α-glucosidase activities (IC50= 38.30 mg/mL) in the presence of their respective substrates in a concentration-dependent manner in comparison with acarbose. Conclusion: Ethanoic extract of M. oleifera flower could be useful as an alternative phytotherapy in the management of DM, having shown a strong antioxidative capacity and substantial inhibition against the activities of key enzymes involved in carbohydrate hydrolysis in vitro.

<p>The current study explores the capacities of Moringa oleifera (L), to overcome and/or tolerate oxidative stress induced by a saline substrate as a constraining factor. For this, Moringa seeds have been treated for 15 days after germination in vitro with increasing concentrations of NaCl, and specifically at 0 (control), 5; 7.5; 10; 12.5 and 15g l-1. Morpho-physiological and biochemical aspects were evaluated which were considered as probable indicators of tolerance or sensitivity to this stress. Our results reveal an increased synthesis of proline, lipids, proteins, CAT, GPX, APX, flavonoids, condensed tannins and a decrease in total polyphenols (-29.97%), mainly due to their high carbon cost. The state of degradation of the cell membranes was evaluated by the MDA assay, which increased by 91.25%. On the other hand, the evaluation of the oxidative damage caused by Reactive Oxidative Species (ROS) was detected by H2O2, which decreased by 37.89%. Additionally, our results revealed a disturbance of the germinative power of the seeds indicating moderate resistance to stress as it was manifested by the development of epicotyledonary axes, even under 15 g l-1 of NaCl without showing symptoms of stress or sensitivity. This result supports our hypothesis of the ability of the species to maintain or even improve its oxidative status.</p>
