Abstract Soybean [Glycine max (L.) Merrill] is one of the most important legume crops and its plant height (PH) is one significant quantitative trait closely related to node number (NN) and internode length (IL) on the main stem, which affect yield together. In this study, we used a recombinant inbred line (RIL) derived from a cross between two semi-determinate stem soybean varieties (Dt1Dt1Dt2Dt2), JKK378 and HXW. A consistent QTL named qPH18 simultaneously controlling PH, NN, and IL was identified, in which this region harbors the semi-determinant gene, Dt2. Sequencing of the promoter of Dt2 from JKK378, identified three polymorphisms relative to HXW, including two SNPs and a 18bp Indel. The expression level of Dt2 in qPH18JKK378 group was lower than that in qPH18HXW group, meanwhile, as a downstream gene, the expression of Dt1 from two groups showed a contrary tendency. Transient transfection assay confirmed that the promoter activity of Dt2 from JKK378 is lower compared to HXW. We speculate that the polymorphisms in the two dominant Dt2 promoter caused difference in expression level of Dt2 and its downstream gene Dt1, hence, affect PH, NN, IL, and grain weight per plant without changing stem growth habit. Compared to PH18HXW allele, qPH18JKK378 allele suppresses the expression of Dt2 gene which releases the expression of Dt1, thus promotes plant node number and enhanced grain yield. These results will be helpful for revealing the mechanism underlying the node number and plant height, the soybean materials and molecular marker will facilitate molecule breeding.

An initial survey of the diversity of early lignotuber development in Eucalyptus and an analytical study of the anatomy of young lignotubers and the seedling stem are presented. Studies of the early stages of the morphological development of the lignotuber in 13 species, representative of five taxonomic groups, resulted in the recognition of four modes of lignotuber initiation. The importance to lignotuber formation of the presence of a suite of accessory buds, adaxial to the axillary bud, is emphasized but lignotuber initiation is not in all cases associated with these buds. Lignotuber buds are derived by branching from existing buds, ultimately from the accessory buds of the node. Following its initiation, the possibilities of later morphological development of the lignotuber are discussed. Lignotuber growth may dominate over stem growth and the lignotubers at a node may then fuse laterally to encircle the stem. Stem growth, on the other hand, may dominate over lignotuber growth and the lignotuber then appears to regress. The consequences for the growth habit of the plant of these alternative pathways of development are outlined. The wood of young lignotubers (and that of the swollen hypocotyl) is shown to be different in composition and in the sizes of its elements from that of seedling stem wood; these differences owe their origin to differences in the nature and performance of the cambia of the lignotuber and stem. In lateral fusion of the lignotubers at a node, and their upward and downwards extension over the stem, e.g. over the hypocotyl, stem cambial initials are either progressively lost or, more likely, converted to lignotuber-type initials. The possibility of the reverse process occumng in stem dominance is discussed.

ABSTRACT We have studied the pathways of regulation of cytokine and cell cycle control proteins during infection of human B lymphocytes by Epstein-Barr virus (EBV). Among 30 cytokine RNAs analyzed by the RNase protection assay, tumor necrosis factor alpha (TNF-α), granulocyte colony-stimulating factor, lymphotoxin (LT), and LTβ were found to be regulated within 20 h of EBV infection of primary B cells. Similar results were obtained using the estrogen-regulated EBNA-2 cell line EREB2.5, in which RNAs for LT and TNF-α were induced within 6 h of activation of EBNA-2. Expression of Notch also caused an induction of TNF-α RNA. The induction of TNF-α RNA by EBNA-2 was indirect, and constitutive expression of either LMP-1 or c-myc proteins did not substitute for EBNA-2 in induction of TNF-α RNA. Cyclin D2 is also an indirect target of EBNA-2-mediated transactivation. EBNA-2 was found to activate the cyclin D2 promoter in a transient-transfection assay. A mutant of EBNA-2 that does not bind RBP-Jκ retained some activity in this assay, and activation did not depend on the presence of B-cell-specific factors. Deletion analysis of the cyclin D2 promoter revealed that removal of sequences containing E-box c-myc consensus DNA binding sequences did not reduce EBNA-2-mediated activation of the cyclin D2 promoter in the transient-transfection assay. The results indicate that cytokines are an early target of EBNA-2 and that EBNA-2 can regulate cyclin D2 transcription in EBV-infected cells by mechanisms additional to the c-myc pathway.

ABSTRACT The recA gene of Mycobacterium tuberculosis is unusual in that it is expressed from two promoters, one of which, P1, is DNA damage inducible independently of LexA and RecA, while the other, P2, is regulated by LexA in the classical way (E. O. Davis, B. Springer, K. K. Gopaul, K. G. Papavinasasundaram, P. Sander, and E. C. Böttger, Mol. Microbiol. 46:791-800, 2002). In this study we characterized these two promoters in more detail. Firstly, we localized the promoter elements for each of the promoters, and in so doing we identified a mutation in each promoter which eliminates promoter activity. Interestingly, a motif with similarity to Escherichia coli σ70 −35 elements but located much closer to the −10 element is important for optimal expression of P1, whereas the sequence at the −35 location is not. Secondly, we found that the sequences flanking the promoters can have a profound effect on the expression level directed by each of the promoters. Finally, we examined the contribution of each of the promoters to recA expression and compared their kinetics of induction following DNA damage.

Soybean consumption here in Indonesia continues to increase each year that is notaccompanied by an increase in soybean production. One way to increase soybeanproduction by using improved varieties. The purpose of this study were (1) Estimating thevalue of the diversity of phenotypes soy F6 generations from crosses between Wilis x Mlg2521,(2) Estimating the heritability estimates soybean F6 generations from crosses Wilis x Mlg2521,(3) Know the numbers expectation of F6 generation crosses Wilis x Mlg2521. The researchwas conducted from March 2014 until June 2014 at the Land Lab Lampung StatePolytechnic and observations made in Laboratory of Seed and Plant Breeding, University ofLampung. Soybean seeds used were F6 generation zuriat from Wilis x Mlg2521, Wilis andMlg2521. The treatment laid out in a randomized block design, 2 replications. The resultsshowed that (1) The diversity of phenotypes population F6 for characters date of flowering,harvesting age, plant height, number of productive branches, total number of pods plant,weight of 100 grains, grain weight plant and number of seeds plant including all the broadcriteria, (2) The value of heritability estimates the population F6 for characters date offlowering, harvesting age, plant height, number of productive branches, total number ofpods plant, weight of 100 grains, grain weight per plant and number of seeds plantbelonging to the high criteria, (3) Number expectations of the population F6 is 7-64-1-8 and7-64-1-3.Keywords: heritability, phenotypic variance, soybean
