Abstract Background: Jiangxiangru, a commonly used clinical Chinese medicine, has been investigated mainly over its effects on Summer heatstroke and the dampness inside the spleen and stomach. This study focuses on the diuretic effect of Jiangxiangru in normal rats as well as the underlying mechanism.Methods: 48 rats were randomly divided into the model group (saline), the positive control group (furosemide 0.02g/kg), low and high dosage of water decoction of Jiangxiangru group (0.2 g/ml and 1g/ml), low and high dosage of water extract of Jiangxiangru group (0.1463g/ml and 0.7317g/ml), and the low and high dosage of volatile oil of Jiangxiangru group (0.1791g/ml and 1.4328g/ml). Each group was assigned 6 rats. Thirty minutes after the successful establishment of the model, the rats were treated orally with corresponding doses of drugs at 10 ml/kg. The control group received an equivalent amount of water. After administration, Urine volume at each time point was measured and urine K+, Na+, Cl- content were measured. The contents of ALD, ANP, ADH, Na+-K+-ATPase, the gene expression and protein levels of AQP1, AQP2 and AQP3 in urine of rats in each group were determined. SPSS22.0 statistical software was used to analyze the experimental data. One way ANOVA and multiple comparisons between LSD groups were performed. The difference is considered significant When p < 0.05 or p < 0.01.Results: Our results showed that Jiangxiangru decoction group has the strongest diuretic effect, followed by water extract, then the volatile oil. The diuretic mechanism of Jiangxiangru may affect the kidney in addition to the hormone level related to the change of body fluid metabolism. The distal convoluted tubules are related to the content of aquaporin AQP in the collecting tube, thereby inhibiting water reabsorption.Conclusion: The contents of Na+, K+, and Cl- and PH in urine of rats don’t change significantly in this experiment. The diuretic effect of Jiangxiangru could not cause electrolyte disturbance and other adverse reactions. This fully reflects the complex composition of traditional Chinese medicine, the role of multi-target characteristics.

Processing of celery as diuretic is done by society in various ways. So far, the most effective preparation process of celery extract has not been known. This research aims to examine the diuretic effect of various celery extract preparation.  This research was conducted using celery water extract which is made in accordance with the people’s habit, using aquades as a negative control and HCT as a positive control. The effectiveness test of celery water extract as diuretic was done on white male rat by measuring the urine volume every hour for six hours. The results showed that the largest urine volume is obtained from the boiling celery extract.

Abstract Background Coix seed is a traditional Chinese medicine with pharmacological effects and food efficacy. It is well-know and widely used in China and other Asian countries. Fayiren is a new artillery material processed from Coix Seed. This is the first study of the diuretic activities of Fayiren and coix seed in normal rats. Methods Forty-eight rats were randomly divided into 6 groups (8 rats/group) according to body mass and urine volume stratification, namely model (physiological saline) group, positive control (furosemide 20 mg/kg) group, coix seed (1666 mg/kg) group, low-dose of Fayiren group (166 mg/kg) and high-dose Fayiren group (1666 mg/kg). Rats were fasted for 16 h before the experiment, with normal water supply. Immediately after administration, each group of rat was placed in a metabolic cage, and urine was collected once after every hour for a total of 6 times. The urine volume at each time point for collection was measured, and then totaled. After the 6 h recording procedure, the treatments were administered once a day and the urine was collected at the 24th, 48th, 72th, 96th, 110th, 134th, and 158th hours from the start respectively. The urine pH and Na +, K +, Ca 2+, Cl - concentrations at each time point of each group of rats were measured. The contents of ALD, ANP, ADH, Na+-K+-ATPase, the gene expression and protein levels of AQP1, AQP2 and AQP3 in urine of rats in each group were determined. SPSS22.0 statistical software was used to analyze the experimental data. One way ANOVA and multiple comparisons between LSD groups were performed. The difference is considered significant When p < 0.05 or p < 0.01. Results After the administration of Fayiren and coix seed, compared with normal rats, the total output of urine in the high-dose coix seed & Fayiren group and the positive control group both increased; In addition, the contents of Na+ and Cl-in urine in these groups increased as well. However, the content of K+ did not increase in all groups. Contents of ALD and ADH was decreased and increased contents of ANP observed in urine in both high-dose and low-dose Fayiren groups with significant differences from model group (P < 0.05). Coix seed significantly reduced ADH and increased ANP levels compared with model group (P < 0.05). There was no obvious change in the urine pH content in urine of all rats. All the aquaporin (AQP1, AQP2, and AQP3) contents were decreased comparing to the model group, the coix seed group, as well as the Fayiren group. The effect of coix seed was particularly notable on AQP2, and the difference was significant against the Furosemide group (P < 0.05). By contrast, Fayiren showed greater effects on AQP1 and AQP3, with statistically significant difference comparing to the model group (P༜0.05). Conclusions This study will provide experimental evidence for explaining the different mechanisms of diuretic effects in natural medicine use of coix seed and artillery material processed from Coix Seed. The Fayiren may affect the reabsorption of water in the kidney through mechanisms related to increased ANP secretion,decreased ALD and ADH secretion in urine, and it significantly affect the expression of aquaporin 1 and aquaporin 3. However, coix seed may increase ANP but decreased ADH secretion, further affecting the expression of aquaporin2 and exert diuretic effect.

Abstract Background The higher levels of low-density lipoproteins (LDL) cholesterol (LDL-C) have been reported to correlate with higher prevalence and risk of colon cancer. Proprotein convertase subtilisin/kexin 9 (PCSK9) function, through reducing protein levels of liver LDL receptor, leads to elevated levels of plasma LDL-C. Purpose Here, we evaluated anti-tumor effect of nanoliposomal anti-PCSK9 vaccine in mice bearing colon carcinoma. Methods To formulate nanoliposomal anti-PCSK9 vaccine, liposome nanoparticles prepared by lipid-film hydration method were covalently attached to immunogenic PCSK9 peptide. The liposomal vaccine formulation was adsorbed to Alum adjuvant (L-IFPTA+) and injected subcutaneously four times with a bi-weekly interval in BALB/c mice. Two weeks after the last immunization, the vaccinated and unvaccinated mice were subcutaneously inoculated with CT26 colon cancer cells into the right flank. After tumor mass was palpable (approximately 10 mm3), the mice were randomly divided into three groups and involved to different treatments: (1) PBS (untreated control), (2) vaccine group, and (3) Doxil®(positive control) group which involved unvaccinated tumor-bearing mice who received Doxil®. To study therapeutic efficacy, mouse body weight, tumor size, and survival were monitored in a 3-day interval for 50 days. Results The nanoliposomal anti-PCSK9vaccine could efficiently induce specific antibodies against PCSK9 in BALB/c mice, and thereby reduce plasma level and function of PCSK9. Tumor size in the vaccinated mice was significantly lower than in Doxil and the control mice. Tumor size analysis revealed that time to reach endpoint (TTE) of the vaccine group was slightly but not significantly higher than that of Doxil and the control groups. The vaccinated mice survived slightly but not significantly longer than Doxil and the control mice. The vaccinated mice's life was prolonged by 24.4% as compared with the control mice, while it was increased by 9.8% in Doxil group (see Figure below). Conclusions Our findings declare that the nanoliposomal anti-PCSK9vaccine not only have no harmful effects, but also can mildly reduce tumor growth, and enhance live span and survival in mice bearing colon cancer.

Fasting 24-hour exposures of altitude-acclimated rats (380 mm Hg, 18,000 ft. simulated) to ground level pressure (750 mm Hg) at either cold (3°C), neutral (24°C), or hot (35°C) temperatures seldom resulted in return of their metabolic functions to preacclimative ‘normalcy.’ Although the control and altitude-acclimated groups both were accustomed to neutral temperatures (24° and 26°C), quantitative differences at ground level and altitude occurred in various indices of water, mineral and nitrogen metabolism. Of the 32 physiologic variables studied, only 4 (ratio of urine volume/ water intake, and urinary excretion of potassium, creatinine and glycine) failed to differentiate the responses of the altitude- and ground-accustomed rats. The temperature response curves of the altitude group tended to parallel the corresponding ones for the control group, but most variables were on higher or lower planes. The difference in plane resulted either from the effects of the return to ground level pressure, or from nonreversible effects of acclimation to altitude per se.

Zinc (Zn) was found to enhance the antidepressant efficacy of imipramine (IMI) in human depression and animal tests/models of depression. However, the underlying mechanism for this effect remains unknown. We measured the effect of intragastric (p.o.) combined administration of IMI (60 mg/kg) and Zn (40 mg Zn/kg) in the forced swim test (FST) in mice. The effect of Zn + IMI on serum, brain, and intestinal Zn concentrations; Zn transporter (ZnT, ZIP) protein levels in the intestine and ZnT in the brain; including BDNF (brain-derived neurotrophic factor) and CREB (cAMP response element-binding protein) protein levels in the brain were evaluated. Finally, the effect of IMI on Zn permeability was measured in vitro in colon epithelial Caco-2 cells. The co-administration of IMI and Zn induced antidepressant-like activity in the FST in mice compared to controls and Zn or IMI given alone. This effect correlated with increased BDNF and the ratio of pCREB/CREB protein levels in the prefrontal cortex (PFC) compared to the control group. Zn + IMI co-treatment increased Zn concentrations in the serum and brain compared to the control group. However, in serum, co-administration of IMI and Zn decreased Zn concentration compared to Zn alone treatment. Also, there was a reduction in the Zn-induced enhancement of ZnT1 protein level in the small intestine. Zn + IMI also induced an increase in the ZnT4 protein level in the PFC compared to the control group and normalized the Zn-induced decrease in the ZnT1 protein level in the hippocampus (Hp). The in vitro studies revealed enhanced Zn permeability (observed as the increased transfer of Zn through the intestinal cell membrane) after IMI treatment. Our data indicate that IMI enhances Zn transfer through the intestinal tract and influences the redistribution of Zn between the blood and brain. These mechanisms might explain the enhanced antidepressant efficacy of combined IMI/Zn treatment observed in the FST in mice.

Aim. To study the effect and mechanism of traditional Chinese medicine Qinbai Qingfei concentrated pellet (QQCP) against Mycoplasma pneumoniae (MP). Methods. Rat airway smooth muscle (ASM) cells were used to examine the antimycoplasmal activity of QQCP via four drug-adding modes: pre- and postadding drugs, simultaneous-adding after drug and MP mixed, and simultaneous-adding drug and MP; taking roxithromycin dispersive tablets (RDT) as positive control, the cellular A570 values were determined by MTT method. Results. All of A570 values in QQCP group were significantly higher than those of the corresponding MP control group (P<0.01) in four drug-adding modes; there was no significant difference in A570 values between the QQCP group and that of the positive control group (P>0.05), confirming that QQCP could significantly inhibit the infectivity of MP to ASM cells. Conclusion. QQCP had significant activity in preventing and treating MP infection, killing MP, and antiabsorption.

Abstract: The objectives of the research were to find out the analgesic effect of giving Gedi (Abelmoschus manihot (L.) Medik) leaf ethanol extract orally on the number of writhing after thermal pain induction of mice. This research using 15 mice which is divided into 5 groups consisted of 1 negative control group given by the aquades, 1 positive control group given by the tramadol, and 3 experiment groups. Experiment group given by Gedi (Abelmoschus manihot (L.) Medik) leaf ethanol extract with the doses which different each other, that is 30 mg/30 g BW, 60 mg/30 g BW and 120 mg/30 g BW. Thermal pain induction was done by placing the mice on hot plate constant temperature of 550C. The mice gave respond in the way of lick its foot or even jumping. The data was collected using table, graphic and analyzed using one direction ANOVA model and it was continued with LSD test to find out the difference every treatment group. The result of analysis showed that gedi’s leaf ethanol extract have the analgesic effect and the maximum effect presented at gedi leaf ethanol extract dosage 60 mg/30 g BW. Medik) peroral pada mencit yang kemudian diamati jumlah geliatnya setelah diinduksi panas. Penelitian ini menggunakan 15 ekor mencit yang dibagi 5 kelompok yang terdiri dari 1 kelompok kontrol negatif yang diberi aquades, 1 kelompok kontrol positif yang diberi tramadol, dan 3 kelompok eksperimen. Kelompok eksperimen diberi ekstrak etanol daun gedi dengan dosis yang berbeda-beda, yaitu 30 mg/30 g BB, 60 mg/30 g BB, dan 120 mg/30 g BB. Induksi nyeri berupa panas dilakukan dengan meletakkan mencit pada hot plate dengan suhu 550C . Mencit memberi respon berupa menjilat kaki dan atau melompat. Data disajikan berupa tabel, grafik dan menggunakan analisis statistik ANOVA yang dilanjutkan dengan LSD untuk menemukan perbedaan dari setiap kelompok. Hasil analisis menunjukkan bahwa ekstrak etanol daun gedi memiliki efek analgesik dan efek maksimumnya didapatkan pada dosis 60 mg/30 g BB. Kata kunci: Daun gedi, efek analgesik

The goal of this study was to determine the sealing ability of MTA, Biodentine, and Fuji IX as root-end materials after ultrasonic retrograde preparation using a dye penetration method. </span></p><p class="Default"><strong><span>Materials and Methods</span></strong><span>. Seventy permanent anterior teeth were used in this study. Root canals were prepared using rotary endodontic files, and obturated using the lateral condensa­tion technique. The resection of the root apices was performed perpendicularly to the long axis, and 3 mm deep retrocavities were made by ultrasonic tips. The samples were randomly divided into three experimental groups (N=20) and two control groups (N=5). The root-end cavities were filled with mineral-trioxide aggregate (Rootdent MTA, TehnoDent, Russia) in Group 1, Biodentine (Septodont, Saint Maur des Fossés, France) in Group 2 and Fuji IX GP (GC Corporation, Tokyo, Japan) in Group 3. The retrocavities of the positive control group were left unfilled, while retrocavities in the negative control group were sealed using a flowable composite material. The samples were made transparent, and dye penetration was assessed under a stereomi­croscope (x10). The data were analyzed with the Kruskal-Wallis test and the Mann-Whitney U-test. </span></p><p class="Default"><strong><span>Results</span></strong><span>. The Biodentine group had a lower mean leakage value than the MTA and Fuji IX groups, and the difference was statistically significant (P&lt;0.01). No statistical difference in apical microleakage was found between the MTA and Fuji IX groups (P&gt;0.05). </span></p><p class="Default"><strong><span>Conclusion</span></strong><span>. Within the limitations of this study, it was concluded that Biodentine provides better sealing ability as a root-end material than MTA and Fuji IX. There was no statistically significant difference between the sealing ability of MTA and Fuji IX as root-end materials.</span></p>

Abstract Aims: The present study aimed to explore the correlation between cholecystectomy and cholangiocarcinoma, and to provide preliminary clinical basis for precise cholecystectomy in China.Methods: We conducted a retrospective analysis of 9744 patients with cholangiocarcinoma, colon cancer, pancreatic cancer, femoral fracture, and hepatic hemangioma diagnosed in Xijing hospital from August 2008 to August 2018. They were divided into three groups: case group (1749 cases of cholangiocarcinoma), positive control group (3137 cases of colon cancer and 1950 cases of pancreatic cancer), negative control group (1794 cases of femoral fracture and 1114 cases of hepatic hemangioma). We collected the general information (gender, age), past medical history, cholecystectomy history from the patients, and these data were analyzed by chi-square test and logistic regression analysis. Results: The cholecystectomy rate of the case group was significantly higher than that of the positive control group and the negative control group by chi-square test (p<0.025). The cholecystectomy rate and the history of cholecystolithiasis were analyzed by logistic multivariate regression analysis. The OR values of cholecystectomy rate were 1.553 (95%CI: 1.311-1.840) and 3.181 (95%CI: 2.561-3.951), respectively, and the difference was statistically significant (p<0.000). The OR values of the history of cholecystolithiasis were 2.460 (95%CI: 2.093-2.890) and 5.426 (95%CI: 4.325-6.809), respectively, and the difference was statistically significant (p<0.000). In case group, the difference between cholecystectomy and cholecystolithiasis was statistically significant (p<0.000) by chi-square test. Conclusions: In conclusion, cholecystectomy is one of the risk factors of cholangiocarcinoma and the patients who undergo cholecystectomy have a higher risk of cholangiocarcinoma than the control groups. Cholecystectomy should be conducted with caution and the precise surgical treatment of gallbladder diseases is advocated.
