Abstract Background Female sex hormone secretion and reproductive ability decrease with ageing. Bone marrow mesenchymal stem cells (BMMSCs) have been postulated to play a key role in treating ovarian senescence; however, the curative effect and mechanism are not clear. Methods We used the macaque ovarian senescence model and observed the structural and functional effects of juvenile BMMSCs in the treatment of ageing macaque ovaries. Moreover, to elucidate the molecular regulatory mechanism by which BMMSCs reverse ovarian senescence, RNA sequencing (RNA-seq) of the ovaries was used to identify key genes and signalling pathways associated with transcriptome profile changes. Results (1) The Rhesus monkey ovarian aging models were an average of 24 years old and had the following sex hormone levels: 0.28 ± 0.11 mIU/mL hFSH, 0.017 ± 0.009 mIU/mL hLH, 0.24 ± 0.042 ng/mL Testo, 51.86 ± 18.37 pg/mL ESTRDL, 0.13 ± 0.012 ng/mL Prog, 0.013 ± 0.012 hCG, and 11.96 ± 2.96 pmol/l AMH. The ovarian organ index was 0.057 ± 0.021, and the HE staining results showed almost no follicular structure, with only local atresia follicles observed. For young rhesus monkeys, the average age was 7 years old, and sex hormone levels were as follows: 0.043 ± 0.03 mIU/mL hFSH, 0.007 ± 0.009 mIU/mL hLH, 0.57 ± 0.15 ng/mL Testo, 123.2 ± 26.26 pg/mL ESTRDL, 0.28 ± 0.014 ng/mL Prog, 0.05 ± 0.012 hCG and 11.96 ± 2.96 pmol/l AMH. The ovarian organ index was 0.011 ± 0.005, and the HE staining results showed all levels of follicles, suggesting that senile ovaries occur in old macaques. (2) P4 generation BMMSCs presented a typical cell morphology, staining positive for Oil Red O, Alizarin Red, and Alcian Blue. The positive rates of CD29, CD34, CD90, and CD105 on the cell surface were 98, 0.98, 98.8, and 99.8%, respectively, in line with mesenchymal stem cell standards. (3) The PET-CT results showed that the ovarian volume in the elderly treatment group increased, the lesions decreased, and the metabolism was vigorous. (4) The level of sex hormone secretion generally recovered to the level of the follicular phase for the 3rd, 6th and 8th months after the treatment of BMMSCs. (5) The HE results showed that all levels of follicles were observed in the young control group, and the medulla and stroma were neatly arranged, whereas primitive, primary, secondary, and atretic follicles were observed in the elderly treatment group. In addition, the medulla and stroma had obvious boundaries with a small amount of calcium nodules in the young control group. while those in the elderly control group had essentially no follicular structure, with only atretic follicles were seen locally that were filled with connective tissue. Masson staining results showed that the proportion of collagen fibres was 10.61 ± 1.83% in the young control group, 56.79 ± 3.58% in the elderly control group, and 23.71 ± 2.4% in the elderly treatment group. TUNEL staining results showed that cell apoptosis was 1.07 ± 0.04%, in the young control group was, 25.93 + 2.49% in the old control group, and 6.98 + 1.35% in the old treatment group. The immunohistochemistry results showed 114 ± 17, 73 ± 6, and 118 ± 18 blood vessels in the young control group, the elderly control group, and the elderly treatment group, respectively. Immunofluorescence staining showed a lack of expression of enhanced green fluorescence protein (E-GFP) in BMMSCs without from the old control group, while green fluorescence was observed in the old treatment group. (6) After the treatment of BMMSCs, 1258 genes were identified as being differentially expressed. The 3D-PCA trace showed that the ovaries of the macaques in the elderly treatment group shifted to that observed in the young group. The genes that were upregulated with age were downregulated after the stem cell treatment. Genes that are downregulated with age were upregulated after stem cell therapy, and the top 20 PPI network genes were enriched for the progesterone-mediated maturation of follicles, oocytes, and cell cycle categories. Conclusions BMMSCs derived from juvenile macaques can reverse ovarian aging in elderly macaques

Background: Hypertension is the silent disease which a person will not know if he has increased blood pressure. The incidence rate of hypertension is very high especially in the elderly population over 60 years. Hypertension management can be done in pharmacological ways such as medicines and can use natural ingredients in lowering blood pressure that is by utilizing the noni fruit.Objectives: This study aimed to analyze the effectiveness of Noni fruit tea in lowering elderly blood pressure with hypertensionMethod: This research was quasy experimental throug with Pretest-Postest Control Group Design design. Determination of research sample by using inclusion and exclusion criteria. The sample of the study was 34 respondents divided into two groups. Intervention of Noni fruit tea given as much as 5 gram for 30 days with frequency of giving 2 times a day in the morning and afternoon. Blood pressuremeasurement was performed at the beginning of the study and at the end of the study. Data were analysed using wilcoxon test and Mann Whitney test.Result: Mann Whitney test results showed that there was significant effect after the intervention of noni fruit tea in treatment group compared with control group with systolic blood pressure value (p =0.000171) and diastolic blood pressure (p = 0.000480) with α = 0.05Conclusions: Blood pressure in the treatment group after the intervention of noni fruit tea decreased blood pressure compared with the control group. It is Suggested of research, need to do periodic nutrition education and giving noni fruit tea as alternative medicine that effectively decrease hypertension, scheduled.ABSTRAKLatar Belakang : Hipertensi merupakan the silent disease dimana seseorang tidak akan mengetahui jika dirinya mengalami peningkatan tekanan darah. Angka insiden hipertensi sangat tinggi terutama pada populasi lanjut usia diatas 60 tahun. Penatalaksaan hipertensi dapat dilakukan dengan cara farmakologis seperti obat-obatan serta dapat menggunakan bahan alami dalam menurunkan tekanan darah yaitu dengan memanfaatkan buah mengkudu.Tujuan : Penelitian ini bertujuan menganalisis efektifitas teh buah mengkudu dalam menurunkan tekanan darah lansia dengan hipertensiMetode : Penelitian ini bersifat quasy eksperimental dengan desain Pretest-Postest Control Group Design. Penentuan sampel penelitian dengan menggunakan kriteria inklusi dan eksklusi. Sampel penelitian sebanyak 34 responden yang dibagi menjadi dua kelompok. Intervensi teh buah mengkudu diberikan sebanyak 5 gram selama 30 hari dengan frekuensi pemberian 2 kali sehari pada pagi dan sore hari. Pengukuran tekanan darah darah dilakukan pada awal penelitian dan di akhir penelitian. Analisis data penelitian menggunakan uji wilcoxon dan uji Mann Whitney.Hasil : Hasil uji Mann Whitney menunjukkan bahwa terdapat pengaruh yang signifikan sesudah intervensi teh buah mengkudu pada kelompok perlakuan dibandingkan kelompok kontrol dengan nilai tekanan darah sistolik (p=0.000171) dan tekanan darah diastolik (p=0,000480) dengan α=0.05Kesimpulan : Tekanan darah pada kelompok perlakuan susudah intervensi teh buah mengkudu mengalami penurunan tekanan darah dibandingkan dengan kelompok kontrol. Saran penelitian, perlu dilakukan pendidikan gizi secara berkala dan pemberian teh buah mengkudu sebagai obat alternatif yang efektif menurunkan hipertensi, secara terjadwal.

Background: Much alternative therapy such as cupping therapy in the community as one of the cheap treatment efforts without side effects makes the challenge and the goal for nursing staff improve nursing services. The unfinished scientific verification of cupping therapy's alternative therapy makes nurses need to prove decreased blood pressure and cholesterol levels in the elderly. Objective: Scientific proof of cholesterol and blood pressure decrease after Cupping Therapy in an elderly group in Wedomartani Village, Sleman Yogyakarta Indonesia. Method: quantitative type with pseudo experimental research design. Pre-test and Post-test nonequivalent control group. The treatment group was given wet cupping therapy, while the control group was given dry cupping. Respondents were selected using a purposive sampling technique. Blood pressure measurement uses a digital tensimeter and cholesterol measurement using a digital blood cholesterol level measuring device. Results: The elderly blood cholesterol levels after the average therapy decreased 42.89 mg/dl, whereas blood cholesterol levels in the control group decreased an average of 20.95 mg/dl. The blood pressure of the elderly systole in the intervention group after the average therapy decreased 10.74 mmHg, whereas the control group's systole blood pressure decreased an average of 1.58 mmHg. Diastole blood pressure in the treatment group decreased 3,48 mmHg, in the diastole blood pressure control group increased 5,26 mmHg. Conclusions: Wet and dry cupping are both significantly associated with decreased blood cholesterol and blood pressure levels, but wet cupping lowers blood cholesterol levels by a mean difference of 21.95 mg/dl and mean systole blood pressure of 3.74 mmHg and pressure diastole blood difference of mean 9.1 mmHg. The pineal hormone melatonin has some circadian regulatory effects and is assumed to have a close relation with sleep initiation and maintenance. Many previous reports have described age-related decreases in melatonin levels, especially in elderly insomniacs (EIs), which may act as causal or exacerbating factors in sleep disturbances in the elderly. Ten elderly residents with psychophysiological insomnia (mean age, 74.2 yr), 10 healthy residents of the same home [elderly control (EC) group; mean age, 72.7 yr], and 10 healthy young control subjects (mean age, 20.9 yr) living at home participated in this study. The elderly persons, especially the EIs, were exposed to significantly less environmental light and simultaneously suffered from significantly diminished nocturnal melatonin secretion. Supplementary exposure to 4 h (1000 to 1200 h, 1400 to 1600 h) of midday bright light in the EI group significantly increased melatonin secretion to levels similar to those in the young control group without circadian phase-shifting. There was a tendency for the magnitude of the increase in nocturnal melatonin secretion stimulated by bright light to parallel amelioration of sleep disturbances in these subjects. The present findings suggest that we need to pay attention to elderly individuals who suffer under conditions of poor environmental light resulting in disorganized circadian rhythms, including the sleep-wake cycle.

Abstract Background Female sex hormone secretion and reproductive ability decrease with ageing. Bone marrow mesenchymal stem cells (BMMSCs) have been postulated to play a key role in treating ovarian ageing. Methods We used macaque ovarian ageing models to observe the structural and functional changes after juvenile BMMSC treatment. Moreover, RNA-seq was used to analyse the ovarian transcriptional expression profile and key pathways through which BMMSCs reverse ovarian ageing. Results In the elderly macaque models, the ovaries were atrophied, the regulation ability of sex hormones was reduced, the ovarian structure was destroyed, and only local atretic follicles were observed, in contrast with young rhesus monkeys. Intravenous infusion of BMMSCs in elderly macaques increased ovarian volume, strengthened the regulation ability of sex hormones, reduced the degree of pulmonary fibrosis, inhibited apoptosis, increased density of blood vessels, and promoted follicular regeneration. In addition, the ovarian expression characteristics of ageing-related genes of the elderly treatment group reverted to that of the young control group, 1258 genes that were differentially expressed, among which 415 genes upregulated with age were downregulated, 843 genes downregulated with age were upregulated after BMMSC treatment, and the top 20 differentially expressed genes (DEGs) in the protein-protein interaction (PPI) network were significantly enriched in oocyte meiosis and progesterone-mediated oocyte maturation pathways. Conclusion The BMMSCs derived from juvenile macaques can reverse ovarian ageing in elderly macaques.

BACKGROUND: Osteoarthritis (OA) is one of the most common diseases among the elderly. OA occurs due to an imbalance between degradation and synthesis in articular joint tissue, causing changes in joint components such as cells, matrices and molecular production. Therefore, knowledge of cartilage-degrading enzymes such as ADAMTS-4 and iNOS is needed. AIM: This study aims to prove the effect of Mesenchymal Stem Cell Wharton Jelly on decreasing ADAMTS-4 levels as cartilage-degrading enzymes and increasing levels of iNOS which showed the immunosuppressive potential of MSC-WJ in cases of osteoarthritis in vivo. MATERIAL AND METHODS: This research is an experimental study with the design of Post-test-Only Control Group Design. The sample consisted of 16 OA rats as a control group and 16 OA rats treated with MSC-WJ as a treatment group. OA induction is done by injection of monosodium iodoacetate (MIA) into the intra-articular right knee. Giving MSC-WJ is done in the third week after MIA induction. The serum ADAMTS-4 and iNOS levels were measured after 3 weeks treated with MSC-WJ using the ELISA method. The statistical test used is an independent t-test. The value of p < 0.05 was said to be statistically significant. RESULT: The result showed that serum ADAMTS-4 levels were lower in the group treated with MSC-WJ than in the control group, but not statistically significant (p > 0.05). Serum iNOS levels were higher in the group treated with MSC-WJ than in the control group (p < 0.05). CONCLUSION: This study concluded that MSC-WJ significantly reduced ADAMTS-4 levels and increased the serum iNOS levels of OA rats.

This study concerns aspects of positive psychology connected to foreign language learning (FLL) in an older healthy generation. The positive psychology perspective stresses the positive aspects of improved wellbeing in participants who engage in various activities, particularly mental and brain-training practices. Therefore, the aim of this study was to explore older people’s subjective feelings connected to their FLL as one of the crucial ways to improve their quality of life (QoL). The objective of the research was to determine the subjective satisfaction level of the participants of a second language (L2) acquisition course. The research sample (experimental group) consisted of 105 respondents who were Czech citizens and 55+ years old. Two control groups were set up. The first (young control) consisted of 102 young adults (university students), also Czech citizens, aged between 19 and 23 years. The second control group (elderly control) consisted of 102 subjects older than 55 years, similar in age to the experimental group. A standardized online questionnaire survey was the principal research method, identical both for the experimental and control groups. The findings clearly showed that language training significantly improved the subjective positive feelings and wellbeing of the older participants, regardless of their objective progress in FLL itself. These results stood in opposition to the young control group and were different from the elderly control group. The results revealed that FLL is an effective tool for enhancing the overall wellbeing of older people, which was shown in their expression of their feelings of happiness, satisfaction, and positive motivation to learn an L2. In addition, FLL objectively affected their mental health in a positive way and expanded their social networks. Moreover, FLL was a meaningful activity for them, despite the weak objective learning outcomes due to the decline of cognitive functions, helping them find their general purpose of life, as well as life motivation as expressed in the survey. These findings are crucial, as it has already been proven that wellbeing is directly connected with good health and longevity. Therefore, national governments and all stakeholders dealing with the present issue of the aging population should pay undivided attention to the enhancement of older people’s wellbeing by all possible intervention approaches, including FLL. There is limited research into the issue and the findings of this investigation could be an impetus for further research into the topic from the perspectives of cognitive science, psychology, and psycholinguistics.

The LCOT is a self-administered test designed to assess olfactory deficits. Altogether, 525 subjects contributed to the validation. Elderly participants were well represented in this sample. In a validation study (study 1), 407 healthy and 17 anosmic volunteers between 15 and 91 years of age underwent threshold, supraliminal detection, and identification testing. Cutoff values for normosmia and hyposmia were calculated and applied in a second study in a group of patients with smell complaints and in a group of Alzheimer patients with age-matched controls. Incidence of smell deficit was estimated at 5.6% in the healthy population of study 1, and at 16% in the elderly control group of study 2. Assessment of the ability of each subtest to discriminate between groups showed that LCOT is relevant to differentiating between perception and identification deficits and between Alzheimer's and hyposmic patients.

Background: Several various physiological functions in elderly people are diminished due to cell or tissue damage. One of the probable causes are oxidative stress yielded by free radicals.Oxidative stress (ROS) induce lipid peroxidation in endothelial cell membrane, which generates atherosclerotic plaque. In a state of oxidative stress, MDA level will increased. The purpose of this study is to determine the effect of SOD supplementation on MDA, total cholesterol and LDL cholesterol plasma levels in the elderly.Methods: This study was open label, a randomized control trial. Subjects were elderly people aged > 60 years (median 75, 60-82 ys, male 10 (24,4%)) institutionalized at Social Rehabilitation Unit Pucang Gading Semarang, Indonesia. The treatment group consisted of 16 people, received SOD (GlisodinR) 1 capsule (250 IU) 1 hour before meals, plus exercise scheduled for 8 weeks. The control group consisted of 15 people, received placebo, and exercise. Plasma MDA levels were examined using TBARS method, while total cholesterol and LDL cholesterol were examined using CHOD-PAP method.Results: This study show a reduction of plasma MDA levels in the treatment group compare to control group ( p = 0.062 ). A significant reduction of total cholesterol and LDL cholesterol levels in the treatment group were found (before 190.00 and 131.47 g/dl, after 182.27 and 121.93 g/dl, p = 0.005 and 0.001).Conclusion: The SOD supplementation significantly reduce Total Cholesterol and LDL level, but not MDA level in the elderly.

Sekitar 90% usia dewasa dengan tekanan darah normal akan berkembang menjadi hipertensi pada usia lanjut.  Hipertensi pada usia lanjut mempunyai beberapa kekhususan, umumnya disertai dengan faktor resiko yang lebih berat. Masage punggung  bermanfaat melancarkan peredaran darah dan memberikan efek tenang sehingga tekanan darah menjadi stabil. Penelitian ini bertujuan untuk mengetahui pengaruh  terapi  masase punggung terhadap tekanan darah  pada lansia dengan hipertensi. Rancangan penelitian menggunakan quasy experiment dengan desain pretest-posttest control group design, jumlah sampel 20 orang tiap kelompok, dengan  tehnik random sampling. Hasil penelitian menunjukkan rata rata  kelompok perlakuan berusia 73 tahun dan kontrol berusia 75 tahun, jenis kelamin tiap kelompok 10 orang laki laki dan 10 orang perempuan. Pada kelompok perlakuan nilai systole pre-post (164-148.5 mmHg), diastole pre-post (85-80 mmHg).  Kelompok control rata rata nilai systole pre-post  (167,7 - 151.2 mmHg), diastole (87.5-77.5 mmHg). Uji Wilcoxon test didapatkan ada perbedaan yang signifikan tekanan darah pada kelompok perlakuan dengan p value systole adalah 0.000, p value diastole 0.025. Tidak ada perbedaan yang signifikan tekanan darah pada kelompok kontrol baik systole dengan p value 0.086 dan p value diastole 0.140. Ada pengaruh yang signifikan terapi komplementer massage punggung  terhadap tekanan darah systole dengan p value 0.000 dan  p value diastole adalah 0,028. Direkomendasikan terapi massage punggung sebagai terapi non farmakologi untuk menstabilkan tekanan darah guna mencegah komplikasi kardiovaskuler Kata Kunci : massage punggung; tekanan darah; hipertensi; lansia. THE INFLUENCE OF COMPLEMENTER THERAPY ON BLOOD PRESSURE IN ELDERLY WITH HYPERTENSION AT GIANYAR SUKAWATI II HEALTH CENTRE ABSTRACT  Approximately 90% of adults with normal blood pressure will develop hypertension in old age. Back massage is useful for blood circulation and provides a calming effect so that blood pressure becomes stable. This study was aimed to determine the effect of back massage therapy on blood pressure in the elderly with hypertension. The research design used a quasi-experimental design with a pretest-posttest control group design, the number of samples were 20 people per group, with random sampling technique. The results showed that the average age of the treatment group was 73 years old and the control group was 75 years old, the sexes of each group were 10 men and 10 women. In the treatment group the value of pre-post systole (164-148.5 mmHg), pre-post diastole (85-80 mmHg). The control group averaged pre-post systole values (167.7 - 151.2 mmHg), diastole (87.5-77.5 mmHg). The Wilcoxon test showed that there was a significant difference in blood pressure in the treatment group with p-value of systole being 0.000, p-value of diastolic 0.025. There is no significant difference in blood pressure in the control group, both systole with p value 0.086 and p value diastole 0.140. There is a significant effect of complementary back massage therapy on systolic blood pressure with a p value of 0.000 and a diastolic p value of 0.028. BACKGROUND: The population of the elderly is increasing rapidly in almost every part of the world. A common problem faced by the elderly is prolonged depression. One of the practical and economic efforts to reduce depression is by walking and consuming chamomile flowers. AIM: The aim of this study is to analyzing the effect of walking exercise and consumption of chamomile (Matricaria recutita) flower infusion on depression in the elderly in Langsa City. METHODS: We used a quasi-experimental design with a pretest-posttest control group design. This research was conducted from September to December 2020 in Karang Anyar Village, Langsa City. The population in this study were all elderly who met the inclusion criteria. The sample was selected as many as 24 elderly. The sample size of the study was calculated using the formula for the sample size for experimental research from Federer, namely (t–1) (r–1) ≥15). Data collection techniques in this study were interviews and observations using the Geriatric Depression Scale (GDS) questionnaire instrument, which was carried out twice, namely at the time before treatment and after treatment in four groups, namely, the control group, the walking exercise group, the chamomile flower consumption group and the combination group of walking and exercise. Consume chamomile flowers. Data analysis used paired t-test and one-way analysis of variance test to see the effect of the intervention on depression. RESULTS: This study’s results indicate that walking can significantly reduce the GDS depression score with a p-value of 0.025. This study indicates that consumption of chamomile Flower can significantly reduce the GDS score of the elderly with a p-value of 0.037. This study showed that the combination of walking and consumption of chamomile tea was able to significantly reduce the GDS depression score with a p-value of 0.017. When viewed from the p-value, this treatment group showed the most significant reduction in the GDS depression score than the other two treatment groups, namely walking and consumption of chamomile tea. In the walking exercise group, the chamomile flowers were the consumption treatment group combined treatment group walking and chamomile tea consumption. All three treatments had the same ability to reduce the GDS score, as evidenced by a p-value of 0.808 (p > 0.05). CONCLUSION: Walking exercise and consumption of chamomile tea can significantly reduce the GDS score of the elderly in Langsa City.
