The accumulation of total soluble sugars (TSS) and starch and their relationship to flower bud hardiness were studied in three Forsythia taxa: Forsythia ×intermedia `Spectabilis', Forsythia ×intermedia `Lynwood', and F. suspensa. Taxon hardiness was based on the mean temperature at which low temperature exotherms (LTEs) occurred during thermal analysis. Ethanol-extracted soluble sugars were quantified with anthrone, and starch was enzymatically digested and quantified with Trinder reagent. Qualitative changes in sugar content were determined with high-performance liquid chromatography and co-chromatography of authentic standards. Quantitative and qualitative changes in sugar content, similar for the three taxa, were observed in conjunction with fluctuations in flower bud hardiness, although neither TSS nor starch were correlated with mean LTE temperature. TSS was higher in acclimated than nonacclimated buds. However, after deacclimation began, sugars continued to increase with mean LTE temperature. Buds lacked starch except for a brief period during deacclimation. Galactose, stachyose, raffinose, and an unidentified carbohydrate were positively correlated with hardiness (P = 0.005, 0.001, 0.005, and 0.001, respectively).

The aim of the present study was to characterize the phenolic acids, flavonoids, and antioxidant properties of monofloral honey collected from five different districts in Bangladesh. A new high performance liquid chromatography (HPLC) equipped with a UV detector method was developed for the identification of the phenolic acids and flavonoids. A total of five different phenolic acids were identified, with the most abundant being caffeic acid, benzoic acid, gallic acid, followed by chlorogenic acid and trans-cinnamic acid. The flavonoids, kaempferol, and catechin were most abundant, followed by myricetin and naringenin. The mean moisture content, total sugar content, and color characteristics of the honey samples were 18.36 ± 0.95%, 67.40 ± 5.63 g/100 g, and 129.27 ± 34.66 mm Pfund, respectively. The mean total phenolic acids, total flavonoid content, and proline content were 199.20 ± 135.23, 46.73 ± 34.16, and 556.40 ± 376.86 mg/kg, respectively, while the mean FRAP values and DPPH radical scavenging activity were 327.30 ± 231.87 μM Fe (II)/100 g and 36.95 ± 20.53%, respectively. Among the different types of honey, kalijira exhibited the highest phenolics and antioxidant properties. Overall, our study confirms that all the investigated honey samples are good sources of phenolic acids and flavonoids with good antioxidant properties.

Thirty-five samples of commercial cheeses, 9 fresh, 9 semicured or semiripened and 17 ripened made with different types of milk (cow, ewe, goat and mixtures of milk of various species) produced in the South of Spain were analyzed for the presence of aflatoxin M1 (AFM1) by high-performance liquid chromatography, In 16 of the 35 samples (45.71%) the presence of AFM1 was detected in concentrations ranging between 20 and 200 ng/g of cheese, In the positive cases, the mean levels of AFM1 were 105.33 ng/g in ripened cheeses, 73.80 ng/g in semiripened cheeses and 42.60 ng/g in fresh cheeses.

The Physalisbelongs to the Solanaceae family and has sweetish fruit that can be eaten fresh or processed. In Brazil only Physalis peruvianais commercialized, it is necessary research to encourage the cultivation of other promising speciesof the genus. The aim of this study was evaluate the productivity and the physical and biochemical characteristics of P. angulata, P.pubescensand P. ixocarpafruits, in different stages,in Feira de Santana, Bahia. Productivity and the number of fruits in the breeding period were evaluated.In the fruits, longitudinal (LD) and transverse (TD) diameters, the LD/TD ratio,the masses of fruit, the protein content,reducing sugars,and total soluble sugars were analyzed. The results showed differences between species, with higher number of fruits for P. angulata(140,5). P. ixocarpashowed higher productivity (9 t ha-1) and fruit with greater mass and diameter,however P. pubescensand P. angulatashowed round fruits and higher sugar content, desirable characteristics for fresh consumption and for industrialization.

The contents of kaempherol, quercetin and L-ascorbic acid in sea buckthorn berries were measured at different maturation stages using High Performance Liquid Chromatography (HPLC) methods. The content of ascorbic acid decreased over time with significant differences between sampling dates for the five cultivars studied. The mean decrease was 25% in 19 days, from 1.48 to 1.10 g kg-1. Quercetin decreased whereas kaempherol increased during maturation. Among three studied cultivars, the decrease in quercetin was significant (from 0.028 to 0.014 g kg-1) in 'Otradnaja', where as the increase (from 0.012 to 0.016 g kg-1) in kaempherol was significant in the others two, 'Prozratnaja' and 'Gibrid Pertjik'.;

HPLC analysis of the true huckleberry species Gaylussacia baccata, G. dumosa, G. frondosa, G. mosieri, and G. ursina identified the 3-monoarabinosides, 3-monogalactosides, and 3-monoglucosides of cyanidin, delphinidin, malvidin, peonidin, and petunidin. Gaylussacia brachycera contained all anthocyanins, except peonidin-3-arabinoside. Gaylussacia brachycera differed from other species in percent delphinidin-3-arabinoside. It was higher than the other species in percent of the aglycone delphinidin and lower in cyanidin, and also higher in percent of the sugar arabinose. There were no detectable differences among the other species for anthocyanins, aglycones, or aglycone-sugars. The phylogenetic implications of the similarities among species of Gaylussacia and Vaccinium in anthocyanins, aglycones, and aglycone-sugars of the fruit were discussed.Key words: High-performance liquid chromatography, huckleberries, blueberries, chemotaxonomy, taxonomy, biosystematics

Abstract A specific radioimmunoassay involving a mouse monoclonal antibody to cyclosporine has been developed for monitoring the parent drug in blood. Pretreatment with methanol removes cyclosporine from the erythrocytes. The limit of detection is about 12 micrograms/L, sample volume is 50 microL of blood, and within- and between-assay CVs are less than 7%. Assay results correlated well with those obtained by "high-performance" liquid chromatography (HPLC) for liver (n = 42), for heart (n = 64), for bone-marrow (n = 36), and for kidney (n = 140). For blood specimens obtained from patients treated with cyclosporine postoperatively for as long as 65 months, the mean RIA/HPLC ratio in all with transplant indications was close to 1. Therefore, the specific radioimmunoassay apparently can be used instead of HPLC to measure the parent drug in blood.

Fusarium oxysporum (F. oxysporum) is the pathogen that caused fusarium wilt diseases on the tomatoes. The rise of the symptom was caused by secondary metabolic produced by F. oxysporum. The associated with the level of secondary metabolic pathogenesis that cause symptoms of wilt on tomatoes, but secondary metabolic excreted by F. oxysporum was not necessarily wilt in a plant. The phenomenon caused F. oxysporum producing secondary metabolic in the different concentration, either its quantity or quality. The nature of physiology being tested, observed by growing 4 isolates using a medium PDA on the temperature of 10, 15, 20, 25, 30, and 35 °C. The content of secondary metabolic measured on the four isolates using the Notz et al., (2002) and analyzed by using High-Performance Liquid Chromatography (HPLC). The results of the analysis of the nature of the physiological (colour colonies) were that every isolate planted for 8 days shows the variation of white, white redness, white yellowness until purpleness. Based on Methuen Handbook of Colour (Kornerup & Wanscher, 1978), generally isolates tested shows a bright colour, signaled with A letter at every code. Analysis HPLC on the results of secondary metabolic, either quantity or quality, sequentially were: isolates BAR (3; 1,997 ppm); ENR (3; 5,105 ppm); SID (4; 2,135 ppm) and MAL (5; 2,065 rpm). If it was seen by the relationship between the colour of colonies with the production of the secondary metabolic compound, it seemed that the older or darker colonies’ colour the more secondary compounds formed, but dark or old colonies’ colour does not determine a high quality of acid fusaric produced.
