PDO Rotonda’s Red Eggplant Extract: In vitro Determination of Biological Properties and Minerals Bioaccessibility

Background: The Rotonda’s Red Eggplant belongs to the family of Solanum aethiopicum and it is cultivated in a specific area of Potenza (Basilicata, South of Italy) including villages of Rotonda, Viggianello, Castelluccio Superiore and Castelluccio Inferiore. The Red Eggplant cultivated in this area has gained the PDO, “Protected Designation of Origin”. Objective: The aim of this research was to evaluate the use of PDO Rotonda’s Red Eggplant extract as a possible nutraceutical supplement. The antioxidant, antihypertensive, hypoglycemic, and hypolipidemic properties were in vitro evaluated. Methods: The antioxidant activity was investigated by evaluating the scavenging properties against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals and by performing the Ammonium Molybdate and Folin-Ciocalteu assay. The hypoglycemic and antihypertensive activity was studied by evaluating the α-Amylase, α-Glucosidase and Angiotensin Converting Enzyme, respectively, inhibiting activity. In order to evaluate the hypolipidemic activity, the pancreatic lipase inhibiting property was determined and Oil Red O staining assay was performed. Finally, to evaluate the possible use of this extract as a minerals supplement, Selenium, Potassium and Chrome bioaccessibility was studied. Results: The obtained results underline the good antioxidant, hypoglycemic, antihypertensive and hypolipidemic in vitro properties of the PDO Rotonda’s Red Eggplant extract. Moreover, the obtained data show a higher minerals bioaccessibility and this higher value could be ascribable to the natural phytocomplex of PDO Rotonda’s Red Eggplant, which increases the minerals bioaccessibility if compare it with a control sample. Conclusion: The obtained results show that PDO Rotonda’s Red Eggplant extract, might be used as a possible nutraceutical supplement, along with traditional therapies, both for its biological properties and for its minerals bioaccessibility value.

Several exciting lines of research have emerged from the study of animal models of mental disorders. This chapter presents seven opportunities for enhancing the diagnosis and treatment of mental disorders. They include improvements to the system for diagnosis of mental disorders, use of induced pluripotent stem cells from patients to generate neuronal cultures for in vitro determination of effective drug therapies for those individuals, use of data-mining techniques for understanding patient variability, a commitment to a greater focus on the prevention of mental disorders, innovative uses of smartphones to track patients and individuals at high risk of developing a mental disorder, and developing next-generation therapies and delivery systems that target a specific area of the brain rather than the entire brain. A common theme in these seven thoughts for the future is a commitment to bringing precision medicine tools to the treatment of patients with mental disorders.

The work is devoted to predicting and studying biological properties of N-substituted analogs of 3,5-diamino-1,2,4-thiadiazole, which, in their turn, include in the composition of many drugs that exhibit a wide range of pharmacological actions. For searching of new alternative drugs with an antibacterial activity, but lacking resistance of microorganism strains to them, a computer screening of 2N-alkyl-substituted 5-amino-3-imino-1,2,4-thiadiazolines previously synthesized by us was carried out. The prediction of the spectrum of biological activity, as well as the determination of the probable toxicity of these compounds, was performed using the freely available computer programs PASS, Anti-Bac-Pred, and GUSAR. The study of the antibacterial activity in vitro against gram-positive (Staphylococcus aureus, Staphylococcus saprophyticus, Staphylococcus epidermidis) and gram-negative (Escherichia coli, Pseudomonas aeruginosae) bacterial strains was performed by the disco-diffusion method. Experimental data roughly correspond to the predictions.

In this study, the antihypertensive activity of Purafect®-smooth hound viscera protein hydrolysate (VPH) and its peptide fraction with molecular weight (MW) below 1 kDa (VPH-I) was investigated. In addition, the lipase inhibitory activity, as well the anticoagulant potential, in vitro, were assessed. The antihypertensive effects of VPH and VPH-I were studied during 24 h (short-term effect) and 30 days (long-term effect) using high-salt (18% NaCl) and -fructose (10%) diet (HSFD)-induced hypertension. Data showed that, 4 h post-administration of VPH and VPH-I (200 mg/kg BW), the systolic blood pressure of rats was reduced by about 6 and 9 mmHg, respectively. These effects were similar to that obtained with Captopril (~9 mmHg at t = 4 h). On the other hand, exposing the rats to daily to HSFD, coupled to the administration of viscera peptides, was found to attenuate hypertension. In addition, the proteins’ treatments were able to correct lipid and glycemic disorders, by reducing the total cholesterol and triglyceride contents and resorting to the plasma glucose level, compared to the HSFD group. Overall, the present findings demonstrated the preventive effect of VPH-peptides from hypertension complications, as a result of their biological properties.

Various methods for evaluation of virulence properties of causative agents of infectious diseases in vitro were analyzed: molecular-genetic, cultural-biochemical, immunologic, physiologic. Predominant use of molecular-genetic methods, expediency of a complex approach, relevance of search of novel informative parameters of virulence are noted. Study of biological properties of pathogens in vitro is the first screening stage of evaluation of their virulence.

Hospital Infection is a major health problem and affects around 1.5 million people annually around the world. The Amazon region has a wide diversity of native palm trees that have fruits and oilseeds. Astrocaryum vulgare, commonly known as Tucumã in Brazil, belongs to the family Arecaceae. This palm has orange, fleshy, single-egg-shaped fruits that are used for therapeutic purposes in diseases of the eyes and skin due to the high content of carotenoids, oil is used in cooking, health treatment and massage. This study evaluated the antimicrobial activity of the Tucumã oil against 18 microorganisms. The antimicrobial activity of Tucumã was measured through the determination of the Minimum Inhibitory Concentration (MIC), as well as the determination of the Minimum Microbicidal Concentration (CMM) aiming to contribute to the discovery of new antimicrobials against pathogenic microorganisms’ human health and may contribute to the treatment of nosocomial infections. The results showed that the oil of Tucumã presented antimicrobial activity against five important bacteria, four Gram - positive bacteria (Enterococcus faecalis, Enterococcus faecium, Staphylococcus epidermidis and Streptococcus agalactiae) and one Gram - negative (Acinetobacter baumannii).

A virus-infected leaf of Mucuna pruriens was collected and tested for an incidence of a virus through biological properties using various plant host range inoculation tests, longevity-in-vitro, dilution-end-point, thermal-inactivation point and electron microscopy. Results of host range inoculation tests showed the following plant species to be susceptible to the virus; Nicotiana tabacum, N. rustica, N. benthamiana, N. glutinosa, N. occidentalis, N. Sylvesris (Speg and Comes) and others. No plant in the family Convolvulacaee, Cucurbitaceae was found to be infected with the virus.  However Chenopodium quinoa and C. murale in the family Chenopodiaceae were infected with symptoms of mottling and necrotic lesions.  In the family Fabaceae, hybrid cowpea line TVu 76, IT84S – 2114 were infected with mosaic, leaf crinkling and severe leaf curl respectively. The virus had a dilution-end-point of 10-6-10-7, thermal-inactivation-point of 95°C in crude sap of TVu 76. The virus is of agricultural importance because of its incidence on seed coats of legumes which are freely exchanged between agricultural stakeholders.

Pentavalent antimony, as meglumine antimoniate (Glucantime® ) or sodium stibogluconate (Pentostam® ), is the main treatment for leishmaniasis, a complex of diseases caused by the protozoan Leishmania, and an endemic and neglected threat in Brazil. Despite over half a century of clinical use, their mechanism of action, toxicity and pharmacokinetic data remain unknown. The analytical methods for determination of antimony in biological systems remain complex and have low sensitivity. Radiotracer studies have a potential in pharmaceutical development. The aim of this study was to obtain a radiotracer for antimony, with suitable physical and biological properties. Meglumine antimoniate was neutron irradiated inside the IEA-R1 nuclear reactor, producing two radioisotopes 122Sb and 124Sb, with high radionuclidic purity and good specific activity. This compound showed the same antileishmanial activity as the native compound. The use of the radiotracers, easily created by neutron irradiation, could be an interesting tool to solve important questions in antimonial pharmacology.

Background. Sensitization to the fruits of the family Rosaceae is widespread in children population, but its diagnostics is often difficult. Methods. The study included 190 children with suspected allergy to the fruit family of Rosaceae. Results. The presence of sensitization was found in 148 patients. Qualitatively, the results of in vitro and in vivo tests with allergens differed approximately one third of cases. Among children with sensitization to the fruits of a family Rosaceae specific IgE to rBet v 1 was identified approximately in 3/4, rPru p 3 — in 2/3, rBet v 2 — in 1/5 of patients. Conclusion. The results indicate a high informative determination of specific IgE (ImmunoCap, Phadia AB) to the fruits of Rosaceae (apple, pear, peach) and a recombinant allergens (rBet v 1, rBet v 2, rPru p 3).
