Preliminary Phytochemical Screening, Evaluation of the Phenolic Compositions and Antioxidant Activities of Four Iranian Alyssum Species

Background: Alyssum L. is a genus of herbaceous perennial or annual plants belonging to the Brassicaceae family. Little is known about biochemical properties of Alyssum species, specially endemics to Iran. In particular, the species were A. homolocarpum (Fisch. & C.A.Mey.) Boiss., A. lepidotum Boiss., A. maritimum (L.) Lam. (now syn. of Lobularia maritima (L.) Desv.) and A. simplex Rudolph. and eight accessions were studied. These diverse species accessions were collected in six different areas. Materials and Methods: In this work, the Total Phenolic Content (TPC), the Total Flavonoid Content (TFD), the Total Flavonol Content (TFL), the Carotenoid content (Car) and the Anthocyanin content (Ant) of the leaf methanolic extracts of four Alyssum L. species collected in Iran, were assessed. The antioxidant activity assay for every extract obtained from the eight accessions was also carried out according to three distinct methodologies including three different methods including: DPPH, Beta carotene/ linoleic acid and phosphomolibdate assays. Results: The highest TPC, TFD and TFL values were separately observed in two different accessions of A. lepidotum whereas the highest carotenoid content was observed in one accession of A. homolocarpum and the highest anthocyanin content was observed in A. maritimum. Different results were observed for different methodolies for antioxidant evaluations methods and some of them were found to have values, expressed in mg/mL, much lower with respect to the control. The least DPPH activity and the highest total antioxidant activity with phosphomolybdate assay was found in A. lepidotum. Conclusion: The preliminary phytochemical screening and the evaluation of their antioxidant activities were reported here for the first time for Iranian Alyssum species. The findings eventually recommends, the use of those accessions in the ethnopharmacological and nutraceutical fields.

Commercial blueberry production has been a viable industry throughout the world for 95 years; because of blueberry is a good source of antioxidant. Blueberries are especially rich in anthocyanin, a flavonoid with potent antioxidant capacity. The aim of this study was to compare the phenolic quantities, antioxidant activities, anthocyanin, sugar and phenolic compounds of blueberries produced in Turkey with those of similar blueberry varieties produced around the world. As a result of the conducted analysis, the total phenolic content (TPC) amount found in the berries was 77.26-215.12 mg GAE/100 g, the total flavonoid content (TFC) was 30.44-91.69 mg QE/100 g and the total anthocyanin content (TAC) was 43.03-295.06 mg c3-GE/100 g. Examining the antioxidant activities of the berries, DPPH between 1.10-5.65 mg/ml, FRAP between 454.93-36832.96 µmol troloks/100 g, β-Carotene between 40.66-86.48%. It was determined that the natural berries contained much more phenolic compounds and higher antioxidant activity than that of the cultivars The result of HPLC analysis, chlorogenic acid is determined to be the dominant compound in all berries. Furthermore, fructose and glucose are found in all fruits in different quantities while sucrose is found in certain varieties of berries as well. At the end of the performed study the data indicate that wild and cultivars of blueberries are rich sources of antioxidants for local as well international industries importing this fruit for food processing and enormous products.

An extract of seeds of pea was prepared using 80% (v/v) acetone. Six fractions (I–VI) were separated from the crude extract on a Sephadex LH-20 column using methanol as the mobile phase. The antioxidant activity of fractions was investigated in a β-carotene-linoleate model system. For individual fractions, UV spectra were recorded and the content of total phenolics was determined. Results of the β-carotene-linoleate model system indicated that antioxidant activities of separated fractions correlated with their content of total phenolic compounds and they decreased in the order of IV = VI > V > III > I > II. The antioxidant activity of fractions IV and VI was very strong as compared with that of butylated hydroxyanisole (BHA). Absorption maxima from UV spectra showed that flavonoids, and not phenolic acids, were the main phenolic compounds in separated fractions.

Exposure of sunlight lead tropical Sargassum sp. to maintain their growth and moreover to bring up their secondary metabollite for life struggling. Sargassum sp. has bioactive compounds that has a potential antioxidant acitivity such as phenolic compounds as well as chlorophyll and carotenoids. This research was conducted to determine antioxidant activities, phenolic compound and pigmens of Sargassum sp. with different solvent that have different polarities. Sample was macerate with n-heksana, ethyl acetate, methanol and aquadest. All the parameters were done spectrophotometrically. IC50 was used to determine the antioxidant activity by antiradical scavenging activity using DPPH (515 nm). Total phenolic compound were tested by Folin-Ciocalteu solution  and used gallic acid as standard (725 nm). The chlorophylls a content were measured at wavelength 662 nm and 645 nm and carotenoids were measured at wavelength 470 nm. The results showed best IC50 is achieved by aquadest extract (72.95 ±0.22 ppm). The highest Total phenolic compound is achieved by ethyl acetat extract (120.29 ±0,404 mg GAE/g sample). The highest chlorophyll a content is achieved by ethyl acetat extract (18.23 ± 0,049 mg/g sample) and the highest carotenoid content is achieved by ethyl acetat extract extract (60.65 ±0,008  µmol/g sample). It can be concluded that aquadest extract can be categorized as the strong antioxidant and antiradical activity, ethyl acetat as a medium antioxidant activity. The simple and save methods of aquadest extract promising that Sargassum  sp. frorn Indonesia is a good candidate compoud for nutraceutical and cosmeceutical approach.

This study aimed to determine the influence of maturity stages and processing methods (blanching and steaming) on the antioxidant profile and in vitro antioxidant activities of bignay (Antidesma bunius (Linn.) Spreng var. Kalabaw) flesh and seeds. Bignay fruits of three maturities (unripe, half ripe, and fully ripe) were collected from Laguna, Philippines. Each maturity stage was subdivided into three lots. One lot underwent blanching at 90 ± 5°C for 2 minutes, and another underwent steaming at 105 ± 5°C for 5 minutes while the last did not undergo treatment. Seeds of the samples were then separated from the flesh. Both seeds and flesh were freeze-dried, extracted, and analyzed for antioxidant contents (total phenolic content, total flavonoid content, and total anthocyanin content) and antioxidant activity by DPPH, ABTS, and FRAP assays. Results show that both the maturity and processing methods significantly affect the antioxidant content and activity of the samples. Moreover, except for the FRAP assay done on flesh samples, all assays showed that there is significant interaction between the effect of maturity and processing method on the antioxidant contents and activity of bignay flesh and seeds. Results also show that fully ripe flesh and seeds yielded greater antioxidant content and antioxidant activity than their half-ripe and unripe counterparts; whereas, blanched flesh and seeds generally had higher antioxidant activities than their unprocessed and steamed counterparts.

ABSTRACT: This study evaluated the composition and the cancer chemopreventive and antioxidant activities of the seed, skin, and pulp of Maximo hybrid grapes (IAC 138-22) harvested at different ripening stages. The content of total phenolic compounds and tannins were determined by the Folin-Ciocaulteau method, and total anthocyanins were determined using the differential pH method. Different chemical methods assessed the antioxidant activity. Cell viability (macrophage and melanoma skin cancer) and cancer chemopreventive activity were tested in cell lines by quinone reductase induction assay, TNF-α-induced NF-κB inhibitory activity assay, and aromatase inhibitory activity assay. The hybrid grape had high phenolic contents in both seeds and skin, significantly reduced upon ripening. The results showed that fruit ripening significantly affected both the composition and the antioxidant and cancer chemopreventive activities. Except for the anthocyanin content, the most significant results were observed for green seeds, with the greenest stage showing the best results. The green seeds showed the highest antioxidant activity, even when compared to free resveratrol. In the assessment of cancer chemopreventive activity, the green seeds showed the ability to inhibit N-FκB and aromatase activity, important factors for the early stage of cancer development. Besides, no extracts showed cytotoxicity. Therefore, this grape’s green seeds showed strong antioxidant and cancer chemopreventive activities confirmed herein, suggesting beneficial health effects.

The effects of maturity (green-unripe, red-ripe, and purple-fully ripe) and heat treatment (blanching at 90±5°C for 2 mins and steaming at 105±5°C for 5 mins) on the antioxidant content and activity of Philippine bignay (Antidesma bunius (Linn.) Spreng cv. ‘common’) flesh and seeds were investigated. The total phenolic content (TPC), total flavonoid content (TFC) and total anthocyanin content (TAC) were determined using the Folin-Ciocalteau assay, aluminum chloride assay, and pH differential technique, respectively, and the antioxidant activity was determined using DPPH, ABTS, and FRAP assays. Results showed that the antioxidant content and activity of bignay increased with maturity. Fully ripe flesh and seeds were found to have the highest antioxidant content and activity regardless of the heat treatment applied. The TPC (8.06±0.15 mg gallic acid equivalents (GAE)/g), TFC (8.10±0.06 mg quercetin equivalents (QE)/g), TAC (4.07±0.03 mg cyanidin-3-glucoside equivalents (CGE)/L extract), and antioxidant activity (ABTS 10.37±0.04 , DPPH 9.60±0.10, FRAP 24.37±0.53 Trolox equivalents (TE)/g) of fully ripe seeds were superior when blanched. On the other hand, the TPC (40.73±0.04 mg GAE/g), TFC (31.54±0.15 mg QE/g), TAC (20.93±0.12 mg CGE/L extract), of fully ripe flesh were superior when not heat-treated while its antioxidant activity (ABTS 34.19±0.19, DPPH 14.01±0.02, FRAP 87.78±1.22 (TE)/g) was highest when blanched but did not vary significantly compared to unheated samples. This study suggested that bignay ‘common’ variety was a good source of antioxidants particularly the flesh at fully ripe stage. Blanching had shown to enhance its antioxidant activities.

Vismia genus is distributed mainly in tropical and subtropical regions of Central, South America and some areas of Africa. According to previous investigations, antioxidant potential of Vismia species might be related to anthrones, anthraquinones, flavonoids and phenol derivatives biosynthesized by these plants. The aim of present study is to evaluate the free radical scavenging capacity, total phenolic and flavonoids content as well as the qualitative phytochemical screening of methanol extracts obtained from Vismia macrophylla (VM) and Vismia baccifera (VB) collected in Táchira and Mérida state, Venezuela. Phytochemical screening of VB and VM methanolic extracts carried out using various chemical assays revealed an abundant presence of anthraquinones in both species analyzed. Glycosides were also present while flavones and dehydroflavones were observed abundantly in VB but moderated in VM. Triterpenes were also detected and steroids showed to be abundant in VM but moderate in VB. On the other hand, antioxidant capacity measured by the DPPH radical scavenging assay showed that VM possesses a stronger antioxidant activity than VB with IC50 5.50 µg/mL. Phenol and flavonoid assays also revealed that methanol extracts of both VM and VB contain high concentrations of these metabolites. A relationship between the antioxidant activity, total phenol and flavonoids content of the extracts analyzed was demonstrated in present investigation since those samples with higher phenolic concentrations showed likewise higher antioxidant activity.

Abstract Objectives: The aim of this study was to assess storage effects on anthocyanin and total polyphenol content in different bilberry and blueberry extracts and to evaluate the antioxidant and antibacterial activity of these extracts. Materials and methods: Total phenolic content, total monomeric anthocyanin content and antioxidant activity were determined in the first month and after three months storage of berries at either -20 °C or -50 °C. Two different solvents were used (methanol and 50% ethanol). Antibacterial activity was determined for the 3 months stored fruits using a microdilution method and was expressed as the minimum inhibitory concentration. Results: There were significant differences between the concentration in the first month and after three months storage in both types of fruit extracts. Regarding the extracting solvent, we noticed that total phenols were better extracted with 50% ethanol, while the total monomeric anthocyanin content was higher in the methanolic extracts. No significant or slightly significant differences were observed between the fruits stored at -20 °C or -50 °C. Ethanolic extracts showed the highest scavenging activity. Good antibacterial activity was observed on gram-positive bacteria. Conclusions: Storage conditions are an important factor that can influence chemical composition of fruits. Although freezing is a good option for preservation, our study showed a high decrease in the concentration of total phenols and anthocyanins after only three months. The fruits have shown a high antioxidant activity and a good antibacterial effect. Further studies are needed for better understanding the changes that can appear during the storage.

Heavy metals (HM) are toxic components present in the earth’s crust that can have a negative impact on plants as well as animals. Andrographis paniculata or ‘King of bitters’ belonging to the family Acanthaceae, is a medicinal herb traditionally used in the treatment of fever, common cold etc. In the present study, the effect of heavy metals (copper, tin and cobalt) on the andrographolide content, biochemical parameters like chlorophyll, carotenoid, protein, Total phenolic content (TPC), Total flavonoid content (TFC) and antioxidant activity in A. paniculata were analysed. Saplings of A. paniculata were treated at 50 and 100 mM concentrations, three different times at a time interval of 7 days. Andrographolide production was found to increase in copper and cobalt treated saplings when compared with the control. From the results, maximum andrographolide concentration was found in the saplings treated with 50 mM copper (8.51 mg/g of DW) and 50 mM tin (8.10 mg/g of DW) respectively. 50 mM cobalt treated plants have shown the highest concentration of TPC (17.21 mg/g of extract) and TFC (6.97 mg/g of extract). Notable variations in other biochemical parameters like total chlorophyll, carotenoid content and antioxidant activities were observed in all treatments compared with the control.
