Ziziphus jujuba Mill. is one of the most important fruit crops and has been cultivated in China for more than 4000 years. Z. jujuba fruit is rich in nutritional and medicinal values. Compared to other wood fruits, Z. jujuba is unique in its flowering and fruiting characteristics. Floral buds differentiation and formation of Z. jujuba is an essential process that affects yield. Z. jujuba ‘Jinsi No.4’ blooms profusely, yet its final yield is low. In this study, the floral bud differentiation and development of ‘Jinsi No.4’ were examined by paraffin section. Results showed that the floral buds of ‘Jinsi No.4’ differentiated in the current year and started from early April. The duration of a single flower differentiation was short, taking only 7 days for maturation of flowers buds. Floral bud differentiation of ‘Jinsi No.4’ can be divided into six stages, i.e., pre-differentiation, initial differentiation, sepal differentiation, petal differentiation, stamen differentiation, and pistil differentiation. Flower development experienced seven stages, i.e., alabastrum, alabastrum break, sepal flattening, petal flattening, stamen flattening, filament withering, and ovule swelling. Dysplasia was observed in some floral organs in Z. jujuba ‘Jinsi No.4’, suggesting that the dysplasia of floral organs may be one of the main reasons for low yields. Our findings on flower bud development in ‘Jinsi No.4’ will contribute to its production and flowering management in Hunan area of southern China.

The induction and differentiation phases of reproductive buds of Coffea arabica L. have not received much attention. In the present paper, axillary buds from five plagiotropic branches that developed in the same growing season without fruits (1st crop branches), and from green segments of five recently grown plagiotropic branches with fruits (2nd crop branches), were collected every two weeks during successive inductive months of the year. This study was carried out with adult arabica trees, Catuaí Vermelho cv. IAC 81, cultivated under normal farming conditions in the region of Campinas, SP, Brazil (22º54' Lat. S). Slides of longitudinal-axial sections of 10-12 mum thickness were mounted for the characterization and quantification of histological stages of bud differentiation. The results indicate that the regulatory signals controlling the phases of induction and differentiation of floral buds are distinct, and that there are differences in the response between branches with and without fruits. In the case of 1st crop branches (no fruits present), induction of floral buds took place in January and February, whereas floral bud differentiation was observed during the months of March and April. In 2nd crop branches (fruits present), the induction of floral buds was observed during any month of the year provided that they had already overcome their juvenile state (October-July, in this study). In these 2nd crop branches, the flower bud differentiation was only observed after harvesting all pre-existing fruits of each branch (after May, in this study), which suggests that floral bud differentiation in Arabica coffee is influenced by the source-sink relationship, i.e. by the presence of developing fruits within each plagiotropic branch.

To investigate whether reproductive disorders exist in the sexual reproduction of Ziziphus jujuba Mill. ‘Zhongqiusucui’ and to understand the reproductive biology of ‘Zhongqiusucui’ and genetic improvements in jujube trees, we used ‘Zhongqiusucui’ flowers at different developmental stages as materials and conducted field and microscopic observations on the developmental pattern of mega- and microsporogenesis, as well as on the development of male and female gametophytes. The results show the following. 1) From the inflorescence development stage to flowering, the grade 0 bud on the inflorescence exhibited an increase in horizontal diameter, longitudinal diameter, peduncle length, and bud weight, but the rates of increase were different. From day 1 to day 5 after the inflorescence had developed, floral buds mostly grew horizontally. Day 5 was the floral bud flattening stage. From day 6 to day 8 after the inflorescence had developed, floral buds mostly grew longitudinally, and day 8 was the floral bud enlarging stage. 2) The stamens of ‘Zhongqiusucui’ had five anthers, and there were four locules per anther. The anther wall consisted of epidermis, endothecium, one- to two-layered middle layer, and a secretory-type tapetum. In addition, the development of the anther wall belonged to the basic type. The cytokinesis of the microsporocytes was synchronous, the tetrads mostly arranged as a tetrahedron, and the mature pollen had three germ pores, three grooves, and was bicellular pollen. During meiosis, the microsporocytes in each locule were at the same phase and therefore exhibited synchrony. Among the different anthers in the same floral bud, as well as the four locules in the same anther, the microsporocytes had asynchronous meiosis. 3) The pistils in the ‘Zhongqiusucui’ had two ovaries, two anatropous ovules, inner and outer integument, crassinucellate tetrads formed by the meiosis of megasporocytes aligned linearly along the nucellus, megaspore at the chalazal end that developed into the functional megaspore, which underwent mitotic division three times and developed into the mature embryo sac containing seven cells and eight nuclei, and embryo sac development of the Polygonum type. 4) The external morphology of the ‘Zhongqiusucui’ floral buds correlated with the internal developmental stage of the male and female gametophyte. Therefore, the internal developmental progress of the stamen and pistil can be determined by the external morphological characteristics of the floral buds.

This research aimed to explore the reproductive characteristics of three species of Psidium (P. guajava L., P. cattleyanum Sabine, and P. guineense Sw.) and estimate a probable reproduction strategy based on the pollen:ovule (P:O) methodology. The number of pollen grains per floral bud (NGPB), number of pollen grains per anther (NGPA), number of anthers per flower bud (NAB), number of ovules per flower bud (NOB), and the P:O ratio of each species were estimated. All species had a P:O ratio over 2,000 and were classified as xenogamous. P.guajava presented the highest values for all characteristics evaluated, with the NGPB at 3,777,519, the NOB at 584.50 and a P:O ratio of 6,462.82. Similarly, P. cattleyanum had a P:O ratio of 5,649.89 (NGPB 762,736 and NOB 135). However, P. guineense was considered facultative xenogamous, with P:O of 2,085.75, the NGPB at 741,484 and the NOB at 355.50. Thus, it was concluded that the studied species have a preference for allogamy and require many pollen grains to fertilize each ovule, demonstrating that the transfer of pollen to the stigma is not very specialized.

AbstractPhyllodiaptomus tunguidus and Heliodiaptomus falxus are dominant copepods species in drinking water processing plants in southern China. With a potential penetration risk, the breeding and leakage of copepods are drawing more and more attention in recent years. The current study provided a thermal treatment method to control copepods and their eggs. Results showed that: (1) the immediate death rates of P. tunguidus and H. falxus after heated to 34–40 °C for 5 min are positively correlated to the treatment temperatures (P < 0.01), and all individuals of the both species were eliminated after heated at 40 °C for 5 min; (2) overall hatching rates of P. tunguidus eggs were negatively correlated with treatment temperatures (P < 0.01) between 39–45 °C, with zero percent hatched after treatment at 45 °C for 5 min; (3) hatching rates of H. falxus were negatively correlated with treatment temperatures (P < 0.01) between 37–41 °C, with no nauplii hatched when treated at 41 °C for 5 min; (4) paraffin section histological examination indicated that thermal treatment caused severe damage to internal organs and egg structure. Finally, based on the experimental data, the application of the thermal treatment method was discussed in ozonation combined with biological activated carbon (O3/BAC) processing of drink water treatment.

A new freshwater goby, Rhinogobius maculagenys sp. nov., was collected from Hunan Province in Southern China. This species can be distinguished from all congeners by a combination of the following features: first dorsal fin with 6 spines; second dorsal fin with a single spine and 7–9 segmented rays; anal fin with a single spine and 6–8 segmented rays; pectoral fin with 16 segmented rays; 32–34 longitudinal scales; 9–13 transverse scales; 11+16=27 vertebrae; pore ω1 missing; head and body yellowish brown; cheek and opercle yellowish brown with over 30 small orange spots, branchiostegal membrane yellow with over 10 small orange spots in males and white and spotless in females; first dorsal fin trapezoidal in males and nearly semicircular in females, with large bright blue blotch in front of second spine; spines 4 and 5 longest, rear tip extending to base of second branched ray of second dorsal fin in males when adpressed, but just reaching or not reaching anterior margin of second dorsal fin in females; caudal fin with 5–6 vertical rows of brown spots; flank with several longitudinal rows of blackish-brown spots; and belly pale white.

Rosa chinensis is one of the most popular flower plants worldwide. The recurrent flowering trait greatly enhances the ornamental value of roses, and is the result of the constant formation of new flower buds. Flower bud differentiation has always been a major topic of interest among researchers. The APETALA1 (AP1) MADS-box (Mcm1, Agamous, Deficiens and SRF) transcription factor-encoding gene is important for the formation of the floral meristem and floral organs. However, research on the rose AP1 gene has been limited. Thus, we isolated AP1 from Rosa chinensis ‘Old Blush’. An expression analysis revealed that RcAP1 was not expressed before the floral primordia formation stage in flower buds. The overexpression of RcAP1 in Arabidopsis thaliana resulted in an early-flowering phenotype. Additionally, the virus-induced down-regulation of RcAP1 expression delayed flowering in ‘Old Blush’. Moreover, RcAP1 was specifically expressed in the sepals of floral organs, while its expression was down-regulated in abnormal sepals and leaf-like organs. These observations suggest that RcAP1 may contribute to rose bud differentiation as well as floral organ morphogenesis, especially the sepals. These results may help for further characterization of the regulatory mechanisms of the recurrent flowering trait in rose.

The effects of three temperature treatments on morphological changes in the apical meristem and contents of GA3 and IAA in leaves during floral bud differentiation in early maturing cultivar of broccoli were studied. Plants went through every stage of flower-bud differentiation at day/night temperatures of 17.3±1/9.3±1°C. At 21.3±1/13.3±1°C, floral bud development ceased after primary axillary scape primordium differentiation and apical meristem entered a reversion stage. The apical meristem remained in the vegetative growth phase in plants growing at 25.3±1/17.3±1°C. Leaf GA3 contents started to increase while IAA contents started to decrease when plants entered the flower bud initiation stage. Getah virus (GETV) has caused many outbreaks in animals in recent years. Monitoring of the virus and its related diseases is crucial to control the transmission of the virus. In the summer of 2018, we conducted routine tests on clinical samples from different pig farms in Guangxi province, South China, and isolated and characterized a GETV strain, named GX201808. Cytopathic effects were observed in BHK-21 cells inoculated with GX201808. The expression of E2 protein of GETV could be detected in virus-infected cells by indirect immunofluorescence assays. Electron microscopic analysis showed that the virus particles were spherical and ~70 nm in diameter with featured surface fibers. The multistep growth curves showed the virus propagated well in the BHK-21 cells. Molecular genetic analysis revealed that GX201808 belongs to Group 3, represented by Kochi-01-2005 isolated in Japan in 2005, and it clustered closely with the recently reported Chinese strains isolated from pigs, cattle, and foxes. A comparison of the identities of nucleotides and amino acids in the coding regions demonstrated that the GX201808 showed the highest amino acid identity (99.6%) with the HuN1 strain, a highly pathogenic isolate resulting in an outbreak of GETV infection in swine herds in Hunan province in 2017. In the present study, GETV was identified and isolated for the first time in Guangxi province of southern China, suggesting that future surveillance of this virus should be strengthened.
