Objective: This research was conducted to determine the potential antidiabetic activity fractions of purified extract Lawsonia inermis leaves in mice (Mus musculus) and identification of the compound. Methods: The method included maceration, purification using ethanol and distilled water was followed by liquid-liquid extraction using ethyl acetate and magnesium sulfate as drying agents. Furthermore, the extract was analyzed using thin layer chromatography (TLC) for testing the purified extract. Fractionation using vacuum liquid chromatography, antidiabetic activity test of fractions at dose 100 mg/kgBW with alloxan induced and compound identification by Liquid Chromatography-Mass Spectrometry (LC-MS/MS) using HPLC connected to a Q-TOF spectrometer equipped with an ESI source, with Phenomenon column C8, and methanol with 0.3% formic acid as solvent. Results: The results showed that from the purification step of L. inermis leaves by vacuum liquid chromatography method, 7 fractions were obtained, i.e. A-G fractions. While the antidiabetic activity of fractions shown by decreasing blood sugar level in mice on the 15th day were 64, 75, 73, 73, 57, 45 and 67%, respectively. The identified compounds from each fraction were the ester groups namely 12-hydroxy-methyl abietate, 9,12-octadecadienoic acid (Z,Z)-(2,2-dimethyl-1,3-dioxolan-4-yl)methyl ester, dehydromorroniaglycone, and (E)-hexadecyl-ferulate; the steroid group namely siraitic acid E; phenylpropanoid groups namely umbelliferone and bletilol C, and the alkaloid groups namely moupinamide and valine. Conclusion: L. inermis leaves had activity in lowering blood sugar levels. LC-MS/MS analysis revealed the presence of ester groups, steroid groups, phenylpropanoid groups and alkaloid groups. The presence of these compounds mostly contribute to antidiabetic activity.

Bioassay-guided isolation of bioactive compound is a modern and efficient technique in metabolites screening. It may shorten the total time of the entire process and reduce some costs of it. The aim of this paper was to fractionate and isolate alkaloids by developing an innovative vacuum liquid chromatography method for a species of Narcissus c.v. ‘Hawera’ rarely investigated so far and establishing the inhibitory activity of acetylcholinesterase (AChE). The studies consisted of the extraction of plant material by modern pressurized liquid extraction (PLE), followed by the isolation of alkaloidal fractions. For this purpose, the pioneering gradient vacuum liquid chromatography (gVLC) technique was employed by using two sorbents in various proportions packed in polypropylene cartridges for the first time. This step was performed in order to pre-clean the samples but also to establish the best combination of sorbents which permits obtaining potentially strong AChE inhibitors. The collected fractions were examined by HPLC/ESI-QTOF-MS in order to compare which combination of sorbents would allow us to obtain the highest concentration of alkaloids. The combination of these techniques confirmed the presence of the alkaloids and enabled the development of a modern method for the fractionation and isolation of the compounds with strong anti-AChE activity.

A rapid and sensitive high performance liquid chromatography method for quantifying simultaneously sucrose, fructose and glucose in fruits and vegetables is reported. Samples were extracted in 95% ethanol, homogenized and treated at 95C for 20 min. The supernatant was stored at -20C overnight and filtered through a G-25 Sephadex column. Aliquots were evaporated, redissolved in water, filtered, and injected. A Sorbex NH2 column operated at room temperature was used for separations. The sugars were detected at 192 nm. The retention times were 4.8, 5.9 and 10.3 min for fructose, glucose and sucrose, respectively. The method was applied to twenty-one fruit and vegetable species with different maturity stages. In addition, quality characteristics such as firmness, pH, acidity, soluble solids and color were evaluated. Main sugars for the different samples varied among species. In temperate fruits, fructose and glucose were the predominant sugars, while in tropical and subtropical fruits, the main sugar was sucrose. On the sampled vegetables, fructose was the primary sugar, although at very low levels. Quality characteristics coincided with sugar levels found among the different species.

Background. Some anorexic agents are used to fraudulent augmentation herbal weight loss formulations. This study was designed to evaluate the potential existence of illicit substances in 63 herbal weight loss formulations collected from local apothecaries in Hamadan, Iran. Methods. The thin-layer chromatography method was applied for the primary screening of potential illicit substances in the samples. The positive samples were analyzed using an isocratic high-performance liquid chromatography method. Results. The results showed that 26.98% of the samples contained 17.76 ± 6.02 mg/cap of sibutramine. Daily therapeutic dose intake of sibutramine is in the range of 5 to 15 mg daily. Conclusion. Since apothecaries have advised consumers to take at least two capsules a day, it seems that the blood concentration of sibutramine will likely rise beyond the therapeutic concentration and become toxic. Therefore, the usage of such products could pose serious risks to consumers’ health.
