Now a days there are numerous applications have been proposed for the use of resealed erythrocytes as carrier for drugs, enzyme replacement therapy etc. Until other carrier systems come of age, resealed erythrocytes technology will remain an active field for the further research. The use of resealed erythrocytes shows potential for a safe and effective delivery of various bioactive molecules for effective targeting. In coming future, erythrocyte based drug delivery system with their capability to afford controlled and site specific drug delivery have been developed for disease management. Erythrocyte carriers are “Nano devices in the field of Nanotechnology”. A large amount of valuable work is needed so as to utilize the potentials of erythrocytes in passive as well as active targeting of drugs in diseases like cancer. At present erythrocytes are most effective carriers in novel drug delivery systems considering their tremendous potential. Hence the present article is reviewed about method of drug loading, evaluation and applications of RSE.

From the past two decades, technological advancements in science and chemistry made possible many new drug delivery systems that have the potential to completely change the course of routine therapeutic ways.  Lipid and polymer-based drug delivery systems are considered to be the pillars of many drug dosage forms, irrespective of their route of administration. With increasing knowledge on their chemistry, lipids and polymers are being modified and used as potential novel drug delivery systems with smart polymers and lipid nanotechnology paving the way for efficient drug delivery into the patient. This review article covers the swing of these drug delivery systems in the current market and interpreting all this from a health care professional’s point of view. Background: Recent past decades have reported emerging of polymeric nanoparticles as a promising technique for controlled and targeted drug delivery. As nanocarriers, they have high drug loading and delivery to the specific site or targeted cells with an advantage of no drug leakage within en route and unloading of a drug in a sustained fashion at the site. These stimuli-responsive systems are functionalized in dendrimers, metallic nanoparticles, polymeric nanoparticles, liposomal nanoparticles, quantum dots. Purpose of Review: The authors reviewed the potential of smart stimuli-responsive carriers for therapeutic application and their behavior in external or internal stimuli like pH, temperature, redox, light, and magnet. These stimuli-responsive drug delivery systems behave differently in In vitro and In vivo drug release patterns. Stimuli-responsive nanosystems include both hydrophilic and hydrophobic systems. This review highlights the recent development of the physical properties and their application in specific drug delivery. Conclusion: The stimuli (smart, intelligent, programmed) drug delivery systems provide site-specific drug delivery with potential therapy for cancer, neurodegenerative, lifestyle disorders. As development and innovation, the stimuli-responsive based nanocarriers are moving at a fast pace and huge demand for biocompatible and biodegradable responsive polymers for effective and safe delivery.

The herbal-based drugs have been getting wide attention over the last few decades. The herbal drugs have been used from the ancient times, but the dose accuracy, efficacy, and treatment was just based on symptomatic observatory. Modern research is focused on pure bioactive compound instead of crude. The researchers are trying to apply the current advanced nanotechnology-based knowledge on the herbal medicines to improve efficacy and reduces the unwanted pharmacological actions' adverse effects of the therapeutic molecules. The many novel drug delivery systems available used for the herbal bioactive molecules include polymeric micelle, injectable hydrogel, nano gel, nano emulsions, microsphere, polymeric nanoparticles, nano capsules, liposomes, phytosomes, transferosomes, dendrimer, and ethosomes. This chapter highlights the existing research and development of current standing of the of various novel formulations especially for the herbal bioactive molecules and summarizes their active ingredients, commercial formulations, route of drug administration, and pharmacological activity.

The development of micro and nanobubbles as theranostics is been an emerging trend in the 21st century. Ultrasound molecular imaging is a real-time non-invasive, cost-effective, promising non-viral tool, which is been widely used in the recent times. These micro and nanobubbles are marked as ultrasound agents for both diagnosis and targeting therapeutic agents. These are designed in order to obtain efficient drug delivery. Micro and nanobubbles are very much used for targeting drug and achieving site specific release. These are stable and have longer residence time in systemic circulation, finally shows efficient and promising drug delivery [1]. These echogenic bubbles are helpful for disease diagnosis and therapy more prominently when compared with other novel drug delivery systems. This review describes about functioning of micro and nanobubbles along with mechanism, preparation and studies which describes the works on micro and nanobubbles.

Nanosuspensions are important carriers to develop novel drug formulations. Nanosuspensions have emerged as a promising strategy for the efficient delivery of hydrophobic drugs because of their versatile features and unique advantages. Nanosuspension technology solved the problem of drugs which are poorly aqueous soluble and less bioavailability. Stability and bioavailability of the drugs can be improved by the Nanosuspension technology. Techniques such as media milling and high-pressure homogenization have been used commercially for producing nanosuspensions. Nanosuspensions can be delivered by oral, parenteral, pulmonary and ocular routes. Nanosuspensions can also be used for targeted drug delivery when incorporated in theOcular inserts and mucoadhesive hydrogels. Currently, efforts are being directed to extending their applications in site-specific drug delivery.

Oral controlled release and site specific drug delivery system has been of great interest in pharmaceutical field to achieve improved therapeutic advantage. Concept of novel drug delivery system arose to overcome certain aspect related to physicochemical properties of drug molecule and the related formulations. Gastro retentive drug delivery system is one of such novel approaches to prolong gastric residence time, thereby targeting site specific drug release in the stomach for local or systemic effects. This approach is useful particularly for the drugs which have narrow absorption window in the upper part of gastro intestinal tract. In this review we have been discussed various approaches of gastro retentive drug delivery system, such as floating and non-floating systems.
