In this study, the antioxidant properties and total phenolic and flavonoid contents of Rubus caesius L. var. aqu?ticus Weihe & Nees leaf extracts were examined. The radical scavenging capacity of four leaf extracts (methanol, ethanol, acetone and aqueous) was estimated against 2,2-diphenyl-1-picrylhydrazil (DPPH) and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assays. Total antioxidant capacity was tested by ferric reducing ability of plasma (FRAP) and total reducing capacity (TRC) procedures. Total contents of phenols and flavonoids were also determined. Total phenol content in the extracts was determined using Folin-Ciocalteu reagent and amounts ranged between 263.74 for ethanol to 366.27 mg gallic acid (GA)/g for acetone extracts. The amounts of flavonoids varied from 10.73 mg quercetin equivalent (QE)/g for aqueous to 75.83 mg QE/g for acetone extract. The highest antioxidant activity against DPPH and ABTS?+ radicals was that of the acetone extract. Values for FRAP varied between 3.00 ?mol Fe+2/mg for the ethanol extract and 5.31 ?mol Fe+2/mg for the methanol extract. The results obtained for total reducing capacity indicate that the acetone extract has the highest antioxidant potential.

Aim: This study investigated the phytochemical constituents and in vitro antioxidant properties of methanol and aqueous leaf extracts of Geophila obvallata using standard methods. Materials and Methods: The in vitro antioxidant assays carried out were 1, 1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging ability, Nitric oxide (NO•) radical scavenging activity assay, 2, 2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS•+) radical cation scavenging assay, ferric reducing properties and hydroxyl radical scavenging assays. Results: Phytochemical analysis revealed the presence of alkaloids, flavonoids, phenolic compounds, steroids, saponins, terpernoids and cardiac glycosides in both extracts. Relative to the aqueous extract, the methanol extract contained a higher amount of the secondary metabolites. However, both extracts exhibited appreciable and dose-dependent capacities for quenching DPPH, ABTS•+ and NO• free radicals, and potent ferric reducing ability to levels comparable to those of ascorbic acid. The crude methanol extract showed significantly increased (P<0.05) antioxidant activity than the aqueous extract. Conclusion: It was concluded that the extract possesses strong antioxidant properties due to its content of phytochemicals, and provides scientific basis for its ethno medicinal applications.

In order to facilitate the quality control of Trifolium repens flowers and leaves, a RP-HPLC method with UV detection was developed for the simultaneous quantitative determination of flavonols and isoflavones. The total flavonoid and phenolic (TPC) contents were determined spectrophotometrically in a visible part of the light spectrum at 425 and 760 nm, respectively. Additionally, evaluation of the antioxidant properties of the plant materials was performed using the DPPH in vitro test. The results showed that the flowers are the richest source of phenolics ranging from 28.7 to 38.8 mg GAE/g, and flavonoids, calculated for hyperoside, up to 20 mg HP/g, which hydrolyzed mainly to flavonols (the quercetin level greater than 6 mg/g). T. repens is poor in isoflavones; similar quantities of ca. 0.2 mg/g were detected in the flowers and leaves. The flower and leaf extracts showed antioxidant activity towards DPPH with EC50 values ranging from 72.3 to 179.3 μg/mL. Significant linear correlations were found between antioxidant potentials of the studied plant materials and total phenolic and flavonoid contents determined by HPLC and spectrophotometric methods ( R2 in the range of 0.97 - 0.99).

AbstractVaccinum myrtillus L., Ribes nigrum L., Rubus fruticosus L., Fragaria vesca L. leaves are considered an agro-waste of the berry industry. Although numerous studies indicate fruit is a rich source of bioactive compounds, the authors prove leaves can also be a valuable source of compounds used in the pharmaceutical and cosmetic industries. The study attempts to assess and compare the antioxidant and cytotoxic properties of berry leaves extracts. The total phenolic compounds, flavonoids, anthocyanins and procyanidins content were determined. Antioxidant potential was evaluated using the DPPH radical scavenging method. Cytotoxicity studies were conducted to evaluate the effect of the extracts on the metabolism and proliferation of keratinocytes and fibroblasts in vitro. The effect on the migration capacity of these cells was also assessed. The obtained results show that the examined extracts are a source of valuable bioactive agents. All tested extracts show significant ability to remove free radicals in higher concentrations. Cytotoxicity assessments have shown that leaf extracts of the analyzed plants differ in cytotoxicity, both for keratinocytes and fibroblasts. The results of the assessment of cell migration capacity correlate with cytotoxicity tests, because the concentration of extracts showing cytotoxic activity towards the tested cells also inhibited their migration.

The purpose of this study was to determine the antioxidant activity of the seed shells and kernels of Jatropha curcas L. The extracts obtained from five solutions (0%–95% ethanol) were tested and compared. Overall, the antioxidant capacity of seed shell extracts was higher than that of seed kernel extracts. The seed shell extract obtained using 95% ethanol exhibited the best antioxidant activity among the five solutions. The half-maximal inhibitory concentration (IC50) of 1,1-diphenyl-2-picrylhydrazyl and free radical scavenging ability of 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) were 13.63 ± 0.15 and 6.75 ± 0.51 μg/mL, respectively. The reduction ability and total phenol content were 95.14 ± 27.04 μg ascorbic acid equivalents/mg of extract and 536.33 ± 8.62 μg gallic acid equivalents/mg of extract, respectively. In in vitro cytotoxicity assays, solutions with less than 250 μg/mL of seed shell extract had no major cytotoxicity. The seed shell of Jatropha curcas L. can be used as an antioxidant material and has potential for biomedical applications.

Echinocystis lobata (Mich.) Torr. et Gray extracts were investigated for their bioactive compounds and their potential for natural antioxidant properties. In vitro antioxidant activity of methanol, ethanol, and water extracts were evaluated by 1,1-diphenyl-picrylhydrazyl (DPPH) radical scavenging activity. Total phenolic content (TPC) and total flavonoid content (TFC) of fruits and seeds were determined. TPC is a fairly constant value and varies weakly both among the different organs of Echinocystis lobata and among the geographical locations of the populations studied. Like other invasive species, Echinocystis lobata is superior to many other plants in terms of the total amount of flavonoids contained in its fruits and seeds. The dried fruits contain more flavonoids than the seeds. Seeds from Slovak populations have higher flavonoid content than those from Russian populations. The results demonstrated that Echinocystis lobata can be a good source of natural antioxidants and have a great potential for commercialization, especially in the pharmaceutical industry.

Crude extracts from a medicinal Tunisian plant,Pergularia tomentosaL., were the investigated natural material. Butanolic extract of roots analyzed with IR spectra revealed the presence of hydroxyl, alcoholic, and carboxylic groups and sugars units. Analysis of some secondary metabolites, total phenolic, flavonoids, flavonols, and procyanidins, was performed using different solvents following the increased gradient of polarity. Fruits and leaves contained the highest amounts of all these compounds. Antioxidant properties were evaluated by the determination of free radical scavenging activity and the reducing power of methanolic extracts. Fruits and leaf extracts were the most powerful antioxidants for the two-assay in vitro system. Stems and fruits extracts exhibit an antifungal activity againstFusarium oxysporum f. sp. lycopersiciwhich could become an alternative to synthetic fungicide to controlSolanumspecies fungal diseases.

Objectives: The objective of this study was to estimate the total phenolic and flavonoid contents and to explore the antioxidant potential of various leaf extracts (chloroform, ethyl acetate, and ethanol) of Canthium coromandelicum, which is considered traditionally as an important medicinal plant.Methods: Antioxidant properties of the extracts were assessed using 1, 1- diphenyl -2- picrylhydrazyl and hydrogen peroxide radical scavenging assays for ascorbic acid equivalents. The total phenolic and flavonoid contents were also investigated to determine their correlation with the antioxidant activity of the leaf extracts and expressed in Gallic acid and quercetin equivalents, respectively.Results: The results showed that the content of total phenols and flavonoids was found to be high in ethyl acetate extract which was recorded as 61.02±1.30 mg Gallic acid equivalent (GAE/g) and 81.72±0.61 QE/g, respectively. Compared to other extracts, ethyl acetate leaf extract was found to possess high antioxidant activity at p<0.05 level, with high percentage of inhibition at 100 μg/ml concentration (82.70%) toward hydrogen peroxide radical scavenging with IC50 value 62.94 μg/ml. Statistically, two-tailed Pearson’s correlation showed strong positive correlations between hydrogenperoxide radical scavenging activity and total phenolic contents (TPC) (r=1.000) at p<0.05 level.Conclusion: The results obtained in this study clearly signifies that the ethyl acetate leaf extract of C. coromandelicum has high content of total phenols which are correlated to its antioxidant activity and thus has the potential to use as a source of natural antioxidants and can be explored as a therapeutic agent in free radical induced diseases.Keywords: Canthium coromandelicum, In-vitro antioxidant activity, Ethyl acetate extract, Pearson’s correlation, 1, 1- diphenyl -2- picrylhydrazyl, Hydrogen peroxide, Nitric oxide, Superoxide.

Background Diabetes mellitus type II (DMT-2) is a widely spread metabolic disorder both in developed and developing countries. The role of oxidative stress is well established in DMT-2 pathogenesis. The synthetic drugs for DMT-2 are associated with serious side complications. Antioxidant and α-glucosidase inhibitory actions of phytochemicals from various plant species are considered as an alternative to synthetic drugs for DMT-2 management. The present study aimed to evaluate the antioxidant activity, α-glucosidase inhibitory potential and phytochemical profiling of Hyophorbe lagenicaulis. Methods The total phenolic and flavonoid contents, in vitro antioxidant activity (α, α-diphenyl-β-picrylhydrazyl (DPPH) free radical scavenging and phosphomolybdenum method) and α-glucosidase inhibition of ultrasonicated hydroethanolic H. lagenicaulis leaf extracts were determined spectrophotometrically. The results of DPPH assay and α-glucosidase inhibition were reported in terms of IC50 value. The phytochemical profiling was accomplished by UHPLC-Q-TOF/MS/MS technique. Results and Discussion Findings leaped 60% ethanolic extract as rich fraction regarding total phenolic and flavonoid contents. The 60% ethanolic fraction was a promising source of natural antioxidants and α-glucosidase inhibitory agents as indicated by anti-radical and enzyme inibitory activities. Kaempferol, rutin, hesperetin 5-O-glucoside, kaempferol-coumaroyl-glucoside, luteolin 3-glucoside, Isorhamnetin-3-O-rutinoside, trimethoxyflavone derivatives and citric acid were identified by UHPLC-Q-TOF-MS/MS. These compounds were believed to be responsible for the strong antioxidant and enzyme inhibitory activity of plant extracts. The extensive metabolite profiling of H. lagenicaulis was carried out the first time as never reported previously. The H. lagenicaulis might be an appropriate choice to manage diabetes mellitus in an alternate way. The findings may be further exploited extensively for toxicity evaluation to proceed with functional food development having antidiabetic attributes.

Cordia macleodii, an important medicinal plant traditionally used for the treatment of various disorders, including wound healing, jaundice, gastric ulcer, hepatoprotective activity and antivenom potential, thus the plant showed the high antioxidant properties. Successive extraction of leaf and barks powder with methanol and water was tested for quantitative determination of total phenolics, total flavonoids and various in vitro antioxidant activities. Phytochemical screening of crude plant extracts revealed the presence of sterols, alkaloid, flavonoid, phenolic, Reducing sugar, glycosides, tannins and saponins. All extracts showed the significant total phenolic and flavonoid contents and as well as radical scavenging and iron-chelating activities. Among all, methanol leaf extract was observed to be higher antioxidant activity than that of other extract. The methanol extracts showed greater antioxidant activity by DPPH scavenging the free radical with IC50 values of 7.63 ± 0.38 μg gm-1 along with scavenged free radical in a concentration depended manner, which is compare to the standard ascorbic acid.  Our results revealed that the presence of grater antioxidant activity of C. macleodii extract may be due to the high level of flavonoid and phenolic content in the plant. These results clearly indicated that C. macleodii is an effective natural antioxidant. Key words: Cordia macleodii, antioxidant, radical scavenging, phenolics, flavonoid, phytochemical.

This experiment aimed to evaluate the antioxidant and hepatoprotective effect of methanolic extract of Helianthus annuus (sunflower) seeds against paracetamol induced liver injury. Four groups of rats (n = 3) were used and administered orally once daily with H. annus methanolic extract (100, 300, and 500 mg/kg) for 7 days, followed by the induction of hepatotoxicity using acetaminophen. The blood and liver samples were subjected to liver function biochemical and lipid peroxidation assay. The extract was also subjected to in-vitro antioxidant study using the 2, 2-diphenyl-1-picrylhydrazyl radical scavenging assay. The total phenolic content and total flavonoid content were also evaluated. The extract was found to have a total flavonoids of 7.72 ± 2.3 mg/g quercetin equivalence and total phenolic content of 40.60244 ± 1.14 mg/g Gallic acid equivalence. H. annus methanolic extract exhibited a significant decrease (P < 0.0001) in Alanine aminotransferase, alkaline phosphatase, and aspartate aminotransferase enzyme levels. The extract exhibited antioxidant activity and contained high total phenol content. H. annus methanol extract was found to possess a potential hepatoprotective activity that could be partly attributed to its antioxidant activity and high phenolic content.
