A media consisting of isatin-Schiff bases and its inclusion complexes with ?-cyclodextrine was developed to maximize the production of antibiotics Hexaene H-85 and Azalomycine ? by Streptomyces hygroscopicus CH-7. The media with ?-cyclodextrine inclusion complex of isatin-3-thiosemicarbazone resulted in the maximum antibiotics concentration of 493 ?g/cm3 for Hexaene H-85 and 191 ?g/cm3 for Azalomycine B. The production of Hexaene H-85 and Azalomycine is higher when ?-cyclodextrine complex is added as a nitrogen source, comparing to pure isatin-Schiff base. The maximum concentration of hexaene H-85 in medium with inclusion complex of isatin-Schiff base is 1.4-2.3 times higher than the basal medium. The maximum production of Azalomycine is 2.1-3.4 times higher in media with inclusion complex of Schiff base. During the fermentation process, the nutrient media with ?-cyclodextrine inclusion complexes with isatin-Schiff bases affect the strain morphology, since it is in the form of compact pellets, which are formed from short and long, branched filaments

In the present work, four derivatives of salicylaldehyde (salicylaldehyde-hydrazone, phenylhydrazone, semicarbazone and thiosemicarbazone) were synthesized using both conventional (95% ethanol) and green (crude glycerol from biodiesel production) solvents. The obtained compounds were identified by elemental microanalysis, as well as FTIR, UV/Vis and 1H-NMR spectroscopic methods. Yields of 93?98% of the compounds in crude glycerol were achieved within 10?25 min. The derivatives of salicylaldehyde and crude glycerol were used as a nitrogen and carbon source, respectively, in the medium for antibiotic (hexaene H-85 and azalomycine B) production by Streptomyces hygroscopicus CH-7. The highest concentrations of hexaene H-85 and azalomycine B were achieved in the medium containing salicylaldehyde-thiosemicarbazone (198 and 69 ?g/cm3, respectively). Derivatives of salicylaldehyde also impacted the strain morphology. In the media with salicylaldehyde- phenylhydrazone and salicylaldehyde-thiosemicarbazone, S. hygroscopicus CH-7 grew like large dispersive pellets with long twisted filaments that produced the highest yield of the antibiotics.

Addition of pure methyl oleate to a Streptomyces hygroscopicus NRRL B-1865 culture improved polyether antibiotic production, particularly of the closely related nigericin and abierixin, more than did technical methyl oleate. Among the fatty acid constituents of technical methyl oleate, the methyl esters of stearate and particularly linoleate were detrimental to growth and antibiotic production, while the methyl esters of palmitate and palmitoleate had the same effect as technical methyl oleate. The presence of Tween 80 or cyclodextrin in S. hygroscopicus cultures supplemented with methyl oleate increased valine uptake and antibiotic production. Antibiotic production was highest with a mixture of 1% methyl oleate, 0.1% Tween 80, and 0.1% cyclodextrin. The results suggested that the stimulatory effect of methyl oleate might be enhanced by a substance increasing the accumulation of branched amino acids known to be direct precursors of polyether antibiotics. The rate of abierixin synthesis was more important than that of nigericin at the end of the culture. The nigericin/abierixin ratio decreased from 6 in basal medium supplemented with methyl oleate to 2 in basal medium supplemented with methyl oleate, Tween 80, and cyclodextrin. A strong decrease in culture pH seen in the presence of Tween 80 and cyclodextrin might lead to a Michael transformation of nigericin to abierixin.Key words: Streptomyces hygroscopicus, nigericin, abierixin, methyl oleate, Tween 80, cyclodextrin.

Influence of a modified media on Hexaene H-85 and Azalomycine B production by Streptomyces hygroscopicus CH-7 was investigated. The amino acid L-tryptophan, as a nitrogen source, was replaced with isatin-3-thiosemicarbazone and its complexes with some divalent metal ions. Isatin-3-thiosemicarbazone was synthesized in crude glycerol obtained as a byproduct in biodiesel production from sunflower oil. The complexes were charac-terized by elemental microanalysis and magnetic susceptibility, as well as, by Atomic absorption(AA), Fourier-transform infrared (FTIR) and Ultraviolet?visible (UV/VIS) spectro-scopic methods. The spectral studies indicated an octahedral geometry for the Mn(II), Fe(II) and Ni(II) complexes and a tetrahedral one for the Zn(II) complex. Comparing to the basal medium, isatin-3-thiosemicarbazone (ITC) and its metal complexes in the concentration of 0.5 g dm-3 showed better results in the antibiotics production. Use of medium supple-mented with the Fe(II) complex resulted in the maximum Hexaene H-85 and Azalomycine B concentrations of 306 ?g cm-3 and 127 ?g cm-3, respectively. Addition of ITC and its complexes changed the morphology of S. hygroscopicus CH-7 from filaments to pellets as a dominant shape in media resulting in higher antibiotic production.

Ezetimibe, the antihyperlipidemic drug of poor bioavailability was complexed with native and derivatized cyclodextrins.The complexes were characterized in terms stability, stoichiometry and structure using various 1D and 2D solution NMR spectroscopic techniques. The complexes were found to be of moderate stability (logK[3). The least stable inclusion complex is formed with b-cyclodextrin, while the ezetimibe-methylated-b--cyclodextrin has a 7-fold higher stability. The results can be useful to improve the poor water-solubility and the concomitant bioavailability of ezetimibe.

Abstract Background The aim of this study was to provide an information about the homogeneity on the level of enterokinase productivity in P. pastoris depending on different suppliers of the media components. Results In previous studies, we performed the optimisation process for the production of enterokinase by improving the fermentation process. Enterokinase is the ideal enzyme for removing fusion partners from target recombinant proteins. In this study, we focused our optimization efforts on the sources of cultivation media components. YPD media components were chosen as variables for these experiments. Several suppliers for particular components were combined and the optimisation procedure was performed in 24-well plates. Peptone had the highest impact on enterokinase production, where the difference between the best and worst results was threefold. The least effect on the production level was recorded for yeast extract with a 1.5 fold difference. The worst combination of media components had a activity of only 0.15 U/ml and the best combination had the activity of 0.88 U/ml, i.e., a 5.87 fold difference. A substantially higher impact on the production level of enterokinase was observed during fermentation in two selected media combinations, where the difference was almost 21-fold. Conclusions Results demonstrated in the present study show that the media components from different suppliers have high impact on enterokinase productivity and also provide the hypothesis that the optimization process should be multidimensional and for achieving best results it is important to perform massive process also in terms of the particular media component supplier .

Triazole ring system has attracted a continuously growing interest of synthetic organic chemists and those dealing with the medicinal compounds due to its versatile potential to interact with biological systems. Schiff bases are also considered as one of the most biologically active compounds.  The aim of the present study was to synthesize new Schiff bases bearing triazole nucleus and to assess their antimicrobial activities. Four new Schiff base derivatives of 1,2,4-triazole-3-thione were synthesized by combining two different pharmacophores viz. triazole nucleus and Schiff base moiety and were characterized by spectral techniques (UV, FT-IR, and NMR). The Schiff bases were evaluated for antibacterial (Staphylococcus aureus, Escherichia coli, and Klebsiella pneumoniae) and antifungal (Candida albicans) activities. The synthesized compounds exhibited good to moderate activities against different strains of bacteria and fungi tested.

Condensation of salicylaldehyde with enantiopure (R)-(1-aryl-ethyl)amines yields the enantiopure Schiff bases (R)-N-(1-aryl-ethyl)salicylaldimine (HSB*; aryl = phenyl, 2-methoxyphenyl, 3- methoxyphenyl, 4-methoxyphenyl (4), 4-bromophenyl (5), 2-naphthyl). These Schiff bases readily react with dinuclear (acetato)(η4-cycloocta-1,5-diene)rhodium(I), [Rh(μ-O2CMe)(η4-cod)]2, to afford the mononuclear complexes, cyclooctadiene-((R)-N-(1-aryl-ethyl)salicylaldiminato-κ2N,O)- rhodium(I), [Rh(SB∗)(η4-cod)] (SB* = deprotonated chiral Schiff base = salicylaldiminate; aryl = phenyl (7), 2-methoxyphenyl, 4-methoxyphenyl, 4-bromophenyl, 2-naphthyl). The complexes have been characterized by IR, UV/vis, 1H/13C NMR and mass spectrometry, optical rotation as well as by single-crystal X-ray structure determination for 4, 5 and 7. The structure of 5 shows C-Br· · ·π contacts. Compound 7 is only the second example of a Rh(η4-cod) complex with a six-membered Rh-N,O-chelate ring
