A total of 39 Fusarium graminearum, F. sporotrichioides, F. semitectum and F. equiseti isolates, originating from wheat and maize samples collected at 10 locations in Serbia, were analyzed by ELISA method for their potential of deoxynivalenol (DON) and zearalenone (ZEA) production under optimal laboratory conditions. Fusarium graminearum isolates with the highest intraspecies variability were the best producers of both deoxynivalenol and zearalenone. In contrast, F. equiseti isolates were the weakest producers of these two toxins. Considering the plant origin of the isolates, wheat-originating F. sporotrichioides isolates were better deoxynivalenol producers, while the maize-originating isolates produced more zearalenone. There was no clear difference in ZEA production between wheat- and maizeoriginating isolates of F. graminearum, while higher average DON concentrations were produced by F. graminearum wheat-originating isolates. Negative correlation was detected between the production of deoxynivalenol and zearalenone by various Fusarium spp.

Maize is an important worldwide commodity susceptible to fungal contamination in the field, at harvest, and during storage. This work aimed to determine the occurrence of Fusarium spp. in maize grains produced in the Tagus Valley region of Portugal and the levels of related mycotoxins in the 2018 harvest and during their storage for six months in barrels, mimicking silos conditions. Continuous monitoring of temperature, CO2, and relative humidity levels were done, as well as the concentration of mycotoxins were evaluated and correlated with the presence of Fusarium spp. F. verticillioides was identified as the predominant Fusarium species. Zearalenone, deoxynivalenol and toxin T2 were not found at harvest and after storage. Maize grains showed some variability in the levels of fumonisins (Fum B1 and Fum B2). At the harvest, fumonisin B1 ranged from 1297 to 2037 µg/kg, and fumonisin B2 ranged from 411 to 618 µg/kg. Fumonisins showed a tendency to increase (20 to 40%) during six months of storage. Although a correlation between the levels of fumonisins and the monitoring parameters was not established, CO2 levels may be used to predict fungal activity during storage. The composition of the fungal population during storage may predict the incidence of mycotoxins.

In in vitro conditions, virulence of 91 isolates of species Fusarium genus (F. oxysporum, F. solani, F. acuminatum, F. equiseti, F. arthrosporioides, F. proliferatum, F. avenaceum, F. semitectum, F. tricinctum, F. sporotrichioides and F. graminearum) towards alfalfa seedlings was investigated. Isolates of investigated species originated from diseased alfalfa plants collected on four locations in Serbia based on symptoms of wilting caused by fusarium and root rotting. Pathogenicity and virulence of investigated isolates of Fusarium spp. were determined by visual evaluation of inoculated seedlings of cultivars K28 in laboratory conditions. All isolated of investigated species had pathogenic effect on alfalfa seedlings, which expressed symptoms such as necrosis of root, moist rotting and "melting of seedlings". Colour of necrotic root tissue varied from light brown, brown, lipstick red to explicit black, depending on the Fusarium species. Strong virulence was established in 48 isolates, medium virulence in 31 and weak virulence in 12 isolates.

Fusarium graminearum Schwabe [teleomorph: Gibberella zeae Schwein. Petch] is the predominant Fusarium species pathogenic to wheat, barley and corn in Ontario and produces the mycotoxin deoxynivalenol (DON) in grain. The first objective of this study was to determine the Fusarium spp. and mycotoxin concentrations (DON, HT-2, and T-2) from commercial oat (Avena sativa L.) fields grown in the same area as wheat, corn and barley in Ontario. The second objective was to measure DON level across cultivars grown in the Ontario Performance Trial (OPT) after spray-inoculation with F. graminearum. Fusarium sporotrichioides, F. graminearum and F. poae were the predominant species identified in oat. The highest level of DON in commercial oat fields were 0.3, 0.1, 0.2 and 1.2 ppm in 2005, 2006, 2007 and 2008, respectively. The highest concentrations of HT-2, and T-2 in 2006 were 0.5 and 0.2 ppm, respectively, whereas the levels in 2007 were both under 0.2 ppm. The relatively low level of DON in commercial oat fields confirmed that oat is in general less affected with F. graminearum than other cereals. However, this study supports the importance of monitoring Fusarium spp. and concentrations of mycotoxins in oat in Ontario, especially during growing conditions, as experienced in 2008, which favored Fusarium infection and DON accumulation. Differences in percent of Fusarium spp. infected kernels and DON level (ppm) were identified amongst the cultivars in the OPT in naturally infected oat samples and after inoculation, respectively. Production and providing of high quality maize grain are of primary importance for livestock production since maize is the main component of livestock feed. Contamination with fungi not only diminishes the quality of grain, but some fungi species can produce highly toxic compounds known as mycotoxins. Considering that maize is economically the most important grown plant in Serbia, content and intensity of frequency of these pathogen fungi species are investigated in maize stored in the storage facilities. Based on studies of the fungi incidence in stored maize intended for nutrition of livestock, it was established that during the period from February 2005 to March 2006, some fungi species were present successively (Fusarium spp., Penicillium spp.), other with interruptions of one (Nigrospora spp., Aspergillus spp., Acremonium spp., Alternaria spp), three (Mucor spp., Chaetomium spp., Rhizopus spp.) to four months (Cladosporium spp.), and also sporadically during three (Epicoccum spp., Rhizopus spp., Trichothecium spp.), two (Gliocladium spp.) or one month (Papulaspora spp., Phialophora spp.). Period June-October is the most critical for preservation of the quality of stored maize since the number of fungi species present (8?14 of total of 17 identified fungi genera) and their frequency during this period are the highest, especially of toxigenic species of Fusarium (43.5?62.5%) and Penicillium (10.0?33.5%) genera. Positive correlation (r = 0.5979**) between dynamics of incidence and frequency of isolated fungi species indicates that fungi with higher frequency of incidence remain longer on the grain during storage. F. verticillioides and F. subglutinans, of total of 6 identified Fusarium species were present on the grain during entire year and with the highest frequency (24.7% and 5.9%, respectively). During single year (2005) F. verticillioides was present in the highest percentage from February to October (22.0?39.5%), and F. subglutinans from April to October (8.0?12.5%), whereas the both species were isolated the least during winter period December-January (4.0?8.0% and 0.5?1.0%, respectively). F. graminearaum is the third toxigenic Fusarium species which from the mycotoxicological aspect can be important for period June-September when it is mostly present (5.0?11.0%). Incidence of other Fusarium species is sporadic (1.3% F. proliferatum, 1.0% F. sambucinum and 0.5% F. poae).

Fusarium species coexist as toxigenic, systemic pathogens in sweet corn seed production in southwestern Idaho, USA. We hypothesized that fungal antagonists of seedborne Fusarium would differentially alter production of Fusarium mycotoxins directly and/or systemically. We challenged the Fusarium complex by in vitro antagonism trials and in situ silk and seed inoculations with fungal antagonists. Fungal antagonists reduced growth and sporulation of Fusarium species in vitro from 40.5% to as much as 100%. Pichia membranifaciens and Penicillium griseolum reduced fumonisin production by F. verticillioides by 73% and 49%, respectively, while P. membranifaciens and a novel Penicillium sp. (WPT) reduced fumonisins by F. proliferatum 56% and 78%, respectively. In situ, pre-planting inoculation of seeds with Penicillium WPT systemically increased fumonisins in the resulting crop. Morchella snyderi applied to silks of an F1 cross systemically reduced deoxynivalenol by 47% in mature seeds of the F2. Antagonists failed to suppress Fusarium in mature kernels following silk inoculations, although the ratio of F. verticillioides to total Fusarium double with some inoculants. Fusarium mycotoxin concentrations in sweet corn seed change systemically, as well as locally, in response to the presence of fungal antagonists, although in Fusarium presence in situ was not changed.

Fusarium species are probably the most prevalent toxin-producing fungi of the northern temperate regions and are commonly found on cereals grown in the temperate regions of America, Europe and Asia. Among the toxins formed by Fusarium we find trichothecenes of the A-type or B-type, zearalenone, fumonisins or nivalenol. The current exposure assessment consists of the qualitative and/or quantitative evaluation based on the knowledge of the mycotoxin occurrence in the food and the dietary habits of the population. This process permits quantifying the mycotoxin dietary intake through deterministic or probabilistic methods. Although these methods are suitable to assess the exposure of populations to contaminants and to identify risk groups, they are not recommended to evaluate the individual exposition, due to a low accuracy and sensitivity. On the contrary, the use of biochemical indicators has been proposed as a suitable method to assess individual exposure to contaminants. In this work, several techniques to biomonitor the exposure to fumonisins, deoxynivalenol, zearalenone or T-2 toxin have been reviewed.

The fungal pathogen Fusarium circinatum is the causal agent of Pine Pitch Canker (PPC), a disease which seriously affects different species of pine in forests and nurseries worldwide. In Europe, the fungus affects pines in northern Spain and Portugal, and it has also been detected in France and Italy. Here, we report the findings of the first trial investigating the susceptibility of Polish provenances of Scots pine, Pinus sylvestris L., to infection by F. circinatum. In a greenhouse experiment, 16 Polish provenances of Scots pine were artificially inoculated with F. circinatum and with six other Fusarium species known to infect pine seedlings in nurseries. All pines proved highly susceptible to PPC and displayed different levels of susceptibility to the other Fusarium spp. tested. The findings obtained indicate the potentially strong threat of establishment of an invasive pathogen such as F. circinatum following unintentional introduction into Poland.

O trabalho teve como objetivos, identificar e quantificar os fungos associados a sementes de azevém, comparar a incidência em diferentes meios de cultura, e determinar o número de escleródios de Claviceps purpurea presentes em amostras de sementes. Foram analisadas 37 amostras de sementes de azevém provenientes de municípios do Rio Grande do Sul, Santa Catarina e Paraná. As sementes foram plaqueadas em três meios de cultura: BDA, semi-seletivo de Reis e semi-seletivo de Segalin & Reis, analisando-se a incidência dos fungos. Para detecção de C. purpurea, foram pesados 100g de sementes por amostra e, através de exame visual, foi determinado o número de escleródios. Os fungos detectados foram Alternaria alternata, Bipolaris sorokiniana, Drechslera spp., D. siccans, Fusarium graminearum, Fusarium spp., Aspergillus spp. e Penicillium sp. A incidência de A. alternata variou de 0,0% a 33,7% e freqüência de 89,2% nas amostras analisadas. Para B. sorokiniana a incidência foi de 0,0% a 2,2% e frequência de 62,2%, Drechslera spp., apresentou incidência de 0,0% a 40,3% e frequência de 78,4%. D. siccans a incidência foi de 0,1% a 20,0% e frequência de 100%.Para Fusarium spp., e F. graminearum a incidência foi de 0,0% a 31,0% e 0,0% a 11,3% e frequência de 81,1% e 64,9%, de 0,0% a 43,7% de incidência e 94,6% de frequência para Aspergillus spp. e Penicillium sp. com incidência entre 0,0% a 51,7% e frequência de 91,9%, respectivamente. O fungo C. purpurea foi encontrado em 81,1% das amostras em estudo.

A total of 41 species of fungi were isolated from seed samples of barley, maize, soybean, and sunflower collected at different locations in Serbia. The majority of detected species occurred on barley (35 of 41 species or 87.8%) comparing to soybean (17 of 41 species or 41.5%), sunflower (16 of 41 species or 39.0%) and maize (15 of 41 species or 36.9%). Species belonging to genera Alternaria, Chaetomium, Epicoccum, Fusarium, Penicillium and Rhizopus were present on seeds of all four plant species. Alternaria species were dominant on soybean, barley and sunflower seeds (85.7%, 84.7% and 76.9%). F. verticillioides and Penicillium spp. were mainly isolated from maize seeds (100 and 92.3% respectively), while other species were isolated up to 38.5% (Chaetomium spp. and Rhizopus spp.). F. graminearum, F. proliferatum, F. poae and F. sporotrichioides were the most common Fusarium species isolated from barley (51.1-93.3%), while on the soybean seeds F. oxysporum (71.4%), F. semitectum (57.1%) and F. sporotrichioides (57.1%) were prevalent. Frequency of Fusarium species on sunflower seeds varied from 7% (F. equiseti, F. graminearum, F. proliferatum and F. subglutinans) to 15.4% (F. verticillioides). Statistically significant negative correlation (r = -0.678
