The work is devoted to modeling the growth of optically dense microalgae cultures in natural light. The basic model is based on the idea of the two-stage photoautotrophic growth of microalgae. It is shown that the increase in the intensity of sunlight in the first half of the day can be described by a linear equation. Analytical equations for the growth of biomass of microalgae and its macromolecular components are obtained. As the initial conditions, it is assumed that at the time of sunrise, the concentration of reserve biomass compounds is zero. The simulation results show that after sunrise, the growth of the microalgae culture is due only to an increase in the reserve part of the biomass, while the structural part practically does not change over six hours. Changes in the ratio of the reserve and structural parts of the biomass indicate a change in the biochemical composition of cells.

An apparatus is described for the continuous growth of bacteria. Brucella abortus S.19 has been grown in continuous culture for periods up to 3 weeks with populations up to 2 × 1011viable cells per ml and without the establishment of nonsmooth variants.Concentrations between 3 × 109and 2 × 1011cells per ml could be maintained as a function of the dilution rate without the requirement of a known limiting factor in the medium. In a series of steady-state conditions, the specific growth rate increased steadily up to 0.28 hour−1with decreasing population levels.Incidence of mutants was governed by the dilution rate and could also be reduced by various chelating substances.In continuous growth combined with continuous dialysis, population levels were approximately twice those obtained in continuous growth without dialysis. The effect of dialysis appears to be the continuous removal of growth-limiting metabolic products.

AbstractConventional microalgae oil extraction applies physicochemical destruction of dry cell biomass prior to transesterification process to produce fatty acid methyl esters (FAMEs). This report presents a simple and rapid direct transesterification (DT) method for FAMEs production and fatty acid profiling of microalgae using freshly harvested biomass. Results revealed that the FAMEs recovered from Chlorella vulgaris were 50.1 and 68.3 mg with conventional oil-extraction-transesterification (OET) and DT method, respectively. While for Messastrum gracile, the FAMEs recovered, were 49.9 and 76.3 mg, respectively with OET and DT methods. This demonstrated that the DT method increased FAMEs recovery by 36.4% and 53.0% from C. vulgaris and M. gracile, respectively, as compared to OET method. Additionally, the DT method recovered a significantly higher amount of palmitic (C16:0) and stearic (C18:0) acids from both species, which indicated the important role of these fatty acids in the membranes of cells and organelles. The DT method performed very well using a small volume (5 mL) of fresh biomass coupled with a shorter reaction time (~ 15 min), thus making real-time monitoring of FAMEs and fatty acid accumulation in microalgae culture feasible.

SUMMARYThe effects of an altered rumen dilution rate (D) upon the molar proportions of volatile fatty acids (VFA) in rumen liquor, VFA production rate, microbial protein synthesis and carbohydrate digestion within the rumen were studied using adult wether sheep.Dilution rate and VFA proportions were unaltered by the infusion of up to 121 water/day into the rumen of sheep fed dried grass and concentrate (9:1). There was a small but significant (P< 0·05) increase in the rumen volume when the infusion rate was increased from 8 to 12 1/day.The intraruminal infusion of artificial saliva (41/day), or artificial saliva containing 4% or 8% w/v polyethylene glycol (PEG) caused a significant increase in D with an associated decline in the molar proportion of propionate (Pr) in the rumen liquor. A similar effect was obtained with the intraruminal infusion of 2·5% w/v sodium bicarbonate. The overall regression of Pr on D was highly significant: Pr = 32·5–82·1D;r= –0·99, P < 0·001.A diet of flaked maize: dried grass (6:4) was offered to three sheep each fitted with a rumen cannula and with a re-entrant cannula at the proximal duodenum. The intraruminal infusion (4 1/day) of artificial saliva containing 4% w/v PEG caused a significant (P< 0·01) increase in D and a significant (P< 0·01) depression in Pr in two animals. The dilution rate and Pr in the third animal were virtually unaltered by infusion. The regression of Pr on D for the three animals was highly significant: Pr = 34·8–136·8D; r = –0·98, P < 0·001. Each increase in D was associated with an increased flow of α-linked glucose polymer, total amino acids and total microbial amino acids into the small intestine and with an increased efficiency of microbial protein synthesis within the rumen.

Abstract The complex magnetic susceptibility χ(f) = χ′(f) - i χ″(f), of a ferrofluid sample with magnetite particles dispersed in kerosene and stabilized with oleic acid, over the range 0.1 GHz to 6 GHz, was determined. The initial sample has been successively diluted with kerosene (with a dilution rate of 2/3), thus obtaining further three samples. Using the complex magnetic susceptibility measurements of each sample, the frequency field and particle concentration dependencies of the heating rate of the ferrofluid samples, were analyzed. The results show the possibility of using the heating rate of ferrofluid samples with different particle concentrations, in hyperthermia applications.
