Red clover (Trifolium pratense L.) is the predominant legume used in northern European agriculture. Official red clover variety trials are conducted by Natural Resources Institute Finland (Luke) to determine the value of field crop varieties. The trials used for the current analysis were conducted in Luke units in southern Finland (Mikkeli) and northern Finland (Ruukki) in two consecutive years. Plant samples for isoflavone analyses were collected from four varieties grown as four replicates and harvested twice during both growing seasons. The four main isoflavones biochanin A, genistein, daidzein and formononetin were analysed using high performance liquid chromatography. Total phytoestrogen content in the varieties varied in the range of 11.2−14.8 mg g-1 dry matter (DM). The variety and the time of harvest had most effect on the isoflavone, especially formononetin, contents of red clover. A more northern growing area and challenging weather conditions were associated with increased isoflavone concentrations.

В данной статье представлены результаты сравнительной оценки 11 сортов клевера лугового и одного образца-дикороса, проведённой в Пермском НИИ сельского хозяйства в 2018–2019 годах. Все селекционные сорта сформировали полноценные первый и второй укосы или отаву в оба года использования. Дикорастущая форма формировала только один укос, демонстрируя особенность позднеспелого одноукосного типа. Сорта Кретуновский и Ранний 2 показали себя как типичные раннеспелые двухукосные. Другие сорта занимали промежуточное положение. Наибольший сбор сухого вещества обеспечивал образец-дикорос в оба года при крайне различных погодных условиях (1,266 и 1,259 кг/м2 соответственно). Дикорастущая форма и сорт Ранний 2 обеспечивали стабильные урожаи зелёной массы и сухого вещества независимо от возраста и погодных условий. Образец дикорастущего клевера имел более однородный фенотип по сравнению с сортом Пермский местный — стандартом для позднеспелого типа, что указывает на большую генетическую однородность. Согласно всесторонней оценке, включающей степень перезимовки, урожайность зелёной и сухой массы, характер цветения, созревания, формирования и конечный урожай семян, образец-дикорос имеет отличительные признаки позднеспелого (одноукосного) типа и может использоваться в качестве исходного генетического материала для дальнейшей селекции позднеспелых одноукосных сортов клевера лугового, характеризующихся экономически ценными свойствами: высокой устойчивостью к неблагоприятным условиям окружающей среды, высокой и стабильной урожайностью зелёной массы и семян. Исследования будут продолжены с оценкой ряда образцов дикорастущих форм клевера лугового, обнаруженных в 2019 году в природных биоценозах Пермского края. This article deals with the competitive trial of 11 varieties and 1 wild genotype of red clover conducted at the Research Institute of Agriculture in 2018–2019. All the varieties provided good yields of green mass both in the first and second cuts. Wild genotype provided only one cut as long-season one-cut type. “Kretunovskiy” and “Ranniy 2” performed as typical short-season two-cut varieties. Wild genotype produced the highest yield of dry matter (DM) under contrast weather conditions (1.266 and 1.259 kg/m2, respectively). Wild genotype and “Ranniy 2” formed stable yields of green mass and DM regardless of time period and weather. Wild genotype showed homogeneous phenotype compared to “Permskiy mestnyy” — long-season standard variety indicating high genetic uniformity. Such traits as winter hardiness, green and dry mass yields, flowering pattern, seed maturation, formation and production were evaluated. According to this evaluation wild genotype performed as long-season (one-cut) type showing high resistance to unfavorable conditions, high and stable yield of green mass and seeds. Therefore, it can be used a source of economically important traits for future breeding programs of red clover. Further investigations will focus on the evaluation of wild genotypes of red clover found in the Perm region in 2019.

Red clover (Trifolium pratense L.) is one of the most important temperate legume species, used to lessen the lack of forage during the critical fall-winter period in Rio Grande do Sul, RS, Brazil. However, its utilization has been restricted mainly because of its lack of persistence. This work evaluates the dry matter yield and persistence of red clover half-sib progenies in two physiographic regions of RS: "Depressão Central", in Eldorado do Sul and "Encosta Superior do Nordeste", in Veranópolis. Experiments were carried out for two growing seasons and results were compared to two red clover commercial checks, Qüiñequeli and Estanzuela 116. The region of Veranópolis was more adequate for red clover forage production, enabling better yield and persistence. The best check for both locations was cultivar Estanzuela 116. At Eldorado do Sul persistence was highly affected and some progenies were superior to the best check. At Veranópolis the best check was very productive, with good persistence. The best progenies at both locations were selected to be propagated and submitted to additional recurrent selection cycles.

Abstract Background Extracts of red clover (Trifolium pratense L.) containing estrogenic and pro-estrogenic isoflavones are used in dietary supplements primarily for the management of menopausal symptoms in women. Objective A UHPLC-MS/MS assay was developed and validated for the quantitative analysis of the six major red clover isoflavones in dietary supplements and in human serum in support of clinical trials. Methods Enzymatic deconjugation of isoflavone glucuronides and sulfate conjugates in human serum specimens was carried out followed by protein precipitation. Isoflavones in red clover dietary supplements were acid hydrolyzed to release aglycons from glycosides. UHPLC separations (< 4 min) were combined with MS/MS using collision-induced dissociation, selective reaction monitoring and deuterated internal standards to measure biochanin A, formononetin, daidzein, genistein, irilone, and prunetin. Results The method was validated with respect to selectivity, specificity, accuracy, linearity, precision, LOD, and LOQ. The calibration curves for all analytes were linear (R2 > 0.998). The mean recovery for low-, medium- and high-quality control standards ranged between 80% and 108%. The precision of the method was assessed using coefficients of variation, which were <15%. Conclusions The UHPLC-MS/MS method is fast, precise, sensitive, selective, accurate, and applicable to the quantitative analysis of red clover isoflavones in different matrices. Highlights This validated UHPLC-MS/MS assay is applicable to the rapid quantitative analysis of red clover isoflavones in human serum and in dietary supplements.

Background. Identification of crop varieties is presently one of the most important aspects due a significant annual increase in the number of newly developed cultivars. Application of molecular markers makes it possible to identify cultivars and secure protection of plant breeders’ rights. Marker techniques based on SSR loci and PawS markers were evaluated for their efficiency in revealing the DNA polymorphism in Russian red clover cultivars, and the research results are presented in this publication.Materials and Methods. The total genome DNA was extracted by a modified SDS method from 30 seedlings per each cultivar. Nine simple sequence repeats (SSR) and 4 PawS markers were used for genotyping. The basic genetic diversity parameters were measured and analyzed using the software resources GelAnalyzer 2010а, MStools v.3, and Statistica 7.0.Results and conclusion. The mean level of intervarietal DNA polymorphism in red clover was 38.6%. Cultivar-specific amplicons were obtained for 4 accessions (cvs. ‘Trifon’, ‘Topaz’, ‘Trio’ and ‘Mars’) with SSR loci RCS1307 and RCS3095. These loci were found appropriate for identification and certification of such cultivars. The tested PawS markers (individually and in combinations) proved non-informative for the analysis of intervarietal DNA polymorphism in red clover. The only primer pair PawS5+PawS16 generated reproducible PCR products, but unique amplicons were absent in the DNA profiles. The data obtained in this study may be helpful for further identification and certification of Russian red clover cultivars and promising breeding materials.

A collection of subterranean clover (Trifolium subterraneum L.) genotypes from Sardinia was evaluated for the content of oestrogenic isoflavones to assess differences in total and individual compounds. Daidzein, genistein, and their respective 4′-methoxy derivatives biochanin A and formononetin, were detected in leaves and quantified by GC/FID and GC/MS methods. A large variation among genotypes for individual compounds and for the total isoflavone concentration was observed. Total isoflavones were measured in the range 0.8-13.4 mg/g fresh weight (0.3-4.5% dry matter). Daidzein was usually present in lower amount compared to the other isoflavones. Genistein and biochanin A were found in most cases in higher concentration than formononetin. Phyto-oestrogens have been shown to have both oestrogenic and anti-oestrogenic properties, and their effects entail a positive role for the human health. T. subterraneum revealed the presence of higher concentration of isoflavones compared to other sources of these compounds, e.g. soybean (Glycine max (L.) Merrill) and red clover (Trifolium pratense L.). This study provided information on the biodiversity of the natural strains from Sardinia, emphasizing the importance of these genetic resources for their variation in the amount of total and individual isoflavones.

Isoflavones, known for their health benefits, are abundant in red clover (Trifolium pratense L.). Total isoflavone concentrations can be 30 times that of soybean, indicating that red clover is a good source of nutraceutical and functional food ingredients. In this study, tissue samples of 13 red clover cultivars were taken at two growth stages (late-bud stage and late-flowering stage) to determine the concentration of individual isoflavones using HPLC. Individual isoflavone concentrations and total isoflavone concentration differed significantly according to red clover cultivar. We found significant genetic variability for total isoflavone concentration and individual isoflavone concentrations; these differences were not related to ploidy level (diploid vs. tetraploid). Broad-sense heritability (H = genetic variance/total variance) ranged from 0 to 83% and was influenced by isoflavone type and sampling date. The results of this study suggest that there is significant genetic variability for isoflavone concentrations among currently available red clover cultivars. Six Trifolium pratense L. cultivars, five diploid and one tetraploid, were grown in the lowland region (123 m altitude) and the mountain region (650 m altitude). Dry matter (d.m.) and green mass (g.m.) yield, stem height and leaf proportion were assessed from each of the three growing seasons. Chemical composition was assessed from the average samples of all cuts in the second year of the experiment. Greater green mass (54.14 t/ha) and dry matter yield (9.86 t/ha), stem height (0.61 m), crude protein (157.6 g/kg), crude fiber (222.9 g/kg), crude ash (100.68 g/kg) and crude fat (30.09 g/kg) content were observed in the mountain region, compared to the lowland region (45.61 t/ha g.m. 8.92 t/ha d.m. 0.59 stem height, 156.38 g/kg crude protein, 216.6 g/kg crude fiber, 94.85 g/kg crude ash, 24.98 g/kg crude fat). The greater leaf proportion (47.2%) and nitrogen free extract content (420.13 g/kg) were observed in the lowland region compared to the mountain region (42.95% and 402.99 g/kg, respectively). A significant location × cultivar interaction was found for all investigated parameters except for leaf proportion.

Red clover consists of the overground parts and inflorescence of Trifolium pratense L., a leguminous plant belonging to the genus Trifolium. It is widely distributed worldwide and has long been used in traditional medicine. In this study, a combination approach using UPLC-MS and network pharmacology was applied to explore the quality control markers for the quality assessments of red clover. Firstly, UPLC-MS was used to identify the compounds in different parts of red clover. Twenty-eight compounds were totally identified. According to the traditional clinical efficacy of red clover, a compound-target-function network was constructed by network pharmacology to discover the main active compounds based on the identified compounds. Nine compounds of chlorogenic acid, daidzin, calycosin-7-O-β-d-glucoside, genistin, ononin, daidzein, genistein, formononetin, and biochanin A were filtrated and further confirmed in rat plasma in view of the blood-absorbed components taking effects. Finally, a novel method for simultaneously detecting the nine quality control markers was developed by UPLC-QQQ-MS in an effort to assess the quality of red clover. For all samples, the average contents of the nine compounds measured from high to low consist of formononetin, ononin, biochanin A, genistin, daidzin, calycosin-7-O-β-d-glucoside, genistein, daidzein, and chlorogenic acid. The samples from Gansu province showed the best quality in the three producing areas This study provides new strategies to explore the quality control markers and develops a novel method for the quality assessment of red clover.
