Relationships among macro-minerals, other selected serum markers of bone profile and milk components of dairy cows during late-lactation

AbstractTesting blood and milk parameters as well as analysing the relationships among these markers is very useful for monitoring the internal homeostasis and health in high-yielding dairy cows during various production periods. The aim of the study was to assess the correlations (relationships) among macro-minerals, such as calcium (Ca), inorganic phosphorus (P), magnesium (Mg), other selected bone profile markers, such as total protein (TP), albumin, activity of alkaline phosphatase (ALP) measured in serum and selected milk components such as number of somatic cells (SCC), colony-forming units (CFU), milk fat (MF), milk protein (MP), milk lactose (ML), dry matter (DM), non-fat dry matter (FDM) and milk production in late-lactation cows. Both blood and milk samples were collected from 11 clinically healthy milking cows during the late-lactation period. The cows were examined once a day for 3 consecutive days resulting in 33 sets of blood and milk samples for laboratory and statistical analysis. Significant correlations were observed between: Mg and MP, Mg and FDM, ALP and SCC, TP and SCC, TP and MP, TP and FDM, albumin and MP, albumin and FDM, P and Mg, Mg and albumin, and between TP and albumin. When monitoring macro-mineral homeostasis and mammary gland health, especially in intensively fed high-yielding dairy cows correlations between these markers should be considered. The revealed correlations can allow for deeper comparative laboratory diagnostics of homeostasis and can be especially useful for laboratory monitoring of the potential risk of subclinical macro-mineral deficiency in high-yielding dairy cows.

Abstract The objective of this study was to examine the effect of feeding cranberry vine (CV) on milk components during early lactation as part of a larger study on the antiparasitic efficacy of CV supplementation on ewes during the periparturient period. Ewes were fed a 50% CV pellet (CVP; n = 12) or a control pellet (CON; n = 13) beginning at 102±1 d of pregnancy until d 65±1 postpartum. The CV pellet fed was formulated to be equivalent in digestible dry matter to the control pellet. BW was determined weekly during the study and milk samples were collected weekly during lactation. Data were analyzed in SAS with repeated measures. Ewe BW were similar at the start of study (180.36lbs ± 4.70lbs; P = 0.43) however by wk 8 postpartum CVP ewes weighed less than CON ewes (CON: 212.31lbs±7.86lbs; CVP: 185.58lbs±8.00lbs; P = 0.04). There was a treatment*wk effect observed for milk fat, protein and MUN (P ≤ 0.05). Ewes fed CVP exhibited reduced milk fat at wks 2 (CON: 6.75%±0.63%; CVP: 6.06%±0.58%; P = 0.03) and 5 (CON: 6.66%±0.37%; CVP: 5.54%±0.26%; P = 0.05), milk protein was reduced in CVP ewes at wk 2 (CON: 4.92%±0.12%; CVP: 4.72%±0.19%; P = 0.05) and MUN was greater at weeks 4 (CON: 22.89±0.7mg/dL; CVP: 27.93±0.85mg/dL; P = 0.01) and 8 (CON: 24.80±0.51mg/dL; CVP: 26.64±0.86mg/dL; P < 0.01) in CVP ewes. Somatic cell analysis is pending. In conclusion, CVP supplementation during lactation affects milk composition in sheep. Studies are underway to determine the effect of CV supplementation on additional metabolic parameters in pregnant and lactating ewes.

Abstract Alfalfa is a commonly grown forage in the Intermountain west and is often included in lactating dairy cow rations. This study investigated the effects of including novel alfalfa products: ProLEAF MAX™ (PLM), an alfalfa leaf pellet; and ProFiber Plus™ (PFP), alfalfa stems, in the diet of lactating dairy cows on dry matter intake, milk yield, and milk components. Holstein cows were housed in a freestall barn and milked in a free-flow automatic milking system. All cows were fed each treatment for 21-days, then switched to the next treatment utilizing a crossover experimental design. The five treatments were: control (CON, typical diet including alfalfa hay; n = 65); low-quality alfalfa hay (LQ+PLM, a diet that replaced alfalfa hay with low-quality alfalfa hay and PLM; n = 62); PLM+PFP (a diet that replaced alfalfa hay with PLM and PFP; n = 65); PLM (a diet that replaced alfalfa hay with PLM; n = 62); and PFP (a diet that replaced alfalfa hay with PFP; n = 60). Cows were group fed a partial mixed ration balanced for 40.8 kg milk, 3.9% milk fat and 3.3% milk protein. Individual milk yield and milk components were recorded daily by the automatic milking system. Dry matter intake was also recorded daily. When fed the PFP diet, cows had decreased (P < 0.01) dry matter intake compared to the other diets. Milk yield was increased (P < 0.01) when cows received the PLM diet when compared to the other diets. When fed the PFP and PLM+PFP diets, milk fat was increased (P < 0.01) when compared to the other diets. Milk protein was decreased (P < 0.01) when cows were fed the PFP diet when compared to the other diets. These data indicate that inclusion of fractionated alfalfa products in the diet of lactating dairy cows has the potential to increase milk yield and milk components.

Abstract The objective of the study was to evaluate the potential of Fourier transform infrared spectroscopy (FTIR) analysis of milk samples to predict body energy status and related traits (energy balance (EB), dry matter intake (DMI) and efficient energy intake (EEI)) in lactating dairy cows. The data included 2371 milk samples from 63 Norwegian Red dairy cows collected during the first 105 days in milk (DIM). To predict the body energy status traits, calibration models were developed using Partial Least Squares Regression (PLSR). Calibration models were established using split-sample (leave-one cow-out) cross-validation approach and validated using an external test set. The PLSR method was implemented using just the FTIR spectra or using the FTIR together with milk yield (MY) or concentrate intake (CONCTR) as predictors of traits. Analyses were conducted for the entire first 105 DIM and separately for the two lactation periods: 5 ≤ DIM ≤ 55 and 55 < DIM ≤ 105. To test the models, an external validation using an independent test set was performed. Predictions depending on the parity (1st, 2nd and 3rd-to 6th parities) in early lactation were also investigated. Accuracy of prediction (r) for both cross-validation and external test set was defined as the correlation between the predicted and observed values for body energy status traits. Analyzing FTIR in combination with MY by PLSR, resulted in relatively high r-values to estimate EB (r = 0.63), DMI (r = 0.83), EEI (r = 0.84) using an external validation. Only moderate correlations between FTIR spectra and traits like EB, EEI and dry matter intake (DMI) have so far been published. Our hypothesis was that improvements in the FTIR predictions of EB, EEI and DMI can be obtained by (1) stratification into different stages of lactations and different parities, or (2) by adding additional information on milking and feeding traits. Stratification of the lactation stages improved predictions compared with the analyses including all data 5 ≤ DIM ≤105. The accuracy was improved if additional data (MY or CONCTR) were included in the prediction model. Furthermore, stratification into parity groups, improved the predictions of body energy status. Our results show that FTIR spectral data combined with MY or CONCTR can be used to obtain improved estimation of body energy status compared to only using the FTIR spectra in Norwegian Red dairy cattle. The best prediction results were achieved using FTIR spectra together with MY for early lactation. The results obtained in the study suggest that the modeling approach used in this paper can be considered as a viable method for predicting an individual cow's energy status.

The aim was to determine the effect of the herbage allowance (HA) and supplement type (ST) on dry matter intake (DMI), milk production and composition, grazing behavior, rumen function, and blood metabolites of grazing dairy cows in the spring season. Experiment I: 64 Holstein Friesian dairy cows were distributed in a factorial design that tested two levels of daily HA (20 and 30 kg of dry matter (DM) per cow) and two ST (high moisture maize (HMM) and cracked wheat (CW)) distributed in two daily rations (3.5 kg DM/cow/day). Experiment II: four mid-lactation rumen cannulated cows, supplemented with either HMM or CW and managed with the two HAs, were distributed in a Latin square design of 4 × 4, for four 14-d periods to assess ruminal fermentation parameters. HA had no effect on milk production (averaging 23.6 kg/day) or milk fat and protein production (823 g/day and 800 g/day, respectively). Cows supplemented with CW had greater protein concentration (+1.2 g/kg). Herbage DMI averaged 14.17 kg DM/cow.day and total DMI averaged 17.67 kg DM/cow.day and did not differ between treatments. Grazing behavior activities (grazing, rumination, and idling times) and body condition score (BCS) were not affected by HA or ST. Milk and plasma urea concentration increased under the high HA (+0.68 mmol/L and +0.90 mmol/L, respectively). Cows supplemented with HMM had lower milk and plasma urea concentrations (0.72 mmol/L and 0.76 mmol/L less, respectively) and tended (p = 0.054) to have higher plasma β-hydroxybutyrate. Ruminal parameters did not differ between treatments.

ABSTRACTAn experiment was carried out to investigate the effects of compound composition and silage characteristics on silage intake, feeding behaviour and productive performance of dairy cows during the first 25 weeks of lactation. Over a period of 3 years, 36 lactating British Friesian cows (12 per year), in their third or later lactations, were divided into two groups. The cows in each group received either compound S, in which the principal energy source was cereal starch, or compound F containing a mixture of high quality digestible fibre. Cows given compound F ate 2·2 kg more silage dry matter per day (P < 0·05). The type of compound had no effect on the frequency of silage feeding and the time spent eating was significantly different only over weeks 10 to 25 of lactation (P < 0·05), with cows on compound F spending on average 20 min longer feeding per day. Cows on compound F produced 1·7 kg more milk per day than cows on compound S. Although there were no significant differences in the concentration of milk constituents, compound F was associated with higher yields of milk constituents. Significant differences were found between the years of experiment in the frequency of feeding (P < 0·05) and also in the concentration of milk protein and milk fat. There were no significant differences in the magnitude of live-weight change between treatments or year of experiment.

In a scientific economic experiment conducted in the experimental farm «Klenovo-Chegodaevo» (Moscow) on three groups of Holsteinized black-and-white cows with a milk yield of 7000 kg of milk per lactation, 10 heads in each, it was found that the inclusion in the diet of cows of the experimental groups tested protected fats (hydrogenated and fractionated) in the amount of 300 g per head per day, contributed to the tendency for more intensive metabolic processes in the rumen due to a slight increase in the concentration of volatile fatty acids by 5,6–7,4% and an increase in the mass of microorganisms in the contents of the rumen by 5,4–14,4% (P≥0,05). At the same time, an increase in the concentration of metabolic energy in the dry matter of the cows ration from 10,7 to 11,0 MJ / kg in the period from 21 to 120 days of lactation, due to the inclusion of protected fats in the diet of cows from the experimental groups, contributed to an increase in milk yield 4% fat content for 100 days of the experiment by 9,7% and 11,0% (P≤0,05), compared with the control, as well as the production of milk fat and protein, respectively by 9,6–11,0% (P≤0,05 in the second case) and 7,4–8,3%, feed costs expressed in ME decreased by 4,9–5,2%.

Abstract The aim of the study was to carry out retrospective and prospective comparative analyses of the pharmacokinetics of CEF after single intramammary (IMM) administration in cows. The prospective study (study A) was conducted on 9 dairy cows of the Polish Black-White race with clinical mastitis during the lactation period. Milk samples were collected at 2, 4, 6, 8, 10, 24, 36, 48, 72 and 84 h after single IMM administration of 250 mg of CEF to one quarter. Drug concentrations in milk samples were determined by HPLC-MS/MS technique and the results of the pharmacokinetic analysis were compared to those obtained in previous studies based on the microbiological (study B) and HPLC-UV methods (study C and D). Pharmacokinetic parameters were calculated based on adapted two-compartment model of drug distribution. One of the findings of the comparison of the analysed investigations is that the CEF kinetics determined with the microbiological method is consistent with the results obtained by the authors of this paper. Both studies yielded similar results of the key pharmacokinetic parameters related to the level of the drug distribution to tissues and elimination half-life. In the pharmacodynamic analysis, the observations in all four studies were entirely consistent and have shown lower values of T>MIC90 in healthy animals and significantly higher values in infected dairy cows. The comparison of studies A, B, C, and D revealed that the time of complete CEF wash-out of 90.90% varied and amounted to 5.7, 8.0, 2.2, and 2.2 days after administration of the drug, respectively. It was confirmed that not only the type of the analytical method but also correct sampling have a significant impact on determination of the correct value of the drug half-life after IMM administration. The comparative analysis of studies in which the milk yield was high and low allows a conclusion that this parameter in the case of CEF has no significant effect on T>MIC90.

Dietary yeast supplementation may improve the digestive efficiency of ruminants, but responses depend on the yeast strain and the diet composition. Corn silage and citrus pulp are usual carbohydrate sources for dairy cows in southeast Brazil. This study evaluated the supplementation of dairy cows fedding on corn silage-citrus pulp-based diets with Saccharomyces cerevisiae CNCM I-1077 (Lallemand SAS, Toulouse, France). Twenty multiparous, midlactation Holstein cows were assigned to two treatments in crossover design. Treatments were: live yeast on oyster meal capable of supplying a daily minimum of 1 × 10(10) CFU per cow or oyster meal top-dressed at 10 g to the morning meal. Diet contained (% of dry matter): 16.8% crude protein, 30.9% neutral detergent fiber, 43.9% corn silage, 2% tifton hay, 14.4% steam flaked corn, 16.9% citrus pulp and 21.7% soybean meal. Yeast supplementation increased daily yields of milk (29.4 vs. 28.5 kg, p = 0.11), protein (0.939 vs. 0.908 kg, p = 0.05), and lactose (1.294 vs. 1.241 kg, p = 0.06), but did not affect milk fat contents (p = 0.59). Daily dry matter intake was 21.4 with yeast and 20.7 kg for the control (p = 0.11). Total tract apparent digestibility of the neutral detergent fiber was 48.1% with yeast and 43.2% for the control (p = 0.08). There was a trend for increased intake of digestible organic matter with yeast supplementation (p = 0.07). The positive milk protein yield response to yeast supplementation may have resulted from the increased fiber digestibility, but the response mechanism could not be elucidated.

The objective of the investigations described in this work was to determine the energy status and to make recommendations for correcting the cow diet at a farm of high-yield dairy cows, on the grounds of values for the concentration of organic components of milk and their ratios in individual milk samples. A total of 147 cows were examined, including 97 in the first and 50 in the second lactation. Average concentrations of milk fat and urea were within the physiological values. Namely, the milk fat concentration in cows in the first lactation was 38.88?5.07 g/l, and it was 36.47?4.82 g/l in cows in the second lactation, while the urea concentration in cows in the first lactation 3.16?0.58 mmol/l and it was 3.72?0.64 mmol/l in cows in the second lactation. The protein concentration in both groups of cows was below the physiological values, being 30.33?2.35 g/l in cows in the first lactation and 30.17?2.27 g/l in cows in the second lactation. Based on the ratio of urea and protein concentrations, as well as of fat and proteins in the individual milk samples, it was concluded that in most examined cows, both in those in the first and those in the second lactation, there is a deficit of energy, along with a deficit or relative surplus of proteins. On the grounds of the obtained results, recommendations were given for correcting the feed rations in the coming period. .
