AbstractSubclinical mastitis is an asymptomatic udder infection distributed worldwide with enormous losses in the dairy industry. The study’s objective was to determine the presence of this pathological condition in small dairy farms in the R. of N. Macedonia and to identify the most common associated bacteria. Milk samples were obtained from 96 dairy cows (378 udder quarters) in seven dairy farms, in 3 consecutive samplings 24–72 hours apart. The samples were cultured on routine bacteriological growth media and incubated for 24–48 hours. The isolates were identified by AximaiD Plus MALDITOF MS Platform. Subclinical mastitis was found in 49 animals (51%) and 104 infected quarters (27%). The most frequent isolated bacteria on cow level were Streptococcus uberis (19.4%), Staphylococcus haemolyticus (13.4%), Staphylococcus aureus (7.4%) and Staphylococcu ssimulans (7.4%). On quarter level, the most isolated pathogen was Streptococcus uberis (35.6%) followed by Staphylococcu shaemolyticus and Staphylococcus aureus (10.3% and 9.2% respectively). Subclinical mastitis was found to be highly present in the selected small dairy farms. The most prevalent bacteria identified in the dairy farms (Streptococcus uberis, Staphylococcus aureus and coagulase–negative staphylococci) indicate that poor management and udder health practices, inadequate milking procedures and lack of mastitis control strategies greatly contribute to occurrence and persistence of subclinical mastitis.

<p>The aim of this research was to identify the occurrence of pathogens causing subclinical mastitis in grade B milk farms of the Jaguapitã county, state of Paraná, Brazil. California Mastitis Test (CMT) were carried out in 400 milk samples from 100 animals and 157 teats from 55 animals (55%) were positive, showed score two or higher to CMT. When these 157 positive samples to CMT were transported for bacterial culture in blood agar, 25.48% (40/157) samples showed no bacterial growth or more than two types of bacterial colonies grew, 28.03% (44/157) were Coagulase-negative staphylococci (CNS), 8.28% (13/157) were <em>Streptococcus uberis</em>, 7.64% (12/157) were <em>Staphylococcus aureus</em>, 7.64% (12/157) were <em>Corynebacterium </em>spp, 7.01% (11/157) were <em>Staphylococcus intermedius</em>, 4.46% (7/157) were <em>Staphylococcus hyicus</em>, 3.82% (6/157) were <em>Bacillus </em>spp., 2.55% (4/157) were <em>Streptococcus dysgalacteae</em>, Enterobacteria and Yeasts. We conclude that CNS is the most relevant subclinical mastitis causative agent.</p><p><strong> </strong></p>

Introduction: The aim of this study was to monitor the occurrence of bacterial agents causing mastitis, forms of mastitis and antibiotic resistance in 300 ewes on a farm in Eastern Slovakia. Material and methods: During the milking season, were performed three complex investigations including clinical examination, California Mastitis Test and laboratory analysis of milk samples. The investigations and milk samples were taken in three phases; the beginning (April), the middle (June) and the end (September) of the milking season. Results: Of all the samples (806), 225 (28.0%) were positive for bacterial pathogens. The highest incidence of mastitis (33.3%) was recorded in September, while April (23.8%) and June (25.3%) had lower incidence. The samples from September showed the highest incidence of subclinical mastitis (20.1%), with 13.2% being an acute form of mastitis. Coagulase-negative staphylococci (CNS) were identified in 61.7% of the positive samples. Especially, S. chromogenes, S. epidermidis and S. schleiferi were most frequently isolated.  Staphylococcus aureus was the causative agent in 20.0% of the positive samples and caused acute or subclinical mastitis in the affected ewes. The tested bacteria showed very high resistance to novobiocin (59.5%) and penicillin (51.4%) and high resistance to amoxycillin (35.1%). We found that 80% of Staphylococcus aureus bacteria tested for antibiotic resistance were resistant to novobiocin and 70% were resistant to penicillin. Of all tested CNS, 56.5% were resistant to novobiocin, 39.1% to penicillin, and 34.7% to amoxicillin. Conclusion: Proper isolation and identification of the causative organism play a significant role in the prevention and control of the intramammary infection. In our study, a combinations of Streptomycin, Ciprofloxacin and Tetracycline were the most effective antibiotics for the control of mastitis.

The effect was quantified of coagulase-negative staphylococci (CNS) intramammary infections on quarter- and cow-level somatic cell count (SCC) and on bulk milk somatic cell count (BMSCC) in different BMSCC cohorts in Dutch dairy herds. Two datasets were used for this purpose. In the first dataset, on 49 randomly selected dairy farms a total of 4220 quarter milk samples of 1072 cows were collected of all cows and heifers with a test-day SCC ⩾250 000 and ⩾150 000 cells/ml, respectively, and of 25% of cows and heifers below these thresholds. In the second dataset, on 39 selected dairy farms a total of 8329 quarter milk samples of 2115 cows were collected of all cows with a test-day SCC ⩾250 000 cells/ml following two consecutive SCC <250 000 cells/ml, and of heifers using the same SCC criteria but with a threshold of 150 000 cells/ml. These cows and heifers were defined as new high SCC. In both datasets, CNS was the most frequently isolated pathogen, 11% in the first dataset and 12% in the second dataset. In both datasets, quarters with CNS IMI had a lower SCC than quarters infected with major pathogens, and a higher SCC than culture-negative quarters. The same was found for SCC at cow level. Coagulase-negative staphylococci were more often found in quarters with SCC ⩾200 000 cells/ml in dairy farms with a BMSCC <150 000 cells/ml compared with dairy farms with a higher BMSCC. Prevalence of CNS in cows and heifers with a high SCC was higher in dairy farms with a BMSCC <150 000 cells/ml compared with dairy farms with a medium or high BMSCC: 30, 19 and 18%, respectively. This indicates that CNS IMI as a cause of subclinical mastitis is relatively more important in dairy farms with a low BMSCC and may become a point of attention in udder health management on that type of farm.

A longitudinal study was conducted from November 2013 to May 2014 in Mekelle town, Northern Ethiopia, to assess the incidence of heifer mastitis, to isolate and identify the causative pathogens and their susceptibility to different antimicrobials and also identify risk factors associated with occurrence of heifer mastitis. Twenty - five heifers were included from 4 d airy farms and 11 cooperative small holder’s dairy farms during the study period. Total of 100 quarters were examined to detect clinical and subclinical mastitis by physical examinations of udder and milk and California mastitis Test (CMT), respectively. T he incidence of mastitis per gland at risk was 64 % and the spontaneous cure rate of infected quarters was 33 %. Incidence of heifers’ intra - mammary infections (IMI) in the study area was highest at calving (28%). Infection per quarter revealed that 49% we re mastitis positive of which 8.2% of their quarters were clinical and 45/49 (91.8%) quarters were sub clinical type of mastitis. The incidence rate per quarters were statistically significant (p <0.05) and higher in rear quarters than front quarters. The univariate logistic regression showed that among potential risk factors considered from the farm attributes, breed, age, practice of milking mastitic cow last, housing nature, hand wash before and in between milking and udder hygiene had significant effect on the prevalence of subclinical mastitis. Coagulase negative Staphylococci (CNS) (54.5%) and Staphylococcus aureus (21.8%) , were the predominant bacteria. Relatively most of the isolates are susceptible to Chloramphenicol, Gentamycin, Ciprofloxacin and V ancomycin but resistant to Penicillin. The study also revealed that S.aureus, E.coli and S.agalactiae were highly incident at calving. It is concluded that prevalence of CNS, lack of hygienic practice and infestation of ticks should give serious attention.Intramammary treatment of prepartum heifer with appropriate antimicrobials may help to reduce the incidence of mastitis at calving and subsequent lactation period.

Abstract Background: Staphylococcus aureus is one of the predominant causative agents of mastitis disease in dairy herds. Mastitis disease has a negative impact in the economic losses in the dairy sector across the globe. The aims of this study were to determine the prevalence and detect antimicrobial resistance genes in the Staphylococcus aureus isolated from milk samples of subclinical bovine mastitis in Central Ethiopia.Methods: A total of 265 lactating dairy cows from various dairy farms in four different geographical locations were screened by California mastitis test (CMT) for bovine subclinical mastitis. One-hundred thirty CMT positive milk samples were collected and transported to laboratory. Different biochemical tests and polymerase chain reaction (PCR) were used for the identification of S. aureus isolates. Finally, phenotypic and genotypic methods were performed for detection of some antimicrobial resistance patterns and genes (mecA, ermA, ermC, and msrA), respectively. Results: From total of 265 lactating dairy cows screened, 49% (n=130) were positive for bovine subclinical mastitis. One-hundred thirty mastitic milk samples were subjected to bacterial culturing, one hundred (76%) S. aureus isolates were identified based on phenotypic characters. Sixty-eight confirmed S. aureus isolates were obtained using PCR. Of the sixty-eight isolates tested 12 samples were contained the methicillin resistance gene A (mecA). No amplification was observed for the erythromycin resistance genes (ermA, ermC, and msrA). Conclusion: The high resistance of Staphylococcus aureus to commonly used antimicrobials contribute in dairy farms may cause health problems in the community consuming raw milk purchased from these farms.

Thirty-two farms (n=535cows) located in the state of Guanajuato, Mexico, were sampled. Pathogens from bovine subclinical mastitis (SCM) and clinical mastitis (CLM) were identified by 16S rDNA and the sensitivity to both antibiotics and bacteriocins ofBacillus thuringiensiswas tested. Forty-six milk samples were selected for their positive California Mastitis Test (CMT) (≥3) and any abnormality in the udder or milk. The frequency of SCM and CLM was 39.1% and 9.3%, respectively. Averages for test day milk yield (MY), lactation number (LN), herd size (HS), and number of days in milk (DM) were 20.6 kg, 2.8 lactations, 16.7 animals, and 164.1 days, respectively. MY was dependent on dairy herd (DH), LN, HS, and DMP<0.01, and correlations between udder quarters from the CMT were around 0.49P<0.01. Coagulase-negative staphylococci were mainly identified, as well asStaphylococcus aureus,Streptococcus uberis,Brevibacterium stationis,B. conglomeratum, andStaphylococcus agnetis. Bacterial isolates were resistant to penicillin, clindamycin, ampicillin, and cefotaxime. Bacteriocins synthesized byBacillus thuringiensisinhibited the growth of multiantibiotic resistance bacteria such asS. agnetis,S. equorum,Streptococcus uberis,Brevibacterium stationis, andBrachybacterium conglomeratum, but they were not active againstS. sciuri, a microorganism that showed an 84% resistance to antibiotics tested in this study.

Little information is available regarding the molecular epidemiology of Staphylococcus aureus–induced mastitis in dairy sheep. In this study, 4 different typing techniques were compared in typing 26 S. aureus isolates, predominantly from cases of subclinical mastitis in dairy ewes. The 4 techniques were pulsed-field gel electrophoresis (PFGE), restriction fragment length polymorphism (RFLP) on 2 genes (coagulase and clumping factor B), randomly amplified polymorphic DNA–polymerase chain reaction (PCR) (RAPD-PCR), and multilocus sequence typing (MLST). On the basis of discriminatory power as the key parameter of typing systems, MLST and PFGE were found to be the most powerful techniques. The MLST and PFGE could contribute to epidemiological surveillance and evaluation of mastitis control programs, by documenting prevalence and dissemination of endemic clones in infected populations. The results of this study show that a single clone of S. aureus is widely distributed in infected ewe mammary glands.

Inflammation of the mammary gland, mastitis in cows, presents one of the most acute problems in intensive dairy production, inflicting huge economic losses. In the course of one year, 80 samples were taken at investigated farms from udder quarters of cows with clinical mastitis and 160 samples from udder quarters of cows with subclinical mastitis. The efficacy of three preparations, A, B, and C, was examined in the treatment of clinical and subclinical mastitis in cows. The investigations indicate that antibiotic preparation A (neomycin, polimixine B, oleandomycin and prednisolone) exhibited a greater efficacy in the treatment of clinical mastitis caused by Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis and Micrococcus sp., but a smaller efficacy in the treatment of subclinical mastitis caused by Staphylococcus aureus. Preparation B (amoxicillin, clavulanic acid and prednisolone) exhibited a higher efficacy in the treatment of clinical mastitis caused by Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis and Micrococcus, but a weaker effect in the treatment of subclinical mastitis caused by Staphylococcus aureus. Preparation C (procaine penicillin G, streptomycin, neomycin sulfate and prednisolone acetate) exihibited efficacy in the treatment of clinical and subclinical mastitis caused by Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, Micrococcus, Staphylococcus aureus and Esherichie coli.
