AbstractThe number of parasites followed the rapid growing of human population worldwide, not only in developing but also in developed countries. Many of them are diagnosed in children and adolescents. The occurrence of selected intestinal endoparasites in children coming from areas with low hygienic and socioeconomic status was studied. Out of 81 faecal samples examined, 46 (56.8 %) were positive for presence of intestinal parasites. From helminths, Ascaris lumbricoides was found to be the leading parasite (24.7 %), followed by Trichuris trichiura (17.3 %). Tapeworm Taenia spp. eggs were detected in 4.9 % of examined children. From protozoan parasites Cryptosporidium spp. was observed in 36 children (44.4 %) and Giardia intestinalis in 20 children (24.7 %). The occurrence of these epidemiologically low risky parasites in Roma children population suggests low hygienic standard in the Roma settlements.

The macaque is a member of the sub family Cercopithecinae and family Cercopithecidae of primate order of mammalian class. Among five species of monkeys reported from Nepal, Assamese Macaque is one of the less common primate species. A total of 85 fresh faecal samples of Assamese Macaque (Macaca assamensis) were collected from Shivapuri Nagarjun National Park (SNNP), Kathmandu, Nepal. All the samples were macroscopically as well as microscopically examined to identify the distribution of gastrointestinal parasites using faecal floatation and Sedimentation techniques. The result revealed the distribution of three species of protozoan and seven species of helminthes parasite among Assamese Macaque of SNNP with the prevalence rate of 72.94% (62/85). Among protozoan parasites, Balantidium coli showed the highest prevalence (28.24%) followed by Entamoeba sp. (20%), and Isospora sp. (3.53%). Ascaris sp. was found to be distributed maximum (10.58%) among helminth parasites followed by Trichuris sp. (9.41%), Strongyloides sp. (8.24%), Moniezia sp. (8.24%), Oesophagostomum sp. (4.7%), Hookworm (4.7%) and Physeloptera sp.(1.17%). The present study highlights the necessity of considering parasitic diseases as one of the threat in conservation of Assamese Macaque.Journal of Institute of Science and Technology, 2014, 19(2): 53-57

Nowadays, many commercial kits allowing the detection of digestive parasites by DNA amplification methods have been developed, including simplex PCR assays (SimpPCRa) allowing the identification of a single parasite, and multiplex PCR assays (MultPCRa) allowing the identification of several parasites at once. Thus, aimed at improving the diagnosis of intestinal protozoal infections, it is essential to evaluate the performances of these new tools. A total of 174 DNA samples collected between 2007 and 2017 were retrospectively included in this study. Performances of four commercial SimpPCRa (i.e., CerTest-VIASURETM) and three MultPCRa (i.e., CerTest-VIASURETM, FAST-TRACK-Diagnostics-FTD-Stool-ParasiteTM and DIAGENODE-Gastroenteritis/Parasite-panel-ITM) were evaluated for the detection of Cryptosporidium spp., Entamoeba spp., and Giardia intestinalis in stool samples compared to our routinely used in-house SimpPCRa. Globally, the SimpPCRa showed better sensitivity/specificity for the detection of G. intestinalis, E. histolytica, E. dispar, and Cryptosporidium spp. (i.e., 96.9/93.6%; 100/100%; 95.5/100%; and 100/99.3%, respectively), compared to the three commercial MultPCRa tested. All in all, we showed that MultPCRa offer an interesting alternative for the detection of protozoans in stool samples depending on the clinical context.

A total of 22 faecal samples of Red panda were collected in the month of May/June 2011 and 21 faecal samples from the same site during the month of May/June 2012. All the samples were microscopically examined by faecal floatation and sedimentation techniques. Out of 43 faecal samples, 40 samples (93.02%) were found positive for gastro-intestinal parasites. Altogether, 12 different species of gastro-intestinal parasites were recovered from Red panda. Among them, two species of protozoan parasites and 10 species of helminthes were observed. In protozoan parasites, Eimeria (67.44%) was more prevalent as compared to Entamoeba (62.79%). Among helminthes, Oxyuris sp. showed the highest prevalence rate followed by Toxoascaris sp. (48.84%), Hookworm (44.19%), Baylisascaris sp., Crenosoma sp. (34.88%), Strongyloides sp., Moniezia sp. (18.60%), Trichuris sp., etastrongylus sp. (4.65%) and Angiostrongylus sp. (2.33%). Only one species of cestoda i.e. Moniezia sp. was found but trematodes and acanthocephalans were not found in Red Panda of Rara National Park. J. Nat. Hist. Mus. Vol. 29, 2015, Page: 78-90

Cryptosporidium is a leading cause of childhood diarrhoea and associated physical and cognitive impairment in low-resource settings. Cryptosporidium-positive faecal samples (n = 190) from children aged ≤ 5 years enrolled in the Global Enteric Multicenter Study (GEMS) in Mozambique detected by ELISA (11.5%, 430/3754) were successfully PCR-amplified and sequenced at the gp60 or ssu rRNA loci for species determination and genotyping. Three Cryptosporidium species including C. hominis (72.6%, 138/190), C. parvum (22.6%, 43/190), and C. meleagridis (4.2%, 8/190) were detected. Children ≤ 23 months were more exposed to Cryptosporidium spp. infections than older children. Both C. hominis and C. parvum were more prevalent among children with diarrhoeal disease compared to those children without it (47.6% vs. 33.3%, p = 0.007 and 23.7% vs. 11.8%, p = 0.014, respectively). A high intra-species genetic variability was observed within C. hominis (subtype families Ia, Ib, Id, Ie, and If) and C. parvum (subtype families IIb, IIc, IIe, and IIi) but not within C. meleagridis (subtype family IIIb). No association between Cryptosporidium species/genotypes and child’s age was demonstrated. The predominance of C. hominis and C. parvum IIc suggests that most of the Cryptosporidium infections were anthroponotically transmitted, although zoonotic transmission events also occurred at an unknown rate. The role of livestock, poultry, and other domestic animal species as sources of environmental contamination and human cryptosporidiosis should be investigated in further molecular epidemiological studies in Mozambique.

Cryptosporidium spp., common parasites of vertebrates, remain poorly studied in wildlife. This study describes the novel Cryptosporidium species adapted to nutrias (Myocastor coypus). A total of 150 faecal samples of feral nutria were collected from locations in the Czech Republic and Slovakia and examined for Cryptosporidium spp. oocysts and specific DNA at the SSU, actin, HSP70, and gp60 loci. Molecular analyses revealed the presence of C. parvum (n = 1), C. ubiquitum subtype family XIId (n = 5) and Cryptosporidium myocastoris n. sp. XXIIa (n = 2), and XXIIb (n = 3). Only nutrias positive for C. myocastoris shed microscopically detectable oocysts, which measured 4.8–5.2 × 4.7–5.0 µm, and oocysts were infectious for experimentally infected nutrias with a prepatent period of 5–6 days, although not for mice, gerbils, or chickens. The infection was localised in jejunum and ileum without observable macroscopic changes. The microvilli adjacent to attached stages responded by elongating. Clinical signs were not observed in naturally or experimentally infected nutrias. Phylogenetic analyses at SSU, actin, and HSP70 loci demonstrated that C. myocastoris n. sp. is distinct from other valid Cryptosporidium species.

AbstractThe objective of this study was to determine the possibility of soil contamination with propagative stages of intestinal endoparasites at different public places (public parks, playgrounds, sandpits, sidewalks, road sides). In a one-year-study, totally 578 dog’s faecal samples from 8 towns (Košice, Trebišov, Veľké Kapušany, Prešov, Snina, Levoča, Zvolen and Trenčín) and 3 villages (Dlhé Stráže, Dravce, Valaliky) were examined for the presence of parasitic germs. 29.9 % of faecal samples were positive. Eight different species of intestinal parasites with following incidence were detected: Toxocara canis (11.9 %), Trichuris vulpis (8.5 %), Ancylostomatidae (8.1 %), Taenia spp. (4.0 %), Toxascaris leonina (3.1 %), Capillaria spp. (1.0 %), Dipylidium caninum (0.2 %) and Coccidia oocysts (0.2 %). Additionally 285 sandpits were examined. Sand samples were collected from Košice (136), Zvolen (38), Trenčín (36), Prešov (30), Veľké Kapušany (24), Snina (15), and village Valaliky (6). The parasitic eggs occurence in the sandpits was as follows: Toxocara spp. (11.8 %), Ancylostomatidae (1.1 %), Taenia type (0.7 %) and Trichuris spp. (0.4 %). The occurrence of Toxocara spp. varied between the urban and rural environments. The highest number of Toxocara spp. was found in village Valaliky (33.3 %) whereas cities showed lesser prevalence (Prešov — 10 %, Košice — 7.4 %, Trenčín — 5.6 %, Zvolen — 5.3 %, Veľké Kapušany — 4.2 %). The epidemiological aspects and health risk factors are also discussed.

The prevalence of Cryptosporidium spp. infection in a cross-sectional study of dairy cattle, from two contrasting dairying regions in Tanzania, were determined by staining smears of faecal samples with the modified Ziehl-Neelsen technique. Of the 1 126 faecal samples screened, 19.7% were positive for Cryptosporidium spp. The prevalence was lower in Tanga Region than in Iringa Region. The prevalence of affected farms was 20% in Tanga and 21% in Iringa. In both regions, the probability of detecting Cryptosporidium oocysts in faeces varied with animal class, but these were not consistent in both regions. In Tanga Region, Cryptosporidium oocysts were significantly more likely to be found in the faeces of milking cows. In Iringa Region, the likelihood that cattle had Cryptosporidium-positive faeces declined with age, and milking cattle were significantly less likely to have Cryptosporidium positive faeces. In this region, 7% of cattle were housed within the family house at night, and this was marginally associated with a higher likelihood that animals had Cryptosporidium-positive faeces. Our study suggests that even though herd sizes are small, Cryptosporidium spp. are endemic on many Tanzanian smallholder dairy farms. These protozoa may impact on animal health and production, but also on human health, given the close associations between the cattle and their keepers. Further studies are required to assess these risks in more detail, and understand the epidemiology of Cryptosporidium spp. in this management system.

Abstract The aim of the research was to determine the prevalence of non-pathogenic protozoa circulating in the human population of Slovakia. We particularly focused on the socially deprived areas with poor sanitation conditions, as they are one of the factors affecting the transmission of these infections. Within this study, 2760 people were coprologically screened for the presence of protozoan cysts. The analyzed group comprised 1173 men and 1587 women from different regions of Slovakia. The total prevalence (2.03%) of non-pathogenic protozoa species was determined. The prevalence of Entamoeba coli was 0.80%, the prevalence of Endolimax nana 0.58%, and the prevalence of Blastocystis hominis was 0.65%. The presence of non-pathogenic protozoa was more frequent in women than that in men, in all age groups. The highest incidence of Entamoeba coli was found in children aged one month – seven years (0.79%), the lowest in the age group of 19–88 years (0.66%). Endolimax nana was most frequent in 8–18 year-olds (0.95%), where the statistical significance was found (p<0.05). The prevalence of Blastocystis hominis by the age group ranged from 0.39 to 0.95%. We did not find any statistical significance (p>0.05) for Entamoeba coli, and similarly for Blastocystis hominis associated with the sex and age. Although the circulation of non-pathogenic protozoa in the human population is far from being limited to the developing countries, their occurrence is also frequent in the population of developed countries. Despite their controversial pathogenicity, they should not be neglected, particularly in the patients with gastrointestinal symptoms.
